Abstract:A new class of chiral phosphoric acids with spirobiindane as scaffold were conveniently synthesized from (S)-1,1'-spirobiindane-7,7'-diol ((S)-SPINOL) and employed to catalyze the asymmetric Friedel-Crafts reaction of indoles with imines to afford 3-indolyl methanamines. High yields (68-97%) and excellent enantioselectivities (up to 99% ee) were obtained.

Abstract:Comprehensive Summary The design and synthesis of spirochromane‐4,1'‐indan‐2‐one‐7'‐ol (SCIOL) and its application for synthesizing chiral monophosphinite ligands is reported. The synthesis features a tandem double Friedel‐Crafts reaction/lactonization to construct the spirocyclic and the desired racemic SCIOL was obtained via 6 steps with total yield of 44.5%. Using an inclusion resolution with N‐benzylcinchonidinium chloride, the optical SCIOL could be obtained in good yields on a gram scale. The preliminary studies indicated that the corresponding chiral spiro monophosphinite ligands exhibit high catalytic activity and enantioselectivity (up to 94% ee) for the rhodium‐catalyzed asymmetric hydrogenation of N‐acetyl dehydroamino esters. These results indicate that SCIOL has a great potential as a useful framework for the development of chiral spiro ligands. This article is protected by copyright. All rights reserved.

Abstract:A new type of robust planar chiral phosphoric acid bearing a [2.2]paracyclophane scaffold was synthesized and shown to be an optimal catalyst in asymmetric aza-Friedel-Crafts reactions for the first synthesis of enantioenriched styryl indolylmethanamine derivatives in good yields with excellent enantioselectivities (93->99% ee) under 0.5-1 mol % catalyst loading.

Abstract:A method of SPINOL-derived chiral phosphoric acid catalyzed asymmetric intermolecular N-hydroxyalkylation of multisubstituted indoles with ethyl glyoxalates is described in this report. This protocol provides an alternative, convenient, and direct strategy for efficient access to structurally unique α-chiral indole N,O-acyclic aminals with a broad substrate scope and good to excellent enantioselectivities. The synthetic utility of this methodology is illustrated by a gram-scale experiment and the subsequent efficient synthesis of more complex chiral N,O-aminal indole derivatives.
