Abstract:The first asymmetric synthesis of 2,3-dihydrofuro[2,3-b]quinolines has been achieved by a cascade asymmetric aziridination/intramolecular ring-opening process of differently substituted 3-alkenylquinolones. Good yields and high enantioselectivities (up to 78% yield and 95% ee) were recorded when employing 2,2,2-trichloroethoxysulfonamide as the nitrene source, PhI(OCOtBu)(2) as the oxidant, and a chiral C-2-symmetric Rh-II complex as the catalyst (1mol%). The catalyst bears two lactam motifs, which serve as binding sites for substrate coordination through supramolecular hydrogen-bonding interactions.

Abstract:The furoquinoline unit is present in many natural products.Here,an approach is presented for the preparing of furo[2,3-b]quinolines from readily available multi-substituted furans in the presence of Br(o)nsted acid via an intramolecular cyclization under the heating conditions.Simple operation,good compatibility,high regioselectivity and morderate yields are the advantages of the method.

Abstract:In one easy step, doubly activated cyclopropanes 1 can be transformed into furoquinoline derivatives 2 through a tandem ring-opening/recyclization reaction mediated by SnCl4⋅5 H2O. A variety of substrates 1 derived from cheap starting materials were converted into the corresponding furoquinolines in good to excellent yields with high chemo- and regioselectivity. Abstract:A new synthesis of furo[3,4-b]quinolin-3(1H)-one derivatives has been established in straightforward fashion in high overall yields. An efficient Hendrickson reagent-initiated cascade annulation, which is composed of mild conversion of the stable amide precursor to the reactive imido-carbonium intermediate and subsequent intramolecular aza Diels–Alder reaction, successfully serves as the key reaction in the synthesis. Final oxidative cleavage of the carbon–carbon double bond of representative tricyclic precursor 8g led to completion of the synthesis of quinoline-lactone 2g in a high yield.

Abstract:C–Si bonds were constructed by utilizing copper hydride-catalyzed asymmetric hydrosilylation of 1,2-dihydroquinolines, affording various chiral 4-silyl-1,2,3,4-tetrahydroquinolines in good yields and enantioselectivity. In addition, the C–Si bonds were transformed into C–O bonds with retention of stereochemistry through the Tamao oxidation, giving a series of useful 4-hydroxy-1,2,3,4-tetrahydroquinolines. This method with the enantioselective introduction of silyl groups provides an option to adjust bioactive properties of tetrahydroquinolines.

Abstract:The construction of novel skeletons through the recombination of natural products ring systems is attractive. Herein, a basicity‐controlled synthesis of (dihydro)furo[2,3‐c]quinolin‐4(5H)‐one derivatives through (3+2) cyclization between 3‐hydroxyquinolin‐2‐ones and chlorinated nitrostyrenes was developed. In addition, preliminary study gave the chiral dihydrofuro[2,3‐c]quinolin‐4(5H)‐one derivatives with up to 83% ee.

Abstract:Via a copper-catalyzed three-component annulation reaction, we herein report a new method for the direct and syn-selective construction of cyclic ether-fused tetrahydroquinolines from readily available secondary anilines, saturated five or six-membered cyclic ethers, and paraformaldehyde. The synthesis features operational simplicity, excellent step and atom efficiency, good functionality and substrate compatibility. In comparison with the reported synthetic protocols capable of synthesizing N-alkyl fused tetrahydroquinolines, this newly developed chemistry allows access to both N-alkyl and N-aryl products. The current work complements the preparation of fused tetrahydroquinolines.

Abstract:An efficient N‐heterocyclic carbene (NHC)‐catalyzed kinetic resolution of 2‐substituted 3‐nitro‐1,2‐dihydroquinolines (DHQs) allowing the synthesis of densely functionalized enantioenriched tetrahydroquinolines through the resolution at remote stereocenter is described. The Re‐face addition of α,β‐unsaturated azolium homoenolate intermediate generated in situ from enal with the more kinetically favored (S)‐enantiomer of the racemic 2‐substituted 3‐nitro‐1,2‐dihydroquinolines (rac‐1), affording the enantiopure tetrahydroquinolines (up to >99:1 dr, >99:1 er) and the opposite (R)‐enantiomer of DHQ (ent‐1) was recovered (up to >99:1 er).
