Abstract:Pestaloficiols F-L (1-7), new isoprenylated chromone derivatives including one heterodimer (7), have been isolated from a scale-up fermentation extract of the plant endophytic fungus Pestalotiopsis fici. The structures of these compounds were elucidated primarily by NMR and MS methods. The absolute configurations of 1 and 4 were assigned using the modified Mosher method. Compounds 1-3, 5, and 6 displayed inhibitory effects on HIV-1 replication in C8166 cells, whereas 4-7 showed cytotoxic activity against the human tumor cell lines HeLa and MCF7.

Abstract:Chemical investigation of the plant endophytic fungus Pestalotiopsis sp. afforded two new butenolides, pestalolides B and C (1 and 2), two new diphenyl ethers, pestalotethers E and F (3 and 4), together with seven known compounds (5-11). Their structures were established by extensive spectroscopic analyses and the absolute configuration of 1 was further confirmed by ECD calculation. Structurally, compound 1 represents the first example of a new type of butenolide derivatives, bearing an unprecedented carbon scaffold having 13 carbon atoms. Compounds 1-4 were evaluated for the cytotoxic activities, and compound 1 showed remarkable inhibitory potency against human tumor HL60, U87MG, MDA-MB-231, and HEP-3B cell lines, with IC50, values of 1.42, 5.90, 5.90, and 4.29 mu M, respectively.

Abstract:Pestalotheols A-D (1-4), four new metabolites, have been isolated from cultures of an isolate of the plant endophytic fungus Pestalotiopsis theae. The structures of these compounds were determined by NMR spectroscopy, and 1 is further confirmed by X-ray crystallography. The absolute configurations of compounds 1 and 3 were assigned by application of the modified Mosher method. Pestalotheol C (3) displayed an inhibitory effect on HIV-1(LAI) replication in C8166 cells with an EC50 value of 16.1 mu M.

Abstract:Eight new caryophyllene sesquiterpenoids named pestaloporinates A-G (1-7) and 14-acetylhumulane (8) have been isolated from the solid cultures of an endophytic fungus Pestalotiopsis sp., which was obtained from the fresh stem bark of Melia azedarach Linn. Their structures as well as absolute configurations were determined by spectroscopic data, ECD experimentation, and single-crystal X-ray diffraction. Among all the isolates, compound 2 displayed potent inhibitory activity with IC50 value of 19.0 μM during the evaluation of nitric oxide (NO) inhibition in lipopolysaccharide (LPS)-induced RAW 264.7 macrophage cells.

Abstract:Three new secondary metabolites pestalothenins A–C ( 1 – 3 ), including two new humulane-derived sesquiterpeniods ( 1 and 2 ) and one new caryophyllene-derived sesquiterpeniod ( 3 ), together with five known compounds ( 4 – 8 ) were isolated from the crude extract of the plant endophytic fungus Pestalotiopsis theae (N635). Their structures were elucidated by the extensive analyses of HRESIMS and NMR spectroscopic data. The absolute configurations of 1 – 3 were determined by comparison of experimental and calculated electronic circular dichroism (ECD) spectra. The cytotoxic effects of these compounds were evaluated in vitro . Compound 6 showed moderate cytotoxicity against T24 and MCF7 cell lines. In addition, compounds 1 – 8 were also evaluated for antibacterial activity.

Abstract:A new isocoumarin derivative pestalotiopisorin B (1), along with six known compounds, (R)-(-)- mellein methyl ether (2), pestalotiopyrone G (3), (R)-mevalonolactone (4), pestalotiollides A-B (5-6) and pestalotiopsol A(7) were isolated from Pestalotiopsis sp., an endophytic fungus obtained from Chinese mangrove plant Rhizophora stylosa. Their structures were elucidated unambiguously by the comprehensive analysis of extensive spectroscopic data. Compound 1 exhibited modest antibacterial activity against Escherichia coli and Pseudomonas aeruginosa with 12.5 mu g/ml, 50 mu g/ml, respectively. Compound 4 showed moderate calcineurin inhibitory activity towards p-nitrophenyl phosphate (IC50 =134.29 +/- 5.377 mu M).

Abstract:Chlorotheolides A (1) and B (2), two new spiroketals possessing the unique [4,7]methanochromene and dispirotrione skeletons, respectively, and their putative biosynthetic precursors, 1-undecen-2,3-dicarboxylic acid (3) and maldoxin (4), were isolated from the solid substrate fermentation of the plant endophytic fungus Pestalotiopsis theae (N635). Their structures were elucidated based on NMR spectroscopic data and electronic circular dichroism calculations. Biogenetically, compounds 1 and 2 could be generated from the co-isolated 3 and 4 via Diels Alder reactions. Chlorotheolide (2) showed an antiproliferative effect against the human tumor cell line HeLa and induced an autophagic process in the cells.

Abstract:A new pair of enantiomeric isoprenylated chromone derivatives, (+/-)-pestaloficiol X [(+/-)-1], along with a known compound pestaloficiol J (2), were isolated from the plant endophytic fungus Pestalotiopsis sp. The racemic mixture 1 was separated through chiral HPLC. The structures of new compounds (+/-)-1 were elucidated on the basis of extensive spectroscopic data and their absolute configurations were further configured through computational analysis of their electronic circular dichroism (ECD) spectra. Compound (+)-1 showed significant inhibitory potency against HL-60 and HEP-3B cell lines, with IC50 values of 1.35 +/- 0.15 and 3.70 +/- 0.33 mu M, respectively, while compound (-)-1 showed significant inhibitory potency against HL-60 and HEP-3B cell lines, with IC50 values of 2.39 +/- 0.26 and 2.99 +/- 0.35 mu M, respectively.

Abstract:A new diphenyl ether derivative, named pestalotilide A (1), along with four known compounds (2-5) have been isolated from the organic extracts of an endophytic fungus Pestalotiposis guepinii. Their structures were elucidated on the basis of HR-ESI-MS, ID and 2D NMR spectroscopic data analyses. These isolates were evaluated for cytotoxicity against three cancer cell lines: Hela, HCT116 and A549; pestalotilide A exhibited weak in vitro anti-proliferative activities against the tested cell lines, with IC50 values of 23.8, 18.4 and 35.2 mu M, respectively.

Abstract:Four new diphenyl ether derivatives, sinopestalotiollides A–D (1–4), one new natural α-pyrone product (11), as well as twelve known compounds (5–1 7), were obtained from the ethyl acetate extract of the endophytic fungus Pestalotiopsis palmarum isolated from the leaves of medicinal plant Sinomenium acutum (Thunb.) Rehd et Wils. The structures were elucidated by HR-ESI-MS and NMR spectrometry data. Bioassay experiments revealed that compounds 1–4 and 11 exhibited strong to weak cytotoxicities against three human tumor cell lines Hela, HCT116 and A549.

Abstract:Four undescribed pymne derivatives, pestalopyrones A-D, containing unusual tricyclic 5/6/6 polycyclic skeletons, were isolated from the ethyl acetate extract of the endophytic fungus Pestalotiopsis neglecta 53 derived from the fresh stems of Rubia podantha Diels (Rubiaceae). Their planar structures were elucidated mainly by NMR and HRESIMS. Pestalopyrones A-D contained six contiguous chiral carbons, and the relative configurations of C-4, C-5, and C-8 in tricyclic 5/6/6 polycyclic skeletons were determined by ROESY spectra. For pestalopyrone B, the absolute configuration of C-16 was determined by the Mosher's method. All isolated compounds were tested for their cytotoxicity against cancer cell lines, antibacterial activity, and inhibitory effect on the lipopolysaccharide (LPS)-induced nitric oxide (NO) production, and the results showed that pestalopyrone A inhibited LPS-induced NO production with an IC50 value of 35.8 mu M.

Abstract:Four new compounds, including two isocoumarins, pestaloisocoumarins A and B (1, 2), one sesquiterpenoid degradation, isopolisin B (4), and one furan derivative, pestalotiol A (5), together with one known isocoumarin, gamahorin (3), and three chlorinated benzophenone derivatives, pestalachloride B (6), pestalachloride E (7) and a mixture of pestalalactone atropisomers (8a/8b), were isolated from a culture of the fungus Pestalotiopsis heterocornis associated with sponge Phakellia fusca. These new chemical structures were established using NMR and MS spectroscopic data, as well as single-crystal X-ray crystallographic analysis and CD Cotton effects. All of the isolated compounds were evaluated for their antimicrobial and cytotoxic activities. Isocoumarins 1–3, showed antibacterial activities against Gram-positive bacteria Staphylococcus aureus and Bacillus subtilis with MIC values ranging from 25 to 100 μg/mL and weak antifungal activities. Chlorinated benzophenone derivatives 6–8 exhibited antibacterial activities against S. aureus and B. subtilis with MIC values ranging from 3.0 to 50 μg/mL and cytotoxicities against four human cancer cell lines with IC50 values of 6.8–87.8 μM.

Abstract:Six new polyketide derivatives, including pestalpolyol I (1), pestapyrones A (2) and B (3), (R)-periplanetin D (6), pestaxanthone (7), norpestaphthalide A (8), and an isolation artifact pestapyrone C (4), were obtained from extracts of the endophytic fungus Pestalotiopsis clavispora isolated from the Mangrove plant Rhizophora harrisonii. In addition, seven known metabolites, including similanpyrone B (5), (R,S)-5,7-dihydroxy-3-(1-hydroxyethyl)phthalide (9), for which we propose the trivial name norpestaphthalide B, pestaphthalides A (10) and B (11), 2-epi-herbarumin II (12), and pestalotiollides A (13) and B (14) were isolated. The structures of the new compounds were unambiguously elucidated on the basis of one- and two-dimensional NMR spectroscopy, as well as by high -resolution mass spectrometry. All compounds were tested for their cytotoxicity against the mouse lymphoma cell line L5178Y. Compound 1 exhibited strong activity with an IC50 value of 4.10 mu M. Abstract:Five new isocoumarin derivatives, pestalactone A-C (1-3) and pestapyrone D-E (4-5), together with two known compounds (6-7) were isolated from the solid cultures of the endophytic fungus Pestalotiopsis sp. obtained from Photinia frasery. Their structures were mainly determined by extensive spectroscopic analysis, Mo2(OCOCH3)4-induced electronic circular dichroism (ECD), and ECD calculation. Compounds 1 and 2 were rare isocoumarin derivatives and derived from distinctive polyketide pathways. Compound 3 exhibited potent antifungal activity against Candida glabrata (ATCC 90030) with an MIC50 value of 3.49±0.21μg/mL.

Abstract:Natural products from fungi have remained a rich resource for drug discovery. Here we report the isolation of three new spirobisnaphthalenes, namely sacrosomycin A-C (1-3), and three known analogues (4-6), from the ethyl acetate extract of a nonsporulating endolichenic fungus derived from Peltigera elisabethae var. mauritzii. The structures of these compounds were elucidated by IR, UV, MS, and NMR. Biological functions of these compounds were evaluated using cultured human cancer cell lines. Short-term cell growth and long-term cell survival assays show that compound 5 demonstrated the strongest cancer cell growth inhibition effect. We reveal that compound 5 induced both cell cycle arrest at the G2/M phase and cell death. Using western blotting, luciferase reporter assay and quantitative PCR (qPCR), we show that compound 5 induced up-regulation of the P53-P21 pathway, supporting the cell cycle arrest and growth inhibition effect of this compound. In contrast, these compounds did not induce cell death in a normal cell line. These results demonstrate a potential anticancer effect of this rare family of spirobisnaphthalene compounds isolated from endolichenic fungi.
