Abstract:3-Hydroxybenzylhydrazine dihydrochloride was synthesized from 3-hydroxybenzaldehyde and methoxycarbonylhydrazine via condensation, hydrogenation, decarboxylation and salt formation in a total yield of 77.1%. The yield of condensation was 94.6% and of hydrogenation was 99.8% by using Pd/C (5%) which can be reused 3 times at the optimal quantity of 3% (W/W). The yield of salt formation was 81.7%. This route is easy to industrialize for its simple operation, mild reacting condition and low cost.

Abstract:8-[4-[4-(2-Pyrimidinyl)-1-piperaziny]butyl]-8-azaspiro [4,5] decane-7,9-dione hydrochloride (buspirone hydrochloride) was obtained in one pot with a 51.8% overall yield. The key intermediate, 1-(2-pyrimidinyl) piperazine, was synthesized through chlorination and cyclization condensation reaction with diethanolamine as initial material. This modified protocol has the notable advantages of mild reaction condition, convenient operation, and high overall yield.

Abstract:Quetiapine hemifumarate wassynthesized in 63.4% overall yield from 2-aminodiphenyl sulfide and triphosgene to give 2-isocyanato diphenyl sulfide, which subjected to cyclization in polyphosphoric acid, chlorination and condensation with piperazine to afford 11-piperazin-1-yl-dibenzo[b,f][1,4]thiazepine followed by N4-alkylation of piperazine and then salification with fumaric acid.

Abstract:A novel synthesis process of 5H-dibenz[b,f]azepine was studied.It is an important intermediate of 5H-dibenz[b,f]azepine-5-carboxamide,used in genetic engineering,materials science and so on.With 10,11-dihydro-5H-dibenz[b,f]azepine as raw material,5-chloroformyl-5H-dibenz[b,f]azepine was prepared by reflux with triphosgene in toluene,brominated with bromine in chlorobenzene,and dehydrobrominated at 130℃,finally,the title compound was obtained by removing chloroformyl group with NaOH in isopropanol with 80.4% overall yield.The optimal reaction conditions and yields (temperature,time,molar yield) were as follows: acyl chlorination,110℃,10 h,93.0%; bromination,dehydrobromination,100℃,2 h,130℃,5 h,88.0%; removal of chloroformyl group,40℃,3 h,98.2%. The melting point of the title compound was the same as literature reported,and its structure was confirmed by 1H NMR.The optimal technology condition to remove chloroformyl group as protection group and its application of protection for some amine compounds were also studied.

Abstract:Octopamine (4-(2-amino-1-hydroxy-ethyl)-phenol hydrochloride) acts as a neurotransmitter, neurohormone,or neuromodulator in invertebrate nerve tissue, and is an ideal target for development of highly active, highly selective,and biorationally designed pesticides. In addition, octopamine has the ability to strengthen body metabolism and curediseases such as neurasthenia. Phenol and aminoacetonitrile hydrochloride undergo nitrile acylation in the presence ofanhydrous aluminum tricloride, followed by hydrolysis to yield a carbonylated product, and then a Raney nickelcatalyzed hydrogenation reaction to give the final product with an overall yield of 65%.

Abstract:2-Chloromethyl-3,5-dimethyl-4-nitropyridine hydrochloride, an important intermediate of omeprazole which is a very effective drug for the treatment of gastric ulcer, was prepared by N-oxidation with phosphotungstic acid as catalyst without solvent、nitration、rearrangement with acetic anhydride、hydrolysis in acidic condition and chlorination. The overall yield was over 70% and the molecular structure of the title compound was confirmed by ()~1H NMR.

Abstract:2, 4-Dimethoxybenzaldehyde was prepared using 1, 3-dimethoxybenzene as raw material through Vilsmeier formylation with DMF directly. The compound was characterized by IR and NMR as a qualified product. Under the optimal conditions the product obtained from the orthogonal experiments could be given in 94.2% yield with purity of 98.64%, after purification the purity of final compound could be reached 99.6%.

Abstract:Purpose To prepare 1-methyl-3-ethyl-3-(3-hydroxyphenyl)-hexahydro-1H-azepin hydrochloride (Ⅰ) from m-methoxyacetophenone. Methods 1-methyl-3-ethyl-3-(3-hydroxyphenyl)-hexahydro-1H-azepin hydrochloride(Ⅰ) was synthesized by eight steps with m-methoxyacetophenone as starting material.The eight steps were Willgerodt rearrangement,formation of acyl chloride,acylation,cyclization,C-ethylation,Wolff-Kishner-Huang reduction and reduction by Lithium aluminium hydride and salt formation. Results 1-methyl-3-ethyl-3-(3-hydroxyphenyl)-hexahydro-1H-azepin Hydrochloride(Ⅰ) was prepared in eight steps from cheap material and reagent on normal condition. Conclusions This method is applicable to industrial production.

Abstract:3-Nitropyrazole was prepared via two steps, nitration and rearrangement, using pyrazole as a raw material. Pyrazole was nitrated in a mixed acid of nitric acid and sulfuric acid below 15 ℃for 3.5 hours with stirring, giving N-nitropyrazole with a yield of 86.6%, which was then refluxed in n-octanol at 185-190℃, giving 3-nitropyrazole with ayield of 87.8%. This nitration reaction was innovated by using sulfuric acid as the proton donor instead of acetic acid and acetic anhydride, and solvent n-octanol could be recycled without further treatment.

Abstract:5,6-Diaminobenzimidazolone-2 is obtained in an overall yield of 62.1% by the nitration of benzimidazolone-2 and followed with hydrogenation.Single-factor experiments were conducted to optimize the reaction condition.5,6-Dinitrobenzimidazolone-2 was synthesized by the nitration of benzimidazolone-2 under 0 ℃,for 6 h,n(nitric acid)/n(benzimidazolone-2)=2.2 in 69% yield.5,6-Diaminobenzimidazolone-2 was prepared in the absolute alcohol in 90% yield by 9 h of the Raney-Ni catalytic hydrogenation of 5,6-dinitrobenzimidazolone-2 under the condition of 100 ℃、2.5 MPa、m(catalyst)/m(5,6-dinitrobenzimidazolone-2)=0.09.MS、HPLC、IR、1HNMR、13CNMR and elementary analysis are properly used for the characterization of the structures of the product.

Abstract:Loratadine was synthesized from 2-cyano-3-methylpyridine via Ritter reaction, alkylation and cyanidation to give 2-cyano-3-[2-(3-chlorophenyl)ethyl]piperidine, which subjected to cyclization in presence of PPA/P2O5 and then coupling with N-ethoxycarbonyl-4-piperidone. Abstract:OBJECTIVE To study the synthetic method of 2-aminoxy-1- aminoethane dihydrochloride. METHOD The objective compound was synthesized by steps of ring-opening, condensation, hydrazinolysis and hydrolysis, starting from phthalic anhydride.RESULTS The chemical structure of synthetic product was confirmed by IR and 1HNMR, the total yield was amount to 39.4%. CONCLUSION This synthetic route is practical.

Abstract:3-Amino-1,3,4,5-tetrahydro-2H-1-benzazepin-2-one in total yield of 90% was synthesized by aminization reaction of 3-bromo-1,3,4,5-tetrahydro-2H-1-benzazepin-2-one with benzylamine,and then through hydrogenation reaction catalyzed by 5%Pd/C.The optimum hydrogenation reaction conditions at 100 ℃/2.5 MPa for 10 h were as follows: absolute alcohol was solvent,amount of 5%Pd/C was 10%.The yield of 3 was 95.5% and the purity was 99.7% under the optimum reaction conditions.The 5%Pd/C catalyst could be reused for 7 times without change in yield(about 95%).

Abstract:The titled compound (1), which is an important intermediate of metoprolol, was synthesized in overall 29% yield from cheap p-nitrotoluene and paraformaldehyde via condensation, alkylation, reduction, diazotisation and hydrolysis. The key step (p-nitrotoluene condensation with paraformaldehyde) was investigated in detail, including effects of catalysts, solvent, and reactants mole ratio on yield. Thus, 2 mol p-nitrotoluenes reacted with 1 mol paraformaldehyde in DMSO, catalyzed by potassium phenol to give p-nitrophenyl ethanol (4) with the conversion of p-nitrotoluene of 31% and selectivity of 72%. The remaining p-nitrotoluene could be recycled.
