Abstract:A Rh(I)‐catalyzed chelation‐assisted C2‐selective C−H decarbonylative alkylation of indoles with readily available, cheap, safe and structurally diverse alkyl carboxylic acids or anhydrides has been developed. A wide variety of primary and secondary alkyl carboxylic acids and differently substituted indoles are compatible with this transformation, allowing facile synthesis of various C2‐alkylated indoles with high efficiency and broad tolerance of diverse functional groups. The reaction proceeds in the absence of any external oxidant, and the presence of readily installable and removable N‐pyrimidyl directing group is critical for catalysis. The process is convenient and scalable, and avoids the use of a dried solvent and an inert atmosphere. Moreover, selective C7‐alkylation and C2, C7‐dialkylation have also been achieved by slightly modifying the reaction conditions.

Abstract:An efficient palladium-catalyzed decarboxylative ortho-acylation of 2-aryloxypyridines with -oxocarboxylic acids is described. In this new transformation, the aromatic C(sp2)H bond was successfully acylated to give diverse aromatic ketones regioselectively in moderate to good yields. The pyridine group can be removed easily after the acylation to give the corresponding 2-hydroxy aromatic ketones.

Abstract:A novel palladium-catalyzed cascade reaction of o-iodostyrenes, internal alkynes and formates to access various indene-1-acetates was developed. The method enables the construction of three C-C and one C-O bonds through sequential double carbopalladation and aryloxycarbonylation under a palladium/air system. This protocol exhibited high regioselectivity and broad functional group tolerance. In addition, the synthetic utility of this protocol has been successfully demonstrated by the gram-scale synthesis and the late-stage modification of a series of complex bioactive molecules.

Abstract:Mild conditions have been developed to achieve Pd-catalyzed homocoupling of indoles with excellent regioselectivity in the presence of Cu(OAc)(2) x H(2)O in DMSO at room temperature in high efficiency. This method provides a simple route to 2,3'-biindolyls from the electron-rich to moderately electron-poor indoles. The reaction tolerates the bromide substituent on indoles. In addition, a one-pot approach to C3-position acetoxylated biindolyls is realized via palladium catalysis by the use of AgOAc under oxygen atmosphere as oxidants.

Abstract:Aldehydes and indoles generally undergo nucleophilic addition reactions to generate alcohols rather than ketones. Here, we report a method for synthesis of 3-acyl indoles from aldehydes by a combination of photocatalysis and palladium catalysis. This mild, robust method is compatible with a wide array of functional groups and has a broad substrate scope. Our findings may inspire the development of new dehydrogenative cross-coupling reactions.

Abstract:A novel palladium-catalyzed decarboxylative coupling reaction of vinyl benzoxazinanones with arynes which may feature an intramolecular nucleophilic attack of an amino group at the central carbon of π-allylpalladium intermediate has been developed. The cis-5,5a,6,10b-tetrahydroindeno[2,1-b]indoles were generated in moderate to good yields. One key to the success of the present reaction was to achieve comparable rates for the palladium-catalyzed decarboxylation and aryne formation steps.

Abstract:The chosen one! A novel Pd-catalyzed direct and selective C3-acetoxylation of indole derivatives has been accomplished (see scheme). This selective CH activation reaction was implemented without the assistance of directing groups and took place under mild conditions. The kinetic study revealed that the reaction was zero-order with respect to the oxidant and first-order with respect to the indole derivatives.

Abstract:We, herein, present the first highly regio- and chemoselective C2 C-H acyloxylation of indole under rhodium catalysis and N-quinolinyl auxiliary. This strategy accommodates a wide range of indoles and structurally diverse carboxylic acids with good reaction efficiency to furnish functionalized indoles. The application utility of this logic was demonstrated by the concise synthesis of a kind of functionalized 2-oxindole derivatives. The preliminary mechanistic studies evidence that a catalyst turnover of RhIII-RhIV/V- RhII/III- RhIII might be involved within this catalytic C-H transformation.

Abstract:Herein, Rh(III)-catalyzed and weak carbonyl coordination-directed C4-selective C-H allylation and prenylation of indoles have been reported in the first time. By employing 5-methylene-1,3-dioxan-2-ones, 4 vinyl-1,3-dioxolan-2-ones and 2-methyl-2,3-butadiene as novel and scalable cross-coupling partners, respectively, these divergent synthesis protocols proceed smoothly under redox-neutral reaction conditions, delivering various allylated and prenylated indoles with excellent C4-position regioselectivity in moderate to excellent yields. This transformation exhibits high functional-groups compatibility and broad substrate scope. Gram-scale experiment and mechanistic studies were also accomplished.

Abstract:A palladium-catalyzed domino Heck cyclization/carbonylative Hiyama-Denmark cross-coupling reaction between alkene-tethered aryl iodides and silylcarboxylic acids is presented. This reaction proceeds well without toxic carbon monoxide (CO) gas and has good functional group tolerance, providing an alternative access to carbonyl-containing oxindoles. In this transformation, silylcarboxylic acids play a dual role as a CO source and a nucleophile.

Abstract:New functionalizations of indoles via palladium-catalyzed reaction of indoles and 2-acetoxymethyl-substituted electron-deficient alkenes are reported. It was found that for N-protected indoles the reaction proceeded smoothly in the presence of 5 mol % of Pd(acac)2 and 10 mol % of PPh3 at 80 degrees C in HOAc, while for N-unprotected indoles, the reaction was carried out by using 5 mol % of Pd(dba)2 or 2.5 mol % of Pd2(dba)3.CHCl3 with 10 mol % of 2,2'-bipyridine as the catalyst in toluene. This strategy allows the selective installation of electron-deficient olefin functionality at the 3-position of indoles, which might be difficult to obtain by other methods and can be further elaborated.

Abstract:A practical process for the synthesis of 2-unsubstituted indoles and azaindoles has been developed by the palladium-catalyzed direct annulation of electron-poor o-chloro/bromoanilines and o-chloroaminopyridines with aldehydes. Coupled with the previous results of Jia and Zhu, this allows rapid access to a variety of 2-unsubstituted indoles and azaindoles starting from simple and easily accessible precursors.

Abstract:A palladium-catalyzed iodine-assisted carbonylation reaction of indoles with readily available ClCF(2)CO(2)Na and alcohols has been developed. This protocol provides a practical and efficient approach to highly regioselective indole-3-carboxylates via a preiodination strategy of indoles. Different from classic carbonylation using toxic and difficult-to-handle carbon monoxide, this operationally simple and scalable reaction employed difluorocarbene as the carbonyl surrogate.

Abstract:Five-carbon (C5) structural units are the fundamental building blocks of many natural products. An unprecedented palladium-catalyzed three-component dehydrogenative cascade coupling of indoles, 2-methylbut-2-ene, and carboxylic acids has been developed. The approach enables the straightforward introduction of a C3'-bonded five-carbon structural unit with a tertiary alcohol quaternary carbon center into indoles. The protocol employs 2-methylbut-2-ene as the C5 source and is featured by a broad substrate scope, atom and step economies, and high chemo- and regioselectivies.
