Abstract:(S)-6-Nitroindoline-2-carboxylic acid, a substructure occurring in numerous biologically active natural products, was synthesized with moderate yield (53%) and high enantiomeric excess (> 99.5%) starting from the nitration of L-phenylalanine, which is a commercially available chiral pool compound, followed by successively bromination and intramolecular cyclization. The route was carried out in gram quantities and it is suitable for industrial application due to its convenient reaction conditions and low cost.

Abstract:A new method for the direct and stereoselective synthesis of 3-substituted isoindolinones via Rh(iii)-catalyzed chiral N-sulfinyl amide directed asymmetric [4 + 1] annulation of benzamides with acrylic esters has been developed. The reaction proceeded through an oxidative C-H olefination and a subsequent cyclization by intramolecular aza-Michael addition, producing a series of diastereoisomeric chiral isoindolinones (20 examples) in generally good yields with a dr value up to 5.5 : 1. The absolute configurations of the newly formed C-stereocenters in the major and minor diastereomers of the catalysis product have been determined by X-ray crystal diffraction analysis to be S and R, respectively. The separation of the major diastereoisomers from the catalysis products and subsequent removal of the N-sulfinyl chiral auxiliary afforded enantiomerically pure (S)-isoindolinones. The application of the obtained (S)-isoindolinones in the synthesis of several biologically active isoindolinones such as (S)-PD172938, (S)-pazinaclone and (S)-pagoclone is presented.

Abstract:Objective:: 3,3-Disubstituted indol-2-one derivatives have wider applications in pharmaceuticals and they are key intermediates for the synthesis of many kinds of drug candidates. The development of an efficient and practical method to prepare this class of compound is highly desirable from both environmental and economical points of views. Methods:: In order to establish an effective synthetic method for preparing 3,3-disubstituted indol- 2-one derivatives, the bis-condensation reaction of isatin and 1H-indene-1,3(2H)-dione was selected as a model reaction. A variety of natural deep eutectic solvents (NADESs) were prepared and used for this reaction. The generality and limitation of the established method were also investigated. Results:: It was found that model reaction can be carried out in natural deep eutectic solvent (NADES) based on choline chloride (ChCl) at 80°C under microwave irradiation. This protocol with a broad substrate applicability afforded various 2,2'-(2-oxoindoline-3,3-diyl)bis(1H-indene- 1,3(2H)-dione) derivatives in high yields. Conclusion:: A simple and efficient procedure has been developed for synthesis of 2,2'-(2-oxoindoline- 3,3-diyl)bis(1H-indene-1,3(2H)-dione), spiro[indoline-3,7'-pyrano[5,6-c:5,6-c']dichromene]- 2,6',8'-trione, and spiro[indoline-3,9'-xanthene]trione via bis-condensation between isatin with 1,3- indandione, 4-hydroxycoumarin or 1,3-cyclohexanedione in natural deep eutectic solvent (NADES) based on choline chloride (ChCl) and glycerol (Gl) under microwave irradiation. The salient features of this protocol are avoidance of any additive/catalyst and toxic organic solvent, clean reaction profiles, non-chromatographic purification procedure, and high to excellent yield. Furthermore, the use of NADES as green reaction medium reduces burden on environment and makes the present method environmentally sustainable.

Abstract:Chiral N-alkylated indoles are privileged structural motifs found in many natural products and pharmaceuticals and a search for effective methods with which to prepare chiral N-alkylated indoles is ongoing. Efficient syntheses of enantioenriched N-alkylated indoles has been developed but not with standing these advances, the development of a facile method to prepare chiral N-alkylated indoles from simple and available starting materials under mild conditions is still needed. In this paper, we report a nitrogen replacement process driven by dearomatization that directly incorporates natural a-amino acids into 2-alkynylanilines leading to the formation of a variety of multi-functional chiral N-alkylated indoles.

Abstract:The indole unit is one of the most abundant and relevant heterocycles in natural products and pharmaceuticals. The synthesis of indoles has been a topic of research for over 100 years, and a variety of well-established classical methods are now available. The venerable Fischer indole synthesis, discovered in 1883, remains one of the most powerful and versatile routes to the indole heterocycle, although this method suffers from several drawbacks. A variety of synthetic modifications have been introduced to increase the practicality and lessen the environmental impact of the Fischer cyclization such as using mild acids, reusable catalysts, and ionic liquids. However, it is important to note that hydrazines are carcinogenic and that certain hydrazines are difficult to prepare, are unstable, or display only moderate functional-group tolerance under acidic conditions (Scheme 1a).

Abstract:A chiral SPINOL derived phosphoric acid-catalyzed asymmetric N-alkylation reaction of indoles with cyclic α-diaryl-substituted N-acyl imines, which are generated in situ from 3-aryl 3-hydroxyisoindolinones, has been demonstrated. The transformation proceeds smoothly with a broad range of indoles and isoindolinone alcohols. A variety of indole derived N-alkylated tetrasubstituted chiral aminals were afforded in moderate to good yields (50-79%) with excellent enantioselectivities (up to 98% ee).

Abstract:Polycyclic indolines and indolenines were synthesized via base‐catalyzed intramolecular dearomatizing 3‐alkenylation reactions of alkynyl indoles 1 at room temperature. The base enhanced the nucleophilicity of the carbon at the 3‐position of the indole moiety, facilitating an exclusive 5‐exo‐dig cyclization reaction with the alkyne to form spiroindolenines 2. The imine functionality of 2 could undergo in situ nucleophilic addition to form spiroindolines 3 when R was a carbamoyl group or reduction to form spiroindolines 4 when R was H.This article is protected by copyright.
