Abstract:A novel strategy for protecting dihydroxyl groups of catechols has been developed. Base-mediated cyclizations of catechols with 1,3-dibromopropane provided the corresponding benzo[b] 1,4-dioxepans, and herefrom the protecting group was easily cleaved by aluminum chloride. The preparation of the antibacterial and antifungal agent 4-(2-aminothiazol-4-yl) benzene-1,2-diol from catechol reliably verified its availability amenable to various harsh reaction conditions.

Abstract:A library of 32 members of 3,4-dihydro-3-oxo-2H-1,4-benzoxazines has been synthesized from two substituted 2-aminophenols via microwave-assisted one-pot regioselective annulation of 2-bromoalkanoates and subsequent Suzuki-Miyaura cross-coupling of the chloro-substituted scaffolds. The latter transformation was carried out using our Aphos-Pd(OAc)(2) catalyst and the coupling of the aryl chlorides with arylboronic acids proceeded under mild reaction conditions at 60-80 degrees C in THF-H2O (10:1) in the presence of K3PO4 center dot 3H(2)O as the base to furnish the corresponding biaryl products in 80-98% yields. Abstract:Pd(II)-catalyzed intermolecular 1,2-aminooxygenation and 1,2-oxyamination of conjugated dienes have been developed. The chemoselective preparation of a variety of 2-functionalized and 3-functionalized 1,4-benzoxazine derivatives was accomplished via the adjustment of a coordinating solvent. Oxygen was successfully used in this oxidative difunctionalization of alkenes. Good yields and selectivities were obtained for most products. A product bearing a spiro structure was also obtained from a 2,3-disubstituted-1,3-diene.

Abstract:A matter of protection: The outcome of a divergent organocatalytic aza-Michael/aldol cascade process toward quinolines and 1,4-dihydroquinolines depends on the choice of the N-protecting group (see scheme; TEA = triethylamine, TMS = trimethylsilyl). Use of an electron-donating sulfonyl group results in an unanticipated aza-Michael/aldol/aromatization cascade to give polysubstituted quinolines (right). In contrast, chiral 1,4-dihydroquinolines are obtained with an electron-withdrawing sulfonyl group (left).

Abstract:A mild and operationally simple protocol for the direct C-H functionalization of the 1-position of tetrahydrocarbazoles is reported. Abstract:Employing a chiral spirodiphosphine monoxide ligand with 1,1'-spirobiindane backbone (SDP(O)), a desymmetrization strategy of Pd-catalyzed intramolecular asymmetric aryl C-O coupling of 2-(2-halophenoxy)propane-1,3-diols, was developed. The SDP(O) ligand shows much better results than its SDP counterpart. The protocol provides an efficient and highly enantioselective method for the synthesis of 2-hydroxymethyl-1,4-benzodioxanes. Density functional theory studies provide a model that accounts for the origin of the enantioselectivity.

Abstract:Various benzyl ethers of carbohydrates have been selectively oxidized to the corresponding benzoate esters 2a-j with tetrabutylammonium peroxydisulfate under neutral conditions followed by debenzoylation with sodium methoxide to afford the debenzylated carbohydrates 3a-j in good overall yields. Noteworthy is that highly chemoselective reactions were achieved with the co-existence of other functionalities.

Abstract:An efficient and convenient cobalt-catalyzed ortho-C(sp(2))-H amidation of benzaldehydes employing dioxazolones as the aminating reagent has been developed. The key feature of this protocol is the use of green and economic earth-abundant metals cobalt as the catalyst with the p-chloroaniline as the transient directing group. Further application of our approach was demonstrated by the synthesis of C1r serine protease inhibitor 45 and elastase inhibitor 49.

Abstract:Benzoxazines are important structures in pharmaceuticals and functional molecules. We herein described an electrochemical oxidative strategy for the synthesis of benzoxazines without transition-metal catalysts and external oxidants. In a undivided cell, a series of olefinic amides can be transferred to difluoromethylated benzoxazines in up to 87% yield via oxydifluoromethylation with CF2HSO2Na as the difluoromethylating reagent.
