Abstract:Aldehydes and indoles generally undergo nucleophilic addition reactions to generate alcohols rather than ketones. Here, we report a method for synthesis of 3-acyl indoles from aldehydes by a combination of photocatalysis and palladium catalysis. This mild, robust method is compatible with a wide array of functional groups and has a broad substrate scope. Our findings may inspire the development of new dehydrogenative cross-coupling reactions.

Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:Indole represents a privileged scaffold extensively used in drug design due to its promise to improve both binding affinity and specificity receptors. Direct functionalization of indole framework is an efficient route to achieve structurally diverse indole derivatives. Herein, highly regio-selective C3-alkylation of indoles via hydrogen borrowing methodology (HBM) has been established using alcohols as the alkylation reagents by palladium catalysis. This method enables an efficient synthesis of C3-alkylated indoles, demonstrating high group adaptability to both indole and alcohol substrates (20 examples, up to 94% yield).

Abstract:A novel palladium-catalyzed cascade reaction of o-iodostyrenes, internal alkynes and formates to access various indene-1-acetates was developed. The method enables the construction of three C-C and one C-O bonds through sequential double carbopalladation and aryloxycarbonylation under a palladium/air system. This protocol exhibited high regioselectivity and broad functional group tolerance. In addition, the synthetic utility of this protocol has been successfully demonstrated by the gram-scale synthesis and the late-stage modification of a series of complex bioactive molecules.

Abstract:We describe here a Cu-catalyzed and 4-OH-TEMPO-mediated sequential dehydrogenation/aza-Michael addition/annulation cascade reaction for the construction of N-alkyl 2-arylindoles from facilely available saturated ketones and 2-arylethynylanilines. This reaction shows high regioselectivity and tolerates a variety of functional groups. Moreover, 3-alkyl-substituted indoles can also be achieved when using 2-alkylethynylanilines as starting materials.

Abstract:New functionalizations of indoles via palladium-catalyzed reaction of indoles and 2-acetoxymethyl-substituted electron-deficient alkenes are reported. It was found that for N-protected indoles the reaction proceeded smoothly in the presence of 5 mol % of Pd(acac)2 and 10 mol % of PPh3 at 80 degrees C in HOAc, while for N-unprotected indoles, the reaction was carried out by using 5 mol % of Pd(dba)2 or 2.5 mol % of Pd2(dba)3.CHCl3 with 10 mol % of 2,2'-bipyridine as the catalyst in toluene. This strategy allows the selective installation of electron-deficient olefin functionality at the 3-position of indoles, which might be difficult to obtain by other methods and can be further elaborated.

Abstract:A palladium-catalyzed intermolecular allenylation of non-strained 2,3-disubstituted indoles and allenyl carbonate has been developed, providing convenient access to indolenines bearing an allene unit by taking advantage of the C-3 nucleophilicity of indoles. Decent yields and good functional group tolerance have been achieved with diverse indoles under mild conditions. Gram-scale reaction and various synthetic transformations have been demonstrated.

Abstract:The chosen one! A novel Pd-catalyzed direct and selective C3-acetoxylation of indole derivatives has been accomplished (see scheme). This selective CH activation reaction was implemented without the assistance of directing groups and took place under mild conditions. The kinetic study revealed that the reaction was zero-order with respect to the oxidant and first-order with respect to the indole derivatives.

Abstract:A practical process for the synthesis of 2-unsubstituted indoles and azaindoles has been developed by the palladium-catalyzed direct annulation of electron-poor o-chloro/bromoanilines and o-chloroaminopyridines with aldehydes. Coupled with the previous results of Jia and Zhu, this allows rapid access to a variety of 2-unsubstituted indoles and azaindoles starting from simple and easily accessible precursors.

Abstract:<p>A new Rh(III)-catalyzed direct C-H alkenylation of 2,3-unsubstitued indoles with alkenyl borates through C-H activation followed by transmetallation was described. This protocol provides an efficient method for the synthesis of terminal C2-alkenylindoles under mild conditions, and also shows broad substrate scope and high functional group tolerance with respect to both components. Abstract:A Rh(I)‐catalyzed chelation‐assisted C2‐selective C−H decarbonylative alkylation of indoles with readily available, cheap, safe and structurally diverse alkyl carboxylic acids or anhydrides has been developed. A wide variety of primary and secondary alkyl carboxylic acids and differently substituted indoles are compatible with this transformation, allowing facile synthesis of various C2‐alkylated indoles with high efficiency and broad tolerance of diverse functional groups. The reaction proceeds in the absence of any external oxidant, and the presence of readily installable and removable N‐pyrimidyl directing group is critical for catalysis. The process is convenient and scalable, and avoids the use of a dried solvent and an inert atmosphere. Moreover, selective C7‐alkylation and C2, C7‐dialkylation have also been achieved by slightly modifying the reaction conditions.

Abstract:An efficient palladium-catalyzed intramolecular deacetylative dearomatization reaction of 3-acetoxyindoles has been developed. A range of tetracyclic indolin-3-ones bearing C2-quaternary stereocenters are achieved in good yields, showing a wide substrate scope for this reaction. A preliminary enantioselective reaction is established to furnish the product in 63% ee by using (R,R,R)-phosphoramide-PE as a chiral ligand.

Abstract:With alanine as a transient directing group, Pd-catalyzed regioselective alkynylation at the indole C4-position was successfully established in a good yield. The total synthesis of the PAF antagonist demonstrated the synthetic utility of this protocol. The regioselectivity was explicitly proven by the prepared C4-selective palladacycle intermediate in the catalytic process and the DFT calculation of the energy barriers of C4- and C2-site-selective C-H activation of indole.
