Abstract:Four previously undescribed iridoid glycosides, including two bis-iridoid glycosides, and three undescribed lignans, together with 16 known analogues, were isolated from the fruits of Gardenia jasminoides Eills. Their structures were elucidated on basis of spectroscopic methods, and the absolute configurations of three of the unknown compounds were determined by interpretation of their electronic circular dichroic (ECD) and [α]20D data. The α-glucosidase inhibitory effects of the isolated compounds were evaluated and all the compounds exhibited slightly inhibitory activity with the values of IC50 greater than 50 μM.

Abstract:A new lignan glucoside, (+)-(7S,8R,8′R)-lyoniresinol 9-O-β-D-(6″-O-trans-sinapoyl)glucopyranoside (1), and a new iridoid glucoside, 10-O-trans-sinapoylgeniposide (2), together with eight known compounds, were isolated from the fruits of Gardenia jasminoides Ellis. The structures of the isolates were elucidated by extensive spectroscopic studies, including UV, IR, 1D and 2D NMR, ESI-MS, HR-ESI-MS, and CD experiments. The short-term-memory-enhancement activities of some compounds were evaluated on an Aβ transgenic drosophila model.

Abstract:Two new iridoid glycosides, 2′-O-cis-coumaroylgardoside (1), and 6′-O-caffeoylioxide (2), were isolated from the fruit of Gardenia jasminoides. The structures of these compounds were elucidated based on spectroscopic analysis (HR-ESI-MS, NMR) and chemical methods. The anti-inflammatory activities of the isolates were evaluated by measuring their inhibitory effects on PGE2 production in LPS stimulated RAW 264.7 macrophages, compounds 1 and 2 could reduce PGE2 levels in LPS-activated RAW 264.7 macrophages with IC50 values of 121.4 and 83.38 μM, respectively.

Abstract:To search for bioactive natural products,we study the chemical constituents from the rhizome of Gardenia jasminoides.The compounds 1-4 were isolated by combining reverse-phase and normal silica gel column chromatography,and their structures were identified by spectral methods.Four known compounds were isolated from the ethanol extracts of the rhizome of Gardenia jasminoides.Their structures were identified by means of spectroscopic analysis as: 10-O-trans-caffeoyl-6α-hydroxygeniposide(1),icariside E5(2),6α-hydroxygeniposide(3),geniposide(4).Compounds 1 and 2 are isolated from the rhizome of Gardenia jasminoides for the first time.

Abstract:Three new 3,4-seco-cycloartane triterpenoids (1-3), including an unusual C-1 oxygenated 3,4-seco-cycloartane triterpenoid lactone (3), along with six known triterpenoids, were isolated from 95% ethanol extract of the flower of Gardenia jasminoides. Their structures were determined by spectroscopic methods, especially 2D NMR and HRESIMS techniques. Compounds 3, 4 (dikamaliartane D), and 7 (ursolic acid) showed promising cytotoxic activity against HepG2 human hepatocellular carcinoma cell line with IC50 values of 9.3, 27.7, and 39.3 mu M, respectively.

Abstract:Seven new iridoid glucosides, 6 "-O-trans-sinapoylgenipin gentiobioside (1), 6 "-O-trans-p-coumaroylgenipin gentiobioside (2), 6 "-O-trans-cinnamoylgenipin gentiobioside (3), 6'-O-trans-p-coumaroylgeniposide (4), 6'-0-trans-p-coumaroylgeniposidic acid (5), 10-O-succinoylgeniposide (6), and 6'-O-acetylgeniposide (7), two new monoterpenoids, 11-(6-O-trans-sinapoylglueopyranosyl)gardendiol (8) and 10-(6-O-trans-sinapoylglucopyranosyl)gardendiol (9), and three known ones, 6'-O-trans-sinapoylgeniposide (10), geniposide (11), and 10-O-acetylgeniposide (12), were isolated from the fruit of Gardenia jasminoides. The structures of these compounds were elucidated on the basis of 1D and 2D NMR spectra analyses. Furthermore, short-term memory assays on an A beta transgenic drosophila model showed that compounds 4 and 6-12 can improve the short-term memory capacity to varying degrees, with compounds 4 and 7 being the most active ones, suggesting that these compounds may have a potential antagonism effect against Alzheimer's disease.

Abstract:Gardenia jasminoides Ellis,one genus of Gardenia,belongs to the family of the Rubiaceae.The fruit of G.jasminoides is widely used as a traditional Chinese medicine named Zhizi.It has significant biologic activities including antipyretic,antianxiety,diuretic effcts,Cooling the Blood and Detoxication.Recently,a large quantity of newly-emerging bioactive constituents from G.jasminoides has been reported.The unique constituents iridoid glucosides,and other types of active principles include organic acid,flavonoids,coumarins,lignans,terpenoids,volatile oils and fatty acids.They entitle the plant to possess several bioactivities,such as anti-inflammatory,antioxidant,cholagogue,diuretic,anticancer,radiological protection,antiphlogistic,antipyretic,analgesic,and antihyperlipidemic effects.In this paper,we reviewed categorization of chemical constituents and pharmacological activities of G.jasminoides from reports since 2004.This review will provide a reference for further studies.

Abstract:Five new pyronane-type monocyclic monoterpenoids, jasminodiol (1), jasminoside H (6), 6'-O-sinapoyljasminoside A (7), 6'-O-sinapoyljasminoside C (8), and jasminoside 1 (9), together with four known analogues, were isolated from the fruit of Gardenia jasminoides. The structures of the new metabolites were characterized using spectroscopic data, and the absolute configurations of 1, 6, and 7 were established using circular dichroism (CD) analysis. Compound 1 showed tyrosinase inhibitory activity (IC(50) 2.2 mM).

Abstract:Two new compounds, ethylconiferin and (-)-lariciresinol 4-(6'''-O-cinnamyl-β-D-glucopyranoside), along with the three known compounds (+)-pinoresinol, (+)-cycloolivil, nobiletin, were isolated from the root of Jasminum girialdii. All these compounds were isolated for the first time from this source. Their structures were elucidated on the basis of extensive spectroscopic analysis and chemical methods. In addition, the in vitro cytotoxic activity of these compounds was evaluated. The results showed that none of the compounds had any significant inhibitory activities on the proliferation of HCT-116 and SW-620 cells.

Abstract:Fragmentation pathways of five iridoid glycosides have been studied by using electrospray ionization multi-stage tandem mass spectrometry (ESI-MSn). The first-stage MS data of the five iridoid glycosides were compared. The MS spectra showed that the adduct ions of iridoid glycosides and the formate anion were diagnostic ions to distinguish iridoid glycosides with a carboxyl group at the C-4 position or an ester group at the C-4 position. The MS fragmentation pathways of the five iridoid glycosides were also studied. Analyzing the product ion spectra of iridoid glycosides, some neutral losses were observed, such as H2O, CO2 and glucose residues, which were very useful for the identification of the functional groups in the structures of iridoid glycosides. Furthermore, specific loss of one molecule of methyl 3-oxopropanoate or 3-oxopropanic acid was firstly discussed, which corresponded to the isomerization of the hemiacetal group in the structure of iridoid aglycone. According to the fragmentation mechanisms and HPLC/MSn data, the structures of five iridoid glycosides in a crude extract of Gardenia jasminoisdes fruit have been identified. Three compounds were compared with standards and the other two were identified as shanzhiside and genipin gentibioside by their MSn data without standard compounds. In order to further validate the veracity of the deduction, genipin gentiobioside was isolated from the extract of Gardenia jasminoisdes fruit using Purification Factory and was further identified by C- and H-NMR. Unveiling molecular mechanisms of pigment synthesis in gardenia (Gardenia jasminoides) fruits through integrative transcriptomics and metabolomics analysis

Abstract:This study conducted a combined transcriptomics and metabolomics analysis in premature and mature developmental stages of Gardenia jasminoides Ellis fruits to identify the molecular mechanisms of pigment synthesis. The transcriptomics data produced high-quality clean data amounting to 46.98 gigabytes, exhibiting a mapping ratio of 86.36% to 91.43%. Transcriptomics analysis successfully identified about 3,914 differentially expressed genes which are associated with pivotal biological processes, including photosynthesis, chlorophyll, biosynthetic processes, and protein-chromophore linkage pathways. Functional diversity was clarified by the Clusters of Orthologous Groups (COG) classification, which focused mainly on pigment synthesis functions. Pathways analysis using the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) revealed critical pathways affecting pigment development. Metabolomics studies were carried out utilizing Ultra Performance Liquid Chromatography and mass spectrometry (UPLC-MS). About 480 metabolites were detected via metabolomics investigation, the majority of that were significantly involved in pigment synthesis. Cluster and pathway analyses revealed the importance of pathways such as plant secondary metabolite biosynthesis, biosynthesis of phenylpropanoids and plant hormone signal transduction in pigment synthesis. Current research advances our comprehension of the underlying mechanisms at the molecular level governing pigment synthesis in gardenia fruits, furnishing valuable insights for subsequent investigations.

Abstract:A high-performance liquid chromatography-diode array detection/electrospray ionization mass spectrometry (HPLC-DAD/ESI-MS) method was applied to the characterization of ten iridoid glycosides in Gardenia jasminoides Ellis, a traditional Chinese medicine. During the process of structural elucidation, two groups of isomers including two epimers were structurally characterized and differentiated according to their distinctive fragmentation patterns which were closely related to their isomeric differentiations. Subsequently, the major compounds were purified by multi-dimensional chromatography and semi-preparative HPLC and the structure identification was confirmed with NMR techniques. The major fragmentation pathways of iridoid glycosides in Gardenia jasminoides Ellis obtained through the MS data were schemed systematically, which provided the best sensitivity and specificity for characterization of the iridoid glycosides especially the isomers so far. Based on the fragmentation patterns of iridoid glycosides concluded, seven major iridoid glycosides were characterized in rat plasma after intravenous administration of Gardenia jasminoides Ellis. Abstract:Gardenia jasminoides Ellis is a perennial evergreen shrub of G. jasminoides of Rubiaceae. Geniposide and Crocin are important components in the fruit of G. jasminoides . In addition to being used as medicinal materials, they are also widely used in food, medicine, cosmetics, and other fields. They have high medicinal value, economic value, and ornamental value. However, at present, the utilization rate of G. jasminoides resources is low, mainly focused on germplasm cultivation, primary processing, and clinical pharmacology, and there are few studies on the quality of Gardenia fruit.

Abstract:Isolation and purification of triterpene acid from fruit of Gardenia Jasminodes Eills were studied,the optimum processing conditions are: the granularity of powder 0.4 mm, ratio of liquid to solid 8∶1, temperature 85 ℃, 3 times of extraction of 2.5h each, and concentration of alcohol 95 %.Extraction rate of ursolic lixiviate is 92.6 %. Structural identifications by DSC,IR, EI-MS ,~1H and ~ 13 C NMR,HPLC show that the triterpene acid is ursolic acid,the average content of ursolic acid is 0.444 5 %。

Abstract:Three new iridoid glycosides, hedycorysides A-C (1-3, resp.), were isolated from the whole plant of Hedyotis corymbosa (LINN.) LAM., along with four known compounds. Their structures were elucidated by extensive 1D- and 2D-NMR analysis, as well as by HR-ESI-MS experiments. The three new compounds are the first benzoylated geniposide derivatives from Hedyotis.

Abstract:A new flavonoid glycoside, 5-methoxy-7-beta-D-glucopyranosyl-( - )-afzelechin (1), together with seven known flavonoids, 7-O-beta-D-glucopyranosyl-( - )-epiafzelechin (2), sakuranin (3), genkwanin (4), 4 ',5-dihydroxy- 3 ',7-dimethoxyflavone (5), luteolin-3 ',7-dimethyl ether-5-beta-D-glucoside (6), and luteolin-7-methyl ether-5-beta-D-glucoside (7), were isolated from the stem barks of Daphne giraldii. Their structures were identified on the basis of spectroscopic analysis. Compounds 2, 3, and 5-8 were first reported from the titled plant.
