Abstract:The optical scanning holography (OSH) technique can capture all the three-dimensional volume information of an object in a hologram via a single raster scan. The digital hologram can then be processed to reconstruct individual sectional images of the object. In this paper, we present a scheme to reconstruct sectional images in OSH with enhanced depth resolution, where a spatial light modulator (SLM) is adopted as a configurable point pupil. By switching the SLM between two states, different Fresnel zone plates (FZPs) are generated based on the same optical system. With extra information provided by different FZPs, a depth resolution at 0.7 μm can be achieved.

Abstract:Holographic techniques enable precise laser manipulation, but suffer from two considerable limitations: speckle and deterioration of axial distribution. Here, we propose a cylindrical quadratic phase (CQP) method with temporal focusing (TF) to generate speckle-free holographic illumination with high axial resolution. TF-CQP utilizes a superposed cylindrical phase as the initial guess to iteratively optimize phase hologram, realizing speckle-free holographic reconstruction on the target focal plane and eliminating secondary focus on the defocused planes. TF-CQP further disperses defocused beams symmetrically by a blazed grating, placed conjugate to the focal plane, which enhances axial confinement. Simulation and experimental results show that TF-CQP reconstructs speckle-free illumination with arbitrary shapes and <10 µm axial resolution. Compared to TF-GS (Gerchberg-Saxton algorithm), widely used in holographic optogenetics, TF-CQP shows increased uniformity of 200% and improved modulation efficiency of 32.33% for parallel holographic illumination, as well as a 10% increment in axial resolution.

Abstract:Digital holographic microscopy has been widely used for the imaging of micro-objects and biological samples. Lensless in-line digital holographic microscopy is capable of wide field-of-view imaging. However the spatial resolution of the reconstructed images is limited by the pixel size of the detector. The relative position shift between the sample and the detector can effectively improve the resolution in the traditional sub-pixel shifting method, but it requires a high precision of translation stage. To overcome this problem, we propose a method based on the point source scanning to realize sub-pixel shifting. High precision sub-pixel shifting is achieved easily by using the geometric between point source and detector. Through moving the point source, multiple holograms with sub-pixel shifts are captured. These holograms are merged together to obtained a high resolution hologram by a synthesizing algorithm. Then, the high resolution reconstructed image of the object can be obtained by the angular spectrum algorithm. The feasibility of the proposed method is demonstrated by simulation and experiments. A USAF resolution test target was used as the object. Compared with the traditional digital holography, a higher resolution reconstructed image is obtained by our method. The proposed method has the advantages of simple recording setup and lower precision requirement of the translation stage. It can achieve the wide field-of-view and high resolution imaging.

Abstract:Employing a single-pixel digital holographic recording technique called optical scanning holography (OSH), we accomplish the formidable task of sectioning with edge extraction in three-dimensional (3D) optical incoherent imaging. OSH is a special variant of generalized two-pupil heterodyning image processing, where one of the pupils used is a delta function with the other being a uniform function. In this study, we investigate the use of an annular pupil and a random-phase pupil for edge extraction during sectioning of a 3-D object. Novel simulation results indicate excellent edge extraction of a multi-section object with good sectioning capability, i.e., with each focused edge-extracted section out-of-focused haze has been eliminated. Abstract:In optical scanning holography, one pupil produces a spherical wave and another produces a plane wave. They interfere with each other and result in a fringe pattern for scanning a three-dimensional object. The resolution of the hologram reconstruction is affected by the point spread function (PSF) of the optical system. In this paper, we modulate the PSF by a spiral phase plate, which significantly enhances the lateral and depth resolution. We explain the theory for such resolution enhancement and show simulation results to verify the efficacy of the approach.

Abstract:Theoretical analysis shows that, to improve the resolution and the range of the field of view of the reconstructed image in digital lensless Fourier transform holography, an effective solution is to increase the area and the pixel number of the recorded digital hologram. A new approach based on the synthetic aperture technique and use of linear CCD scanning is presented to obtain digital holographic images with high resolution and a wide field of view. By using a synthetic aperture technique and linear CCD scanning, we obtained digital lensless Fourier transform holograms with a large area of 3.5 cm x 3.5 cm (5000 x 5000 pixels). The numerical reconstruction of a 4 mm object at a distance of 14 cm by use of a Rayleigh-Sommerfeld integral shows that a theoretically minimum resolvable distance of 2.57 microm can be achieved at a wavelength of 632.8 nm. The experimental results are consistent with the theoretical analysis.

Abstract:We propose an optical scanning holography system with enhanced axial resolution using two detections at different depths. By scanning the object twice, we can obtain two different sets of Fresnel zone plates to sample the same object, which in turn provides more information for the sectional image reconstruction process. We develop the computation algorithm that makes use of such information, solving a constrained optimization problem using the conjugate gradient method. Abstract:Using conventional refraction-based optical lens, it is challenging to achieve both high-resolution imaging and long-working-distance condition. To increase the numerical aperture of a lens, the working distance should be compensated, and vice versa. Here we propose and demonstrate a new concept in optical microscopy that can achieve both high-resolution imaging and long-working-distance conditions by utilising a scattering layer instead of refractive optics. When light passes through a scattering layer, it creates a unique interference pattern. To retrieve the complex amplitude image from the interference pattern without introducing a reference beam, we utilised a speckle-correlation scattering matrix method. This property enables holographic microscopy without any lens or external reference beam. Importantly, the proposed method allows high-resolution imaging with a long working distance beyond what a conventional objective lens can achieve. As an experimental verification, we imaged various microscopic samples and compared their performance with off-axis digital holographic microscopy.

Abstract:Digital holography is the product of the optical holography, computer technology and photoelectric detection technology, and has the advantage of high-speed, real-time, full field of view, non-contact and quantitative phase contrast imaging. However, the numerical aperture of the hologram is limited due to the smaller sensitive area of the photoelectric sensor and the larger pixel size, and it is uneasy to meet the practical requirement on the imaging resolution. An approach is presented to achieve the high-resolution digital holographic imaging based on a spatial light modulator(SLM). An amplitude spatial light modulator is placed between the object and the CCD in the lensless Fourier transform digital holographic imaging system. The distribution of a diffraction grating is loaded into the SLM. In this way, more light including the high-frequency content, diffracted from the object, can be collected by the CCD. The standard resolution target is used as the object. The reconstructed image is obtained by the Fresnel diffraction propagation algorithm, which exhibits three diffraction orders. The results show that the resolution is improved from 62.5 μm to 31.3 μm.

Abstract:The aspheric light emitted from a pinhole restrains the reconstruction quality of a digital in-line hologram. Herein, the Fresnel-diffracted spot from the first step converging spherical wave diffracted at a rough circular aperture is collimated and expanded to generate an even plane wave, which is converged again by an objective lens and matching a minimum aperture while the central spot is varying from light to dark. We observed that the collected background hologram is filled with a round spot with high contrast as an ideal spherical wave. The resolution board and biology experimental results demonstrated a distinctively reconstructed image without any image processing in a single exposure. The adjustable field of view and magnification, single exposure, and noncontact make it suitable for an online microscope.

Abstract:Pixelation, blur and additional noise of imaging system limit the resolution of final images acquired. Many pixel super-resolution algorithms have been applied to enhance the resolution of imaging system by merging a sequence of low-resolution holograms with different type of imaging system, for example by shifting illumination source or using wavelength scanning. Most of these pixel super-resolution imaging systems can only be implemented to single-layer sample. For multi-layer imaging system and volumetric imaging scenarios, the relative displacement of various sample at different layers will disturb each other. Herein, we report a portable, cost-effective, lensless wide-filed digital in-line holographic microscopy imaging system based on in-line hologram segmentation and pixel super-resolution algorithm, which can separate target sample from the background and improve the resolution of the sample. We demonstrated the effectiveness of our system with numerical simulation and experiment with volumetric sample. In numerical simulation, we applied a very simple two-layer sample model that samples in two layers have various moving speed and directions and also did the volumetric imaging experiment with cuvette containing algae floating in. In our system, the sensor captured a sequence of low-resolution diffraction patterns. The target sample mix with background disturbance, which will invalidate the direct pixel super-resolution technique. We applied segmentation algorithm to the reconstructed images from holograms, separating target sample from background and generating a sequence of sub-images containing only target sample with same resolution and numerical aperture as original holograms. Finally the enhanced resolution reconstructed image of target sample was obtained with pixel super-resolution algorithm, which can go beyond pixel limitation and get sub-pixel perspective microscopy. This imaging system has the advantages of wide-field, portable and lensless.

Abstract:Based on the point spread function of holographic system, the lateral resolution of digital holographic imaging system without any pre-magnification is studied. The expression of resolution limitation of holographic imaging system is thus presented. We investigate the possibilities to improve the lateral resolution. The simple experimental setup with an off-axis arrangement is built. By using a U.S. Air Force (USAF) test target as microscopic object, the recorded holograms are reconstructed digitally based on the principle of Fresnel diffraction. The lateral resolution of 2.76 mu m without any pre-magnification is demonstrated experimentally, which matches the theoretical prediction well.

Abstract:We present a snapshot multi-frame parallel holographic microscopy system through a reconfigurable optical comb source, which consists of a digital micromirror device (DMD) based spectrum filter system and a spectroscopic Michelson interferometric system. The proposed system allows arbitrarily tuning comb spacing and comb number, and the capturing of multi-frame images without overlap in one exposure. As a result, high-quality spectral holograms can be obtained with less acquisition time. The performance of the system is detailed in the experiment and 45-wavelengths holographic imaging for perovskite micro-platelets is conducted, which proves the system has the ability to realize high-performance four-dimensional (4D) imaging.

Abstract:We propose to use sparsely sampled line scans with a sparsity-based reconstruction method to obtain images in a wide field of view (WFOV) multifocal scanning microscope. In the WFOV microscope, we used a holographically generated irregular focus grid to scan the sample in one dimension and then reconstructed the sample image from line scans by measuring the transmission of the foci through the sample during scanning. The line scans were randomly spaced with average spacing larger than the Nyquist sampling requirement, and the image was recovered with sparsity-based reconstruction techniques. With this scheme, the acquisition data can be significantly reduced and the restriction for equally spaced foci positions can be removed, indicating simpler experimental requirement. We built a prototype system and demonstrated the effectiveness of the reconstruction by recovering microscopic images of a U.S. Air Force target and an onion skin cell microscope slide with 40, 60, and 80% missing data with respect to the Nyquist sampling requirement.

Abstract:Digital micro holography offers an in-situ, non-contact and three-dimensional way to explore the microscopic world. However, as it is difficult to focalize the whole object in one single reconstructed image, the application of digital micro holography to cases with a large longitudinal object volume is limited by the microscopes depth of field. By extending the depth of field in reconstructed micro holograms in the wavelet domain, this paper fully takes advantage of numerical reconstruction algorithms to solve this problem. First, a recorded hologram is rebuilt using the wavelet transform approach by setting up an appropriate longitudinal interval to obtain a series of reconstructed hologram planes. Then each plane is decomposed with wavelet into its sub-images of both high and low frequencies. Furthermore, the local variance of the maximum intensity gradients of the high- and low-frequency coefficients is calculated and utilized as the focus criterion. Finally, the image planes are fused into a single one with the depth of field extended to a large extent. The feasibility and robustness of this reconstruction procedure for both continuum and particle fields are investigated. One of the demonstrations is made in an experiment of a tilted continuum:carbon fiber. It is different from most of the previous applications where the interrogated is the particles and where the area involved is parallel to the CCD. The carbon fiber gets successfully reconstructed in three dimensions, and the measurement errors of its diameter are presented together with the reconstruction distances. Another is an experiment of a dispersed particle field:micro transparent particles are generated by an ultrasonic atomizer, for which the reconstruction procedure achieves an extended depth of field. In addition, a numerical model based on generalized Lorenz-Mie theory is used to simulate the holograms of both opaque and transparent particles of 1-15 m in diameter. Variations of the longitudinal location errors with the Fraunhofer number are analyzed, and comparisons are made between the results of opaque and transparent particles. Both the experimental and simulation outcomes show that this reconstruction procedure is a reliable one to acquire an extended-depth-of-field hologram for both the continuum and the dispersed particle fields, and then to accurately measure the objects.

Abstract:For lensless diffraction imaging, it is a challenging dilemma to achieve a large field of view (FOV) and high resolution with a small amount of data at the same time. Ptychography can reconstruct the high-resolution image and illumination light simultaneously. But the illumination is limited to a small size by a probe in typical ptychography. For large samples, it takes much time to collect abundant patterns and has strict requirements for the computing power of computers. Another widely applied method, multi-height measurement, can realize a wide FOV with several holograms. But, the recovered image is easily destroyed by the background noise. In this Letter, a lensless diffraction imaging method by three-dimensional scanning is proposed. All positions of the object are different in three directions instead of scanning schemes only on a plane or along the optic axis, so more diversity of diffraction information is obtained. We apply the illumination without the limit of a confined aperture, which means that the imaging FOV of a pattern is equal to the size of the utilized image sensor. In comparison with the multi-height method, our method can separate the illumination background noise from the retrieved object. Consequently, the proposed method realized high resolution and contrast, large FOV, and the removal of background noise simultaneously. Experimental validations and comparisons with other methods are presented.

Abstract:Conventional multi-height microscopy techniques introduce different object-to-detector distances to obtain multiple measurements for phase retrieval. However, surpassing the diffraction limit imposed by the numerical aperture (NA) of the objective lens remains a challenging task. Here, we report a novel structured modulation multi-height microscopy technique for quantitative high-resolution imaging. In our platform, a thin diffuser is placed in between the sample and the objective lens. By translating the diffuser to different axial positions, a sequence of modulated intensity images is captured for reconstruction. The otherwise inaccessible high-resolution object information can thus be encoded into the optical system for detection. In the construction process, we report a ptychographic phase retrieval algorithm to recover the existing wavefront of the complex object. We validate our approach using a resolution target, a phase target, and various biological samples. We demonstrate a ∼4-fold resolution gain over the diffraction limit. We also demonstrate our approach to achieve a 6.5 mm by 4.3 mm field of view and a half-pitch resolution of 1.2 µm. The reported methodology provides a portable, turnkey solution for quantitative high-resolution imaging with potential applications in disease diagnosis, sample screening, and other fields.
