Abstract:Comprehensive Summary Trifluoromethyl group is one of the most frequently available and promising functional groups in pharmaceutical, agrochemical as well as material molecules. Developing synthetic methods for the synthesis of trifluoromethyl functionalized compounds by trifluoromethylation reactions occupies significant position in modern organic chemistry. On other hand, as a typical tool of sustainable synthesis, transition metal‐free synthesis possesses unique advantages in atom economy, easy product purification as well as excellent eco‐tolerance. Consequently, transition metal‐free trifluoromethylation reactions constitute the most important approaches for accessing trifluoromethylated molecules. Herein, the advances in the transition metal‐free C‐H trifluoromethylation reactions are reviewed for the first time. According to the updated state‐of‐art, both the catalytic reactions by conventional heating chemistry and the more sustainable photocatalysis are included, covering the trifluoromethylation reactions of C(sp3)‐H bond, aromatic C(sp2)‐H bond as well as the non‐aromatic C(sp2)‐H bond in different substrates over the last five years (2017‐2021). Abstract:The introduction of a trifluoromethyl (CF3) group can dramatically improve a compound's biological properties. Despite the well-established importance of trifluoromethylated compounds, general methods for the trifluoromethylation of alkyl C–H bonds remain elusive. Here we report the development of a dual-catalytic C(sp3)–H trifluoromethylation through the merger of light-driven, decatungstate-catalysed hydrogen atom transfer and copper catalysis. This metallaphotoredox methodology enables the direct conversion of both strong aliphatic and benzylic C–H bonds into the corresponding C(sp3)–CF3 products in a single step using a bench-stable, commercially available trifluoromethylation reagent. The reaction requires only a single equivalent of substrate and proceeds with excellent selectivity for positions distal to unprotected amines. To demonstrate the utility of this new methodology for late-stage functionalization, we have directly derivatized a broad range of approved drugs and natural products to generate valuable trifluoromethylated analogues. Preliminary mechanistic experiments reveal that a 'Cu–CF3' species is formed during this process and the critical C(sp3)–CF3 bond-forming step involves the copper catalyst.

Abstract:In the past 5 years, transition-metal-mediated trifluoromethylation for the construction of various CF3-containing building blocks has been the focus of recent research in both industrial and academic communities. Progresses in the construction of C(sp(2))-CF3 bonds and C(sp)-CF3 have been well reviewed. This Letter will focus on the cases of transition-metal-mediated C(sp(3))-CF3 bond formation, which involves the trifluoromethylation of sp(3)-hybridized C-X bonds, alkyl organometallic reagents, sp(3)-hybridized C-H bonds, and alkene derivatives. Abstract:The selective C‐F bonds functionalization in available trifluoromethyl compounds is an economic route for obtaining high‐value organic fluorine compounds. However, due to the high C‐F bonds dissociation energy in trifluoromethyl group, the strength of the bond decreases continuously during defluorination, posing a challenge in modifying such molecules without over‐conversion. Over the past few years, there has been significant progress in developing efficient methods for constructing difluoromethylene unit, using approaches such as radical chemistry, photochemistry, electrochemistry, and organometallic chemistry. These methods involve difluorocarbon radicals, difluorocarbon cations, difluorocarbon anions, and trifluoromethyl carbenes as intermediates. In this review, we summarized the recent five years of research achievements in this field for selective single C‐F bond cleavage in trifluoromethyl aromatic and carbonyl compounds to gain diverse difluoromethylene‐containing organic molecules via different intermediate strategies.

Abstract:The incorporation of fluorine-containing moieties into organic compounds is of great importance in pharmaceutical, agricultural, and materials science. Within these organofluorides, the trifluoromethyl group is one of the most important motifs. In recent years, the trifluoromethyl group has attracted more and more attention, and many trifluoromethylated compounds have been found to possess special activities. However, until now, only a few methods have been developed to achieve this efficiently using Umemoto's reagents. This review highlights recent developments in the direct introduction of a trifluoromethyl group into organic compounds with Umemoto's reagents. Seven approaches to the trifluoromethylation of organic compounds are summarized: (i) trifluoromethylation of arenes, (ii) trifluoromethylation of alkenes, (iii) trifluoromethylation of terminal alkynes, (iv) deoxygenative trifluoromethylation of benzylic xanthates, (v) trifluoromethylation of ketoesters, (vi) trifluoromethylation of aryl boronic acids and aromatic amines (synthesis of ArCF3) and (vii) trifluoromethylation of biphenyl isocyanide derivatives.

Abstract:Transition-metal-catalyzed direct CH functionalization utilizing unsaturated hydrocarbons have been established as a robust strategy to increase molecular complexity without the need of substrate prefunctionalization. Especially, the CH functionalization mode involving several innovative fluorinated π-systems, including difluorinated alkynes, cyclopropenes as well as allenes, has been intensively investigated in recent years, which not only enriches the new connotation of this hot topic but also opens up a new field of vision on how to promote organic integration of transition-metal-catalyzed CH functionalization with fluorine chemistry, even with pharmaceutical science. Induced by their inherently distinctive fluorine effects, these fluorine-functionalized π-compounds have efficiently expanded the scope of the CH functionalization reactions by introducing enhanced chemoselectivity, regioselectivity and stereoselectivity, which greatly enriches the connotation of CH activation chemistry, and ultimately, promoting the development of organic chemistry. In this review, we provide a comprehensive summary of the latest progress in transition-metal-catalyzed CH functionalization with recently developed fluorinated π-systems since 2017, with particular focus on a rational analysis on reaction development and the elucidation of their mechanisms revealed by integrating computational investigation with experimental data. Finally, several challenges and future perspectives on this topic are also discussed. We believe that this review can offer a profound insight into the synergistic combination of experimental and theoretical studies for the future design of unexplored CH functionalization reaction modes driven by diverse CH activation substrates and fluorinated π-partners. This, in turn, is expected to spur the discovery and development of innovative reaction paradigms in this field of transition-metal-catalyzed CH activation/functionalization.

Abstract:Selective functionalization of C–F bonds in trifluoromethyl groups has recently received a growing interest, as it offers atom- and step-economic pathways to access highly valuable mono- and difluoroalkyl-substituted organic molecules using simple and inexpensive trifluoromethyl sources as the starting materials. In this regard, impressive progress has been made on the defluorinative functionalization reactions that proceed via radical intermediates. Nevertheless, it is still a great challenge to precisely control the defluorination process, due to the continuous decrease of the C–F bond strength after the replacement of one or two fluorine atoms with various functionalities. This review article is aimed to provide a brief overview of recently reported methods used to functionalize C–F bonds of CF3 groups via radical intermediates. Abstract:The introduction of trifluoromethylsulfinyl group into the required target molecules has become an indispensable strategy for pharmaceutical chemists to improve the success of new drug discovery. Compared with the indirect methods, direct trifluoromethylsulfinylation by employing available trifluoromethylsulfinylating reagents is an important and elegant strategy for the rapid assembly of valuable CF3S(O)‐derived organic molecules. This review focuses on the recent synthetic approaches and future perspectives of trifluoromethylsulfinyl compounds based on the indirect and direct methods in this area. These transformations proceed effectively with high yields and good functional group compatibility under mild conditions.

Abstract:Palladium-catalyzed CF bond formation via directed CH activation has recently emerged as a significant method to introduce fluorine atoms into organic molecules in an atom- and step-economical path. At the interface between fluorine chemistry and synthetic organic chemistry, the development of direct fluorinations of the CH bond remains sluggish, due to the major challenge of reductive elimination to afford the CF bond. One strategy is to oxidize palladium(II) species to more electron-deficient palladium(IV or III) fluorides to promote the CF bond forming reductive elimination, which opens up palladium-catalyzed CH fluorination as a powerful transformation. This focused review gives a brief summary of the mere handful of examples, including C(sp(2))F and C(sp(3))F bond formations, in this field so far.

Abstract:Trifluoroethyl group served as the bioisosteres of ethyl or ethoxy group widely exists in the fields of pharmaceuticals and agrochemicals. Developing novel strategies of introducing trifluoroethyl group into various skeletons has become a hot topic in organic synthetic chemistry. In this review, the trifluoroethylation reaction of various partners such as alkynes, alkenes, functionalized arenes, amines and so on were summarized. This review was categorized into four parts according to the bond formation: C(sp)-CH2CF3, C(sp2)-CH2CF3, C(sp3)-CH2CF3, and X (X = N, O, S, Se)-CH2CF3. Some representative examples along with reaction mechanisms were also discussed.

Abstract:With the Cu(I) catalyst and the base, alkyl chlorides could react with TMSCF3to realize the trifluoromethylation of alkyl chlorides under irradiation in DMAc. This green and effective method avoids high temperature, poisonous and harmful reaction conditions. The trifluoromethylation of alkyl chlorides can successfully construct unactivated- C(sp3)-CF3bond,which is of great value in organic chemistry.

Abstract:Abstract The structural motifs of trifluoromethylated aromatics (Ar‐CF3) have been widely employed in various industrial fields. The high lipophilicity, strong electron‐withdrawing ability, and characteristic size of the trifluoromethyl group is a key influence in the design of pharmaceuticals, agrochemicals, dyes, liquid crystals, and polymers. Therefore, a great deal of attention has been paid to convenient protocols of introducing trifluoromethyl groups into aromatic rings. With respect to the high regiochemical fidelity in aromatic substitution, the cross‐coupling of aryl halides with CF3Cu is one of the most powerful and versatile methods to construct trifluoromethylated aromatics.
