Abstract:A palladium-catalyzed domino cyclization/cycloisomerization reaction of alkyne-tethered carbamoyl chlorides with (E)-β-chloroenones is reported. This reaction proceeds via a syn-carbopalladation of the alkyne, followed by a vinyl-PdII-catalyzed cycloisomerization of the (E)-β-chloroenone cascade, which provides an efficient method to synthesize furan-linked methylene oxindoles. The reaction features stereodefined vinyl-PdII species, high to excellent 5-exo/6-endo selectivity, excellent Z/E selectivity, and the sequential formation of three bonds and bis-heterocycles. The strategy for the synthesis of furan-containing benzofurans has also been demonstrated.

Abstract:A highly regio- and stereoselctive palladium-catalyzed domino reaction of functionalized aryl allyl ethers has been developed. Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:Highly arylated heteroarenes constitute an intriguing class of molecular scaffolds for material science applications. Among these, tetraarylated furans have demonstrated differentiated properties as compared to other similar heterocyclic cores. Among tetraarylated furans, the synthetic complexity increases drastically with increasing number of different peripheral aryl groups. There are only a very limited number of methodologies developed to access furans with four different (hetero)aryl substituents, none of which involve oxidative coupling on furan core. Herein, we report the first methodology based on a sequential two-fold oxidative C-C coupling of furans with indoles to access bis(indolyl)furans - a new class of tetra-(hetero)arylated furans with up to four different substituents. Moreover, the reaction is mediated by inexpensive, earth-abundant FeCl3.6H2O and displays wide substrate scope and aqueous compatibility. Through the characterization of the photophysical and electrochemical properties of this novel class of furans, we present the first validation of the extensively studied aggregation-caused quenching (ACQ) property of tetraarylated furans beyond phenyls as peripheral groups.

Abstract:We explored the reactivity of various furans towards 2‐azidobenzaldehydes, heterocyclic azidoaldehydes, or substituted 2‐azidobenzyl alcohols to form 2‐(2‐azidobenzyl)furan derivatives, unveiling the remarkable compatibility of the azide group with conditions typical of Friedel‐Crafts reactions. Moreover, we demonstrated that compounds of this type can be successfully synthesized by diazotization/azidation sequence from 2‐(2‐aminobenzyl)furans bearing electron‐donating substituents, while effectively circumventing undesired side reactions. Additionally, we presented a synthetic methodology for the preparation of 2‐(2‐acylvinyl)indoles, that involves thermal generation of nitrenes to initiate furan ring opening. Remarkably, our approach entails a nonconjugated configuration of the azidoaryl group and the furan ring, setting it apart from classical indole syntheses. The broad applicability and high efficiency of our method, which utilizes readily available starting materials, underscore its versatility as a synthetic tool of significant promise.
