Abstract:<p>Self-assembled nanostructures are highly promising for controlled delivery of therapeutic and diagnostic agents. However, for conventional drug delivery nanosystems, there exist some intrinsic limitations such as low drug-loading contents, premature and burst release, nanocarrier matrix-associated toxicity and immunogenicity, and poor shelf stability. To address these issues, the covalent integration of active drug molecules into prodrug and polyprodrug amphiphiles and fabrication of self-delivery nanomedicines via controlled molecular self-assembly have emerged as a new paradigm. Moreover, it is crucial to achieve on-demand and selective activation of prodrugs and polyprodrugs in a spatiotemporally controlled manner, thus considerably reducing the occurrence of systemic toxicities. In this context, dynamic variations of reductive/oxidative (redox) milieu across normal and pathological tissues, cells, and cytoplasmic compartments provide accessible biochemical stimuli for triggered release of intact drugs. In this review, we highlight recent progresses on emerging applications of redox-activatable nanostructures self-assembled from prodrug and polyprodrug amphiphiles.</p>

Dually Responsive Nanoparticles for Drug Delivery Based on Quaternized Chitosan

Abstract:In this work, we report the fabrication and functional demonstration of a kind of dually responsive nanoparticles (NPs) as a potential drug delivery vector. The pH value, corresponding to the acidic microenvironment at the tumor site, and mannitol, to the extracellular trigger agent, were employed as the dually responsive factors. The function of dual responses was achieved by breaking the dynamic covalent bonds between phenylboronic acid (PBA) groups and diols at low pH value (pH 5.0) and/or under the administration of mannitol, which triggered the decomposition of the complex NPs and the concomitant release of anticancer drug of doxorubicin (DOX) loaded inside the NPs. The NPs were composed of modified chitosan (PQCS) with quaternary ammonium and PBA groups on the side chains, heparin (Hep), and poly(vinyl alcohol) (PVA), in which quaternary ammonium groups offer the positive charge for the cell-internalization of NPs, PBA groups serve for the formation of dynamic bonds in responding to pH change and mannitol addition, PVA furnishes the NPs with diol groups for the interaction with PBA groups and the formation of dynamic NPS, and Hep plays the roles of reducing the cytotoxicity of highly positively-charged chitosan and forming of complex NPs for DOX up-loading. A three-step fabrication process of drug-loaded NPs was described, and the characterization results were comprehensively demonstrated. The sustained drug release from the drug-loaded NPs displayed obvious pH and mannitol dependence. More specifically, the cumulative DOX release was increased more than 1.5-fold at pH 5.0 with 20 mg mL−1 mannitol. Furthermore, the nanoparticles were manifested with effective antitumor efficient and apparently enhanced cytotoxicity in response to the acidic pH value and/or mannitol.

Abstract:Reduction-responsive biodegradable polymeric micelles based on functional 2-methylene-1,3-dioxepane (MDO) copolymers are developed and investigated for triggered doxorubicin (DOX) release. The MDO-based copolymers P(MDO-co-PEGMA-co-PDSMA) are synthesized via the simple one-step radical ring-opening copolymerization of MDO, poly(ethylene glycol) methyl ether methacrylate (PEGMA), and pyridyldisulfide ethylmethacrylate (PDSMA). The copolymers can self-assemble to form micelles in aqueous solution. DOX, a model anticancer drug, is loaded into the micelles with the drug loading content (DLC) of 11.3%. The micelles can be disassembled under a reductive environment (10 x 10(-3)m glutathione), which results in a triggered drug release behavior. The glutathione-mediated intracellular drug release of DOX-loaded micelles is investigated against A549 cells. Confocal laser scanning microscopy (CLSM) results demonstrated that DOX-loaded micelles exhibits faster drug release in glutathione monoester (GSH-OEt)-pretreated A549 cells, compared with untreated and buthionine sulfoximine (BSO)-pretreated A549 cells. Based on the facile synthetic strategy, the reduction-sensitive biodegradable micelles with triggered intracellular drug release are promising for anticancer drug delivery.
Biomimic Ph/reduction Dual-Sensitive Reversibly Cross-Linked Hyaluronic Acid Prodrug Micelles for Targeted Intracellular Drug Delivery

Abstract:Biomimic hyaluronic acid (HA) prodrug micelles with pH and reduction responsiveness were prepared by chemical graft of phosphorylcholine and doxorubicin (DOX) to the backbone of HA. DOX was conjugated to HA by pH-sensitive hydrazone bonds which could be hydrolyzed under acidic conditions and thus resulted in controlled release of DOX. Besides, the HA prodrug micelles were cross-linked via reduction-responsive disulfide bonds to improve the stability of the micelles. The HA prodrug can form remarkable core shell micelles with a quite narrow size distribution which was confirmed by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The in vitro drug release studies showed that there was a dramatic release under endosome pH (pH = 5.0) and reductive environment (10 mM of dithiothreitol) than that at normal physiological environment (pH = 7.4). Greater uptake of DOX from HA prodrug micelles was observed in the CD44 receptor highly expressed MDA-MB-231 cell line, compared to the results from the CD44-negative cell line, NIH3T3, which was demonstrated by the flow cytometry and fluorescence microscopy. The glutathione (GSH)-responsive behaviors were studied as well. The micelles showed higher cellular proliferation inhibition against glutathione monoester (GSH-OEt) pretreated MDA-MB-231 cells than that of the non-pretreated and buthionine sulfoximine (BSO) pretreated ones. These results suggested that such pH/reduction dual-responsive prodrug micelles may enhance the selective inhibition to cancer cells which exhibit higher levels of GSH concentration and provide favorable platforms for cancer therapy. Reduction/photo Dual-Responsive Polymeric Prodrug Nanoparticles for Programmed Sirna and Doxorubicin Delivery.

Abstract:Dual and multi-stimuli responsive polymeric nanoparticles that respond to two or more signals can further improve drug release performance compared with nanoparticles that respond to a single stimulus. However, usage of such nanoparticles to deliver siRNA and chemotherapeutic drugs in a sequential manner are currently very rare; meanwhile, this technology is vital to optimize the efficacy of chemotherapy towards cancer cells with multidrug resistance. By loading o-nitrobenzyl ester derivative caged DOX (DOC) into the inner poly(lactic-co-glycolic acid) (PLGA) core and adsorbing siRNA of P-gp protein onto the cationic polymeric shell derived from a disulfide-containing alkyl modified polyethylenimine (C16-S-S-PEI), here, a reduction/photo dual responsive device (RPDRD) is successfully designed for programmed P-gp siRNA and doxorubicin delivery. The dual-stimuli design of the RPDRD allows tumor microenvironment-specific and rapid release of P-gp siRNA triggered by the enrichment of reducing agent glutathione (GSH, up to 10 mM) for reversal of drug resistance by initially suppressing P-gp protein expression in MCF/ADR cells and then selectively triggering drug release by external light for chemotherapy afterwards. The sequential release behavior of P-gp siRNA and DOX can be demonstrated both in vitro and in vivo, thus enhancing the intracellular drug retention and optimizing the chemotherapy efficacy of DOX by silencing P-gp; this strategy may have extensive application prospects in MDR cancer treatment in future.
Reduction-Responsive Drug Delivery Based on Mesoporous Silica Nanoparticle Core with Crosslinked Poly(Acrylic Acid) Shell

Abstract:A novel reduction-responsive drug delivery system was successfully constructed with mesoporous silica nanoparticle (MSN) core as a drug carrier and poly(acrylic acid) (PAA) shell crosslinked by disulfide linkages as a drug release switcher. To keep the pore structure of MSN intact, PAA was covalently attached to the exterior surface of MSN before removing structure-template via radical polymerization. After removing structure-template and loading doxorubicin (DOX), the PAA shell was crosslinked by cystamine dihydrochloride through amidation reaction. The loading content and the entrapment efficiency of DOX could reach up to 40.2% and 80.4%, respectively. Because that the dissociation of disulfide linkage is reduction-responsive, the release behavior of DOX could be controlled by varying the concentration of reductant, and the release rate was 49.4% after 24h with the existence of 2 mM glutathione (simulated environment of cancer cells), about three times higher than that of without glutathione (corresponding to normal human cells), which was only 16.9%. The in vitro cell assays demonstrated that the disulfide linkages crosslinked MSN-PAA (MSN-PAA-crosslinked) was highly biocompatible and suitable to use as drug carrier, and the DOX loaded MSN-PAA-crosslinked showed remarkable cytotoxicity to HeLa cells (human cancer cells), and relatively lower cytotoxicity to 293 cells (human normal cells). These results imply that the MSN-PAA-crosslinked is a promising platform to construct reduction-responsive controlled drug delivery system for cancer therapy.
Dual-Responsive Controlled Drug Delivery Based On Ionically Assembled Nanoparticles

Abstract:Ionically assembled nanoparticles (INPs) have been formed from poly(ionic liquid-co-N-isopropylacrylamide) with deoxycholic acid through electrostatic interaction. The structure and properties of the INPs were investigated by using H-1 NMR, Fourier transform infrared (FTIR), transmission electron microscopy (TEM), dynamic light scattering (DLS), and so on. Due to pH-responsive deoxycholic acid (pK(a) = 6.2) and thermo responsive N-isopropylacrylamide included in the ionic complex, the INPs exhibit highly pH and thermal dual-responsive properties. The potential practical applications as drug delivery carriers were demonstrated using doxorubicin (DOX) as a model drug. With a lower pH (pH 5.2) and higher temperature (above 37 degrees C), structural collapse of the INPs occurred as well as release of DOX owing to protonated DA departure from the INPs and a lower LCST (lower critical solution temperature) at the pathological conditions. The result shows that 80% of DOX molecules were released from INPs within 48 h at pH 5.2, 43 degrees C, but only 30% of the drug was released within 48 h at 37 degrees C and pH 7.4. Moreover, drug-loaded INPs exhibit an inhibitory effect on cell growth.

Abstract:INTRODUCTION: Biodegradable micellar nanoparticles are one of the most promising platforms for targeted cancer chemotherapy. In particular, reduction-sensitive micellar nanoparticles that actively deliver and release drugs into cancer cells have recently appeared as an advanced drug release system. The in vitro and in vivo studies have pointed out that reduction-sensitive micellar drug formulations afford significantly enhanced antitumor activity as compared with 'traditional' reduction-insensitive counterparts.AREAS COVERED: This review provides an overview of recent advances in reduction-sensitive degradable micellar nanoparticles for triggered anticancer drug delivery. The design and preparation of reduction-sensitive micellar nanoparticles, redox-triggered in vitro and intracellular drug release, in vitro as well as in vivo antitumor performances of drug-loaded nanoparticles are discussed.EXPERT OPINION: Reduction-sensitive degradable micellar nanoparticles with unique advantages of high stability against hydrolytic degradation, fast response to intracellular reducing environment and triggered drug release right in the cytosol and cell nucleus have demonstrated a tremendous potential in targeted and active intracellular delivery of potent and poorly soluble anticancer drugs. These intracellular-activatable fast drug releasing nanosystems may eventually be developed as 'drug bomb' for effective treatment of various tumors including multidrug-resistant tumors.

Abstract:The applications of bioresponsive materials are limited by the low levels of physiological triggers and spatiotemporal barriers in vivo. To address these issues, a light‐controlled “Trojan horse” strategy is proposed by encapsulating a photo‐caged reducing agent within reduction‐responsive polymer vesicles. The polymersomes act as an enzyme‐inspired nanoreactor for efficient photo‐synthesis of dithiothreitol in situ, which attacks the disulfide bonds in polymer backbones and disintegrates the vehicles from the inside in a nanoconfined space. The assembly‐catalyzed photoreduction reaction overcomes the temporal and spatial barriers for hyper‐responsivity and enables ultrafast drug release even at 0.014 mm of dithiothreitol residues, a concentration 715 times lower than that required to cleave disulfide bonds. In addition, in both vitro and vivo, the equipment of upconverting nanoparticles and photothermal agents creates a near‐infrared light‐activated and self‐heating nanoreactor, which allows for efficient intracellular drug delivery and excellent photo‐chemo‐immunotherapy of tumors. This work presents a new approach to designing smart materials and a promising platform for on‐demand drug delivery applications. Abstract:Polymeric micelle systems for drug delivery, monitor and chemotherapy have gained significant attention, and reductive polymeric micelle systems have become particularly attractive due to their controlled release behavior without additional assistance. However, there are challenges in accurately controlling drug and probe release from the nanoparticles and determining the loading content of drug and probe. To address these issues, we have developed a reduction-responsive Pt(IV) prodrug-based polymeric delivery system that can be dynamically monitored using aggregation-induced emission luminogens (AIE) based bioprobes. These polymeric micelle can self-assemble into nanoparticles and release both bio-active Pt(II) drug and bio-probe upon reduction activation. TPE molecules released in the inner endo/lysosomal microenvironment aggregate and fluoresce upon irradiation, thus allowing real-time tracking of drug biodistribution without additional contrast agents. Advantages of this system include position-specific chemical bond cleavage, control of platinum content, and monitoring of drug reduction and biodistribution.

Abstract:Significance: The therapeutic effects of current micellar and vesicular drug formulations are restricted by slow and inefficient drug release at the pathological site. The development of smart polymeric nanocarriers that release drugs upon arriving at the target site has received a tremendous amount of attention for cancer therapy. Recent advances: Taking advantage of a high reducing potential in the tumor tissues and in particular inside the tumor cells, various reduction-sensitive polymeric micelles and vesicles have been designed and explored for triggered anticancer drug release. These reduction-responsive nanosystems have demonstrated several unique features, such as good stability under physiological conditions, fast response to intracellular reducing environment, triggering drug release right in the cytosol and cell nucleus, and significantly improved antitumor activity, compared to traditional reduction-insensitive counterparts. Critical issues: Although reduction-sensitive micelles and polymersomes have accomplished rapid intracellular drug release and enhanced in vitro antitumor effect, their fate inside the cells including the mechanism, site, and rate of reduction reaction remains unclear. Moreover, the systemic fate and performance of reduction-sensitive polymeric drug formulations have to be investigated. Future directions: Biophysical studies should be carried out to gain insight into the degradation and drug release behaviors of reduction-responsive nanocarriers inside the tumor cells. Furthermore, novel ligand-decorated reduction-sensitive nanoparticulate drug formulations should be designed and explored for targeted cancer therapy in vivo.
Reduction-Triggered Transformation of Disulfide-Containing Micelles at Chemically Tunable Rates.

Abstract:A dilemma exists between the circulation stability and cargo release/mass diffusion at desired sites when designing delivery nanocarriers and in vivo nanoreactors. Reported herein are disulfide-crosslinked (DCL) micelles exhibiting reduction-triggered switching of crosslinking modules and synchronized hydrophobic-to-hydrophilic transition. Tumor cell targeted DCL micelles undergo cytoplasmic milieu triggered disulfide cleavage and self-immolative decaging reactions at chemically adjustable rates, generating primary amine moieties. Extensive amidation reactions with neighboring ester moieties then occur because of the high local concentration and suppression of the apparent amine pKa value within the hydrophobic cores, thus leading to the transformation of crosslinking modules and formation of tracelessly crosslinked (TCL) micelles, with hydrophilic cores, inside live cells. We further integrate this design principle with theranostic nanocarriers for selective intracellular drug transport guided by enhanced magnetic resonance (MR) imaging performance.
Preparation of Ph- and Reductive-Responsive Prodrug Nanoparticles Via Polymerization-Induced Self-Assembly

Abstract:pH- and reductive-responsive prodrug nanoparticles are constructed via a highly efficient strategy, polymerization-induced selfassembly (PISA). First, reversible addition-fragmentation chain transfer (RAFT) polymerization of 2-(diisopropylamino) ethyl methacrylate (DIPEMA) and camptothecin prodrug monomer (CPTM) using biocompatible poly(N-(2-hydroxypropyl) methacrylamide) (PHPMA-CPDB) as the macro RAFT agent is carried out, forming prodrug diblock copolymer PHPMA-P (DIPEMA-co-CPTM). Then, simultaneous fulfillment of polymerization, self-assembly, and drug encapsulation are achieved via RAFT dispersion polymerization of benzyl methacrylate (BzMA) using the PHPMA-P(DIPEMA-co-CPTM) as the macro RAFT agent. The prodrug nanoparticles have three layers, the biocompatible shell (PHPMA), the drug-conjugated middle layer (P(DIPEMA-co-CPTM)) and the PBzMA core, and relatively high concentration (250 mg/g). The prodrug nanoparticles can respond to two stimuli (reductive and acidic conditions). Due to reductive microenvironment of cytosol, the cleavage of the conjugated camptothecin (CPT) within the prodrug nanoparticles could be effectively triggered. pH-Induced hydrophobic/ hydrophilic transition of the PDIPEMA chains results in faster diffusion of GSH into the CPTM units, thus accelerated release of CPT is observed in mild acidic and reductive conditions. Cell viability assays show that the prodrug nanoparticles exhibit well performance of intracellular drug delivery and good anticancer activity.

Abstract:The development of intelligent drug delivery system that can efficiently deliver drug to targeted site and release the cargo in a controlled manner is of great significance for cancer chemotherapy. In the present study, dual redox/pH-sensitive amphiphilic copolymer, cholesterol modified poly (β-amino esters)-grafted disulfide poly (ethylene glycol) methyl ether (PAE(-ss-mPEG)-g-Chol), is successfully synthesized via Michael-type step polymerization using disulfide linkage-containing PEG segment. PEGylated lipid is introduced to fabricate the doxorubicin (DOX)-loaded hybrid polymer-lipid nanoparticles (NPs) with (DOX-HDPLNPs) or without (DOX-HPLNPs) disulfides to receive stable NPs with enhanced loading capacity and improved cellular uptake ability as well as serum stability. The HDPLNPs show not only pH-sensitivity because of poly(β-amino esters) (PAE) residues in main chain but also the redox-responsivity due to the disulfides on the side chains with PEG segments. As expected, the DOX-HDPLNPs exhibited the extended blood circulation time and higher serum stability in normal physiological environment, but once deposited in the tumor microenvironment (e.g. low pH value and high GSH concentration), they underwent a dramatic size transition and charge reversal. These features not only can facilitate the NPs accumulation through the enhanced permeability and retention (EPR) effect and endocytosis by tumor cells but also realize the drug controlled release and more efficient tumor inhibition. The in vitro and in vivo results demonstrate that the DOX-HDPLNPs possess the highest therapeutic efficacy with high cellular uptake and negligible cytotoxicity compared to the controls and free drug. Therefore, the HDPLNPs have good potential as smart delivery platforms for hydrophobic anticancer drug delivery due to high uptake by tumor cells and redox/pH-triggered drug release.
Reduction Responsive Self-Assembled Nanoparticles Based on Disulfide-Linked Drug-Drug Conjugate with High Drug Loading and Antitumor Efficacy.

Abstract:Most anticancer drugs are poorly soluble and nonspecific, which restricts their clinical application. Drug conjugates, as a prodrug strategy, provide the possibility to overcome these shortcomings, especially combined with nanotechnology. Drug conjugate nanoparticles possess the advantages of high drug loading capacity and passive tumor targeting ability. Here, we prepared doxorubicin drug-drug conjugate nanoparticles (DOX-SS-DOX NPs) based on disulfide-linked doxorubicin drug-drug conjugate (DOX-SS-DOX). Dynamic light scattering (DLS) and transmission electron microscope (TEM) characterization indicated that DOX-SS-DOX NPs were spherical with a uniform size distribution around 89 nm. DLS and in vitro release experiment revealed that DOX-SS-DOX NPs possessed reduction responsive activity. In vitro cellular uptake studies reflected that DOX-SS-DOX NPs could increase the uptake level substantially compared with DOX liposomes. Endocytosis mechanism assay demonstrated that DOX-SS-DOX NPs internalized into cells through a clathrin-mediated endocytosis pathway in an energy-dependent manner. In this manner, the amidase in lysosomes could break the amide bond to release free DOX, which would be helpful to antitumor activity. The in vitro cytotoxicity of DOX-SS-DOX NPs was a bit weaker than that of DOX liposomes, which might be the result of the slow cleavage of the disulfide bridge; but the antitumor efficacy of DOX-SS-DOX NPs evaluated in MCF-7 bearing mice was demonstrated to be higher than that of DOX liposomes. This might be because of the long lasting effect resulting from the slow cleavage of the disulfide bond. In summary, DOX-SS-DOX NPs, prepared nearly totally with drug, provide a good strategy for cancer therapy.

Abstract:Herein, we report a co-delivery system based on poly(ethylene glycol)-b-poly(5-methyl-5-propargyl-1,3-dioxan-2-one) (PEG-b-PMPMC) for cancer treatment. A reduction-sensitive PTX prodrug (azido-SS-PTX) was grafted onto PEG-b-PMPMC by azide-alkyne click reaction to give a reduction-sensitive polymeric prodrug, PEG-b-PMPMC-g-PTX, with a well-defined structure and narrow molecular weight distribution. Interestingly, the polymeric prodrug could self-assemble into micelles or polymersomes in aqueous solution based on the particular graft ratio of PTX on the polycarbonate backbone, which endowed it with a possibility to encapsulate a hydrophobic drug (e.g., doxorubicin, DOX) or a hydrophilic drug (e.g., doxorubicin hydrochloride, DOX center dot HCl) for combination chemotherapy. To promote the stability and on-demand drug release, the self-assemblies of PEG-b-PMPMC-g-PTX were further crosslinked by a reduction-sensitive crosslinker to prepare reversibly crosslinked nanoparticles. The drug release from the carriers was suppressed under normal physiological conditions, whereas markedly accelerated under 10 mM dithiothreitol (DTT) conditions. The dual drug-loaded nanoparticles exhibited significant growth-inhibition for HeLa cells and drug-resistant MCF-7/ADR cells compared with the single drug-loaded nanoparticles, especially for drug resistant tumor cells. More importantly, DOX center dot HCl-loaded polymersomes of the polymeric prodrug exhibited a synergistic effect of cell-growth inhibition on HeLa cells. These results indicated that the co-delivery system based on reduction-sensitive polymeric prodrugs would be a promising technology in cancer therapy to overcome multidrug resistance (MDR).
Design and Synthesis of Redox and Oxidative Dual Responsive Block Copolymer Micelles for Intracellular Drug Delivery

Abstract:The need for smart materials in the area of biotechnology has accelerated the development of stimuli-responsive copolymer micelles. Here, we reported a novel dual-stimuli responsive block copolymer PEG-DMTK-SS-PLA with both dimethyl thioketal (DMTK) and disulfide linkage incorporated into the backbone, capable of triggering fast drug release properties upon both oxidative (H2O2) and reductive (GSH) environment. The CMC values of these copolymer micelles ranging from 0.051 to 0.087 mg mL(-1), the average diameters are from 34 nm to 55 nm and 65 nm to 198 nm for blank and DOX-loaded micelles respectively. MTT assay conducted in NIH 3T3 cells showed the low cytotoxicity of these micelles even when the concentration reached up to 500 mu g/mL. Considering tumor microenvironment's diverse in kinds of tumor cells, fluorescence microscopy, flow cytometry and MTT activity analysis were conducted in several cancer cells (e.g., cervix, lung, gastric, and colon cancer cells), and further confirmed that the dual responsive PEG-DMTK-SS-PLA micelles were degraded much faster than that of non-responsive PEG-PIA and single responsive PEG-SS-PLA and PEG-DMTK-PLA micelles. Abstract:A redox-responsive amphiphilic polymeric prodrug was synthesized in a facile way by polycondensation of oligo(ethylene glycol) with dicarboxylic acids including malic acid and 3,3'-dithiodipropionic acid, followed by esterification with ibuprofen, which was used as a model drug. Because of its amphiphilic nature and relatively high molecular weight, this polymeric prodrug can form stable micelles in aqueous media with a low critical micellar concentration (CMC). Free ibuprofen molecules can be steadily incorporated into the core of these micelles with a surprisingly high loading content (38.9 wt%), owing to hydrophobic interaction and pi-pi stacking with the ibuprofen moieties in the copolymer. The in vitro release results indicate that there was a relatively slow and sustained release of the conjugated ibuprofen moieties, while encapsulated ibuprofen molecules showed a rapid release. Furthermore, for both the conjugated ibuprofen and the encapsulated ibuprofen there was an accelerated release in the presence of 10 mM DL-dithiothreitol due to cleavage of the disulfide bonds, which lead to disassociation of the micelles. Notably, this prodrug was revealed to have excellent cell compatibilities via a cell counting kit-8 (CCK-8) assay. Confocal laser scanning microscope observations indicated that the micelles based on the polymeric prodrug can be taken up quickly by cells and present a redox-responsive drug release in cytoplasm. This kind of polymeric nanocarrier with a high drug loading content, low CMC, excellent biocompatibility and rapid response to a reductive environment may have tremendous scope in the area of controlled drug delivery.
