Abstract:1-(2-Alkynylphenyl) ketoximes react with Lawesson's reagent catalyzed by InCl3 and cyanuric chloride leading to 4-methylene-4H-benzo[d][1,3] thiazines in good yields. This tandem reaction proceeds with high efficiency through Beckmann rearrangement, thioamide formation, and intramolecular nucleophilic cyclization.

Abstract:A convenient and efficient synthesis of 2-substituted benzo[b]thiophenes is described. This one-step reaction of alkylthiobenzaldehyde and activated electrophiles proceeded smoothly in toluene at 120 degrees C with magnesium oxide as a catalyst. The target 2-substituted benzo[b]thiophenes are obtained in moderate yields and the side reaction is inhibited via azeotropic distillation. According to the experiments, a plausible mechanism is presented.

Abstract:A facile and highly efficient three-component reaction of thiazole or benzothiazole carbenes, disubstituted ketenes, and activated alkynes is disclosed. With this methodology, a polysubstituted ring system containing furo[2,3-c]thiazepine core can be constructed from simple and readily accessible starting materials in good yields. The scope and limitation of this transformation were investigated in detail by using various thiazole carbene, ketene, and alkyne components. Furthermore, the synthetic utilities of these unique polyheterocyclic compounds were demonstrated via their Diels-Alder reactions with benzynes to furnish thiazepine-fused 7-oxanorbornadiene derivatives in excellent yields.

Abstract:The core of benzo[f][1,2]thiazonin-5(4H)-one 3,3-dioxide is efficiently assembled through a palladium-catalyzed tandem reaction of 2-alkynylbenzenesulfonamides with 2-(2-bromobenzylidene)cyclobutanones. A range of polycycles with a nine-membered sultam ring are generated in moderate to good yields. During the transformation, three new bonds are formed via a tandem process with the construction of a nine-membered sultam ring. Double carbometallation is believed to be involved with the ring-opening of cyclobutanone.

Abstract:Photoredox-catalyzed aminoarylation and thioamination of unactivated alkenes have been developed, providing novel synthetic routes to access synthetically challenging quaternary carbon-centered benzoindolizidinones and trifluoromethylthiolated piperidines using readily available starting materials. Notably, these transformations were enabled by merging amidyl radical generation from N-alkyl benzamides with oxidant incorporation. Density functional theory calculations were performed to understand the reaction mechanism and to rationalize the regioselectivities. Moreover, the newly developed catalytic aminoarylation provided a convenient synthetic route for natural product tylophorine and its gem-dimethyl analogues with greatly improved drug-like properties such as enhanced solubility and stability. Abstract:A visible light catalyzed three-component tandem annulation strategy for the synthesis of multiply substituted 1,2,4-triazole-3,5-diamines from phenylhydrazines, isothiocyanatobenzenes and 1,1,3,3-tetramethylguanidine is reported herein. The transformations proceed under simple and mild reaction conditions without the requirement of any external metals, ligands, and reductants. This one-step procedure provides facile access to highly substituted 1,2,4-triazole-3,5-diamines, which could be used in the field of medicinal chemistry.
