Abstract:To provide a computational efficient forward model with moderate accuracy for rapid 3D optical tomography in small volumes, radiative transport in the delta-P1 approximation combined with the approximation of the reciprocity was examined. Perturbations of optical signals caused by absorption and fluorescence heterogeneities submerged in a resin-based liquid phantom with background parameters close to rat brain tissues were measured using a recently constructed laminar optical tomography system. These measured perturbations were used to examine the theoretically calculated fluence perturbations based on the delta-P1 approximation and the reciprocity approximation. Results show that the errors between the predicted and measured data are acceptable, especially for fluorescence perturbations.

Abstract:Radiative transport in the delta-P1 and delta-P3 approximations with an extrapolated boundary condition combined with the reciprocity was demonstrated providing accurate estimations for optical perturbations by comparing with experimental data in a small volume of a liquid tissue phantom. For an absorbing target submerged in a homogeneous medium with an albedo of 0.75, both approximations estimated the optical perturbation with acceptable accuracy when the depth of the target is larger than 1.5 mm. For a fluorescent target, the two approximations estimated the perturbations with high accuracy for both shallow and deep regions. Therefore, these two approximations combined with the reciprocity were proposed for rapid tomographic image reconstruction in a small volume or superficial tissues.

Abstract:The diffusion approximation to the transport equation is commonly used in biomedical optical diagnostic techniques, but constrains its applicability to highly scattering system. The generalized diffusion approximation was developed by Venugopalan can be used to quantify optical properties of turbid media using small source detector separations and allow the measurement of media with highly absorption. Unfortunately, the simulated result from this theory was larger than the real value because delta-Eddington phase function contained too much forward scattering. Here a new independent control parameter is introduced to delta-Eddington phase function so as to modify the generalized diffusion approximation presented. The solution is presented in the stationary case for infinite media with a collimated source of finite size exhibiting spherical symmetry. The solution is compared to results given by the conventional diffusion theory, the generalized diffusion approximation as well as to the Monte-Carlo simulation in steady state diffusion equation for slab boundary condition. The simulation results show that the modified generalized diffusion approximation with an appropriate control parameter is more closed to Monte-Carlo simulation. The modified generalized formulation of diffusion theory presented here may enable the quantitative application of present optical diagnostic techniques to turbid systems which are more highly absorbing and allow these systems to be probed using smaller source-detector separations.

Abstract:Accuracy of measurement data has direct bearing on the quality of optical tomography, and in some special tissues, the region of interested is not at the center. Therefore, a new optimization strategy of fiber positions is proposed for the special case. In the new strategy, sources and detectors are located nearest to the surface of region of interested. On the other hand, the transport model of photon also has important effect on the reconstructed results, and diffuse equation is often used in many researches. However, diffusion model has great disadvantages for many cases, so the equation of radiative transfer is imported in this paper. In the reconstruction process, a gradient tree algorithm is put forward to implement the gradient computation of objective function with respect to optical parameters. Experimental results show that the quality of reconstructed images and convergence of reconstruction process are greatly improved based on the equation of radiative transfer with the optimized strategy, and the computation complexity is also significantly reduced at the same time.
A Hybrid P 1 -DP 0 Diffusion Theory for Optical Imaging

Abstract:In optical imaging, although the standard P1 diffusion theory is widely used, its angular flux at boundary is discontinuous, and this model is not incapable of exactly modeling light transport in biological tissue with partially-reflective boundary. In this work, we present a hybrid P1-DP0 (P1 spherical harmonics-double DP0 spherical harmonics) diffusion theory in 3D environment, which effectively interpolates between the P1 and DP0 approximation by a space-dependent weight factor α(r) that controls the local angular approximation. Comparing to the P1 model, the solutions of our model are consistently accurate over a broad range of optical properties. Moreover, with the same reduced scattering and absorption properties, the hybrid model for high anisotropic scattering which is the common case for mammal tissue is more accurate than the low one. Finally, this theory is validated by Monte Carlo simulations.

Abstract:In this paper we experimentally investigate the effect of non-scattering and low scattering regions in turbid media on diffusion-approximation-based image reconstruction in diffuse optical tomography. Using tomographic dc measurements, we demonstrate that both absorption and scattering images of non-scattering and low scattering heterogeneities embedded in a highly scattering background can be simultaneously reconstructed with our diffusion-approximation-based algorithm. The results show that the reconstructions of non-scattering and low scattering targets are quantitatively accurate in terms of the target size, location, and scattering coefficient value, while the absorption coefficient value of the recovered targets is unphysically underestimated under the conditions we examined.

Abstract:Based on the hybrid frequency-domain (FD) and time-domain (TD) radiative transfer equation (RTE), a novel reconstruction strategy is developed to retrieve the optical parameter fields in the participating medium. The discrete ordinate method is employed to solve the forward FD and TD RTE to simulate the radiative transfer process. The sequential quadratic programming is utilized to solve the inverse problem for obtaining the optical parameters combined with an adjoint method. The Otsu algorithm is applied to the FD reconstructed results to distinguish the target region and the background region. To overcome the ill-posedness of the inverse problem, the generalized Gaussian Markov random field model is employed as the regularization term in the objective function. Various numerical experiments are studied and all results demonstrate that the proposed hybrid reconstruction scheme is suitable for efficient and accurate optical tomography by combining the FD and TD radiative transfer models. Meanwhile, the crosstalk of the ill-posed inverse problem can be alleviated effectively.

Abstract:An efficient and robust method based on the complex-variable-differentiation method (CVDM) is proposed to reconstruct the distribution of optical parameters in two-dimensional participating media. An upwind-difference discrete-ordinate formulation of the time-domain radiative transfer equation is well established and used as forward model. The regularization term using generalized Gaussian Markov random field model is added in the objective function to overcome the ill-posed nature of the radiative inverse problem. The multi-start conjugate gradient method was utilized to accelerate the convergence speed of the inverse procedure. To obtain an accurate result and avoid the cumbersome formula of adjoint differentiation model, the CVDM was employed to calculate the gradient of objective function with respect to the optical parameters. All the simulation results show that the CVDM is efficient and robust for the reconstruction of optical parameters.

Abstract:Image reconstruction in optical tomography in the so-called transport regime, where the diffusion approximation is not valid, requires modeling of light transport using the radiative transfer equation. In this work, we approach this problem by utilizing the Monte Carlo method for light transport. In this work, we propose a methodology for absolute imaging of absorption and scattering in this regime utilizing a Monte Carlo method for light transport. The image reconstruction problem is formulated as a minimization problem that is solved using a stochastic Gauss-Newton method. In the construction of the Jacobian matrix for scattering, a perturbation approximation for Monte Carlo is utilized. The approach is evaluated with numerical simulations using an adaptive approach where the number of photon packets is adjusted during the iterations, and with different fixed numbers of photon packets. The simulations show that the Monte Carlo method for light transport can be utilized in the absolute imaging problem of optical tomography and that the absorption and scattering parameters can be estimated simultaneously with good accuracy.

Abstract:Optical tomography is a typical non-invasive medical imaging technique, which aims to reconstruct geometric and physical properties of tissues by passing near infrared light through tissues for obtaining the intensity measurements. Other than optical properties of tissues, we are interested in finding locations of small inclusions inside the object from boundary measurements, based on the time-dependent diffusion model. First, we analyze the asymptotic behavior of the boundary measurements weighted by the fundamental solution of a backward diffusion equation as the diameters of inclusions go to zero. Then, we derive an efficient algorithm for locating small inclusions by finite boundary measurements. This algorithm is direct, simple and easy to be implemented numerically, since it only involves matrix operations and has no iteration process. Finally, some numerical results are presented to illustrate the feasibility and robustness of the algorithm. A new observation of the algorithm is that we can take the source points and test points independently and increase the resolution of numerical results by taking more test points.
Diffuse correlation tomography in the transport regime: A theoretical study of the sensitivity to Brownian motion

Abstract:Diffuse correlation tomography (DCT) uses the electric-field temporal autocorrelation function to measure the mean-square displacement of light-scattering particles in a turbid medium over a given exposure time. The movement of blood particles is here estimated through a Brownian-motion-like model in contrast to ordered motion as in blood flow. The sensitivity kernel relating the measurable field correlation function to the mean-square displacement of the particles can be derived by applying a perturbative analysis to the correlation transport equation (CTE). We derive an analytical expression for the CTE sensitivity kernel in terms of the Green's function of the radiative transport equation, which describes the propagation of the intensity. We then evaluate the kernel numerically. The simulations demonstrate that, in the transport regime, the sensitivity kernel provides sharper spatial information about the medium as compared with the correlation diffusion approximation. Also, the use of the CTE allows one to explore some additional degrees of freedom in the data such as the collimation direction of sources and detectors. Our results can be used to improve the spatial resolution of DCT, in particular, with applications to blood flow imaging in regions where the Brownian motion is dominant.

Abstract:Using the algebra transformation method, we develop and demonstrate the use of the delta-P, approximation to improve steady-state radiative-transfer estimates on spatial scales comparable to the mean free path. We show that the delta-P, approximation agrees well with Monte Carlo simulation from source to infinity when we choose an appropriate parameter f (fractional portion that scatters directly forward) in the delta-Eddington phase function. We also provide the empirical formula to determine the parameter f. (c) 2007 Optical Society of America.

Abstract:Optical tomography is an emerging and important molecular imaging modality. The aim of optical tomography is to reconstruct optical properties of human tissues. In this paper, we focus on reconstructing the absorption coefficient based on the radiative transfer equation (RTE). It is an ill-posed parameter identification problem. Regularization methods have been broadly applied to reconstruct the optical coefficients, such as the total variation (TV) regularization and the L1 regularization. In order to better reconstruct the piecewise constant and sparse coefficient distributions, TV and L1 norms are combined as the regularization. The forward problem is discretized with the discontinuous Galerkin method on the spatial space and the finite element method on the angular space. The minimization problem is solved by a Jacobian-based Levenberg-Marquardt type method which is equipped with a split Bregman algorithms for the L1 regularization. We use the adjoint method to compute the Jacobian matrix which dramatically improves the computation efficiency. By comparing with the other imaging reconstruction methods based on TV and L1 regularizations, the simulation results show the validity and efficiency of the proposed method.

Abstract:Diffuse optical tomography is a novel molecular imaging technology for small animal studies. Most known reconstruction methods use the diffusion equation (DA) as forward model, although the validation of DA breaks down in certain situations. In this work, we use the radiative transfer equation as forward model which provides an accurate description of the light propagation within biological media and investigate the potential of sparsity constraints in solving the diffuse optical tomography inverse problem. The feasibility of the sparsity reconstruction approach is evaluated by boundary angular-averaged measurement data and internal angular-averaged measurement data. Simulation results demonstrate that in most of the test cases the reconstructions with sparsity regularization are both qualitatively and quantitatively more reliable than those with standard L-2 regularization. Results also show the competitive performance of the split Bregman algorithm for the DOT image reconstruction with sparsity regularization compared with other existing L-1 algorithms.

Abstract:Image reconstruction for diffusion optical tomography often involves repeatedly solving the forward problem, wherein the diffusion equation is typically utilized to describe the light propagation. However, the diffusion equation may not be valid in regions of either high absorption or low scatter. In this paper, a new method is proposed to improve the forward model. This method is based on the modelling error between the Monte Carlo method and the diffusion equation. To further improve the efficiency of image reconstruction, we propose the grouping strategy of source-detector configuration. Simulation results demonstrate that the proposed method can significantly speed up the reconstruction process with the high reconstruction quality.

Abstract:An edge-guided TVp (0 < p < 1) regularization is introduced to recover scattering and absorption coefficients simultaneously in optical tomography, which combines the TVp minimizing scheme with an edge-guided strategy. The TVp minimizing scheme consists of a data fidelity term and an l(p )-norm (0 < p < 1) of the gradients of underlying optical coefficients. To make the minimization problem numerically tractable, the Huber function is utilized to locally smooth the l(p)-norm to obtain a differential objective function. The lagged diffusivity-Newton iteration is applied to find the minimizers of the above Huberized objective function. An edge-guided strategy is inserted into each iteration in the form of a weighted matrix. In addition, a normalizing technique is incorporated into our algorithm to reduce the cross-talk. Compared with l(p) regularization, the proposed edge-guided TVp regularization presents superiorities on keeping the shape as well as the size of targets and removing background undulations. Then the proposed method is applied to image the tissue in the presence of a non-scattering or a low-scattering layer. It is shown that our method is capable of imaging the targets in the tissue containing a non-scattering layer using reduced measurements data. Moreover, it holds promise for jointly imaging both targets and the low-scattering layer under a certain noise level.

Abstract:Optical tomography belongs to the promising set of non-invasive methods for probing applications of semi-transparent media. This covers a wide range of fields. Nowadays, it is mainly driven by medical imaging in search of new less aggressive and affordable diagnostic means. This paper aims at presenting the most recent research accomplished in the authors’ laboratories as well as that of collaborative institutions concerning the development of imaging algorithms. The light transport modelling is not a difficult question as it used to be. Research is now focused on data treatment and reconstruction. Since the turn of the century, the rapid expansion of low cost computing has permitted the development of enhanced imaging algorithms with great potential. Some of these developments are already on the verge of clinical applications. This paper presents these developments and also provides some insights on still unresolved challenges. Intrinsic difficulties are identified and promising directions for solutions are discussed.
