Abstract:A palladium(II)-catalyzed C-H arylation directed by tetrazole, a metabolically stable surrogate for the carboxylic acid group in drug design, has been developed. Excellent mono-/di-selectivity was achieved through adjustment of the protecting site on the tetrazole ring. The synthetic utility of this new transformation was demonstrated in the concise total synthesis of Losartan.

Abstract:The reactivity of 2‐benzyl‐1,2,3‐triazole in palladium‐catalyzed direct arylation was studied. The reaction conditions for the selective synthesis of 2‐benzyl‐4‐aryl‐1,2,3‐triazoles in moderate to high yields using phosphine‐free Pd(OAc)2 catalyst and inexpensive KOAc base have been found. Then, from these 4‐aryl‐1,2,3‐triazoles, the palladium‐catalyzed C−H bond functionalization of the C5‐position allowed the synthesis of the corresponding 4,5‐diarylated 2‐benzyl‐1,2,3‐triazoles. This selective 4,5‐diarylation of 2‐benzyl‐1,2,3‐triazole was successfully applied for the straightforward building of the π‐extended polycyclic heteroaromatic structures phenanthro[9,10‐d][1,2,3]triazoles through Pd‐catalyzed C4‐ and C5‐intermolecular arylations followed by Pd‐catalyzed C−H intramolecular arylation.

Abstract:In this paper, tetrazole ring systems were sythesized by using readily available materials via anodic cyanations of suitably N-substituted pyrrole as key step. The electrooxidation of several 1-arylpyrroles was carried out in methanol containing sodium cyanide at a Pt anode in a divided cell. In all instances, replacement of a heteroaromatic hydrogen by a cyano group occurred regio-selectively. The corresponding pyrrole cyanides were obtained in yields ranging from 76 to 85%. The advantages of electrochemistry synthesis of pyrrole cyanides are as follows: the reaction condition is simple, the cost is low and the products is highly pure. The new tetrazole derivatives which may be non-peptidic compounds with I potential angiotensin U antagonist properties, were synthesized by the formal [2 + 3] cycloaddition of NaN3 and pyrrole cyanides.

Abstract:Oxidant-controlled selective mono-/di-alkenylation of aryl C-H bonds via Pd-catalysis is reported. The substrate scopes for both the mono- and di-alkenylation were good. Thioether was used as the directing group and the product can be transformed to a useful sulfoxide-olefin ligand by simple oxidation in quantitative yield.

Abstract:Herein, we present a novel and highly efficient strategy for the synthesis of dihydropyrazoles via Pd‐catalyzed carboamination reaction of alkenyl hydrazones and aryl triflates. The simultaneous formation of C–C and C–N bonds in a single step is particularly appealing. The methodology provides a practical and efficient synthesis of various diversely aryl substituted dihydropyrazoles in good yields.

Abstract:Non-directed C–H arylation is one of the most efficient methods to synthesize biaryl compounds without the need of the prefuctionalization of starting materials, or the installment and removal of directing groups on the substrate. A direct C–H arylation of simple arenes as limiting reactants remains a challenge. Here we disclose a non-directed C–H arylation of anisole derivatives as limiting reagents with aryl iodides under mild reaction conditions. The arylated products are obtained in synthetically useful yields and the arylation of bioactive molecules is also demonstrated. Key to the success of this methodology is the use of a one-step synthesized S,O-ligand.

Abstract:We report herein an efficient Pd-catalyzed direct C-H bond functionalization of heteroarenes via an isocyanide insertion strategy for the synthesis of di-(hetero)aryl ketones and di-(hetero)aryl alkylamines. The methodology involves a three component reaction between an azole, a haloarene and an isocyanide resulting in the formation of an imine, which in turn is either hydrolyzed or reduced to get the desired product.
