Abstract:An efficient difluorohydroxylation of substituted indoles leading to 3,3-difluoroindolin-2-ols with good yields by using Selectfluor as the electrophilic fluorinating reagent has been developed. In this methodology, the indole rings were difluorinated highly regioselectively at the C3 carbon site. This protocol is practically convenient, easily handled under mild conditions, and provides an efficient way to produce the unique difluorinated indolin-2-ol structure. When alcohols were used as the nucleophiles instead of H(2)O, the corresponding products were obtained in moderate yields. Based on the experimental observations, a plausible mechanism is proposed.

Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:An oxa-6 pi-electrocyclization of difluoroenoxysilanes with diaryl 2-indolylmethanols has been developed. In addition, a rarely reported C3-nucleophilic [3+2] cycloaddition of difluoroenoxysilanes with dialkyl 2-indolylmethanols has been disclosed. This divergent cycloaddition approach affording readily available difluoroenoxysilanes as three-atom and C2 synthons provides rapid access to fluoro 2H-pyrano[3,4-b]indoles and gem-difluoro cyclopenta[b]indoles in good to excellent yields with good functional group tolerance. The metal-free and mild conditions using only HFIP as the solvent without any external acid catalyst illuminate practical and environmentally benign advantages.

Abstract:Indoles as important N-heteroaromatic skeletons are widely used in the fields of pharmaceuticals, agrochemicals and biological sciences, but the N-trifluoromethyl indoles have been rarely explored. So far, the main accesses to N-trifluoromethyl indoles include oxidative desulfurization-fluorination of dithiocarbamates and Fisher indole synthesis via the reaction of N-CF3 hydrazine and ketones. We report herein the efficient straightforward strategies for the synthesis of N-trifluoromethyl indoles through desulfurization-fluorination/cyclization of 2-alkynylaryl isothiocyanates or 2-alkynylanilines under mild conditions. These cascade reactions possess good substrate scope and functional group compatibility, providing a variety of N-trifluoromethyl indoles in moderate to high yileds.

Abstract:Presented herein is a selective synthesis of 3-(α-fluorovinyl)indoles and 3-acylindoles via the coupling of 1-phenylpyrazolidinones with α,α-difluoromethylene alkynes. Mechanistically, the formation of 3-(α-fluorovinyl)indoles is resulted from a cascade process including Rh(III)-catalyzed ortho-C–H bond cleavage, regioselective triple bond insertion, pyrazolidinone ring-opening, indole ring-formation and HF elimination. Interestingly, when this reaction was carried out in CH3OH/H2O instead of CH3CN, the in situ formed 3-(α-fluorovinyl)indoles readily undergo a hydration process to afford 3-acylindole derivatives. Of note, this protocol features with controllable selectivity, simultaneous formation of both the heterocyclic scaffold and the monofluoroalkenyl moiety, atom-economy, substrate diversity, good functional group tolerance and redox-neutral reaction conditions. Furthermore, anticancer screening of selected products against two human cancer cell lines demonstrated their potential as lead compounds for drug development.

Abstract:A metal-free, highly regioselective C3 difluorination and C2 oxidation of N-substituted indoles, with the aid of in situ generated PhIF2 in the presence of two types of fluorinating agents, was reported. Abstract:Here, we have successfully achieved the addition of difluoroacetate radicals to 2‐aryl‐3H‐indol‐3‐ones, enabling various difluoroalkylations of C2‐tetrasubstituted 2‐aryl indolin‐3‐ones in a highly efficient and economical manner. It is worth mentioning that these difluoroalkylation compounds can be easily transformed into various derivatives under mild reaction conditions. Control experiments suggest the involvement of ethyl difluoroacetate radical species in the reaction.

Abstract:A convenient and efficient method for the synthesis of optically active difluoro-substituted indoline derivatives starting from the corresponding 3 H -indoles by chiral phosphoric acid-catalyzed transfer hydrogenation was developed. Using Hantzsch ester as the hydrogen source under mild reaction conditions, the target products can be obtained with excellent yield and enantioselectivity.

Abstract:A highly selective nickel-catalyzed C-H trifluoroethylation of heteroarenes was developed with the assistance of a monodentate directing group. This protocol provides efficient access to various trifluoroethyl-substituted heteroarenes, including indoles, pyrroles, furans, and thiophenes, with commercially available CF3CH2I as an alkylation reagent. This robust catalytic procedure is scalable and tolerates a broad range of functional groups. Moreover, multifluoroalkylation of indoles is also viable.
