Abstract:A sandwich-type photoelectrochemical (PEC) immunosensor was developed for cytokeratin 19 fragment 21-1 (CYFRA21-1) ultrasensitive detection relying on a multiple signal inhibition strategy. In this work, Bi10O6S9 nanosheet arrays (BiOS) sensitized strontium titanate nanocubes (SrTiO3 NCs) was used as photoactive matrix. Bi10O6S9 with the large specific surface area and narrow band gap could improve the utilization of light to provide strong initial PEC signal. Besides, bovine serum albumin stabilized gold nanoparticles (BSA@Au NPs) anchored on Cu7S4 polyhedron composite (BSA-Au@Cu7S4 polyhedron) was utilized as an effective PEC signal regulator. As a duple nanozyme with the glucose oxidase (GOx)-like activity and the peroxidase-like activity, BSA@Au NPs induced intriguing tandem biocatalytic precipitation (BCP). On the one hand, the BCP could produce precipitations which obstructed the electron transfer between the photoelectrode and electron donor, leading a decrease of PEC signal; On the other hand, the competitive light absorption between BSA-Au@Cu7S4 polyhedron and photoactive matrix caused a further reduction of PEC signal. Generally, the PEC signal was quenched sharply taking advantage of multiple signal inhibition strategy including tandem BCP strategy, competitive absorption of light and steric hindrance, which improved the detection sensitivity of PEC immunosensor. The suggested PEC immunosensor displayed a good linearity in the range of 0.1 pg/mL -50 ng/mL for CYFRA21-1 detection with a low detection limit of 1.12 fg/mL (S/N = 3). The skillful strategy has infinite potential in biological samples analysis.

Abstract:The ideal photoelectrode and efficient signaling strategy are pivotal to achieve sensitive photoelectrochemical (PEC) analysis. Here, a multipath collaborative signal amplification-based PEC immunosensor was constructed for the ultrasensitive detection of cytokeratin 19 fragment 21-1. Specifically, the photoelectrode fabricated by Z-scheme In2O3/g-C3N4 heterojunction showed enhanced photocurrent intensity in response to visible light. Meanwhile, the signal probe, horseradish peroxidase functionalized dopamine-melanin nanosphere@Au nanoparticles (HRP-Dpa-melanin NS@AuNPs), were introduced into the system. When the target exists, the signal probe can induce multiple quenching of the photocurrent due to the competition of light absorption, steric hindrance and HRP-mediated biocatalytic precipitation, which effectively inhibit light, electron donor, and electron access to the photoelectrode. The fabricated immunosensor exhibits a wide linear range from 1.0 × 10-3 - 1.0 × 102 ng mL-1 with the detection limit of 0.35 pg mL-1 (S/N = 3) for cytokeratin 19 fragment 21-1 detection. The study enhances sensitivity for PEC detection by utilizing the superior Z-scheme heterojunction photoelectrode, providing a valuable method that combines multiple signal pathways for a synergistic effect in bioanalysis.

Abstract:Sensitive but with simple, inexpensive detection of disease-related biomarkers in real biological samples is of quite necessity for early diagnosis and disease surveillance. We herein first introduced high-activity Fe3O4 nanozyme as signal amplifier to develop an ultrasensitive photoelectrochemical (PEC) immunoassay, which meanwhile has the distinct merits of both simplicity and low cost compared with previously reported enzyme-labeling PEC immunoassays. In the proposal, to illustrate and describe the PEC platform, prostate-specific antigen (PSA, Ag) was used as a target model. Specifically, ZnO nanorods (ZnO-NRs) grown vertically on a bare indium-tin oxide (ITO) electrode was deposited with ZnIn2S4 nanocrystals, producing ZnIn2S4/ZnO-NRs/ITO photoelectrode as the PEC matrix to modify capture PSA antibody (Ab1). Histidine-modified Fe3O4 (his-Fe3O4) nanozyme as signal amplifier was linked with signal PSA antibody (Ab2) to form his-Fe3O4@Ab2 conjugate, and was anchored through specific sandwich immunoreaction. The labeling his-Fe3O4 nanozyme acted as a peroxidase to induce the generation of the insoluble and insulating precipitation, resulting in an evident decrease in the photocurrent signal. On account of combined effects of high catalytic efficiency of the his-Fe3O4 nanozyme and excellent PEC properties of the ZnIn2S4/ZnO-NRs/ITO photoelectrode, ultralow detection limit of 18 fg/mL for target Ag detection was achieved. Besides, as high-activity his-Fe3O4 nanozyme has substituted natural enzyme as signal amplifier, simplicity and low cost of the PEC immunoassay was realized.

Abstract:Recently emerged liposomal photoelectrochemical (PEC) bioanalysis has brought new opportunities for biosensor development. This work presents the new concept of liposome-assisted enzymatic modulation of plasmonic photoelectrochemistry for PEC bioanalysis, which was exemplified by an Au nanoclusters (NCs)-sensitized nanoporous TiO2 nanotubes (Au NC@TiO2 NT) photoelectrode and an alkaline phosphatase (ALP)-loaded liposomal immunoassay of heart-type fatty acid binding protein in a 96-well plate. After sandwich immunorecognition and subsequent lysis treatment, enzymatically generated ascorbic acid by the released ALP was directed to reduce Au3+ into Au nanoparticles using the Au NCs as seeds, leading to the in situ change of the photoelectrochemistry of the electrode and corresponding reduction of the photocurrent. The depressed signal could be correlated with the target concentration with good analytical performance in terms of sensitivity and selectivity. This work features the liposome-assisted enzymatic modulation of plasmonic photoelectrochemistry, which provides a new protocol for general PEC bioanalysis development.
Enzyme-catalyzed high-performing reaction with in-situ amplified photocurrent on carbon-functionalized inorganic photoanode for immunosensing

Abstract:An enzyme-catalyzed high-performing reaction with in-situ amplified photocurrent was innovatively designed for the quantitative screening of carcinoembryonic antigen (CEA) in biological fluids by coupling with carbon-functionalized inorganic photoanode. A split-type photoelectrochemical (PEC) immunoassay was initially executed with horseradish peroxidase (HRP)-labeled secondary antibody on the capture antibody-coated microtiter. Then, the photocurrent of carbon-functionalized inorganic photoanode were improved through enzymatic insoluble product. Experimental results revealed that introduction of the outer carbon layer on the inorganic photoactive materials caused the amplifying photocurrent because of the improving light harvesting and separation of photo-generated e-/h+ pairs. Under optimum conditions, the split-type photoelectrochemical immunosensing platform displayed good photocurrent responses within the dynamic range of 0.01 - 80 ng mL-1 CEA, and allowed the detection of CEA as low as a concentration of 3.6 pg mL-1 at the 3Sblank level. The strong attachment of antibodies onto nano label and high-performing photoanode resulted in a good repeatability and intermediate precision down to 9.83%. No significant differences at the 0.05 significance level were encountered in the analysis of six human serum specimens between the developed PEC immunoassay and the commercially available CEA ELISA kits.

Abstract:Recently developed photoelectrochemical (PEC) sensing systems represent a unique potential detection method for real-time analysis of chemical/biological molecules, while the low absorption of TiO2 nanomaterials in the visible wavelength region and the slow surface charge transfer efficiency limit the ultimate sensitivity. Here we develop a gold nanoparticle-decorated TiO2 nanowire sensor for PEC detection of protein binding. The direct attachment of Au nanoparticles to TiO2 nanowires offers strong surface plasmon resonance for electrochemical field effect amplification, yielding a similar to 100% increase of photocurrent density. In addition, the surface functionalization of gold nanoparticles allows for direct capturing of target proteins near the Au/TiO2 interface and thus substantially enhances the capability of attenuation of energy coupling between Au and TiO2, leading to much-improved sensor performance. As a proof of concept, cholera toxin subunit B has been robustly detected by the TiO2-Au nanowire sensor functionalized with ganglioside GM!, with a high sensitivity of 0.167 nM and excellent selectivity. Furthermore, the real-time feature of photoelectrochemical sensing enables direct measurement of binding kinetics between cholera toxin subunit B and GM!, yielding association and disassociation rate constants and an equilibrium constant K-d of 4.17 nM. This surface plasmon resonance-enhanced real-time, photoelectrochemical sensing design may lead to exciting biodetection capabilities with high sensitivity and real-time kinetic studies.

Abstract:The development of rapid screening sensing platforms to improve pre-screening mechanisms in community healthcare is necessary to meet the significant need for portable testing in biomarker diagnostics. Here, we designed a portable smartphone-based photoelectrochemical (PEC) immunoassay for carcinoembryonic antigen (CEA) detection using Cu-doped ultrathin porous Bi2WO6 (CuBWO) nanosheets as the photoactive material. The CuBWO nanosheets exhibit a fast photocurrent response and excellent electrical transmission rate under UV light due to their surface plasmon resonance effect (SPR). The method uses glucose oxidase-labeled secondary antibody as a signal indicator for sandwich-type immune conjugation. In the presence of the target CEA, the electrons and holes generated at the surface of the photo-excited ultrathin porous CuBWO were rapidly consumed by the production of H2O2 from glucose oxidase oxidizing glucose, resulting in a weakened photocurrent signal. The photocurrent intensity increased logarithmically and linearly with increasing CEA concentration (0.02-50 ng mL-1), with a detection limit of 15.0 pg mL-1 (S/N = 3). The system provides a broader idea for inferring the electron-hole transport mechanism in ultrathin porous nanosheet layer materials and developing efficient PEC sensors.

Abstract:Nowadays, lung cancer is one of the most dangerous cancers threatening human life all over the world. As a crucial biomarker, cytokeratin 19 fragment 21-1 (CYFRA 21-1) is extraordinary important for diagnosis of non-small cell lung cancer (NSCLC). In this work, we synthesized hollow SnO 2 /CdS QDs/CdCO 3 heterostructured nanocubes with high and stable photocurrents, which applied to construction of a sandwich-typed photoelectrochemical (PEC) immunosensor for detection of CYFRA 21-1, integrated by an in-situ catalytic precipitation strategy with home-built PtPd alloy anchored MnCo–CeO 2 (PtPd/MnCo–CeO 2 ) nanozyme for synergistic amplification. The interfacial electron transfer mechanism upon visible light irradiation was investigated in details. Further, the PEC responses were seriously quenched by the specific immunoreaction and precipitation catalyzed by the PtPd/MnCo–CeO 2 nanozyme. The established biosensor showed a wider linear range of 0.001–200 ng mL −1 and a lower limit of detection (LOD = 0.2 pg mL −1 , S / N = 3), coupled by exploring such analysis even in diluted human serum sample. This work opens a constructive avenue to develop ultrasensitive PEC sensing platforms for detecting diverse cancer biomarkers in clinic.

Abstract:Enzyme-like nanomaterials (nanozyme) has been used to generate amplified response signals in biosensors instead of the bio-enzyme, on account of their higher stability and comparable catalytical activity. Especially the metal-organic framework (MOF) materials with ultra-high porosity and high specific surface area, which is more suitable for deriving superior nanozyme. In order to deal with the increasing demand of healthcare diagnostics and biomedical research, here we propose a novel approach for ultrasensitive photoelectrochemical (PEC) immunoassay. The bio-inspired MOF-derived nanozyme Hemin/BSA@ZIF-8 was introduced into the target biorecognition and sensing processes to accelerate the oxidization of substrate to produce insoluble precipitate, which will impede the electron transfer and lead to a significant reduction in photocurrent. The proposed PEC sensor exhibits a wide linear range of 0.1 pg mL −1 ~ 100 ng mL −1 , and a low detection limit of 11.9 fg mL −1 for cardiac troponin I (cTnI) detection. This work brings a new insight of the bio-inspired MOF nanozyme-mediated PEC immunoassay designing, and offers a novel sensing strategy for accurate human biomarker analysis.

Abstract:Photoelectrochemical (PEC) immunoassays exhibiting high sensitivity and decent operability have considerable potential in areas such as cancer diagnostics. In particular, cathodic PEC configurations can prevent interference from reductive substances, which can occur in biological samples; however, challenges remain in terms of sensitivity and operability. In this study, separate-type PEC immunoassays were developed for carcinoembryonic antigen (CEA) by combining microplate-based immune recognition and off-on cathodic PEC detection. Polydopamine (PDA)-coated Prussian blue (PB) nanoparticles (PB@PDA NPs) were used as signal tags to label the detection antibody. The PB NPs and PDA captured on the microplates both disassembled under strongly alkaline conditions to generate redox-active electron acceptors. The disassembled products were quantitatively transferred to PEC detection cells and synergistically enhanced the PEC current with microstructured BiOI, which operated as a cathodic semiconductor electrode. As proof of principle, carcinoembryonic antigen (CEA) was applied to elucidate the potential application of PEC immunoassay in clinical diagnosis, and the obtained linear range of the sensor was 0.001-100 ng mL-1 with the detection limit of 54.9 fg mL-1 (S/N = 3). The proposed separate-type off-on PEC strategy showed high sensitivity and decent operability for CEA detection, indicating its potential for the identification of other tumor markers.
Building of a Three-dimensional Apta-nano Interface Using Silver Nanoflower for Photoelectrochemical Detection of Carcinoembryonic Antigen

Abstract:Enhancing the sensitivity of photoelectrochemical (PEC) assays poses a challenging task. This study proposes a method to improve the sensitivity of PEC assays by creating a three-dimensional (3D) apta-nano interface using silver nanoflowers (AgNFs) with a 3D flower-like structure as a substrate. The aptamer (Apt) was immobilized on the surface of the AgNF-loaded gold nanoparticles (AuNPs) through the dual action of polyA blocking and Au-S bonding. Despite using cadmium sulfide quantum dots (CdS QDs) as the sole photoactive material, the as-prepared photoelectrode still showed excellent sensitivity. Using carcinoembryonic antigen (CEA) as a model target, the variation value of photocurrent (Δ I ) exhibited good linear correlation ( R 2 = 0.9970) with the logarithm of CEA concentration in a wide detection range from 0.001 to 100 ng mL -1 , and the detection limit was as low as 0.64 pg mL -1 . In the assay of CEA in 1% serum, the recoveries reached 98.70 ~ 99.63% with a low relative standard derivation (RSD) ranging from 1.11 to 2.05%. This study introduces novel concepts for the development of PEC sensors, potentially enhancing their utility in the clinical detection of disease markers.

Abstract:Benefiting from the maximum atom-utilization efficiency and distinct structural features, single-atom catalysts open a new avenue for the design of more functional catalysts, whereas their bioapplications are still in their infancy. Due to the advantages, platinum single atoms supported by cadmium sulfide nanorods (Pt SAs-CdS) are synthesized to build an ultrasensitive photoelectrochemical (PEC) biosensing platform. With the decoration of Pt SAs, the PEC signal of CdS is significantly boosted. Furthermore, theory calculations indicate the positively charged Pt SAs could change the charge distribution and increase the excited carrier density of CdS. Meanwhile, it also suggests that Cu2+ can severely hinder the photoexcitation and electron–hole separation of CdS. As a proof of concept, prostate-specific antigen is chosen as the target analyte to demonstrate the superiority of the Pt SAs-CdS-based PEC sensing system. Abstract:A signal-on photoelectrochemical (PEC) biosensor was successfully established for the sensitive monitoring of carcinoembryonic antigen (CEA) by using copper indium disulfide-sensitized graphitic-like carbon nitride (g-C3N4/CuInS2) as the photosensitive material and cobalt oxyhydroxide (CoOOH) as the light-blocking material, coupling target-triggered hybridization chain reaction (HCR) with hydrolysate-induced dissolution/etching of CoOOH nanosheets. Initially, a sandwiched reaction occurred between capture aptamer-conjugated magnetic bead and trigger aptamer in the presence of CEA. Then, the carried trigger aptamer initiated HCR between two hairpin sequences to produce long double-helix strand for capturing alkaline phosphatase. The generated ascorbic acid reduced/etched CoOOH nanosheets into divalent cobalt ions, which decreased the amount and thickness of CoOOH and exposed the underlying g-C3N4/CuInS2, thus leading to a distinct increase in the photocurrent. Under optimum conditions, PEC sensor showed high sensitivity toward CEA with a dynamic range of 0.02-40 ng mL-1 and a detection limit of 5.2 pg mL-1. Also, it possessed favorable selectivity, high stability as well as good precision. The accuracy of PEC approach was well consistent with commercial CEA ELISA kit. These exceptional analytical performances of PEC biosensor indicated that it might have a broad application prospect in the diagnosis of CEA.
Ultrasensitive photoelectrochemical biosensing platform based target-triggered biocatalytic precipitation reactions on a flower-like Bi2O2S super-structured photoanode

Abstract:Herein, we reported a novel photoelectrochemical immunoassay method based on a target-triggered on/off signal of the ultra-structured Bi2O2S (BOS) photoanode system for the sensitive testing of carcinoembryonic antigens (CEAs) in serum samples. Well-defined three-dimensional sheet-like self-assembled flower-like Bi2O2S superstructures were obtained using a time-controlled hydrothermal method. Such well-shaped multifaceted surfaces were considered to be good laser cavity mirror surfaces for multifaceted reflection and refraction of excitation light in the material. An elegant enzyme biocatalytic strategy was introduced into the constructed detection model to sensitively detect CEAs. The substrate 4-chloro-1-naphthol (4-CN) was oxidized to 4-chloro-hexadienone (4-CD) under the formation of target-triggered immune complexes against mAb1 and peroxidase-modified mAb2. Subsequently, 4-CD produced by the biocatalytic precipitation reaction was transferred to the photoanodes of Bi2O2S nanoflowers (BOS NFs) to burst their photoelectric signals, thus achieving the quantification of CEAs. Through optimization of the conditions of the immunization protocol, a good negative photocurrent response to the target CEA was found in the wide range of 0.02-50 ng mL-1 with a detection limit of 11.2 pg mL-1. Impressively, the reported biocatalytic PEC sensing strategy on superstructures is comparable, or superior, to the gold standard ELISA kit in terms of sensitivity and the target response range. This study presents a target-mediated PEC immunoassay for biocatalytic precipitation based on a self-assembled superstructure of Bi2O2S, providing a fresh scheme for the analysis of disease-related markers.

Abstract:The tert-butylation of the tryptophan indole ring during the removal of tert-butyloxycarbonyl groups under different conditions was studied. The incorporation of tert-butyl groups was measured by a 1H-NMR technique. It was established, that the rate and specificity of the tert-butylation is highly dependent on the character of the cleaving reagents used and on the environment of the tryptophan residue in the peptide chain.
CdS Quantum-Dots-Decorated V2O5 Nanosheets as Chemically Etchable Active Materials for Sensitive Photoelectrochemical Immunoassay of Carcinoembryonic Antigen

Abstract:We report here CdS quantum-dots (QDs)-decorated V2O5 nanosheets as high-performance and chemically etchable photoelectric active materials for constructing a photoelectrochemical (PEC) immunoassay platform. CdS QDs-decorated V2O5 nanosheets as new photoelectric materials can show superior photocurrent to V2O5 nanosheets and CdS QDs under visible-light irradiation because of the promoted photogenerated electron-hole separation and the increased visible-light absorption. V2O5 nanosheets can be etched by ascorbic acid (AA) because of the reduction of V2O5 to V4+, and the photocurrent of CdS/V2O5-nanocomposite-modified indium tin oxide electrode decreases significantly after being etched by AA. Inspired by this phenomenon, a PEC immunoassay platform is constructed for carcinoembryonic antigen (CEA) detection by using CdS/V2O5 nanocomposite as the photoelectric material and AA-encapsulated liposome immunonanocapsules as labels. The linear detection range for detecting CEA is from 0.5 pg mL-1 to 1 ng mL-1, with a limit of detection of 0.1 pg mL-1. The proposed method also shows good selectivity, excellent reproducibility, and satisfactory recovery in detection of CEA in human serum samples. We believe that this work will lay the foundation for the future development of V2O5-based materials for PEC analysis, and also provide a reasonable design and implementation for the development of PEC immunoassay.

Abstract:A spatial-resolved and self-calibrated photoelectrochemical (PEC) biosensor has been fabricated by a multifunctional CeO 2 /CdS heterostructure, achieving portable and sensitive detection of carcinoembryonic antigen (CEA) using a homemade 3D printing device. The CeO 2 /CdS heterostructure with matched band structure is prepared to construct the dual-photoelectrodes to improve the PEC response of CeO 2 . In particular, as the photoactive nanomaterial, the CeO 2 also plays the role of peroxidase mimetic nanozymes. Therefore, the catalytic performance of CeO 2 with different morphologies (e.g., nano-cubes, nano-rods and nano-octahedra) have been studied, and CeO 2 nano-cubes (c-CeO 2 ) achieve the optimal catalytic activity. Upon introducing CEA, the sandwich-type immunocomplex is formed in the microplate using GOx-AuNPs-labeled second antibody as detection antibody. As a result, H 2 O 2 can be produced from the catalytic oxidization of glucose substrate by GOx, which is further catalyzed by CeO 2 to form •OH, thus in situ etching CdS and decreasing the photocurrents. The self-calibration is achieved by the dual-channel photoelectrodes on the homemade 3D printing device to obtain the photocurrents ratio, thus effectively normalizing the fluctuations of external factors to enhance the accuracy. This integrated biosensor with a detection limit as low as 0.057 ng mL −1 provides a promising way for ultrasensitive immunoassay in clinic application in complex environments.

Abstract:Herein, a novel silver ion-loaded gold microemulsion assemblies (Au/Ag + MAs) mediated multifunctional signal amplification strategy was proposed to construct a sensitive immobilization-free photoelectrochemical (PEC)/colorimetric biosensor for carcinoembryonic antigen (CEA) detection. Through the sandwiched reaction among CEA, the CEA aptamer (DNA1) loaded on the Au nanoparticles (NPs) functionalized iron oxide (Fe 3 O 4 ) nanospheres and another CEA aptamer (DNA2) immobilized on Au/Ag + MAs, a complex is formed and acquired by magnetic separation. Then, Au/Ag + MAs of the complex are disassembled into Au NPs and Ag + ions driven by an acetone response, and the obtained demulsification solution is transferred to the cadmium sulfide/cadmium telluride (CdS/CdTe) photoactive composites modified electrode. Based on the multiple inhibition functions (blocking effect of oleylamine; energy transfer effect of Au NPs; and electron snatching effect of Ag + ), the photocurrent of the electrode decreases obviously, resulting in the ultrasensitive detection of CEA (a detection limit of 16 fg mL −1 ). Interestingly, the ion-exchange reactions between CdS/CdTe composites and Ag + ions generate silver sulfide/silver telluride (Ag 2 S/Ag 2 Te) composites, and a color change of composites can be distinguished directly, leading to a quick visual detection of CEA. Compared with the traditional single-modal assay for CEA, such dual-modal PEC/colorimetric assay is a more accurate and reliable due to different mechanisms and independent signal conversion. This work will offer a new perspective for the applications of various self-assemblies in PEC bioanalysis.

Abstract:Cathode photoelectrochemical immunoassay usually shows better anti-interference capacity toward real samples than anode photoelectrochemical immunoassay. However, its poor photocurrent response has greatly restricted the detection sensitivity. Herein, a promising ultrasensitive cathode photoelectrochemical immunoassay was developed based on TiO2 photoanode-enhanced 3D Cu2O nanowire array (NWA) photocathode, and coupled with signal amplification by horseradish peroxidase (HRP)-induced biocatalytic precipitation (BCP). Carcinoembryonic antigen (CEA, Ag) was used as a detection model, TiO2 nanoparticle-modified indium tin oxide (ITO) electrode served as the photoanode, and Cu2O NWAs grown in situ on Cu mesh was both the photocathode and photoelectrochemical matrix to immobilize the capture CEA antibodies (Ab1). The signal CEA antibodies (Ab2) were labeled with HRP to form Ab2-HRP bioconjugates, and employed as signal amplifiers when the specific immunoreaction occurred. The developed photoanode-enhanced cathode photoelectrochemical immunoassay has good anti-interference capability, outstanding photocurrent response, and high sensitivity for target Ag detection, which was attributed to the synergistic effects of the 3D nanostructure of Cu2O NWA photocathode, the introduction of TiO2 photoanode as counter electrode, and the signal amplification of Ab2-HRP bioconjugate-induced BCP. The developed cathode photoelectrochemical immunoassay showed a low limit of detection (0.037 pg mL−1) with a wide linear range (from 0.1 pg mL−1 to 50 ng mL−1) for CEA detection.
