Abstract:A palladium bromide and copper(I) bromide co-catalyzed reaction of 4-tosyloxy coumarin with benzo[d]oxazole/oxazole/benzo[d]thiazole via C–H activation is described, which generates 4-substituted coumarins in moderate to good yields. 4-Tosyloxy quinolin-2(1H)-one and 2H-pyran-2-one could be applied in this transformation as well.

Abstract:With a strategy that makes use of palladium activation of both C-I and vicinal C-H bonds of diaryliodonium salts, an unprecedented approach in diarylation of coumarins was reported. As such, a wide range of 4,5-benzocoumarins with potential fluorescence properties have been synthesized in good yields. A series of experiments suggested that the formation of two carbon-carbon bonds proceeded in a synergetic manner.

Abstract:Tetracyclic coumarins are a class of important compounds with diverse and superior pharmacological activities. However, a direct stereoselective method from simple and readily-made coumarins derivatives remains challenging due to the inertness of coumarins as dienophiles. Herein, we develop a decarboxylative asymmetric [4+2] cycloaddition of 3-cyanocoumarins with vinyl benzoxazinones, affording the coumarin-derived condensed rings bearing three continuous stereocenters in high yields with excellent diastereoselectivities (>20/1 d.r.) and enantioselectivities (up to 99% ee ). This direct enantioselective reaction was achieved by a Pd(0)/Cu(I) bimetallic catalytic system. The mechanism studies indicated that the synergistic activation effect, in which chiral Cu(I) as an available Lewis acid catalyst activates 3-cyanocoumarin and chiral Pd(0) complex activates benzoxazinone by the formation of π-allyl-palladium intermediate, plays an important role on the stereoselective control. The current work provides a new activation modes of Cu catalyst in the Pd/Cu bimetallic catalytic system. Abstract:4-Hydroxycoumarin derivatives represent one of the most important scaffolds in biologically active substances, pharmaceuticals and functional materials. Herein, we describe an efficient Pd/amine/Brønsted acid ternary-catalytic multicomponent reaction for the rapid construction of substituted 4-hydroxycoumarin derivatives with adjacent quaternary and tertiary stereocenters via convergent assembly of two in situ generated active intermediates. Furthermore, the late-stage transformations of coumarin derivatives and their in vitro trial of antitumor activity successfully demonstrated the potential utilities of the products as platform molecules.

Abstract:Four tridentate [C,N,S] thiosemicarbazone Pd(II) complexes were applied as catalyst precursors for the Suzuki–Miyaura coupling of a variety of aryl halides and aryl boronic acids. After screening various catalytic conditions, it was found that one dinuclear complex IV containing the bis(diphenylphosphino) ferrocene spacer was able to efficiently catalyze these reactions with yields up to 99% irrespective of the type of aryl substituent on the substrate.
