Abstract:Nanoparticles have been widely developed and shown to have intrinsic enzymatic ability, and are used in biosensors. Compared to biological enzymes used in biosensors, which are expensive and tedious to harvest, enzyme-mimic nanoparticles or nanozymes are both more stable and sensitive. An important area in this work is the development of a simple detection principle of immunosensor based on the one-step synthesis of silver nanoparticle seeded onto a gold core. The gold-silver core-shell nanoparticle acts as a peroxidase mimic, which enables them to oxidise 3,3',5,5'-tetra- methylbenzidine (TMB) with H2O2, giving a colourimetric response. Herein, the analytical performance of the nanozyme is exploited to detect haptoglobin as a model analyte in a 96-well plate and measured the colourimetric product using spectrophotometer. The sensitivity of the immunosensor was as low as 100 pg mL–1. The viability of our immunosensor was shown to have good selectivity and satisfactory recovery in real serum samples.

Fabrication of a Novel Electrochemical Immunosensor Based on the Gold Nanoparticles/colloidal Carbon Nanosphere Hybrid Material

Abstract:The gold nanoparticles/colloidal carbon sphere hybrid material was used for the immobilization of protein, and was developed in biosensing. The hybrid material was fabricated by the assembly of gold nanoparticles onto the surface of colloidal carbon spheres, which constructed a 3D antibody immobilization matrix on the glass carbon electrode and made the immobilized biomolecules hold high stability and bioactivity. After the sandwich-type immunoreaction, the formed HRP-labeled immunoconjugate showed good enzymatic activity for the oxidation of o-phenylenediamine by H2O2. The approach provided a linear response range between 5 and 250ng/mL with a detection limit of 1.8ng/mL. The immunosensor showed good precision, acceptable stability and reproducibility and could be used for the detection of human IgG in real samples, which provided a potential alternative tool for the detection of protein in clinical laboratory.

Abstract:Recently developed photoelectrochemical (PEC) sensing systems represent a unique potential detection method for real-time analysis of chemical/biological molecules, while the low absorption of TiO2 nanomaterials in the visible wavelength region and the slow surface charge transfer efficiency limit the ultimate sensitivity. Here we develop a gold nanoparticle-decorated TiO2 nanowire sensor for PEC detection of protein binding. The direct attachment of Au nanoparticles to TiO2 nanowires offers strong surface plasmon resonance for electrochemical field effect amplification, yielding a similar to 100% increase of photocurrent density. In addition, the surface functionalization of gold nanoparticles allows for direct capturing of target proteins near the Au/TiO2 interface and thus substantially enhances the capability of attenuation of energy coupling between Au and TiO2, leading to much-improved sensor performance. As a proof of concept, cholera toxin subunit B has been robustly detected by the TiO2-Au nanowire sensor functionalized with ganglioside GM!, with a high sensitivity of 0.167 nM and excellent selectivity. Furthermore, the real-time feature of photoelectrochemical sensing enables direct measurement of binding kinetics between cholera toxin subunit B and GM!, yielding association and disassociation rate constants and an equilibrium constant K-d of 4.17 nM. This surface plasmon resonance-enhanced real-time, photoelectrochemical sensing design may lead to exciting biodetection capabilities with high sensitivity and real-time kinetic studies.

Abstract:This work designed a simple, sensitive, and low-cost immunosensor for the detection of protein marker by using a carbon sphere/gold nanoparticle (CNS/AuNP) composite as an electrochemical label. The nanoscale carbon spheres, prepared with a hydrothermal method by using glucose as raw material, were used to load AuNPs for labeling antibody by electrostatic interaction, which provided a feasible pathway for electron transfer due to the remarkable conductivity. The disposable immunosensor was constructed by coating a polyethylene glycol (PEG) film on a screen-printed carbon-working electrode and then immobilizing capture antibody on the film. With a sandwich-type immunoassay format, the analyte and then the CNS/AuNP-labeled antibody were successively bound to the immunosensor. The bound AuNPs were finally electro-oxidized in 0.1?M HCl to produce AuCl4- for differential pulse voltammetric (DPV) detection. The high-loading capability of AuNPs on CNS for the sandwich-type immunorecognition led to obvious signal amplification. By using human immunoglobulin G (IgG) as model target, the DPV signal of AuNPs after electro-oxidized at optimal potential of +1.40 V for 40 s showed a wide linear dependence on the logarithm of target concentration ranging from 10 pg?mL-1 to 10 ng?mL-1. The detection limit was around 9 pg?mL-1. The immunosensor showed excellent analytical performance with cost effectivity, good fabrication reproducibility, and acceptable precision and accuracy, providing significant potential application in clinical analysis.

Abstract:A rapid microwave-hydrothermal method has been developed to prepare monodisperse colloidal carbon nanospheres from glucose solution, and gold nanoparticles (AuNPs) are successfully assembled oil the surface of the colloidal carbon nanospheres by a self-assembly approach. The resulting AuNP/colloidal carbon nanosphere hybrid material (AuNP/C) has been characterized and is expected to offer a promising template for biomolecule immobilization and biosensor fabrication because of its satisfactory chemical stability and the good biocompatibility of AuNPs. Herein, as an example, it is demonstrated that the as-prepared AuNP/C hybrid material can be conjugated with horseradish peroxidase-labeled antibody (HRP-Ab(2),) to fabricate HRP-Ab(2)-AuNP/C bioconjugates, which can then be used as a label for the sensitive detection of protein. The amperometric immunosensor fabricated on a carbon nanotube-modified glass carbon electrode was very effective for antibody immobilization. The approach provided a linear response range between 0.01 and 250 ng mL(-1) with a detection limit of 5.6 pg mL(-1). The developed assay method was versatile, offered enhanced performances, and could be easily extended to other protein detection as well as DNA analysis.
Enzyme-nanoparticle conjugates at oil-water interface for amplification of electrochemical immunosensing

Abstract:In the current research, the preparation of biocatalytic capsule (Ab(2)-catalytic capsule) was based on utilizing "oil" (organic compound) to entrap the nano-Au/HRP/HAFP bioconjugates, which were produced by horseradish peroxidase (HRP) and HRP labeled anti-AFP (HAFP) co-conjugated on gold nanoparticles (nano-Au). For the first time, a signal amplification strategy was introduced by the prepared biocatalytic capsule as labels for electrochemical immunodetection of α-1-fetoprotein (AFP). The transmission electron microscopy (TEM) images indicated that the biocatalytic capsules were successfully synthesized. The cyclic voltammetry (CV) experiments provided the evidence that the prepared capsule exhibited high catalytic activity for the reduction of H(2)O(2). Besides, a biocompatible monolayer (collagen-TiO(2)) was introduced to immobilize a nano-Au layer to fabricate the primary antibody on the electrode surface. After the prepared biocatalytic capsule was captured to the electrode surface to form the sandwich type immunoassay, the enhanced sensitivity was obtained with a detection limit of 29 pg/mL. The proposed immunoassay also presented good stability, selectivity and storage stability.

Abstract:In this work, novel multifunctional patchy gold coated Fe3O4 hybrid nanoparticles (PG-Fe3O4 NPs) have been successfully synthesized in aqueous medium via a facile adsorption-reduction method. A rational formation mechanism has been proposed by monitoring the morphological evolution. The PG-Fe3O4 NPs retained the good magnetic property and exhibited excellent catalytical effeciency towards the electrochemical reduction of hydrogen peroxide. Chronoamperometric and amperometric experiments indicated a relatively high catalytic rate constant of 3.13 × 105 M-1 s-1, a high sensitivity of 578.87 µA mM-1 cm-2 and a low Michaelis-Menten constant of 462 µM. Meanwhile, the introduction of patchy gold could help biofunctionalization via Au-S bond for different biodetection and biosensing purposes. Here, as an example, thiol-terminated aptamers were immobilized onto the patchy gold part as a signal probe to detect carcinoembryonic antigen (CEA). A related paper-based bipolar electrode-electrochemiluminescence (pBPE-ECL) aptasensor was fabricated as the low-cost, disposable and miniature platform. To improve the sensitivity, Au nanodendrites were electrodeposited at the BPE cathode as the matrix for Apt1 immobilization. This aptasensor showed a wide linear range of 0.1 pg mL-1-15 ng mL-1 with a low detection limit of 0.03 pg mL-1, remaining competitive against other ones, and also demonstrating the PG-Fe3O4 NPs have promising potential for catalysis and bioassays.
Proximity hybridization-regulated electrochemical stripping of silver nanoparticles via nanogold induced deposition for immunoassay.

Abstract:A simple and disposable electrochemical immunosensor was developed for sensitive and selective detection of a protein biomarker via target-induced proximity hybridization and electrochemical stripping analysis of silver nanoparticles (AgNPs). The immunosensor was prepared by assembling single-stranded DNA modified gold nanoparticles (ssDNA@AuNPs) on a graphene oxide modified disposable screen-printed carbon electrode (SPCE). In the presence of a target protein and two DNA-labeled antibodies, the proximate complex formed in homogeneous solution could hybridize with the assembled DNA to take away AuNPs, which decreased AuNP-catalyzed deposition of AgNPs on the immunosensor surface, and thus the anodic stripping signal. The proposed method avoided the interference of dissolved oxygen. Using the carcinoembryonic antigen (CEA) as a model analyte, this method showed a linear range of four magnitude orders with a detection limit down to 3.9 pg mL(-1). The electrochemical immunosensor possessed preparation convenience, good stability and high sensitivity, and could be extended to sensitive biosensing of other analytes, showing potential application in point-of-care testing.
C-dots assisted synthesis of gold nanoparticles as labels to catalyze copper deposition for ultrasensitive electrochemical sensing of proteins

Abstract:We report an ultrasensitive protocol for electrochemical sensing using the hydroxyl-rich C-dots assisted synthesis of gold nanoparticles (C-dots@AuNP) as labels with copper depositon reaction. The C-dots catalyzing copper deposition reaction was implemented for the first time. We constructed a sandwich-type immunosensor on the chitosan modified glassy carbon electrode (GCE) by glutaraldehyde (GA) crosslinking, with C-dots@AuNP as biolabels. Copper was deposited on the catalytic surfaces of second antibody-conjugated C-dots@AuNP nanoparticles through CuSO 4 -ascorbic acid reduction, because both C-dots and AuNPs could strongly catalyze the CuSO4 and ascorbic acid to form Cu particles, which amplified the detection signal. Then the corresponding antigen was quantified based on simultaneous chemical-dissolution/cathodic-preconcentration of copper for in-situ analysis using anodic stripping square wave voltammetry (ASSWV) directly on the modified electrode. Under optimized conditions, these electrodes were employed for sandwich-type immunoanalysis, pushing the lower limits of detection (LODs) down to the fg mL −1 level for human immunoglobulin G (IgG) and cardiac troponin I (cTnI), a cardiac biomarker. These novel sensors have good stability and acceptable accuracy and reproducibility, suggesting potential applications in clinical diagnostics.
Nanozyme-Activated Synergistic Amplification for Ultrasensitive Photoelectrochemical Immunoassay

Abstract:At present, enzyme-mediated signal amplification strategies have been widely applied in photoelectrochemical (PEC) biosensing systems, while the introduction of natural enzymes onto the surface of photoelectrodes inevitably obstructs the electron transfer due to their insulating properties as proteins, leading to severe damage to photocurrent. In this work, the PdPt bimetallic nanozymes with the efficient peroxidase-like activity were used as alternatives to natural enzymes and amplified PEC biosensing signals via their efficient enzymatic reaction and remarkable enhancement in photocurrent. As a result, photoactive CdS nanorods modified with PdPt bimetallic nanozymes showed a boosted PEC performance compared with the pristine CdS nanorods due to the localized surface plasmon resonance effect and Schottky junction. On the basis of the as-prepared CdS/PdPt photoelectrode, a sensitive split-type glucose oxidase-mediated PEC immunoassay for carcinoembryonic antigen (CEA) detection was successfully constructed. Along with the sandwich immunocomplexing, the subsequently produced hydrogen peroxide (H2O2) can oxidize 4-chloro-1-naphthol into insoluble precipitates to inhibit photocurrent and simultaneously trigger the bio-etching of CdS to further restrain photocurrent signals due to the excellent peroxidase-mimicking activity of PdPt nanozymes. Owing to the synergistic signal amplification fulfilled by PdPt nanozymes, an ultrasensitive immunoassay of CEA was realized with a wider linear range from 1 to 5000 pg/mL and a low detection limit of 0.21 pg/mL, opening a new avenue for building ultrasensitive PEC biosensors with nanozymes.

Abstract:The construction of simple, stable, low-cost and reproducible enzyme-free electrochemical biosensors can effectively avoid the problem of signal attenuation caused by enzyme inactivation. Hererin, we prepared a novel nanoenzymes PdPtCu mesoporous nanocubes (MNCs) to construct a label-free sandwich electrochemical immunosensor for the highly sensitivity detection of HIV-p24. PdPtCu MNCs have excellent peroxidase activity against hydrogen peroxide (H2O2) due to their synergistic ternary composition, large surface area and ability to penetrate mesoporous channels. Moreover, highly conductive and biocompatible gold nanoparticles@graphene oxide (AuNPs@GO) was introduced as a substrate to modify a glassy carbon electrode (GCE). Owing to the excellent electrochemical performance of the PdPtCu MNCs and AuNPs@GO, the developed immunosensors exhibited a good linear response from 0.04 pg/mL to 100 ng/mL with a low detection limit of 20 fg/mL. In addition, the established method exhibited excellent practical performance in human serum. This novel strategy provides a promising platform for ultrasensitive detection of the HIV-p24 in the field of clinical diagnostics.
Gold Nanoclusters with Two Sets of Embedded Enzyme Nanoparticles for Applications as Electrochemical Sensors for Glucose

Abstract:Enzyme-based electrochemical biosensors have been widely used because of their sensitivity, rapidity, and highly target-specific reactions. However, the immobilized enzymes can hinder electron transfer on the surface of the electrode and can decrease the sensitivity of electrochemical biosensors. To overcome this problem, we synthesized conductive gold nanoclusters with two sets of embedded enzyme nanoparticles, which are called gold nanocluster-embedded dual-enzyme nanoparticles (AuNC-DENPs) composed of a glucose oxidase-stabilized gold nanocluster (GOX-AuNC) and a horseradish peroxide-stabilized gold nanocluster (HRP-AuNC). Moreover, the application of these nanoparticles as an enzyme-based highly sensitive electrochemical sensor was investigated. Owing to the effect of AuNCs, these nanoparticles have good conductivity compared to bare protein nanoparticles. In addition, the synthesized AuNC-DENPs enabled the combination of a two-enzyme cascade reaction, in which the GOX-AuNC component of the nanoparticle oxidized glucose to generate hydrogen peroxide, which then reacted with the adjacent HRP-AuNC component on the nanoparticle. Given the proximity of the two enzyme components in a single nanoparticle, the AuNC-DENPs markedly reduced the diffusion and decomposition of H2O2 during the cascade reaction and showed an enhanced catalytic reaction compared to a mixture of enzymes. As a result, the biosensor exhibited high sensitivity (18,944 μA/mM cm2) and could detect very low concentrations of glucose ranging from 5 to 320 nM and a LOD of 2.58 nM. In addition, by analyzing nontarget materials and serum together with glucose, it was confirmed that the sensor has good selectivity for glucose.

Enzymatic deposition of gold nanoparticles at vertically aligned carbon nanotubes for electrochemical stripping analysis and ultrasensitive immunosensing of carcinoembryonic antigen

Abstract:Herein we combine the sandwich immunoreaction at a vertically aligned single-walled carbon nanotube (SWCNT)-based immunosensor and the enzymatically catalytic deposition of gold nanoparticles (Au NPs) by a gold nanoprobe to develop a novel electrochemical immunosensing method. The vertically arranged nanostructure was prepared through the covalent linking of terminally carboxylated SWCNTs at an aryldiazonium-modified electrode. It not only provides an excellent platform for the high density immobilization of antibodies to obtain the immunosensor but also serves as useful molecular wires to accelerate electron transfer during the electrochemical immunosensing process. Meanwhile, the enzymatic reaction of the nanoprobe prepared by surface functionalization of the nanocarrier of Au NPs by high-content glucoamylases can catalyze the deposition of a large number of Au NPs at the immunosensor. The electrochemical stripping analysis of these nanoparticles enabled the convenient signal transduction of the method. Due to the sensitive gold stripping analysis at the vertically aligned SWCNTs and the multi-enzyme signal amplification of the nanoprobe, the electrochemical signal response was greatly enhanced. Thus, the method can be used for the ultrasensitive detection of the tumor biomarker of carcinoembryonic antigen in a wide linear range of 5 orders of magnitude with a low detection limit of 0.48 pg mL-1. Considering its obvious performance superiorities, this immunosensing method exhibits an extensive prospect for practical applications.
The Preparation of Hollow AgPt@Pt Core-Shell Nanoparticles Loaded on Polypyrrole Nanosheet Modified Electrode and Its Application in Immunosensor

Abstract:The designed synthesis of efficient materials can significantly enhance the performance of electrochemical immunoassay in the detection of diseases, pesticide residues and environmental pollutants. The hollow AgPt@Pt core-shell nanoparticles (AgPt@Pt HNs) have exhibited high catalytic efficiency to the hydrogen peroxide (H2O2) reduction for its high mass activity from their hollow structure. Their limitation of instability can be overcome by loading on polypyrrole nanosheet (PPy NS). Besides, PPy NS exhibits good conductivity, and there exists environmentally-friendly method for its synthetic. Thus, AgPt@Pt HNs loaded on PPy NS (AgPt@Pt HNs/PPy NS) exhibits high catalytic efficiency to the reduction of H2O2 and good stability. Furthermore, the quick electron transfer of AgPt@Pt HNs/PPy NS modified glassy carbon electrode has been evidenced by the finding that the large constant of apparent electron transfer rate has also enlarged the current signal when the amount of electron is invariant. The modified electrode has fabricated a label-free amperometric immunosensor to detect sensitively prostate-specific antigen (PSA) with H2O2 as the electroactive material. The immunosensor in hollow core-shell nanosheet structure exhibiting good detection performance of PSA shows its promising applications in the clinical diagnosis.
Weak Interaction-Tailored Catalytic Interface of Ultrasmall Gold Nanoclusters as Enzyme Mimics for Enhanced Colorimetric Biosensing.

Abstract:Gold nanoclusters (AuNCs) represent an emerging type of engineered nanomaterials with intrinsic enzymatic activity for both chemical and biological applications, but the catalytic activity of most reported AuNCs remains rather limited. Herein, we report a new, efficient strategy of promoting the peroxidase-mimic activity of AuNCs by tailoring their catalytic interfaces via small molecule-mediated weak interactions. Inspired by the presence of imidazole structures in many biocatalytic centers, we screened a series of imidazole-containing small molecules to evaluate their impact on the enzymatic activity of AuNCs. Through monitoring the absorbance change of 3,3',5,5'-tetramethylbenzidine, 1-methyl-2-imidazolecarboxaldehyde (MCA) was identified to possess the most significant effect on enhancing the peroxidase-mimic activity of glutathione-stabilized AuNCs (GSH-AuNCs) among all the examined molecules. Interestingly, the enhancement effect of MCA on the catalytic activity of these AuNCs was found to be highly reversible and can be switched on/off by simply adding MCA/dialysis treatment. Molecular dynamics simulations and further experimental analysis confirmed that these MCA molecules were adsorbed on the surface of GSH-AuNCs through weak non-covalent interactions. The underlying mechanism analysis suggested that the presence of MCA can efficiently promote the production of •OH in the GSH-AuNC system. As a proof of example, we then demonstrated that the presence of MCA can greatly increase the bioanalytical performance of AuNC-based peroxidase mimics, as evidenced by a 65-fold lower LOD for glucose detection of AuNCs@MCA than that using AuNCs only. Finally, the present system has been successfully applied for sensing the blood glucose level of both healthy people and diabetics with promising results.

Abstract:A palladium nanoparticle decorated carbon nanotube was designed as a label for preparation of a highly sensitive disposable immunosensor. The immunosensor was constructed by assembling the capture antibody on gold nanoparticles decorated graphene nanosheets modified screen printed carbon working electrode. With a sandwich immunoassay mode, the palladium nanoparticle decorated carbon nanotubes were captured to the immunocomplex and showed strong electrocatalytic activity toward oxygen reduction. The use of carbon nanotube carrier offered a high amount of palladium nanoparticles on each immunoconjugate, hence amplified the detectable signal from the electro-reaction of dissolved oxygen. The graphene nanosheets and gold nanoparticles improved the electronic conductivity and the hydrophilicity of electrode surface for immobilization of the capture antibody, respectively. Under optimal conditions, a linear detection range from 50pg/mL to 10ng/mL and a limit of detection of 44pg/mL (0.3pM) were achieved for human IgG. Using dissolved oxygen as a signal reporter, the detection process avoided deoxygenation. The immunosensor showed acceptable stability, precision and accuracy, indicating potential applications in clinical diagnostics.

Abstract:A cheap and portable electrochemical immunosensor based on silver. enhanced colloidal gold electrochemical stripping technique and gold nanoclusters (AuNCs) labels was proposed for the detection of human IgG. Inspired by the adhesive proteins that secreted by marine mussels, the stable and robust film. poly (dopamine) was used to immobilize biomolecules (Ab(1)) on an indium tin oxide (ITO) electrode for the fabrication of immunosensor. Then, human IgG and AuNCs-Ab(2) were successively immobilized to the Ab(1) modified ITO electrode. The process for the fabrication of sandwich immunosensor was monitored by electrochemical impedance spectroscopy. Through the immunoreaction between antigen and antibody, IgG could be quantitatively determined by anodic stripping voltammetry. On the basis of the dual signal amplification of AuNCs labels and silver enhancement technique, the sensitivity was improved to 0.5 ng/L. Moreover, the proposed immunosensor could be used in clinical diagnosis.

Abstract:This paper reports an amplification procedure application of multi-functionalized gold nanoparticles (MFAuNPs) which was used for electrochemical impedance immunosensor. The modified electrode which was utilized for the affinity adsorption of protein A, was fabricated by stepwise immobilization of 1, 6-hexanedithiol, AuNPs, and IgG on the gold electrode via self-assembling technique. The interfacial properties of the modified electrodes were evaluated in the presence of Fe(CN)64-/3- couple redox as a probe by cyclic voltammetry and electrochemical impedance spectroscopy. An equivalent circuit model with a constant phase element was used to interpret the obtained impedance spectra. The electrochemical behavior of the redox probe was affected by the accumulation of treated substances on the electrode surface. There is no significant increase in the electron transfer resistance after the binding of protein A to IgG on the electrode surface without amplification. The changes in the electron transfer resistance on the IgG-modified electrodes became more sensitive after the MFAuNPs were introduced for signal amplification. The increments of amplified impedance showed good correlation of the detection of protein A in the range of 5–1000pg/mL with a detection limit of 1pg/mL. The development of a rapid, facile, and sensitive amplification method capable to detect biomolecules in the picogram range, utilizing EIS measurement, may be achieved in this study.

Abstract:Nanocatalysts are a promising alternative to natural enzymes as the signal labels of electrochemical biosensors. However, the surface modification of nanocatalysts and sensor electrodes with recognition elements and blockers may form a barrier to direct electron transfer, thus limiting the application of nanocatalysts in electrochemical immunoassays. Electron mediators can accelerate the electron transfer between nanocatalysts and electrodes. Nevertheless, it is hard to simultaneously achieve fast electron exchange between nanocatalysts and redox mediators as well as substrates. This work presents a scheme for the design of electrochemical immunosensors with nanocatalysts as signal labels, in which pyrroloquinoline quinone (PQQ) is the redox-active center of the nanocatalyst. PQQ was decorated on the surface of carbon nanotubes to catalyze the electrochemical oxidation of tris(2-carboxyethyl)phosphine (TCEP) with ferrocenylmethanol (FcM) as the electron mediator. With prostate-specific antigen (PSA) as the model analyte, the detection limit of the sandwich-type immunosensor was found to be 5 pg/mL. The keys to success for this scheme are the slow chemical reaction between TCEP and ferricinum ions, and the high turnover frequency between ferricinum ions, PQQ. and TCEP. This work should be valuable for designing of novel nanolabels and nanocatalytic schemes for electrochemical biosensors.

Abstract:Gold nanoparticles (AuNPs) have been widely employed for the fabrication of electrochemical biosensors. In most cases, AuNPs are immobilized on the surface of an electrode, so they are difficult to be regenerated, making the use of the biosensor unfriendly. In this work, by adopting AuNPs directly as the electrolytes, we have developed a novel AuNPs-based electrochemical detection system. In brief, AuNPs-catalyzed oxidation of glucose is combined with a HRP-catalyzed reaction as well as an electrocatalytic reaction to compose cascade reactions in the electrolyte. Thus, the intensity of the electrocatalytic signals has quantitative relation with the concentration of glucose, and favors the sensitive detection of glucose. Furthermore, because the catalysis of AuNPs may be blocked under the interaction with single-stranded DNA and unblocked in the presence of a complementary sequence, detection of DNA and even single-nucleotide polymorphism can thereby been achieved. This one-pot detection system can be operated and regenerated very easily, since all the components are integrated in the electrolytes of AuNPs, and the unmodified electrode can be reused after being rinsed. This concept by integrating the advantages of sensitive electrochemical detection with the easy-to-operate nanocolloidal system may also promote the development of other kinds of electrochemical biosensors.
