Abstract:A new, practical and efficient method for the synthesis of anhydrovinblastine AVBL (1f) by oxidative coupling of vindoline and catharanthine in the presence of ceric ammonium nitrate (CAN) was developed. Under the optimized reaction conditions, we synthesized a new series of amide anhydrovinblastine analogs substituted at the vindoline moiety of C-23 site and, evaluated for their proliferation inhibition against HeLa cell. The aryl-substituted derivatives showed loss of potency, while alkyl-substituted derivatives retained some of its cytotoxic potency. The iso-butylamide compound 10b and 2-furancorboxamide compound 18b displayed a similar cytotoxic potency compared to the positive control AVBL.

Abstract:A series of 3-demethoxycarbonyl-3-acylamide methyl vinorelbine derivatives (compounds 7a–7z) were designed, synthesized, and evaluated for their inhibition activities against human non-small cell lung cancer cell line (A549). Most of the amide derivatives exhibited potent cytotoxicity, with the size of the introduced substituents being the foremost factor in determining the resultant cytotoxic activity. Test results in vivo against nude mice bearing A549 xenografts indicated that 7y showed comparable activities compared to the parent NVB.

Abstract:A series of 4 beta N-indole-substituted podophyllotoxin derivatives were synthesized. Nine target compounds were evaluated against human cancer cell lines (HeLa, K562, and K562/A02) using MTT assay including three imine derivatives 8, 9, and 10in vitro. The result showed that the three compounds had higher antitumor activity than their reduced forms. Among them, compounds 8, 9, 11, and 16 were superior to the positive control VP-16.

Abstract:Seventeen new flavone derivatives substituted at the 4′-OH position were designed, synthesized and evaluated for their anticancer and antibacterial activities. Among them, compounds 3, 4, 6f, 6e, 6b, 6c and 6k demonstrated the most potent antiproliferative activities against a human erythroleukemia cell line (HEL) and a prostate cancer cell line (PC3). The results also showed that the IC50 value of compounds 3, 4, 6f, 6e, 6b, 6c and 6k were close to that of the anticancer drug cisplatin (DDP) and lower than that of apigenin. All of the derivatives did not present antibacterial activities. The structure–activity relationships evaluation showed that the configuration of methyl amino acid might affect their biological activities.

Abstract:A general and transition-metal-free method for the construction of the carbazol-4-amine motif via a vinylogous Michael addition/cyclization/isomerization/elimination reaction of 3-nitroindoles with alkylidene malononitriles has been developed. This novel methodology allows the facile synthesis of a series of di- and trisubstituted carbazol-4-amine derivatives in moderate to good yields. A gram-scale experiment was successfully performed, highlighting the practicability of this method. Moreover, this strategy is also applicable to 3-nitrobenzothiophene, affording the corresponding dibenzo[ b, d]thiophen-1-amine derivatives in moderate yields.

Abstract:An efficient synthesis method for fusing triazole ring onto the naphthalimide core was described. The anti-cancer activities of the generated triazolonaphthalimide derivatives were evaluated with five cancer cell lines. The compounds generally displayed higher potency than amonafide. 4d,4e carrying two amino side chains showed the strongest cytotoxicities. N-oxide 5, a prodrug of 4a, was designed and synthesized. The agent was expected to be activated under the hypoxic condition in tumor tissue. Compared with 4a, 5 manifested much lower cytotoxicity both in cancer cell lines and human normal cells in the in vitro assays. However, N-oxide 5 performed potent anti-cancer activity in vivo using S-180 sarcoma bearing mice. All the results suggested that 5 was a promising anti-cancer agent.

Abstract:A new series of vinorelbine analogues are designed and synthesized to explore the vindoline C-16 substituent effects on the biomimetic coupling with catharanthine, and the structure–activity relationships of these vinorelbine analogues as cytotoxic agents are also studied. The results show that introduction of severe steric hindrance and/or electron-withdrawing group at C-16 site are not propitious to improving the yields of the coupling reaction, and the SAR information collected so far suggests that small alkyl groups substituted at C-16 of vindoline are conductive to maintaining the cytotoxicity.

Abstract:Inspired by the diverse bioactivities of α-amino phosphine oxides, an efficient strategy for the synthesis of less researched α-(hydroxyamino)diarylphosphine oxides has been developed and their antitumor activities are explored. Under water as a solvent and catalyst-free conditions, the addition of nitrones and diphenylphosphine oxide occurs smoothly to afford α-(hydroxyamino) diarylphosphine oxides in high yields. This reaction features a wide substrate scope, facile starting materials, atom economy, and easy purification. Moreover, the biological evaluation revealed that two synthesized derivatives 5e and 5f could serve as interesting anti-cancer agents for further development.

Abstract:An efficient and novel strategy for the enantioselective syntheses of various iboga alkaloids has been developed. The salient features include a gold-catalyzed oxidation of a terminal alkyne followed by cyclization, a Stevens rearrangement and a tandem sequence that combines the gold-catalyzed oxidation, cyclization and [1,2]-shift. The catharanthine analogs provided by our approach were further converted to the vinca alkaloid vinblastine and its analogs, which confirmed the remarkable sensitivity of the cytotoxicity to the C20' substituent of vinblastine.

Abstract:A series of new combretastatin A4 derivatives (6a-6c, 9a-12c) containing nitrogen atoms and nitrogen heterocycles were synthesized by condensation reaction, reduction reaction, hydrolysis reaction and Crutius rearrangement, and the structures were confirmed by 1H NMR, 13C NMR and Mass spectrum. The antitumor cytotoxicity of a series of newly synthesized compounds were evaluated in vitro toward the cell line of hepatoma cell with erianin as positive control. Abstract:A series of new 1,3,4-oxadiazin-5(6H)-one derivatives (6a-n) of dehydroabietic acid were designed and synthesized as potential antimicrobial and antitumor agents. Their structures were characterized by IR, H-1 NMR, C-13 NMR, MS, and elemental analyses. All the title compounds were evaluated for their antimicrobial activity against four bacterial and three fungal strains using the serial dilution method. Among them, compound 6e showed the highest antibacterial activity against Bacillus subtilis with a minimum inhibitory concentration (MIC) value of 1.9 g/mL. In addition, the in vitro cytotoxic activities of the title compounds were also assayed against three human carcinoma cell lines (MCF-7, SMMC-7721, and HeLa) through the MTT colorimetric method. As a result, compounds 6b, 6g, 6k, and 6m exhibited significant inhibition against at least one cell line with IC50 values below 10 M. Compound 6m was especially found to be the most potent derivative with IC50 values of 2.26 +/- 0.23, 0.97 +/- 0.11, and 1.89 +/- 0.31M against MCF-7, SMMC-7721, and HeLa cells, respectively, comparable to positive control etoposide.

Abstract:A series of novel nitrogen-containing macrocyclic bisbibenzyl derivatives was designed, synthesized, and evaluated for antiproliferative activity against three anthropic cancer cell lines. Among these novel molecules, the tri-O-alkylated compound 18a displayed the most potent anticancer activity against the A549, MCF-7, and k562 cancer cell lines, with IC50 values of 0.51, 0.23, and 0.19 mu M, respectively, which were obviously superior to those of the parent compound riccardin D, and were 3-10-fold better than those of the clinical used drug ADR. The bis-Mannich derivative lib also exhibited significantly enhanced antiproliferative potency, with submicromolar IC50 values. Structure-activity relationship analyses of these newly synthesized compounds were also performed. Mechanistic studies indicated that these compounds could target the lysosome to induce lysosomal membrane permeabilization, and could also induce cell death that displayed features characteristic of both apoptosis and necrosis. Synthesis and biological evaluation of new mono naphthalimide platinum(IV) derivatives as antitumor agents with dual DNA damage mechanism

Abstract:Naphthalimide has emerged as an interesting DNA intercalator and possessed attracting antitumor properties. In this context, naphthalimide group was linked to platinum(IV) core to construct a series of new mono naphthalimide platinum(IV) derivatives. The title compounds exert effective antitumor activities to the tested tumor cells lines in vitro, especially the one with propionyl chain displays comparable or even better bioactivities than platinum(II) reference drugs cisplatin and oxaliplatin. Moreover, the mono naphthalimide platinum(IV) derivative displays comparable tumor growth inhibitory competence against CT26 xenograft tumors in BALB/c mice in vivo without severe toxic effects in contrast to oxaliplatin. A dual DNA damage mechanism was proven for the title complex. Both naphthalimide ligand and the liberated platinum(II) moiety could generate DNA lesions to tumor cells synergistically and active the apoptotic pathway by up-regulating the expression of caspase 9 and caspase 3. Meanwhile, the conversion of platinum(II) drug into tetravalent form by incorporating naphthalimide moiety increases the uptake of platinum in whole cells and DNA remarkably. All these facts might be the factors for the title platinum(IV) complexes to overcome platinum(II) drug resistance. Additionally, the mono naphthalimide platinum(IV) complex could interact with human serum albumin by hydrogen bond and van der Waals force which would further influence their storage, transport and bioactivities.

Abstract:Tertiary nitroalkanes, as well as its reduced products, α-tertiary amines, play an essential role in drug discovery either as key synthetic precursors or final incorporation in targeted molecules. Existing methods to prepare tertiary nitro compounds generally rely on polar-bond disconnections, in which strong bases or highly active electrophiles are needed. Here, we report the development of an anomeric nitroamide-based reagent that enables exquisitely selective MHAT-based Co-catalyzed alkene hydronitration for the preparation of valuable tertiary nitro compounds. This mild, scalable reaction shows broad functional group tolerance, is applied to a variety of structures. Late-stage nitration of complex contexts (9 examples) derived from drugs and natural products is also pursued. Its high prowess is further highlighted in simplifying the synthesis of a rare naturally occurring nitro sugar. Simple access to isotopically labeled 15N-containing nitro compounds is also disclosed. The anomeric nitroamide reagent was deemed safe by energetic measurements and its unique reactivity rationalized based on X-ray crystallographic analysis.

Abstract:The 2-Aminonicotinonitrile derivative was reacted with different bi-functional reagents such as formamide, thiourea, acetic anhydride, and phthalic anhydride under optimized conditions to give pyrimidine, thiourea, acetamide, and isoindoline derivatives, respectively, When it was treated with active methylene reagents as malononitrile, phenacyl bromide, and ethyl bromoacetate under varied experimental modulation, it afforded 1, 8-naphthyridine, ethyl, and methylamino nicotinonitrile derivatives, respectively. Also, it was reacted with p-toluene sulfonyl chloride, chloroacetyl chloride, and benzoyl chloride to give sulfonamide, 2-chloro-N-acetamide, and benzamide derivatives, respectively. Likewise, it was reacted with diethyl malonate, ethyl cyanoacetate, and cyano acetic acid to give ethylpropanoate, naphthyridine, and cyano acetamide derivatives, respectively. However, treatment of ethylpropanoate and cyano acetamide derivatives with hydrazine hydrate gave pyrazole and 5-amino-pyrazole nicotinonitrile derivatives, respectively. In addition, it was reacted with p-anisaldehyde, phenyl isocyanate, and triethyl orthoformate to give benzylamino nicotinonitrile, phenyl urea, and N-formamide derivatives, respectively. Furthermore, it was reacted with nitrous acid then coupled with aniline; it was also reacted with isatine and 1,3- dibromo propane to give oxoindoline derivative, and the dimer. Elemental analyses, together with spectroscopic data including IR, 1H-NMR in addition to 13C-NMR and mass spectra submit proofs for the chemical structures for all compounds.Graphical

Abstract:The oxime or hydrazone formation is a classic condensation reaction between aldehydes/ketones and hydroxyl amine or hydrazine.It is a simple,yet fundamental coupling reaction that has been widely applied in the ligations and conjugations of biomolecules and material synthesis.However,the reactions are usually sluggish and normally require acidic conditions with large excess of substrates to facilitate conversion,which limit their wide applications.Recent studies have shown that aniline as a nucleophilic catalyst can significantly accelerate the oxime/hydrazone formation reaction,preliminarily solving the reaction rate issue under mild and bio-compatible conditions.Therefore,it has been the most common coupling method in the modification of biological macromolecules.In this review,the design and development of aromatic amine catalyst in recent years as well as the catalytic mechanism and structure activity relationship are summarized.The application of highly active aniline catalysis in bioconjugations and material synthesis was also included together with a prospect for future development.

Abstract:Novel acylhydrazone compounds were synthesized and characterized by spectroscopic techniques. The cytotoxic effects were examined on cancer cell lines, and some compounds show better activity than the standard drug. Molecular docking studies investigated the interaction between the inhibitors and the protein. A series of new acylhydrazone derivatives were synthesized from the condensation of different substituted salicylaldehydes with 4‐nitrobenzohydrazide at a molar ratio of 1 : 1 in a one‐step reaction. These compounds have been characterized by spectroscopic techniques such as FT‐IR, NMR, ESI‐MS and CHN analysis. All compounds were examined for anticancer activity in three cancer cell lines viz. melanoma (A375), colon (HT‐29) and lung (A549). Compound 3 exhibits good cytotoxicity activity with an IC50 value of 0.38±0.08 μM against A375 and compound 2 exhibits better cytotoxicity 1.34±0.02 μM and 1.48±0.04 μM against HT‐29 and A549, respectively. Similarly, other compounds have also shown significant cytotoxicity activity against three cell lines. The cytocompatibility assay on the normal mouse fibroblast cell line (L929) and the hemolysis assay on human red blood cells were used to confirm the nontoxic activity. DAPI(4′,6‐diamidino‐2‐phenyliindole) staining experiments demonstrated that compounds induced apoptosis in the A375 cell line. Further, in silico molecular docking was performed against three various targets. In silico studies demonstrated that compounds 1(ΔGb=−9.3 kcal/mol), 2 (ΔGb=−8.8 kcal/mol), and compound 3(ΔGb=−9.0 kcal/mol) have a good binding energy score with an active pocket of protein inhibitor kinases. The outcomes of the current study showed that these compounds have anticancer action and might be used to create more potent drugs to treat melanoma, colon, and lung cancers.
