Abstract:Low-density lipoprotein (LDL) gains worldwide attention for decades as the key risk factor to atherosclerosis that progressively deteriorating into cardiovascular diseases. Until recent years, LDL-apheresis comes to be extensively used as a direct and efficient LDL removal method, with LDL adsorption materials particularly important. In this paper, a new strategy based on the co-deposition of polydopamine (PDA) with polyethylenimine (PEI) onto polysulfone (PSf) membranes, then subsequent heparinization by amino-carbonyl reactions, to achieve LDL selectivity and simultaneous biocompatibility, is proposed. Surface properties of modified PSf membranes are characterized by ATR-FTIR, XPS, FESEM, Zeta potential and WCA measurements. LDL adsorption ability is investigated by ELISA, while blood biocompatibility is evaluated by platelet adhesion experiments. Results suggest that heparin-modified PSf membranes show high selectivity for LDL removal and fine biocompatibility in contact with plasma, as excellent potential materials for LDL-apheresis. Abstract:Heparin has been considered to be a potentially useful ligand for low-density lipoprotein(LDL) detection and analysis in a clinical context. In order to construct an affinity surface for preferential adsorption of LDL, heparin-modified gold surface(GS-Hep) was fabricated by a self-assembling method and hydrophobic-modified gold surfaces(GS-Hydro) was used as a control. The morphologies of the modified gold surfaces were investigated by atomic force microscopy(AFM) and the quantity of heparin bound to gold surface was assayed by the toluidine blue(TB) colorimetric method. Water contact angles were determined to investigate wettability on GS-Hep and GS-Hydro. Surface plasmon resonance(SPR) technique was used subsequently to detect the selective binding of LDL with heparin. And the investigation on the effect of pH on LDL adsorption suggests that lower pH lead to higher quantities of LDL adsorption on GS-Hep. Compared with GS-Hydro, GS-Hep is selective for LDL from both single and binary protein solutions. Moreover, adsorbed LDL on GS-Hep could be washed off by injecting elution solution, such as NaCl solution, for the purpose of the regeneration of GS-Hep for further LDL adsorption.
Immobilization of Heparin on Polysulfone Surface for Selective Adsorption of Low-Density Lipoprotein (LDL)

Abstract:A versatile method was developed to immobilize heparin covalently on polysulfone sheets (PSu) to achieve selective adsorption of low-density lipoprotein (LDL). This was achieved by activation of PSu with successive treatments of chlorodimethyl ether and ethylenediamine, and subsequent chemical binding of heparin with bifunctional linker molecules. A heparin density up to 0.86μgcm−2 on a dense PSu film was achieved. The modified PSu films were characterized by attenuated total reflectance Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy. The hydrophilicity of the PSu film was improved greatly by covalent immobilization of heparin. The water contact angle of PSu film was decreased from 86.6±3.7° to 50.5±3.2° after binding of 0.36μgcm−2 heparin. An enzyme-linked immunosorbent assay was used to measure the binding of LDL on plain and modified PSu films. It was found that the heparin-modified PSu film could selectively recognize LDL from binary protein solutions. Furthermore, it was possible to desorb LDL from heparinized PSu, but not from plain PSu, with heparin, sodium chloride or urea solution, which indicates a selective but reversible binding of LDL to heparin. The results suggest that heparin-modified PSu membranes are promising for application in simultaneous hemodialysis and LDL apheresis therapy.
SURFACE HEPARINIZATION OF POLYPROPYLENE MICROPOROUS MEMBRANES FOR SELECTIVE ADSORPTION OF LOW-DENSITY LIPOPROTEIN

Abstract:To endow polypropylene microporous membrane (PPMM) with adsorption selectivity to low-density lipoprotein (LDL),a versatile method was developed to covalently immobilize heparin on the PPMM surface. This was achieved by grafting acrylic acid (AA) on the PPMM surface by UV irradiation, and subsequent chemical binding of heparin with ethylenediamine as a spacer. The membrane surfaces were characterized by attenuated total reflectance Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy. The hydrophilicity and blood compatibility of the PPMM surface were improved obviously by the surface heparinization. Water contact angle of the membrane surface was decreased from (138 +/- 7. 2)degrees for the nascent PPMM to (35. 1 +/- 2. 7)degrees for the heparinized PPMM with 7. 39 mu g/cm(2) heparin. An enzyme-linked immunosorbent assay (ELISA) was employed to analyze the adsorption of the LDL on the nascent and heparinized PPMMs. It was found that the surface heparinization greatly enhanced the adsorption of LDL both from single protein solution and binary protein solution containing human serum albumin (HSA), and the heparinized PPMM could selectively adsorb LDL from binary protein solution. In addition, the adsorbed LDL could be desorbed from the heparinized PPMM with sodium chloride solution, which indicated the adsorption was of reversibility.
Covalent Heparin Modification of a Polysulfone Flat Sheet Membrane for Selective Removal of Low-Density Lipoproteins: A Simple and Versatile Method

Abstract:A simple, convenient and economical method for the heparinization of PSf membranes is described, with the aim of preparing an LDL adsorber for simultaneous LDL apheresis and hemodialysis. An atmospheric pressure glow discharge generator is used to activate the PSf membrane surface, with subsequent chemical binding of heparin in the presence of EDC and NHS. ATR-FTIR spectroscopy and XPS measurements confirm successful surface modification. The PSf-Hep membrane shows good blood compatibility, with a relatively low amount and normal morphology of adherent platelets. ELISA results indicate that the PSf-Hep membrane exhibits excellent selective affinity for LDL in single and binary protein solutions, suggesting potential applications in hemodialysis with simultaneous LDL removal.
Fabrication and Study of Dextran/Sulfonated Polysulfone Blend Membranes for Low-Density Lipoprotein Adsorption

Abstract:The abnormal increase in low-density lipoprotein (LDL) in human blood is a main independent risk factor for the pathogenesis of atherosclerosis, whereas a reduced LDL level effectively lowers morbidity. It is important to develop LDL adsorption materials with high efficiency and selectivity, as well as to simplify their fabrication processes. In this paper, polysulfone (PSF), sulfonated polysulfone (SPSF), and sulfonated polysulfone/dextran (SPSF/GLU) membranes were successfully fabricated for LDL adsorption using a solution casting technique. Attenuated total reflectance Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy measurements confirmed the success of the preparation. The water contact angle decreased from 89.7 & PLUSMN; 3.4 & DEG; (PSF) to 76.4 & PLUSMN; 3.2 & DEG; (SPSF) and to 71.2 & PLUSMN; 1.9 & DEG; (SPSF/GLU), respectively. BSA adsorption testing showed that the SPSF/GLU with surface enrichment of sulfonate groups and glycosyl groups possessed higher resistance to protein solution. The adsorption and desorption behaviors of the studied samples in single-protein or binary-protein solutions were systematically investigated by enzyme-linked immunosorbent assay (ELISA), The results showed that SPSF/GLU, which had excellent resistance to protein adsorption, possessed a similar adsorption capacity to that of PSF. SPSF membrane exhibited excellent selective affinity for LDL in single and binary protein solutions, suggesting potential applications in LDL removal.

Abstract:A new and convenient approach for surface immobilization of heparin onto PE porous membranes is reported. 3,4-dihydroxy-L-phenylalanine (L-DOPA) a catecholic amino acid, is an important component of mussel adhesive proteins (MAPs). Based on the strong adhesive behavior of poly(DOPA) on solid surface, hydrophobic PE porous membranes was surface-modified by self-polymerization of L-DOPA in aqueous solution. Subsequently, heparin was immobilized covalently onto PE/poly(DOPA) composite membranes by the coupling between heparin and poly (DOPA) layer. The immobilization of poly (DOPA) and heparin was confirmed by X-ray photoelectron spectroscopy (XPS). The results of water contact angle measurements showed that the hydrophilicity of PE membranes was significantly improved by the introduction of poly(DOPA) and heparin. The effects of poly(DOPA) and heparin on membrane surface morphologies were also investigated by scanning electron microscopy (SEM). Despite the decreased pore amount and pore size on membrane surface after poly(DOPA) and heparin immobilization,the improved hydrophilicity would be beneficial to promote permeability and biocompability of PE membranes.
Selective and Regenerable Surface Based on Β-Cyclodextrin for Low-Density Lipoprotein Adsorption

Abstract:Cyclodextrins (CDs) are a family of cyclic oligosaccharides, whose unique hydrophilic outer surface and lipophilic central cavity facilitate the formation of inclusion complexes with various biomolecules, such as cholesterol and phospholipids, via multi-interactions. Low-density lipoprotein (LDL) is the main carrier of cholesterol in bloodstream and is associated with the progression of atherosclerosis. The surface of LDL is composed of a shell of phospholipids monolayer containing most of the free unesterified cholesterol as well as the single copy of apolipoprotein B-100. To date, various LDL adsorbents have been fabricated to interact with the biomolecules on LDL surface. Owing to its elegant structure, CD is considered to be a promising choice for preparation of more economical and effective LDL-adsorbing materials. Therefore, in this study, interaction between β-CD and LDL in solution was investigated by dynamic light scattering, circular dichroism, and ultraviolet spectroscopy. Further, a supramolecular surface based on β-CD was simply prepared by self-assembled monolayer on gold surface. The effect of hydrogen bond and the cavity of β-CD on the interaction between β-CD and LDL was particularly explored by surface plasmon resonance (SPR) analysis. The SPR results showed that such β-CD-modified surface exhibited good selectivity and could be largely regenerated by sodium dodecyl sulfate wash. This study may extend the understanding of the interaction between LDL and LDL adsorbent or the design and development of more efficient and lower-cost LDL adsorbents in the future.
Functionalized Periodic Mesoporous Silica Nanoparticles for Inhibiting the Progression of Atherosclerosis by Targeting Low-Density Lipoprotein Cholesterol

Abstract:Atherosclerotic disease is a substantial global burden, and existing treatments, such as statins, are recommended to lower low-density lipoprotein cholesterol (LDL-C) levels and inhibit the progression of atherosclerosis. However, side effects, including gastrointestinal unease, potential harm to the liver, and discomfort in the muscles, might be observed. In this study, we propose a novel method using periodic mesoporous silica nanoparticles (PMS) to create heparin-modified PMS (PMS-HP) with excellent biocompatibility, enabling selective removal of LDL-C from the blood. In vitro, through the introduction of PMS-HP into the plasma of mice, we observed that, compared to PMS alone, PMS-HP could selectively adsorb LDL-C while avoiding interference with valuable components such as plasma proteins and high-density lipoprotein cholesterol (HDL-C). Notably, further investigations revealed that the adsorption of LDL-C by PMS-HP could be well-fitted to quasi-first-order (R2 = 0.993) and quasi-second-order adsorption models (R2 = 0.998). Likewise, in vivo, intravenous injection of PMS-HP enabled targeted LDL-C adsorption (6.5 ± 0.73 vs. 8.6 ± 0.76 mM, p < 0.001) without affecting other plasma constituents, contributing to reducing intravascular plaque formation (3.66% ± 1.06% vs. 1.87% ± 0.79%, p < 0.05) on the aortic wall and inhibiting vascular remodeling (27.2% ± 6.55% vs. 38.3% ± 1.99%, p < 0.05). Compared to existing lipid adsorption techniques, PMS-HP exhibited superior biocompatibility and recyclability, rendering it valuable for both in vivo and in vitro applications.

Abstract:Polydopamine was used as a crosslinking agent to assist the immobilization of heparin on the medical polyurethane. The heparinized polyurethane by polydopamine exhibited greater hydrophilicity, hemocompatibility and cytocompatibility. The polydopamine could remain the bioactivity and stability of heparin on the PU membranes. Thus, the heparinized polyurethane by polydopamine has greater potential for small‐diameter vascular grafts. Surface heparinization is an effective solution to resolve low endothelialization, poor anticoagulation, and hemocompatibility of polyurethane (PU) used as materials of small‐diameter vascular grafts. Here, the effects of polydopamine (PDA) and poly (acrylic acid) (PAA) as crosslinking agents on the surface heparinization were explored. The PU membranes grafted with heparin (Hep) via dopamine (PU/PDA‐Hep) showed better hydrophilicity and stability, compared to heparinized PU membranes via acrylic acid (PU/PAA‐Hep). The results of X‐ray photoelectron spectroscopy demonstrated that heparin was successfully grafted onto the PU surface and the grafting efficiency was high when PDA as a cross‐linking agent. The grafted heparin aggregated and formed nanoparticles, and increased the surface roughness of PU membranes. The heparinized membranes demonstrated good anti‐adhesion of bovine serum albumin and fibrin protein. In addition, no activated platelets or educts on heparinized PU were found by platelet adhesion tests, implying that heparin‐immobilized surfaces had good hemocompatibility. Moreover, the in vitro cytocompatibility assessment showed that the PU/PDA‐Hep significantly improved the proliferation of L929 cells and was superior to PU/AA‐Hep. These results demonstrated that PDA‐assisted surface heparinization was an effective method to improve the anticoagulant and biocompatibility of PU small‐diameter vascular materials and could be extended to other implantable materials.

Abstract:A novel method for the immobilization of sodium alginate sulfates (SAS) on polysulfone (PSu) ultrafiltration membranes to achieve selective adsorption of low-density lipoprotein (LDL) was developed, which involved the photoinduced graft polymerization of acrylamide on the membrane and the Hofmann rearrangement reaction of grafted acrylamide followed by chemical binding of SAS with glutaraldehyde. The surface modification processes were confirmed by attenuated total reflectance Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy characterization. Zeta potential and water contact angle measurements were performed to investigate the surface charge and wettability of the membranes. An enzyme-linked immunosorbent assay was used to measure the binding of LDL on plain and modified PSu membranes. It was found that the PSu membrane immobilized with sodium alginate sulfates (PSu-SAS) greatly enhanced the selective adsorption of LDL from protein solutions and the absorbed LDL could be easily eluted with sodium chloride solution, indicating a specific and reversible binding of LDL to SAS, mainly driven by electrostatic forces. Furthermore, the PSu-SAS membrane showed good blood compatibility as examined by platelet adhesion. The results suggest that the PSu-SAS membranes are promising for application in simultaneous hemodialysis and LDL apheresis therapy.

Abstract:Heparin is covalently combined to SPEO or ODO through reaction of SPEO or ODO and heparin in the presence of DCCI. The heparinized surfaces could be obtained by SPEO-heparin or ODO-heparin THF/water mixture solution treatment. Heparin and PEO chain may move to the upper surface region of the modified PS films when the films contacts with water. Thus obtained surfaces exhibited heparin-like antithrombogenic activity. Hydrophilic spacer is helpful for the biological activity of the immobilized heparin on the polymer surfaces.
A New Method of Heparinizing PLLA Film by Surface Entrapment

Abstract:A new method of surface heparinizing biodegradable polymers was designed. A heparin-modified poly(L-lactic acid) (PLLA) system was developed by physically entrapping the heparin on the PLLA surface. The surface characterization and biological performance of these materials were carried out by SEM, attenuated-total-reflection spectroscopy, contact angle measurements, and platelet adhesion evaluations. The modification strategy was performed by reversible swelling of the PLLA surface following exposure to a solvent-nonsolvent mixture. This process resulted in the localized physical entrapment of the diffused heparin. X-ray photoelectron spectroscopy was used to confirm that control over the heparin surface density can be achieved by using set polymer treatment times. Platelet adhesion tests showed significant improvement in blood compatibility by the PLLA surfaces after modification.
Highly efficient one-step selective separation of heparin via multi-functional adsorptive membranes

Abstract:Commercial heparin production, a widely applied anticoagulant, requires a series of time-consuming and costly separation units to achieve isolation and purification and the challenging removal of macromolecular impurities, such as proteins. We report a novel separation strategy for the first time that can capture heparin while simultaneously recovering proteins through an efficient one-step separation. Novel adsorptive membranes were prepared by grafting polymer brushes of (3-acrylamidopropyl) trimethylammonium chloride (APTMAC) onto the surface of poly(vinylidene fluoride) (PVDF) membrane via atom transfer radical polymerization (ATRP) by controlling the polymerization time. The prepared membrane exhibited excellent hydrophilicity with water contact angle of 26.57° and adsorption capacity for heparin (598.6 μg/cm 2 ). XPS, FTIR measure and density functional theory (DFT) simulation demonstrated that the adsorptive membrane could effectively adsorb heparin on the surface via the electrostatic interaction between -N(CH 3 ) 3 + groups of the grafted APTMAC on the membranes and the reactive functional groups (OSO 3 − , NHSO 3 − , COO − ) of heparin. Membrane filtration results showed that the adsorptive membranes created one-step separations that could selectively capture heparin via electrostatic interaction while rejecting and concentrating bovine serum albumin (BSA) because of size sieving and hydrated layer repulsion. Besides, the purity of crude heparin increased from 60 % to 93 % through using adsorptive membrane filtration. Moreover, the heparin adsorption capacity remained at 82.3 % of its original capacity after six cycles. This study provides a novel strategy for efficient, economic and selective multi-functional separations for heparin and its similar biomedical medicines.

Abstract:Heparin, an important polysaccharide, has been widely used for coatings of cardiovascular devices because of its multiple biological functions including anticoagulation and inhibition of intimal hyperplasia. In this study, surface heparinization of a commonly used 316L stainless steel (SS) was explored for preparation of a multifunctional vascular stent. Dip-coating of the stents in an aqueous solution of dopamine and hexamethylendiamine (HD) (PDAM/HD) was presented as a facile method to form an adhesive coating rich in primary amine groups, which was used for covalent heparin immobilization via active ester chemistry. A heparin grafting density of about 900 ng/cm(2) was achieved with this method. The retained bioactivity of the immobilized heparin was confirmed by a remarkable prolongation of the activated partial thromboplastin time (APTT) for about 15 s, suppression of platelet adhesion, and prevention of the denaturation of adsorbed fibrinogen. The Hep-PDAM/HD also presented a favorable microenvironment for selectively enhancing endothelial cell (EC) adhesion, proliferation, migration and release of nitric oxide (NO), and at the same time inhibiting smooth muscle cell (SMC) adhesion and proliferation. Upon subcutaneous implantation, the Hep-PDAM/HD exhibited mitigated tissue response, with thinner fibrous capsule and less granulation formation compared to the control 316L SS. This number of unique functions qualifies the heparinized coating as an attractive alternative for the design of a new generation of stents.

Abstract:A facile method for the immobilization of β-cyclodextrin on polysulfone membranes with the aim of selectively adsorbing low-density lipoprotein (LDL) was established, which is based on the self-assembly of dopamine on the membrane followed by the Schiff base reaction with mono-(6-ethanediamine-6-deoxy)-β-cyclodextrin. The surface modification processes were validated using X-ray photoelectron spectroscopy and attenuated total reflectance Fourier-transform infrared spectroscopy. Surface wettability and surface charge of the membranes were investigated through the water contact angle and zeta potential analysis. The cyclodextrin-modified polysulfone membrane (PSF-CD) showed good resistance to protein solutions, as shown by the measurement of BSA adsorption. The assessment of BSA adsorption revealed that the cyclodextrin-modified polysulfone membrane (PSF-CD) exhibited excellent resistance to protein solutions. To investigate the adsorption and desorption behaviors of the membranes in single-protein or binary-protein solutions, an enzyme-linked immunosorbent assay was employed. The results revealed that the PSF-CD possessed remarkable adsorption capacity and higher affinity for LDL in both single-protein and binary-protein solutions, rendering it a suitable material for LDL apheresis.

Abstract:Based on the self-polymerization and strong adhesion characteristic of dopamine in wet conditions, the hydrophobic polyethylene (PE) porous membranes were surface-modified via simply immersing them into dopamine aqueous solution for 24h. Subsequently, heparin was immobilized covalently onto the resultant membrane by the coupling between heparin and reactive polydopamine layer. Attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy and X-ray photoelectron spectroscopy (XPS) were utilized to determine the chemical compositions of membrane surface, which confirmed the successful introduction of polydopamine and immobilization of heparin molecules. Scanning electronic microscopy (SEM) and atomic force microscopy (AFM) were employed to investigate the changes in surface morphologies after surface modification. The data of water contact angle measurements indicated that the hydrophilicity of PE membranes was remarkably improved after polydopamine coating and heparin immobilization. The results of in vitro hemocompatibility test proved that surface heparinization significantly suppressed the adhesion of platelet and enhanced the anticoagulation ability of PE membranes. This work offered a convenient approach to improve the permeability and biocompatibility of inert PE porous membranes for their biomedical and blood-contacting applications.

Abstract:Based on the strong adhesive behavior of poly(3,4-dihydroxy-l-phenylalanine) (or poly(DOPA)) on solid surface, poly(vinylidene fluoride) (PVDF) microporous membranes were surface-modified by the self-polymerization of DOPA in aqueous solution. Subsequently, heparin was immobilized covalently onto the obtained PVDF/poly(DOPA) composite membranes by the coupling between heparin and poly(DOPA) coating. The modified membranes were subjected to a long-term washing, and the firm immobilization of poly(DOPA) and heparin was confirmed by X-ray photoelectron spectroscopy (XPS). The results of water contact angle measurements showed that the hydrophilicity of PVDF membranes was significantly improved by the incorporation of poly(DOPA) and heparin. The effects of poly(DOPA) and heparin on membrane surface morphologies were also investigated by scanning electron microscopy (SEM).
Immobilized Heparin and Its Anti-Coagulation Effect on Polysulfone Membrane Surface

Abstract:Polysulfone has been widely used as hemodialysis membrane material because of its excellent physiochemical performance. There is still a need to further improve its anti-coagulation property in clinical practice. In this work, we covalently immobilized heparin onto polysulfone membrane to improve its anti-coagulation performance. Low temperature plasma technique with environmentally friendly nitrogen as the gas source, as well as N-ethyl-N '-[3-dimethylaminopropy] carbodiimide hydrochloride/hydroxy-2,5-dioxopyrolidine-3-sulfonicacid sodium chemistry were utilized to immobilize heparin onto the surface of polysulfone membrane. X-ray photoelectron spectroscopy, attenuated total reflectance Fourier-transform infrared spectroscopy, as well as water contact angle results confirmed successful binding of heparin to the membrane surface. Only slight permeability differences were observed between the immobilized surface and the unmodified surface, while the polysulfone membrane had become more hydrophilic after immobilization. The blood coagulation time was greatly prolonged after modification and less platelets adhesion was observed on the heparin immobilized surface. Also, compared with heparin injection doses in clinical, the heparinized process in our work consumed less heparin. Our study suggests that the immobilized heparin has local anti-coagulation effect, while reducing the doses.

Abstract:The influence of different pH values between assembly and application environment on the surface properties of layer-by-layer assembled heparin and chitosan multilayer films was investigated. The high adsorption amount of cationic dye and high wettability on the chitosan terminated multilayer films at high environment pH may indicate the excess of free sulfate group existed on the chitosan surface. Since the ionization degree of chitosan decreased with increasing of pH, the multilayer films assembled at low pH will lead to some excess free unpaired sulfate group in higher pH application environment, which paired with ionized amino group under prepared conditions. The blood-compatibility tests performed with these films clearly demonstrated that the good anticoagulant activity of low pH assembled heparin/chitosan even on the chitosan-terminated surface. Such an easy and controlled processing by the difference of pH values between assembly and application environment may have a good potential for fabrication of biomedical device coating.
