Abstract:The bulky and electron-rich MOP-type ligand was efficient for the Pd-catalyzed amination of aryl tosylates. The in situ generated Pd(0) was a more efficient catalyst precursor than Pd(dba)(2). In the presence of Pd(OAc)(2), PhB(OH)(2), and a hindered and electron-rich MOP-type ligand, a variety of primary aryl amines reacted with various aryl tosylates to form the corresponding secondary aryl amines in high yields with high selectivity. Furthermore, the catalyst system was also efficient for the arylation of indoles and hydrazones with aryl tosylates.

Abstract:We report the amination of aryl carbamates using nickel-catalysis. The methodology is broad in scope with respect to both coupling partners and delivers aminated products in synthetically useful yields. Computational studies provide the full catalytic cycle of this transformation, and suggest that reductive elimination is the rate-determining step. Given that carbamates are easy to prepare, robust, inert to Pd-catalysis, and useful for arene functionalization, these substrates are particularly attractive partners for use in synthesis. The sequential use of carbamate functionalization/site-selective cross-coupling processes highlights the utility of this methodology.

Abstract:Amines are common scaffolds in bioactive molecules as well as building blocks for chemical synthesis. Despite significant advances in palladium-catalyzed C(sp3)−H functionalization of amines over the past decades, it remains challenging to perform directed C−H metalation with native amine groups rather than amine-derived or transient directing groups (DGs). Recently, our group developed the Pd(II)/sulfoxide-2-hydroxypyridine (SOHP) catalytic system, in which SOHP ligands play a pivotal role as a crucial functional module, enabling regioselective C(sp2)−H and enantioselective C(sp3)−H functionalization reactions. In this work, we demonstrate that this chemistry provides an ideal solution for native primary amine-directed γ-C(sp3)-H arylation. Primary amines of varying degrees of complexity, encompassing amino acid esters and aminol silyl ethers, were found to be compatible with the established methodology. Additionally, we achieved a preliminary implementation of the asymmetric variant of this γ-C−H arylation reaction by employing a chiral SOHP ligand. Moreover, the range of applicable substrates could be extended to pyridine, oxime ether, and pyridine-N-oxide.

Abstract:Although nucleophilic benzylation-type reaction to introduce various aromatic systems into molecules has been widely explored, the related pyrrolylmethylation version remains to be disclosed. Reported herein is a palladium-catalysed multiple auto-tandem reaction between N-Ts propargylamines, allyl carbonates and aldimines in the presence of an acid, proceeding through sequential allylic amination, cycloisomerisation, vinylogous addition and aromatisation steps. A diversity of formal pyrrolylmethylated amine products were finally furnished efficiently. In addition, switchable regiodivergent 3-pyrrolylmethylation and 4-pyrrolylmethylation were realised by tuning catalytic conditions. Moreover, remote chirality transfer with readily available enantioenriched starting materials was well achieved with an achiral ligand, relying on diastereoselective generation of η2-Pd(0) complexes between Pd(0) and chiral 1,3-diene intermediates in the key vinylogous addition step. A few control experiments was conducted to elucidate the palladium-involved tandem reaction and regiodivergent synthesis.

Abstract:A reliable method is introduced for the synthesis of branched α‐arylated amines from simple linear N‐sulfonyl amines and aryl boroxines utilizing dual palladium catalysis involving amine dehydrogenation. Bromobenzene serves as a hydride acceptor, and the ensuing imine intermediate undergoes a novel umpolung arylation. Given the wide availability of primary amines, this method can be expected to be useful for the synthesis of structurally complex amines with varied functionality. Functionalization of C(sp3)−H bonds adjacent to a nitrogen atom is an efficient means of preparing structurally complex amines and has long been of interest to synthetic chemists. Herein, dual palladium catalysis is used to achieve α‐C−H arylation of linear N‐sulfonyl amines with aryl boroxines, rather than N‐arylation (C−N bond formation). This method has a broad scope with respect to both the amine and the boroxine, providing convenient access to α‐branched amines. During the dehydrogenation step of the catalytic cycle, bromobenzene accepts a hydride from the α‐carbon on the amine, producing an imine that subsequently undergoes Pd‐catalyzed C‐arylation with the boroxine. Mechanistic studies revealed that imine arylation proceeds through an unusual Pd0/PdII catalytic cycle, in which oxidative cyclization of a palladium η2‐imine complex generates a three‐membered palladacycle that subsequently undergoes transmetallation reaction with the boroxine.

Abstract:The Pd(II)/sulfoxide-2-hydroxypyridine catalytic system shows promising activity in C-H activation chemistry. In this study, we showcase how this catalytic system solves the problem of native primary amine-directed γ-C(sp3)-H arylation. Primary amines with different complexities are compatible with the established methodology, and the range of applicable substrates can be expanded to include pyridine, oxime ether, and pyridine N-oxide.

Abstract:Since the discovery of the palladium/norbornene (Pd/NBE)-catalyzed ortho amination in 2013, escaping the limitation of only yielding tertiary anilines has been a long-standing challenge in the past decade. Here, we describe that, by carefully choosing the phosphine ligand and NBE mediator, installation of a N-mono-alkylamino group becomes feasible. The reaction tolerates a wide range of aryl iodide substrates and various N-mono-tertiary alkylamine-derived electrophiles. Both ipso alkenylation and alkynylation can be realized. The synthetic utility of this method is exemplified in the formation of primary amino group via selective deprotection and streamlined access to N-heterocycles. Preliminary success of installing a bulky N-secondary alkylamino group and mechanistic understanding of the decomposition pathways of mono N-alkylamine electrophiles have been obtained.
