Abstract:Homotaurine and a series of 1-substituted homotaurines were readily synthesized in satisfactory to good yields via the Michael addition of thioacetic acid to aliphatic and aromatic alpha,beta-unsaturated nitriles followed by lithium aluminum hydride mediated reduction and performic acid oxidation. The synthesis of 1,1-disubstituted homotaurines was attempted with beta,beta-disubstituted acrylonitriles as starting materials but failed due to steric hindrance. The current process is an efficient method for the synthesis of 1-substituted homotaurines.

Abstract:Various substituted homotaurines (=3-aminopropane-1-sulfonic acids) 6 were readily synthesized in satisfactory to good yields via the Michael addition of thioacetic acid to alk-2-enamides 3 (4), followed by LiAlH4 reduction (5) and performic acid oxidation (Scheme1). The configuration of anti'-disubstituted homotaurine anti'-6h was deduced from the 3-(acetylthio)alkanamide (=S-(3-amino-1,2-dimethyl-3-oxopropyl) ethanethioate)anti'-4h formed in the Michael addition, which was identified via the Karplus equation analysis, and confirmed by X-ray diffraction analysis. The current route is an efficient method to synthesize diverse substituted homotaurines, including 1-, 2-, and N-monosubstituted, as well as 1,2-, 1,N-, 2,N-, and N,N-disubstituted homotaurines (Table).

Abstract:Taurine and substituted taurines present a group of important structural elements in many natural products. Various substituted taurines, including 1- and 2-substituted, 1,1-, syn-1,2-, and anti-1,2-disubstituted taurines, were synthesized from the corresponding nitroolefins via Michael addition with thioacetic acid, oxidation with peroxyformic acid, and the catalytic hydrogenation under the catalysis of palladium on carbon or platinum dioxide. It is a general, versatile, and salt-free method for the preparation of substituted taurines, especially for syn- and anti-1,2-disubstituted taurines and some taurines with more bulky substituents. The stereostructures of both syn- and anti-1,2-disubstituted taurines were deduced from the nitroalkyl thioacetates in the Michael addition, which were identified via the Karplus equation analysis and computational analysis, and finally confirmed by the XRD single crystal analysis. The diastereoselectivity in the Michael addition was rationalized with the Cram rule.

Abstract:2-Bromo-5-hydroxy-4-methoxybenzaldehyde(I)and 5-hydroxy-4-methoxy-2-nitrobenzaldehyde(II)were synthesized.The intermediates 2-bromo-4,5-dimethoxybenzaldehyde and 4,5-dimethoxy-2-nitrobenzaldehyde were synthesized firstly from veratraldehyde through bromination and nitration respectively.In the presence of molar quantity methionine,the 5th-position methoxys of the intermediates were in turn hydrolysed to hydoxy by concentrated sulfuric acid to give I and II.The chemical structures of the titled compounds were confirmed by 1H-NMR,IR and MS spectrum.The yields of compounds I and II are 85.0% and 65.0% respectively.The orientations of the bromination and nitration steps were suggested and discussed.The mechanism of methionine in the hydrolysis step may involve in the formation of the multiple hydrogen bonded association complex,and the calculated single point energy(E=-1532.5 hartree)of the multiple hydrogen bonded association complex seems to show it is possibly existed at the RHF/6-31G level.

Abstract:Thiiranes are important starting materials for the synthesis of substituted taurines. The regioselectivity of ring-opening reactions of thiiranes with ammonia in the presence of silver nitrate was investigated. The results of the ring-opening reaction and subsequent peroxy acid oxidation indicate that alkyl-substituted thiiranes give rise to 1-monoalkyl- and 1,1-dialkyltaurines, whereas aryl-substituted thiiranes produce 2-aryl-, 2-alkyl-2-aryl-, and 2,2-diaryltaurines. This shows that alkyl-substituted thiiranes were attacked on their less-substituted ring carbon atoms, while aryl-substituted thiiranes were attacked oil their more substituted ring carbon atoms. The current method is all effective and atom-economic route for the synthesis of mono- and disubstituted alpha-alkyl- and beta-aryl-substituted taurines.

This method was also used for the synthesis of trans-cyclic taurines.

Abstract:Various substituted taurines have been synthesized expeditiously and practically in satisfactory to good yields directly from amino alcohols using a two-step, one-pot procedure, in which the amino alcohols undergo sulfuric acid esterification and subsequent sodium sulfite substitution. Treatment of amino-substituted secondary alcohols using the same procedure gave 2-substituted or 1,2-disubstituted taurines, indicating the formation of aziridines as intermediates during the substitution step. This method is one of the most efficient routes for preparing structurally diverse 2-substituted and 1,2- and 2,2-disubstituted taurines.
