Abstract:To develop a feasible and efficient nanocarrier for potential clinical application, a series of photo and redox dual responsive reversibly cross-linked micelles have been developed for the targeted anticancer drug delivery. The nanocarrier can be cross-linked efficiently via a clean, efficient, and controllable coumarin photodimerization within the nanocarrier, which simplify the formulation process and quality control prior clinical use and improve the in vivo stability for tumor targeting. At the same time, cross-linking of nanocarrier could be cleaved via the responsiveness of the built-in disulfide cross-linkage to the redox tumor micro-environment for on-demand drug release. Coumarin and disulfide bond was introduced into a linear-dendritic copolymer (named as telodendrimer) precisely via peptide chemistry. The engineered nanocarrier possesses good drug loading capacity and stability, and exhibits a safer profile as well as similar anticancer effects compared with free drug in cell culture. The in vivo and ex vivo small animal imaging revealed the preferred tumor accumulation and the prolonged tumor residency of the payload delivered by the cross-linked micelles compared to the non-cross-linked micelles and free drug surrogate because of the increased stability.
Nanoemulsion-Templated Shell-Crosslinked Nanocapsules As Drug Delivery Systems

Abstract:A novel biocompatible shell-crosslinked nanocapsule system was developed based on nanoemulsion templates stabilized by a class of food proteins. The nanoemulsion templates were prepared using a combination of mechanical mixing and high-pressure homogenization, while the nanocapsule shell formed simultaneously through calcium ions-induced crosslinking of the food proteins. These core–shell structured nanocapsules with a particle size of about 200nm showed high drug-loading capacity and well improved stability in comparison with their nanoemulsion counterpart. The nanocapsule suspension can be freeze-fried directly; and the solidified nanocapsules can be well reconstituted in water, retaining their original particle size. It is concluded that the nanoemulsion-templated core–shell structured nanocapsules can be used as novel drug delivery systems with high loading capacity for poorly water-soluble drugs as well as well improved long-term and storage stability. Furthermore, the presence of surface food proteins introduces carboxyl and amine moieties, which enables the nanocapsules to anchor ligands, suggesting its potential application in targeting drug delivery, bioimaging and therapeutics.
Preparation of pH and redox dual-sensitive core crosslinked micelles for overcoming drug resistance of DOX

Abstract:In this paper, we first synthesized an amphiphilic block copolymer poly(ethylene glycol)-poly(2,4,6-tri-methoxybenzylidene- pentaerythritol carbonate-co-5-methyl-5-propargyl-1,3-dioxan-2-one), i.e., PEG-P (TMBPEC-co-MPMC), with pendant reactive alkynyl groups as well as pH-sensitive acetal groups. Next, core crosslinked (CCL) micelles were prepared by the introduction of 1,6-diazidohexane and bis(azido-ethyl) disulfide into micelles via azide-alkyne click chemistry, which were denoted as CCL/CC and CCL/SS, respectively. The CCL micelles had superior stability and drug loading efficiency to the uncrosslinked (UCL) micelles. In comparison with free DOX, drug-loaded CCL micelles exhibited lower cell viability in MCF-7/ADR cells due to their "stealth" endocytosis effect that might be beneficial for overcoming delivery barriers of drug resistance. More interestingly, as compared with CCL/CC micelles, CCL/SS micelles were found to further enhance cytotoxicity in MCF-7/ADR cells because of their better on-demand drug release capability of pH and redox dual-sensitive CCL/SS micelles. These results suggest that the self-assembled pH and redox dual-sensitive CCL/SS micelles have promising applications to overcome multi-drug resistance in tumor treatments.

Abstract:Polymeric prodrugs have attracted great interest in the field of anticancer drug delivery owing to their integrated advantages of prodrugs and nanoparticles. However, the ambiguous chemical composition of polymeric prodrugs is still a major obstacle to their clinical applications. In this study, an amphiphilic poly(ethylene glycol) derivative with multiple thiols was first synthesized on a large scale through the melt co-polycondensation of oligo(ethylene glycol), 1,4-butanediol and mercaptosuccinic acid. Then an adamantane modified doxorubicin (DOX) with a pH-sensitive hydrazone bond and a beta-cyclodextrin derivative with multiple acryloyls were precisely synthesized. Finally, novel supramolecular polymeric prodrug (SPP) micelles with a precise drug molecular structure and drug loading content were facilely prepared using the above three components via simultaneous self-assembly, a thiol-ene "click" reaction and a host-guest supramolecular interaction in water. The resultant pH-sensitive SPP micelles with a stable core-crosslinked structure and strong drug "bonding" show long blood circulation time, effective cellular uptake and fast DOX release in tumor cells, which significantly improved the antitumor efficacy and security compared with free DOX. This work established a promising drug delivery platform for cancer chemotherapy.

Abstract:In this work, we report the fabrication and functional demonstration of a kind of dually responsive nanoparticles (NPs) as a potential drug delivery vector. The pH value, corresponding to the acidic microenvironment at the tumor site, and mannitol, to the extracellular trigger agent, were employed as the dually responsive factors. The function of dual responses was achieved by breaking the dynamic covalent bonds between phenylboronic acid (PBA) groups and diols at low pH value (pH 5.0) and/or under the administration of mannitol, which triggered the decomposition of the complex NPs and the concomitant release of anticancer drug of doxorubicin (DOX) loaded inside the NPs. The NPs were composed of modified chitosan (PQCS) with quaternary ammonium and PBA groups on the side chains, heparin (Hep), and poly(vinyl alcohol) (PVA), in which quaternary ammonium groups offer the positive charge for the cell-internalization of NPs, PBA groups serve for the formation of dynamic bonds in responding to pH change and mannitol addition, PVA furnishes the NPs with diol groups for the interaction with PBA groups and the formation of dynamic NPS, and Hep plays the roles of reducing the cytotoxicity of highly positively-charged chitosan and forming of complex NPs for DOX up-loading. A three-step fabrication process of drug-loaded NPs was described, and the characterization results were comprehensively demonstrated. The sustained drug release from the drug-loaded NPs displayed obvious pH and mannitol dependence. More specifically, the cumulative DOX release was increased more than 1.5-fold at pH 5.0 with 20 mg mL−1 mannitol. Furthermore, the nanoparticles were manifested with effective antitumor efficient and apparently enhanced cytotoxicity in response to the acidic pH value and/or mannitol.

Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.

Abstract:Ideal drug delivery systems are required to be stable in circulation, while instable at targeted sites to induce rapid drug release. To address this dilemma, novel light cross-linkable and pH de-cross-linkable micelles were developed as drug nanocarriers. Poly(ethylene glycol)-poly-(hydrazone umbelliferone)aspartamide (mPEG-OMC) with light responsive coumarin moieties and pH responsive hydrazone groups in the side chains was synthesized. The polymers can self-assemble into micelles, which can be conveniently stabilized via UV-light induced cross-linking based on the dimerization of coumarin moieties. While under slight acidic conditions, the cross-linked micelles can also be de-cross-linked by the cleavage of hydrozone bonds in the cross-linking segments, accelerating the release of encapsulated drugs. Thus the benefits of pH and light stimuli-responsiveness can be combined and their disadvantages are avoided. The doxorubicin-loaded micelles showed high mortality towards HepG2 cells. We have demonstrated the UV light cross-linkable micelles with pH-triggered de-cross-linking properties as a new approach for designing smart drug delivery systems.

Abstract:A core cross-linked polymeric micellar cisplatin(IV) conjugate prodrug is prepared by attaching the cisplatin(IV) to mPEG-b-PLL biodegradable copolymers to form micellar nanoparticles that can disintegrate to release the active anticancer agent cisplatin(II) in a mild reducing environment. Moreover, in vitro studies show that this cisplatin(IV) conjugate prodrug displays enhanced cytotoxicity against HepG2 cancer cells compared with cisplatin(II). Further studies demonstrate that the high cellular uptake and platinum-DNA adduct of this cisplatin(IV) conjugate prodrug can induce more cancer-cell apoptosis than cisplatin(II), which is responsible for its enhanced anticancer activity.

Abstract:Multifunctional core-shell nanoparticles are widely used for biomedical and catalytic applications. In this work, bilayers of chitosan (Cs) and phosphorylated polyvinyl alcohol (PPVA) were sequentially deposited on 3-Aminopropyltri-ethoxysilane-modified SiO2 nanoparticles via layer-by-layer electrostatic self-assembly. The good spherical shape and size distribution were observed by DLS and transmission electron microscope analysis. 7-Hydroxycoumarin (7-HC) and rhodamine B (RhB) as model drugs were loaded in the core and shell of the nanoparticles separately. Confocal laser scanning microscopy shows the core-shell structure of HC-SiO2(PPVA/Cs)(n)-RhB nanoparticles and the embedded location of 7-HC and RhB. The pH-sensitive release investigation of RhB indicates that the release profiles of RhB from HC-SiO2(PPVA/Cs) 3PPVA-RhB core-shell nanoparticles are totally different at pH values of 2.0, 7.4, and 9.2. These results predict that the multifunctional nanoparticle SiO2(PPVA/Cs)(n) has a great potential for drug delivery.
Fabrication of Core Crosslinked Polymeric Micelles as Nanocarriers for Doxorubicin Delivery: Self-Assembly, In Situ Diselenide Metathesis and Redox-Responsive Drug Release

Abstract:Polymeric micelles (PMs) have been used to improve the poor aqueous solubility, slow absorption and non-selective biodistribution of chemotherapeutic agents (CAs), albeit, they suffer from disassembly and premature release of payloads in the bloodstream. To alleviate the thermodynamic instability of PMs, different core crosslinking approaches were employed. Herein, we synthesized the poly(ethylene oxide)-b-poly((2-aminoethyl)diselanyl)ethyl l-aspartamide)-b-polycaprolactone (mPEG-P(LA-DSeDEA)-PCL) copolymer which self-assembled into monodispersed nanoscale, 156.57 ± 4.42 nm, core crosslinked micelles (CCMs) through visible light-induced diselenide metathesis reaction between the pendant selenocystamine moieties. The CCMs demonstrated desirable doxorubicin (DOX)-loading content (7.31%) and encapsulation efficiency (42.73%). Both blank and DOX-loaded CCMs (DOX@CCMs) established appreciable colloidal stability in the presence of bovine serum albumin (BSA). The DOX@CCMs showed redox-responsive drug releasing behavior when treated with 5 and 10 mM reduced glutathione (GSH) and 0.1% H2O2. Unlike the DOX-loaded non-crosslinked micelles (DOX@NCMs) which exhibited initial burst release, DOX@CCMs demonstrated a sustained release profile in vitro where 71.7% of the encapsulated DOX was released within 72 h. In addition, the in vitro fluorescent microscope images and flow cytometry analysis confirmed the efficient cellular internalization of DOX@CCMs. The in vitro cytotoxicity test on HaCaT, MDCK, and HeLa cell lines reiterated the cytocompatibility (≥82% cell viability) of the mPEG-P(LA-DSeDEA)-PCL copolymer and DOX@CCMs selectively inhibit the viabilities of 48.85% of HeLa cells as compared to 15.75% of HaCaT and 7.85% of MDCK cells at a maximum dose of 10 μg/mL. Overall, all these appealing attributes make CCMs desirable as nanocarriers for the delivery and controlled release of DOX in tumor cells.

Abstract:A novel redox-sensitvie core cross-linked micelles based on disulfide-linked PEG-polypeptide hybrid polymer were prepared and demonstrated for anticancer drug delivery, where N,N-bis(acrylate) cystamine (BAC) served as cross-linker, allyl-terminated poly(gamma-benzyl-L-glutamate) (PBLG) and polyethylene glycol (PEG) methyl ether methacrylate acted as comonomers. The molecular structure and characteristics of the cross-linked micelle and the precursor were confirmed by H-1 NMR and FT-IR. The cross-linked micelles could be easily degraded into individual linear short chains (M-n = 1800) in the presence of 20 mM glutathione (GSH) by the cleavage of the disulfide linkages from the cross-linker BAC. Doxorubicin (DOX) was selected as a model anticancer drug and encapsulated into the micelles with a decent drug loading content of 21.6 wt%. Compared to the burst release of free DOX in first 6 hours, the in vitro release studies revealed that the micelles exhibited a sustained and high cumulative drug release in GSH (up to 86%) within 24 h, rather than the relatively low release rate of 62% in PBS (pH 7.4). Cell cytoxicity experiments showed that the obtained micelles exhibited nontoxic, and the drug-loaded micelles exhibited high anti-cancer efficacy. All the results showed that the designed cross-linked PEG-polypeptide hybrid micelles may be a promising vehicle for anticancer drug delivery with stimuli-triggered drug release behavior in reducing environment.
Dual-Responsive Polyprodrug Nanoparticles with Cascade-Enhanced Magnetic Resonance Signals for Deep-Penetration Drug Release in Tumor Therapy

Abstract:Smart transformable nanocarriers are promising to treat deep-seated diseases but require adaptable diagnostic/imaging potency to reflect the morphology change and therapeutic feedback, yet their design and synthesis remains challenging. Herein, stimuli-responsive polyprodrug nanoparticles (SPNs) are formulated from the co-assembly of negatively charged corona and positively charged polyprodrug cores, exhibiting high loading content of camptothecin (CPT, ∼28.6 wt %) tethered via disulfide linkages in the core. SPNs are sequentially sensitive to tumor acidic condition and elevated reductive milieu in the cytosol for deep-penetration drug delivery. Upon accumulation at acidic tumor sites, SPNs dissociate to release smaller positively charged polyprodrug nanoparticles, which efficiently enter deep-seated tumor cells to trigger high-dosage parent CPT release in the reductive cytosolic milieu. Meanwhile, the polyprodrug cores of SPNs labeled with DTPA(Gd), a magnetic resonance imaging contrast agent, can trace the cascade degradation and biodistribution of SPNs as well as the resulting intracellular CPT release. The longitudinal relaxivity of SPNs increases stepwise in the above two processes. Reduction-Responsive Drug Delivery Based on Mesoporous Silica Nanoparticle Core with Crosslinked Poly(Acrylic Acid) Shell

Abstract:A novel reduction-responsive drug delivery system was successfully constructed with mesoporous silica nanoparticle (MSN) core as a drug carrier and poly(acrylic acid) (PAA) shell crosslinked by disulfide linkages as a drug release switcher. To keep the pore structure of MSN intact, PAA was covalently attached to the exterior surface of MSN before removing structure-template via radical polymerization. After removing structure-template and loading doxorubicin (DOX), the PAA shell was crosslinked by cystamine dihydrochloride through amidation reaction. The loading content and the entrapment efficiency of DOX could reach up to 40.2% and 80.4%, respectively. Because that the dissociation of disulfide linkage is reduction-responsive, the release behavior of DOX could be controlled by varying the concentration of reductant, and the release rate was 49.4% after 24h with the existence of 2 mM glutathione (simulated environment of cancer cells), about three times higher than that of without glutathione (corresponding to normal human cells), which was only 16.9%. The in vitro cell assays demonstrated that the disulfide linkages crosslinked MSN-PAA (MSN-PAA-crosslinked) was highly biocompatible and suitable to use as drug carrier, and the DOX loaded MSN-PAA-crosslinked showed remarkable cytotoxicity to HeLa cells (human cancer cells), and relatively lower cytotoxicity to 293 cells (human normal cells). These results imply that the MSN-PAA-crosslinked is a promising platform to construct reduction-responsive controlled drug delivery system for cancer therapy.
Supramolecular Modular Approach toward Conveniently Constructing and Multifunctioning a pH/Redox Dual-Responsive Drug Delivery Nanoplatform for Improved Cancer Chemotherapy

Abstract:Because heterogeneity affects many functional aspects of a tumor, a way to overcome it is to arm nanosized drug delivery systems (nanoDDS) with diverse functions required to shatter heterogeneity. However, it remains technically challenging to fabricate a nanocarrier possessing all required functions. Here, we propose a modular strategy for generating a supramolecular, multifunctional, and stimuli-responsive nanoDDS through docking a parental core nanoDDS with various daughter function-prebuilt modules. Doxorubicin (DOX)-loaded mesoporous silica nanoparticles (MSNs) as the parental nanocore are wrapped by poly(β-cyclodextrin) (PCD) as a gatekeeper through host-guest interactions between cyclodextrin units and pyridine groups of pyridine-disulfide bonds that confers pH/redox dual responsiveness, thus constructing stimuli-responsive nanoDDS (DOX@PRMSNs). Meanwhile, PCD's free cyclodextrin is tightly caged by adamantyl (Ad)-terminated daughter modules via host-guest interactions, achieving convenient multifunctionalization of this nanoDDS. DOX@PRMSNs rapidly released DOX in lysosomal pH/redox microenvironment, potently killing drug-resistant cancer cells. Further, three different types of Ad-terminated daughter modules, including two targeting ligands (Ad-PEG-FA and Ad-PEG-LA), a cationic polymer (Ad-PEI), and a fluorescence agent (Ad-FITC), are utilized to functionalize PRMSNs via cyclodextrin-Ad self-assembly, endowing the nanoDDS with cell-targeting capability, gene codelivery property, and imaging function. Thus, this work develops a supramolecular modular self-assembly approach for constructing and multifunctionalizing stimuli-responsive "smart" nanoDDSs.
Mechanistic Study of the Covalent Loading of Paclitaxel Via Disulfide Linkers for Controlled Drug Release

Abstract:A novel controlled drug-delivery system (CDDS) based on fluorescent mesoporous silica nanoparticles (FMSN) covalently linked with paclitaxel (PTX) via a disulfide linker was designed and characterized. A PTX prodrug based on a disulfide linker was synthesized, and its drug-delivery mechanism was determined through HPLC characterization. Utilizing the carboxyl group of the prodrug, PTX was covalently conjugated to the surface of amino-functionalized FMSN, with a disulfide linker as a spacer to bridge between PTX and FMSN, and the loading content of PTX reached as high as 13% by weight. The most important feature of this nanoscale CDDS is that the PTX prodrug modules conjugated with FMSN can be activated to its cytotoxic form inside the tumor cells upon internalization and in situ drug release. To prove the efficacy of this CDDS, glutathione-mediated intracellular drug delivery was investigated against the HeLa cell line, and the results indicated that our CDDS showed higher cellular proliferation inhibition against glutathione monoester pretreated cells than against untreated cells and the cytotoxicity increased with increasing intracellular glutathione concentration. The result indicates that CDDS can release PTX molecules to kill cancer cells and the release behavior is GSH-dependent. Furthermore, the in vitro evaluation revealed that the FMSN-PTX conjugate could be effectively taken up by HeLa cells. All of these results suggest that this redox-responsive CDDS is potentially useful as a drug-delivery system that can reduce the toxic and side effects of anticancer drugs.
Reversible Core-Crosslinked Nanocarriers with Ph-Modulated Targeting and Redox-Controlled Drug Release for Overcoming Drug Resistance

Abstract:Herein, a pH and redox dual-sensitive core-crosslinked targeting nanocarrier was prepared and used for co-delivery of doxorubicin (DOX) and tariquidar (TQR). The nanocarrier not only had excellent stability but also prevented the leakage of the drug in the normal physiological environment efficiently. Meanwhile, the targeting function of nanocarriers could also be suppressed in the normal physiological environment, protecting nanocarriers from being captured by RAW264.7 cells. Under mild acidic conditions, the targeting function was regained, leading to an effective tumor cell uptake of the nanocarrier. Furthermore, reduction-responsive drug release would occur in the cytoplasm due to the collapse of the reduction-sensitive crosslinked structure in the nanocarrier. By means of ligand-receptor mediated endocytosis and TQR-mediated glycoprotein (P-gp) inhibition, the IC50 value of DOX to MCF-7/ADR cells reduced from more than 100 μg mL-1 to 8.55 μg mL-1, exhibiting great potential in overcoming drug resistance.
ROS-responsive core crosslinked micelles by combination of enzymatic polymerization and thiol-ene click chemistry for anticancer drug delivery

Abstract:The reactive oxygen species (ROS) responsive amphiphilic poly(ethylene glycol)- b -poly(thioketal) (mPEG- b -PTK) copolymer was synthesized by enzyme-catalyzed polymerization for the first time. Core crosslinked (CCL) micelles were prepared via thiol-ene click reaction between the double bond of the side chain of mPEG- b -PTK and the thiol group of pentaerythritol tetrakis (3-mercaptopropionate) as a crosslinker under UV light. The success of crosslinking was verified by NMR and IR. CCL micelles showed better stability in stability against dilution and storage stability experiments compared with the uncrosslinked (UCL) micelles. The ROS-responsiveness of CCL micelles was characterized by DLS, TEM, NMR, AFM and SEM. In vitro release studies, CCL micelles encapsulating doxorubicin (DOX) showed a faster release rate of nearly 100% within 32 h in PBS containing 5 mM H 2 O 2 compared to the control without H 2 O 2 . Cytotoxicity test demonstrated good biocompatibility of CCL micelles and low toxicity with cell viability above 82% and 86% in HeLa cells and HUVEC cells, respectively. And CCL DOX-loaded micelles exhibited great anticancer activity with IC 50 value about 4.8 mg/L for Hela cells. Finally, flow cytometry and confocal laser scanning microscope (CLSM) showed that DOX-loaded micelles had excellent internalization efficiency in HeLa cells. The obtained ROS-responsive CCL micelles held promise for anticancer drug delivery.

Abstract:The extracellular stability versus intracellular drug release and the long blood stream circulation versus enhanced cell uptake are two dilemmas for micellar drug delivery systems. To resolve the above two problems, a novel core cross-linked polyurethane micelles with redox sensitive and pH-responsive surface charge switchable properties were prepared and investigated as anti-cancer drug carriers. Firstly, amphiphilic multi-blocked polyurethane with pendant carboxyl groups and disulfides in the hard segment (PU-SS-COOH) was synthesized. Afterwards, 1,6-diaminohexane was connected onto some of the pendant carboxyl groups via dicyclohexylcarbodiimide (DCC) condensation, which resulted in polyurethane with pendant amine and carboxyl groups (PU-SS-COOH-NH2). Then the PU-SS-COOH-NH2 was assembled into micelles and the hexamethylene-1,6-diisocyanate (HDI) was added into the micelle solution. The hydrophobic diisocyanate penetrated into the hydrophobic core of PU micelles and reacted with amine groups, which resulted in core crosslinked PU micelles (PU-CCL). PTX and doxorubicin hydrochloride (DOX) were chosen as model hydrophobic drugs to evaluate the loading and redox-triggered release of the PU micelles. The drug release from the cross-linked polyurethane micelles (PU-CCL) demonstrated that the PU-CCL has superior particle stability compared with the uncross-linked polyurethane micelles in a phosphate buffer saline (PBS) solution without reducing agents, whereas the drug release rate is markedly accelerated with the addition of a reducing agent, glutathione (GSH). Notably, PU micelles with both amine and carboxyl groups, i.e. PU-SS-COOH-NH2 and PU-CCL micelles, showed pH-responsive charge switchable properties. The in vitro cytotoxicity and cell uptake of the PTX (or DOX used to illustrate the cell uptake) loaded micelles was also assessed in HepG2 cells. It was found that the redox-sensitive polyurethane micelles can rapidly enter tumor cells. In vitro cytotoxicity studies demonstrated that the DOX-loaded PU-CCL display lower cytotoxicity in vitro compared to either free DOX or DOX-loaded uncrosslinked PU micelles. The cross-linked reduction responsive biodegradable micelles, having superior extracellular stability and providing rapid intracellular drug release, may hold great potential as a bio- triggered drug delivery system for cancer therapy.
