Abstract:D-glucopyranosid e and stenophylline B-3-0-~D-glucopyranoside) and two novel steroidal alkaloids . The structures of the new compounds have been established by a combination of spectroscopic methods as 3angeloylzygadenine-I3-N-oxide and (20S,25S)-22,26-epiminocholesta-5,22-diene-3I3,12a.-diol (named veramitaline). The chemotaxonomic significance of the isolates are discussed briefly. . Most of Veratrum species, distributing mainly in the temperate regions of the Northern Hemisphere, are well known for their pharmacological potentials. Although a lot of Veratrum plants have been examined chemically to lead to the characterization of more than ninety steroidal alkaloids, much renewed attention is being paid to biologically active and/or structurally novel alkaloidal principles presumably due to their diversified biological actIvIties and structural novelties as reported recently. 1-4 As a follow-up to the phytochemical attention to Veratrum taliense Loes. f. (Liliaceae) , a frequently used traditional Chinese medicine. S • 6 The reinvestigation of the roots and rhizomes of the title species was undertaken. Abstract:Phytochemical investigation of roots and rhizomes of Veratrum taliense yielded a new and six known steroidal alkaloids as well as a new and one reported stilbene derivative. By a combination of spectral methods (IR, MS, H-1- and C-13-NMR, COSY, HMQC, HMBC, and NOESY), the structure of the new alkaloid was established as 15-angeloylgermine while the known ones were identified as 15-(2-methylbutyroyl)germine, jervine, 3-veratroylzygadenine, germine, veramiline 3-O-beta-D-glucopyranoside and stenophylline B-3-O-beta-D-glucopyranoside. The new stilbenoid, named veraphenol, was determined to be 2-(3',5'-dihydroxyphenyl)-6-hydroxybenzofuran, and the known one was shown to be resveratrol. The in vitro enzyme assay indicated that 3-veratroylzygadenine and resveratrol are inhibitors of xanthine oxidase. The enzyme inhibitory action of resveratrol, the most active compound found so far in V. taliense, is dose-dependent with the IC50 value at 30 mu M (the IC50 value of allopurinol used as a positive control in the study is 10 mu M).

Abstract:Two new 18-norschiartane-type schinortriterpenoids, namely wuwezidilactones Q (1) and R (2) were isolated from the stems of Schisandra lancifolia. Their structures were determined on the basis of extensive spectroscopic analysis. The absolute configurations of the new compounds were further determined by ROESY and an empirical comparison of their experimental ECD spectra with literature.

Abstract:Two new steroidal alkaloids (1–2), together with seven known related steroidal alkaloids (3–9), were isolated from the rhizomes of Veratrum nigrum L. Their structures were elucidated by extensive spectroscopic analysis, and by comparison with literature data. Compound 1 possessed a rare 1, 3-oxazolidine unit within varazine-type alkaloids, and 2 was a 9-hydroxy-4-one derivative of 3-veratroylgermine. All isolates were evaluated inhibit tomato yellow leaf curl virus (TYLCV) activity. Compounds 5 and 7 (40 μg/mL) showed a significant anti-TYLCV activity in the host Nicotiana benthamiana with inhibition rates 74.6% and 63.4%, respectively, which are higher than that of the positive control ningnamycin (51.4%).

Abstract:OBJECTIVE:To study the steroidal alkaloids in Veratrum dahuricum. METHOD:The compounds were isolated and purified by various column chromatographic methods. Their structures were identified by spectroscopic analysis. RESULT:Five compounds were isolated and identified as (20R, 22S, 25S)-veratra-5,13-dien-3beta-ol (1), angeloylzygadenine (2), 15-O-(2-methylbutanoyl)-3-O-veatroylprotoverine (3), 20-isoveratramine (4), veratramine (5). CONCLUSION:Compound 1 was a new compound, compounds 24 were obtained from the plant for the first time.

Abstract:Objective: To investigate the chemical constituents of the branches and leaves of Uvaria kurzii. Method: Compounds were isolated and purified by silica gel and Sephadex LH-20 column chromatography. Their chemical structures were identified on the basis of physicochemical properties and spectral data. Result: Nineteen compounds were isolated and identified as:β-sitosterol hexadecanoate ( 1 ) , stigmasterol hexadecanoate (2) ,β-acetylsitosterol (3) ,β-acetylstigmasterol (4) , tetratriacontanol (5) , dotriacontanoic acid (6), β-sitosterol (7), stigmasterol (8), 5α-stigmast-3,6-dione (9), 5α-stigmast-22-ene-3,6-dione (10), vanillic acid ( 11 ), protocatechuic acid ( 12 ), N-( p-trans-coumaroyl ) tyramine ( 13 ) , kaempferol-3-O-β-D-( 6"-O-p-coumaryoyl ) galactopyrannoside (14) , kaempferol-3-O-rutinoide (15) , rutin (16) , daucosterol ( 17 ) , L-quebrachitol( 18 ) , allantoin ( 19 ) , respectively.Conclusion: Compounds 1-6, 9, 10, 13, 18, 19 were isolated from Annonaceae plants; Compounds 14-16 were obtained from the genus Uvaria; and 7, 8, 11, 12, 17 were separated from this plant respectively for the first time.

Compound 3-5 showed inhabitation activities against tumor cell lines A549, Bel7402, BGC823, HCT-8, A2780, respectively.

Abstract:Bioassay-guided fractionation of the ethanol extract of the roots and rhizomes of Veratrum taliense yielded two new and thirteen known steroidal alkaloids. The structures of the two new compounds, neoverataline A and B, were established by extensive spectroscopic analyses to be 3,4-secocevane-4,9-olid-14,15,16,20-tetra-ol-3-oic acid and 3,4-secocevane-4,9-olid-7,14,15,16,20-penta-ol-3-oic acid, respectively, and are a novel carbon skelton. All of the fifteen alkaloids were subjected to in vitro antifungal assays, which showed that the verazine- (veramitaline, stenophylline B, stenophylline B-3-O-β-d-glucopyranoside, veramiline-3-O-β-d-glucopyranoside) and jerveratrum-type (jervine, jervine-3-O-β-d-glucopyranoside) alkaloids exhibited strong antifungal activities against the phytopathogenic fungus Phytophthora capisis with MICs of 160, 120, 160, 80, 80 and 120 μg·L−1, respectively. Furthermore, the verazine-type alkaloids stenophylline B, stenophylline B 3-O-β-d-glucopyranoside and veramiline 3-O-β-d-glucopyranoside were shown to also inhibit the growth of another fungal phytopathogen Rhizoctonia cerealis with MICs of 160, 120 and 120 μg mL−1. The MICs of triadimefon (an antifungal agrochemical used herein as a positive control) against P. capisis and R. cerealis were 120 and 80 μg mL−1, respectively. A preliminary structure–activity relationship regarding these alkaloids has been formulated.

Abstract:Two distinctive alkaloids with 6/6/6/5/6/6 fused rings, in which a previously unidentified linkage of C-12/23 generates a rigid skeleton, resulting in a new subtype of steroidal alkaloid, were isolated from Veratrum grandiflorum. Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.

Abstract:Two new iridoids, 3-(5-(methoxycarbonyl)-2-oxo-2H-pyran-3-yl)butanoic acid, named verbeofflin I (1), and 7-hydroxydehydrohastatoside (2), were isolated from the aerial part of Verbena officinalis L, along with three known iridoids, verbenalin (3), 3,4-dihydroverbenalin (4), hastatoside (5) by means of various column chromatography steps. The structures of these compounds were elucidated through analysis of their spectroscopic data obtained using 1D and 2D NMR and MS techniques. Verbeofflin I (1) is the new class of secoiridoid in the family Verbenaceae.

Abstract:Veratrazine A (1), a steroidal alkaloid with a unique 6/5/5 triheterocyclic scaffold as the side chain, was isolated from Veratrum stenophyllum, and its structure was established via spectroscopic analyses and X-ray diffraction. A plausible biosynthetic pathway for 1 is proposed. Bioassy exhibits moderate anti-inflammatory activities in vitro and in vivo.

Abstract:Four new zwitterionic monoterpene indole alkaloids, rhynchophyllioniums A-D (1-4), together with eight known alkaloids (5-12), were isolated from the hook-bearing stems of Uncaria rhynchophylla. Their structures were elucidated by extensive spectroscopic data analysis of MS, 1D and 2D NMR, and ECD, and the zwitterionic forms and absolute configurations of 1 and 2 were unambiguously confirmed by single crystal X-ray diffraction analysis. All the isolates, including the monoterpene indole alkaloids with free C-22 carboxyl group and those with C-22 carboxyl methyl ester, were proved to be naturally coexisting in the herb by LC-MS analysis. This is the first report of monoterpene indole alkaloids that exist in the form of zwitterion. Additionally, the cytotoxic activities of all isolates against A549, HepG2, and MCF-7 cell lines are reported.

Abstract:Two new lignans, named zuihonins E (1) and F (2), were isolated from the stems of Schisandra bicolor Cheng var. tuberculata Law. The structures of the new lignans were elucidated on the basis of extensive spectroscopic analysis, including 1D, 2D NMR, and MS experiments, and their absolute stereochemistry was determined by circular dichroism spectrum. Compounds 1 and 2 did not inhibit the growth of hepatoma carcinoma cell (HepG2), lung carcinoma cell (A549), and human breast carcinoma (MCF-7) cell lines.

Abstract:AIM:To study the chemical constituents of the root of Stellera chamaejasme L.METHODS:Various column chromatographies on silica gel and RP-18 were employed for isolation and purification. Structures of compounds were elucidated by spectral analysis.RESULTS:Eight lignans and three biflavonoids possessing a C-3/C-3" linkage were isolated. They are ruixianglangdusu A (1) and B (2), 4',4'",5,5",7,7"-hexahydroxy-3,3"-biflavone (3), (+)-kusunokinin (4), lirioresinol-B (5), magnolenin C (6), (-)-pinoresinol monomethyl ether (7), (-)-pinoresinol (8), (+)-matairesinol (9), isohinokinin (10) and (-)-eudesmin (11).CONCLUSION:Compounds 1 and 2 are new biflavanones, 1 is enantiomeric to known chamaejasmenin C, 4, 6, 8, 9, 10 and 11 were isolated from this plant for the first time, and 7 was isolated from natural resources for the first time. In vitro bioassays showed that 3 and 8 exhibited antibacterial activity, and 1, 2, 9 and 11 exhibited immunomodulatory activity.

Abstract:Eight undescribed steroidal alkaloid derivatives, including three cevanine-type isosteroidal alkaloids (two N -oxide glycosides and one D-ring aromatization) ( 1 – 3 ), one verazine-type steroidal alkaloid derivative ( 4 ), three solanidine-type steroidal alkaloid glycosides ( 5 – 7 ), and one veratramine-type analogue ( 8 ), along with three known compounds ( 9 – 11 ) were isolated from the bulbs of Fritillaria sinica . Their structures were elucidated by comprehensive analysis of spectroscopic data, acidic hydrolysis, and X-ray crystal diffractions. In the in vitro bioassay, the anti-cancer effect, anti-oxidation and anti-inflammatory activities for the isolates were evaluated at a concentration of 10 μM.
