Abstract:A growing body of evidence indicates that the Pestalotiopsis genus represents a huge and largely untapped resource of natural products with chemical structures that have been optimized by evolution for biological and ecological relevance. So far, 196 secondary metabolites have been encountered in this genus. This review systematically surveys the taxonomy, biology and chemistry of the Pestalotiopsis genus. It also summarises the biosynthetic relationships and chemical synthesis of metabolites from this genus. There are 184 references.

Abstract:Six undescribed caryophyllene sesquiterpenoids named pestalotiopsins O-T, along with eight known analogues, were obtained from the fungus Pestalotiopsis chamaeropis. Their structures and absolute configurations were assigned by NMR spectroscopic analyses, HRESIMS, single-crystal X-ray diffraction, electronic circular dichroism (ECD) calculations, Mo2(OAc)4-induced ECD, and chemical derivatization. Pestalotiopsin P represents the first example of a caryophyllene sesquiterpenoid possessing an oxatricyclo [7.2.2.03.6]tridecane decorated with a rare bridgehead double bond, while pestalotiopsin Q has an oxatricyclic [6.3.1.01,4]dodecane skeleton with an un-usual ether bridge between C-1 and C-5. These undescribed caryophyllene sesquiterpenoids were screened for their cytotoxic and anti-inflammatory activities.

Abstract:Seven new β-caryophyllene derivatives, pestalotiphains A–G (1–7), along with six known analogues (8–13), were isolated from the plant-associated Pestalotiopsis hainanensis. Compound 1 represents the first example of a caryophyllene–adenine hybrid, and 2 contains a novel oxatricyclo[4.3.1.0] system. Their structures and absolute configurations were assigned by interpretation of a combination of spectroscopic data and electronic circular dichroism calculations. Compound 8 exhibited moderate inhibition of HL-60 and THP-1 cell lines (IC50, 6.2 and 2.0 μM, respectively). Abstract:Chemical investigation of the EtOAc extract of an insect-associated fungus Letendraea sp. 5XNZ4-2 cultured in Potato Dextrose Broth (1/2 PDB) medium lead to the isolation of three new polyketides, named letendronol D (1), phomopsiketones H-I (2-3). The structures of new compounds were elucidated by the analysis of HRESIMS and NMR spectroscopic data, and the absolute configurations were determined by modified Mosher’s method, ECD calculation and single-crystal X-ray diffraction. Cytotoxicity and antibacterial activities of 1 were assayed and regrettably 1 didn’t display any cytotoxicity and antibacterial activity. 3 was the first phomopsiketone derivative obtaining the lactone.

Abstract:Nine undescribed side chain containing azaphilones, pestaphilones A-I, were isolated from the Anoectochilus roxburghii endophytic fungus Pestalotiopsis oxyanthi. The structures of these isolates were identified by spectroscopic data, electronic circular dichroism (ECD) calculations and comparisons, quantum-chemical C-13 NMR calculations with DP4+ probability analysis, Rh-2(OCOCF3)(4)-induced ECD, acetonide formation, selective oxidation reaction and X-ray crystallographic data. Structurally, pestaphilones A-I were the first azaphilones characteristically formed via a methyl group at C-9 in the C-7 side chain. More importantly, a selective oxidation reaction was firstly set up to resolve the absolute configuration of flexible side chain containing azaphilones, and an acetonide formation based Rh-2(OCOCF3)(4)-induced ECD experiment was performed to identify the configurations of the oxygenated pyranoquinone core in the azaphilones. In bioassay, pestaphilones A-F displayed potential immunosuppressive activity in concanavalin A (Con A)-induced T lymphocyte proliferation, with IC50 values ranging from (9.36 +/- 1.14) mu M to (35.21 +/- 3.25) mu M.

Abstract:Three new secondary metabolites pestalothenins A–C ( 1 – 3 ), including two new humulane-derived sesquiterpeniods ( 1 and 2 ) and one new caryophyllene-derived sesquiterpeniod ( 3 ), together with five known compounds ( 4 – 8 ) were isolated from the crude extract of the plant endophytic fungus Pestalotiopsis theae (N635). Their structures were elucidated by the extensive analyses of HRESIMS and NMR spectroscopic data. The absolute configurations of 1 – 3 were determined by comparison of experimental and calculated electronic circular dichroism (ECD) spectra. The cytotoxic effects of these compounds were evaluated in vitro . Compound 6 showed moderate cytotoxicity against T24 and MCF7 cell lines. In addition, compounds 1 – 8 were also evaluated for antibacterial activity.

Abstract:Three new a-pyrones, scirpyrones AC (13), and four new pyranes, scirpyranes AD (47), were isolated from solid cultures of the plant pathogen Pestalotiopsis scirpina. Their structures were elucidated primarily by NMR experiments. The absolute configurations of C-7 and C-2' in 1, C-7 in 2, and C-6 and C-7 in 3 were deduced on the basis of circular dichroism (CD) data, whereas those of the secondary alcohols in 47 were determined by a modification of Mosher's method. Compound 1 is the first described 6-(5-oxotetrahydrofuran-2-yl)-2H-pyran-2-one. Compounds 46 showed significant cytotoxicity towards MCF-7 cells.

Abstract:Six new polyketide derivatives, including pestalpolyol I (1), pestapyrones A (2) and B (3), (R)-periplanetin D (6), pestaxanthone (7), norpestaphthalide A (8), and an isolation artifact pestapyrone C (4), were obtained from extracts of the endophytic fungus Pestalotiopsis clavispora isolated from the Mangrove plant Rhizophora harrisonii. In addition, seven known metabolites, including similanpyrone B (5), (R,S)-5,7-dihydroxy-3-(1-hydroxyethyl)phthalide (9), for which we propose the trivial name norpestaphthalide B, pestaphthalides A (10) and B (11), 2-epi-herbarumin II (12), and pestalotiollides A (13) and B (14) were isolated. The structures of the new compounds were unambiguously elucidated on the basis of one- and two-dimensional NMR spectroscopy, as well as by high -resolution mass spectrometry. All compounds were tested for their cytotoxicity against the mouse lymphoma cell line L5178Y. Compound 1 exhibited strong activity with an IC50 value of 4.10 mu M. Abstract:Two new 10-oxo-10H-phenaleno[1,2,3-de]chromene-2-carboxylic acids, xanalteric acids I (1) and II (2), and 11 known secondary metabolites were obtained from extracts of the endophytic fungus Alternaria sp., isolated from the mangrove plant Sonneratia alba collected in China. The metabolites were confirmed to be of fungal origin, and the structures of the new natural products were unambiguously elucidated on the basis of extensive one- and two-dimensional NMR spectroscopic studies and mass spectrometric analysis. The two new compounds 1 and 2 exhibited weak antibiotic activity against multidrug-resistant Staphylococcus aureus. Altenusin (3) displayed broad antimicrobial activity against several additional multidrug-resistant bacterial and fungal strains.

Abstract:Two new alkenyl phenol derivatives, namely pestalol F ( 1 ) and pestalol G ( 2 ), along with two known compounds, pestalachloride A ( 3 ) and pestalotiopsin J ( 4 ), were isolated from the culture of the fungus Pestalotiopsis clavata JSQ 12. The structures of these compounds were primarily elucidated by MS, NMR and specific rotation data analysises. These secondary metabolites of Pestalotiopsis clavata were reported for the first time. Compound 2 displayed interesting cytotoxic activity against MCF-7 cell line with the IC 50 value of 29.16 μM, whereas compound 3 exhibited moderate activity towards A549 cell line with the IC 50 value of 35.71 μM. The positive control 5-FU showed cytotoxic effects on MCF-7 and A549 cell lines with the respective IC 50 values of 26.70 and 26.07 μM. Compounds 1 and 2 displayed mild antibacterial activities against Staphylococcus aureus with MIC values of 128 and 64 μg/mL (MIC of positive control, penicillin, was 0.016 μg/mL), respectively.

Abstract:Pestalopyrones A–D are four unusual tricyclic pyrone derivatives with flexible chiral structures, isolated from the endophytic fungus Pestalotiopsis neglecta S3. The full elucidation of their structures was a challenging task, and remained unsolved in the original article. Herein, the relative configurations of pestalopyrones A and pestalopyrones B were unambiguously assigned by detailed analyses on spectroscopic data and GIAO 13C NMR calculation method with sorted training sets (STS). The planar structures of pestalopyrones C and pestalopyrones D were revised by reinterpretation of their reported spectroscopic data, and then their relative configurations were deduced by STS GIAO 13C NMR calculation and NOE analysis. The absolute configurations of all the mentioned compounds were determined by the comparison of their experimental and calculated ECD curves.

Abstract:Two new caryophyllene-type sesquiterpenes pestalotiopsins U and V ( 1 and 2 ) and three known compounds pestalotiopsin B ( 7 ), pestaloporinate B ( 8 ), and pestalotiopsin C ( 9 ) were isolated by the cultivation of the endophytic fungus Pestalotiopsis lespedezae on solid rice medium, while four additional new caryophyllene pestalotiopsins W–Z ( 3 – 6 ) were obtained when 3.5% NaI was added to the fungal culture medium. The structures of the new compounds were determined by HRESIMS and 1D/2D nuclear magnetic resonance data. Compounds 1 – 9 were tested for cytotoxicity against the mouse lymphoma cell line L5178Y, but only 6 displayed significant activity with an IC 50 value of 2.4 μM.

Abstract:Marine-derived fungi are a treasure house for the discovery of structurally novel secondary metabolites with potential pharmaceutical value. In this study, a pair of new nor-bisabolane derivative enantiomers (±)−1 and two new phthalides (4 and 5), as well as four known metabolites, were isolated from the culture filtrate of the marine algal-derived endophytic fungus Penicillium chrysogenum LD-201810. Their structures were established by detailed interpretation of spectroscopic data (1D/2D NMR and ESI-MS). The optical resolution of compound (±)−1 by chiral HPLC successfully afforded individual enantiomers (+)−1 and (−)−1, and their absolute configurations were determined by TDDFT-ECD calculations. Compound (±)−1 represents the first example of bisabolane analogs with a methylsulfinyl substituent group, which is rare in natural products. All of the isolated compounds 1–7 were evaluated for their cytotoxic activity against A549, BT-549, HeLa, HepG2, MCF-7, and THP-1 cell lines, as well as for antifungal activity against four plant pathogenetic fungi ( Alternaria solani , Botrytis cinerea , Fusarium oxysporum , and Valsa mali ). Compound 2, a bisabolane-type sesquiterpenoid, was shown to possess excellent activity for control of B. cinerea with half-maximal inhibitory concentration (IC 50 ) of 13.6 μg/mL, whereas the remaining investigated compounds showed either weak or no cytotoxic/antifungal activity in this study.
