Abstract:A series of 4-(1 H-indol-3-yl-methylene)-1 H-pyrazol-5(4 H)-one derivatives have been synthesised. The Z structure of 4-[(1-methyl-1 H-indol-3-yl)methylene]-3-phenyl-1 -p-tolyl-1 H-pyrazol-5-one was determined by X-ray crystallography. The antitumour activity was evaluated against five cancer cells by MTT assay. [(1 H-Indol-3-yl)methylene]-1-(2,4-dini-trophenyl)-3-methyl-1 H-pyrazole-5-one and 4-{4-[(1-benzyl-1 H-indol-3-yl)methylene]-3-methyl-5-oxo-4,5-dihydro-1 H-pyrazol-1-yl}-benzoic acid have similar anticancer activity with 5-UF on the test cancer cells (exception of A375). Almost all the target compounds displayed antitumour activity against A549 and PC-9, and those with benzyl at 1-position of indole had higher activity against PC-9 (IC50 value lower than 30 μM). Those with benzyl at the indole and carboxyl at the phenyl part of of pyrazole were more active against PC-9 and A549 cells, providing a good indication for subsequent optimisation as lung cancer inhibitory agents.

Abstract:A novel class of acetylhydrazone derivatives (5a-x) containing 2-(phenylthiomethyl)-1H-benzo-[d]-imidazole moieties are synthesizer, and their antitumor activities against A549, HCT116, HepG2, PC-9, and A375 were determined by the MTT assay. Among them are N-(2,4-dihydroxybenzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5a) and N-(5-bromo-2-hydroxy-benzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5d) which displayed excellent cancer inhibitory activity against the tested cancer cells (IC(50) 4-17 μM), compared with 5-FU and SU11248. The others have moderate to weak inhibitory activity against the tested cancer cell lines.

Abstract:2-Fluoro-6-morpholinobenzonitrile(1) was prepared by reaction of 2,6-difluorobenzonitrile with morpholine.4-Morpholino-1H-indazol-3-amine(2) was synthesized by cyclization of 1 with hydra-zine.2 was coupled with various carboxylic acids to afford eight novel indazole derivatives(4a~4h). The structures were characterized by 1H NMR, IR and HR-ESI-MS.The antitumor activities were in-vestigated.The results showed that 4-(4-methylpiperazin-1-yl)-N-(4-morpholino-1H-indazol-3-yl)-4-oxobutanamide(4a) exhibited potent antitumor activities against K-562, SMMC7721 and T-47D tumor cell lines with IC50 of 0.056 μmol· L-1 , 0.062 μmol· L-1 and 0.086 μmol· L-1 , respectively.

Abstract:To discover novel compounds with antitumor activities, a series of indolylmaleimide derivatives were designed, synthesized and evaluated for their antitumor activities in vitro. Starting with indole and 2-chloroacetamide, followed by substitution and condensation, monoindolyl derivatives 5a similar to 5t were synthesized. Starting with 5-nitroindole and different N-(3-chloropropyl)tertiary amines, followed by substitution, reduction and condensation, diindolyl derivatives 9a similar to 9f were synthesized. Twenty six novel compounds were synthesized, and their structures were confirmed by ESI-MS, elemental analysis and H-1 NMR. The antitumor activities of the target compounds were evaluated with HL60, ECA-109, A549, SMMC-7721 and PC-3 in vitro. The in vitro antitumor activity showed that some compounds displayed inhibition to a certain degree against tested cells.

Abstract:A series of (E)-3-(aryl(arylamino)methylene)indolin-2-one derivatives were synthesised using an efficient synthetic approach. The method involved reaction of 3-bromo-3-(bromo(aryl)methyl)indolin-2-one with substituted anilines through nucleophilic substitution and a simultaneous elimination using NaHCO3 in DMF. The anticancer activity of the products against four cell lines, HCT-116, A549, SKOV3 and MDA-MB-231, was also evaluated, and several compounds showed moderate inhibitory activity.

Abstract:In order to find new indolin-2-one derivatives as antitumor agents, a series of 3-pyrrole substituted 1-(5-formyl-2-furanylmethyl) indolin-2-one derivatives were designed and synthesized. 5-Formyl-2 ,4-dimethyl-lH-pyrrole-3-carboxylic acid ethyl ester was condensed with 5-substituted indolin-2-one 2a-2d to afford 3-[(pyrrol-2-yl) -methylidenyl] indolin-2-ones 3a-3d. Through N-alkylation, 1-(5-formyl-furfuryl) -indolin-2-one 4a-4d were prepared. Compounds 4a-4d were then condensed with indolin-2-one to afford bis-indolin-2-one derivatives 5a-5d. The structures of the synthesized compounds were determined by 1H NMR, MS and element analysis. Antitumor activities of all the synthesized compounds in vitro were tested. All the 12 synthesized compounds possess antitumor activities against SPC-A1 strain. Especially the compounds 5a-5d possess potent antitumor activities better than sunitinib. Their IC50 are all below 5 micromol x L(-1).

Abstract:A series of novel 2-amino-3-cyano-6-(1H-indol-3-yl)-4-phenylpyridine derivatives were synthesized and their cytotoxic activity against A549, H460, HT-29 and SMMC-7721 cell lines was evaluated in vitro. Among them, ten compounds (10, 11, 14, 16, 17, 26, 27, 29, 30 and 31) displayed excellent anti-tumor activity against different cell lines. The most promising compound 27 showed strong anti-tumor activity against A549, H460, HT-29 and SMMC-7721 cell lines with IC50 values of 22, 0.23, 0.65 and 0.77nM, which were 2.6-, 83-, 1.1×103- and 2.0×103- fold more active than MX-58151 (IC50 values of 0.058, 0.019, 0.70 and 1.53μM), respectively.

Abstract:A new series of 1,5-disubstituted indolin-2,3-diones was synthesized and their inhibition of the growth of a human acute promyelocytic leukemia (HL-60) cell line was evaluated. These compounds had promising inhibition of HL-60 cell growth in vitro. Results indicated that compounds with a benzyl substituent at the N-1 position on the indolin-2,3-dione ring had more potent antiproliferative activity than those with a (4-fluorobenzyl) amino-2-oxoethyl substituent at the N-1 position. Among the compounds synthesized, compound 8l inhibited half of cell growth at a concentration of 0.07 mu M and compound 8p did so at a concentration of 0.14 mu M. These compounds may serve as lead compounds for further optimization in order to develop novel anticancer agents.

Abstract:Three novel indolo[1,2-b] indazole triflate derivatives(3a~3c) were synthesized by salification of indoloindazole derivatives with methyl trifluoromethanesulfonate,then coupling with p-dimethylaminobenzaldehyde.The structures were characterized by NMR and IR.The anticancer activities of 3 against cancer cell lines HL-60 and K562 were tested by the MTT method in vitro.The results showed that 3 exhibited potent anticancer activities.
