Abstract:To avoid the rapid degradation of magnesium alloy AZ31B after implantation in the body, the addition of a zinc oxide (ZnO) film was examined. A rough surface was fabricated by femtosecond laser on AZ31B at frequencies of 100, 200, 300, 500, and 800 kHz, before adding a ZnO film by wet metallurgy. Scanning electron microscopy (SEM) indicated that 500 kHz femtosecond laser frequency produced a coral-like surface morphology on AZ31B and the ZnO film had a needle-like structure. X-ray diffraction (XRD) revealed that the film mainly consisted of Zn and ZnO. Potentiodynamic polarization curves revealed that the ZnO film exhibited a better corrosion resistance than bare AZ31B. It also possessed superior cell proliferation ability and higher suppression of a representative device-related infection of Escherichia coli strain than bare AZ31B. This study provides a novel film strategy for manufacturing biodegradable magnesium alloys with good corrosion resistance, biocompatibility, and antibacterial properties.

Abstract:Biomedical magnesium is an ideal material for hard tissue repair and replacement. However, its rapid degradation and infection after implantation significantly hindersclinical applications. To overcome these two critical drawbacks, we describe an integrated strategybased on the changes in pH and Mg2+ triggered by magnesiumdegradation. This system can simultaneously offer anticorrosion and antibacterial activity. First, nanoengineered peptide-grafted hyperbranched polymers (NPGHPs) with excellent antibacterial activity were introduced to sodium alginate (SA) to construct a sensitive NPGHPs/SA hydrogel. The swelling degree, responsiveness, and antibacterial activity were then investigated,indicating that the system can perform dual stimulation of pH and Mg2+ with controllable antimicrobial properties. Furthermore, an intelligent platform was constructed by coating hydrogels on magnesium with polydopamine as the transition layer. The alkaline environment generated by the corrosion of magnesium reduces the swelling degree of the coatingso that the liquid is unfavorable for contacting the substrate, thus exhibiting superior corrosion resistance. Antibacterial testing shows that the material can effectively fight against bacteria, while hemolytic and cytotoxicity testing suggest that it is highly biocompatible. Thus, this work realizes the smart integration of anticorrosion and antibacterial properties of biomedical magnesium, thereby providing broader prospects for the use of magnesium.

Abstract:In this study, we applied the method of nanosecond laser treatment for the fabrication of superhydrophobic and superhydrophilic magnesium-based surfaces with hierarchical roughness when the surface microrelief is evenly decorated by MgO nanoparticles. The comparative to the bare sample behavior of such surfaces with extreme wettability in contact with dispersions of bacteria cells Pseudomonas aeruginosa and Klebsiella pneumoniae in phosphate buffered saline (PBS) was studied. To characterize the bactericidal activity of magnesium samples with different wettability immersed into a bacterial dispersion, we determined the time variation of the planktonic bacterial titer in the dispersion. To explore the anti-bacterial mechanisms of the magnesium substrates, a set of experimental studies on the evolution of the magnesium ion concentration in liquid, pH of the dispersion medium, surface morphology, composition, and wettability was performed. The obtained data made it possible to reveal two mechanisms that, in combination, play a key role in the bacterial decontamination of the liquid. These are the alkalization of the dispersion medium and the collection of bacterial cells by microrods growing on the surface as a result of the interaction of magnesium with the components of the buffer solution.

Abstract:Magnesium and its alloys have drawn interest as antibacterial biomaterials, owing to their ability to alkalize the surrounding medium during degradation. The antibacterial effect of pure Mg and Mg alloys in vitro has previously been reported. However, the antibacterial property of Mg in vivo might be different because of the apparently dissimilar corrosion characteristics. In this study, pure Mg rods were implanted and bacterial suspension were injected into rat femurs to investigate the antibacterial property of Mg in vivo. The results showed that contrary to the high antibacterial rate in vitro, Mg exhibited a dramatic drop in antibacterial effect in vivo. Bacteria proliferated on the surface of the Mg rods as well as in the femur. Inflammatory cells filled cavities in the cortical bone of the femur, which was demonstrated by histological and micro-CT examination after 2 and 4 weeks of implantation. It is suggested that a reduced corrosion rate in vivo would result in insufficient pH value. In addition, the deposition layer would prevent further corrosion of Mg and provide a favorite site for bacteria adhesion. Hence, the dramatically reduced antibacterial property of Mg needs to be noticed when it is used as a biomaterial.

Abstract:Algicidal bacteria offer a promising option for killing cyanobacteria. In this study, a newly isolated strain of Bacillus methylotrophicus, ZJU, was used to control Microcystis aeruginosa. Analyses of relative reactive oxygen level, malondialdehyde content, superoxide dismutase activity, and fluorescence staining indicated that oxidative damage caused by the algicidal supernatant of strain ZJU mainly affected the cell membrane and consequently the membrane permeability and membrane potential of M. aeruginosa cells. Furthermore, an embedded immobilization technique was employed to improve the practical application of strain ZJU as an algicidal agent. On this basis, we proposed a novel concept of enhancing the algicidal properties of immobilized ZJU by adding Fe3O4 nanoparticles and wheat bran in the process of immobilization. Our studies showed that Fe3O4 nanoparticles conferred the immobilized bacteria with a magnetization of 30.87 emu/g, and this magnetization enabled efficient re-collection of the immobilized bacteria by magnetic means. Moreover, wheat bran endowed the immobilized bacteria with 10.34% higher algicidal activity than immobilized bacteria without wheat bran. The results indicate a novel concept of enhancing the algicidal property of bacteria against M. aeruginosa by adding Fe3O4 nanoparticles and wheat bran.
In vitro and in vivo antibacterial performance of Zr & O PIII magnesium alloys with high concentration of oxygen vacancies

Abstract:The effects of dual Zr and O plasma immersion ion implantation (Zr & O PIII) on antibacterial properties of ZK60 Mg alloys are systematically investigated. The results show that a hydrophobic, smooth, and ZrO2-containing graded film is formed. Electrochemical assessment shows that the corrosion rate of the plasma-treated Mg alloy decreases and the decreased degradation rate is attributed to the protection rendered by the surface oxide. In vitro and in vivo antibacterial tests reveal Zr & O PIII ZK60 presents higher antibacterial rate compared to Zr PIII ZK60 and untreated control. The hydrophobic and smooth surface suppresses bacterial adhesion. High concentration of oxygen vacancies in the surface films are determined by X-ray photoelectron spectroscopy (XPS), UV-vis diffuse reflectance spectra (UV-vis DRS) and electron paramagnetic resonance (EPR) and involved in the production of reactive oxygen species (ROS). The higher level of ROS expression inhibits biofilm formation by down-regulating the expression of icaADBC genes but up-regulating the expression of icaR gene. In addition, Zr & O PIII improves cell viability and initial cell adhesion confirming good cytocompatibility. Dual Zr & O PIII is a simple and practical means to expedite clinical acceptance of biodegradable magnesium alloys.

Abstract:Magnesium alloys are potential biodegradable materials and have received increasing attention due to their outstanding biological performance and mechanical properties. However, rapid degradation in the physiological environment and potential toxicity limit clinical applications. Recently, special magnesium–calcium (Mg–Ca) and magnesium–strontium (Mg–Sr) alloys with biocompatible chemical compositions have been reported, but the rapid degradation still does not meet clinical requirements. In order to improve the corrosion resistance, a rough, hydrophobic and ZrO2-containing surface film is fabricated on Mg–Ca and Mg–Sr alloys by dual zirconium and oxygen ion implantation. Weight loss measurements and electrochemical corrosion tests show that the corrosion rate of the Mg–Ca and Mg–Sr alloys is reduced appreciably after surface treatment. A systematic investigation of the in vitro cellular response and antibacterial capability of the modified binary magnesium alloys is performed. The amounts of adherent bacteria on the Zr–O-implanted and Zr-implanted samples diminish remarkably compared to the unimplanted control. In addition, significantly enhanced cell adhesion and proliferation are observed from the Zr–O-implanted sample. The results suggest that dual zirconium and oxygen ion implantation, which effectively enhances the corrosion resistance, in vitro biocompatibility and antimicrobial properties of Mg–Ca and Mg–Sr alloys, provides a simple and practical means to expedite clinical acceptance of biodegradable magnesium alloys.

Abstract:Implant-related infections are one of the major challenges faced by orthopedic surgeries. Developing implants with inherent antibacterial properties is an effective strategy to address this issue. Biodegradable magnesium and magnesium alloys have become a research hotspot due to their good bioactivity, mechanical properties, biocompatibility, and excellent antibacterial ability. However, magnesium and its alloys have rapid corrosion, and the difficulty in expelling harmful magnesium ions and hydrogen gas produced by degradation from the body. This review summarizes the mainstream surface modification techniques such as laser surface modification, friction stir processing, and micro-arc oxidation, along with their impact on the antimicrobial properties of magnesium-based materials. This paper reviews the latest research progress on improving the antibacterial properties of magnesium alloys through alloying and introduces the antibacterial effects of mainstream magnesium alloys and also elaborates on the antibacterial mechanism of magnesium alloy materials. It is expected to provide more basis and insights for the design of biodegradable magnesium alloys with antibacterial properties, thereby promoting their development and clinical application.

Effect of anionic groups on the antibacterial activity of magnesium oxide nanoparticles

Abstract:MgO nanoparticles were obtained by liquid-phase precipitation of three Mg sources produced from magnesite in this study. The influence of anionic groups in solution on the physical properties and bacterial inhibition of nanomaterials during their formation is discussed. The analysis demonstrates that the process of MgO and Mg(OH)2 precursor generation exhibits a visible anionic effect. As the electronegativity of the anion increases, the degree of ionization becomes higher, the growth environment becomes freer, and the crystal morphology tends towards a desirable hexagonal lamellar structure. The inhibition effect of the three MgO nanomaterials on E. coli and S. aureus was validated by growth curve method and colony counting method. It was found that MgO(C), prepared using MgCl2.6 H2O as the magnesium source, had the best bactericidal performance, with more than 98% bactericidal activity against both E. coli and S. aureus within 2 h at a sample concentration of 600 ppm, which was closely related to the larger electronegativity of Cl-. This research has contributed to the preparation and production of antibacterial MgO nanoparticles.

Abstract:Magnesium (Mg), a potential biodegradable material, has recently received increasing attention due to its Unique antibacterial property. However, rapid corrosion in the physiological environment and potential toxicity limit clinical applications. In order to improve the corrosion resistance meanwhile not compromise the antibacterial activity, a novel Mg alloy; Mg-Nd-Zn-Zr (Hereafter, denoted as JDBM), is fabricated by alloying with neodymium (Nd), zinc (Zn), zirconium (Zr). pH value, Mg ion concentration, corrosion rate and electrochemical test show that the corrosion resistance of JDBM is enhanced. A systematic investigation of the in vitro and in vivo antibacterial capability of JDBM is performed. The results of microbiological counting, CLSM, SEM in vitro, and microbiological cultures, histopathology in vivo consistently show JDBM enhanced the antibacterial activity. In addition, the significantly improved cytocompatibility is observed from JDBM. The results suggest that JDBM effectively enhances the corrosion resistance, biocompatibility and antimicrobial properties of Mg by alloying with the proper amount of Zn, Zr and Nd. Abstract:Mg and its alloys have shown potential application as a bio-absorbable biomaterial, such as for bone screws and plates, however, their fast corrosion rate may result in the sudden failure of the implants. In this paper, compact and dense films are formed on AZ31 magnesium alloy by alkali treatment. The microstructure and composition of the films are examined by SEM, EDS and XRD, respectively. The bonding strength and corrosion behaviour of the films are characterized by tensile strength test, potentiodynamic polarization and electrochemical impedance spectroscopy (EIS) studies. The results show that the main composition of the films is Mg(OH)(2), the bonding strength between the film and the substrate is stronger than 60 MPa. And the corrosion resistance is improved by the alkali treatment compared with that of the bare AZ31 magnesium alloy in the simulated body fluid (SBF) solution. It is increased with increasing the treatment time.
Mg2+ Reduces Biofilm Quantity in Acidithiobacillus ferrooxidans through inhibiting Type IV Pili Formation.

Abstract:Bioleaching is a promising process for 350 million tons of Jinchuan low-grade pentlandite. But high concentration of Mg2+ is harmful to bioleaching microorganisms. Interestingly, biofilm formation can improve leaching rate. Thus, it is actually necessary to investigate the effect of Mg2+ stress on Acidithiobacillus ferrooxidans biofilms formation. In this study, we found that 0.1 and 0.5 M Mg2+ stress significantly reduced the total biomass of biofilm in a dose-dependent manner. The observation results of extracellular polymeric substances and bacteria using confocal laser scanning microscopy showed that the biofilm became thinner and looser under Mg2+ stress. Whereas 0.1 and 0.5 M Mg2+ stress had no remarkable effect on the bacterial viability, the attachment rate of Acidithiobacillus ferrooxidans to pentlandite was reduced by Mg2+ stress. Furthermore, sliding motility, twitching motility and the gene expression level of pilV and pilW were inhibited under Mg2+ stress. These results suggested that Mg2+ reduced biofilm formation through inhibiting pilV and pilW gene expression, decreasing Type IV pili formation and then attenuating the ability of attachment, subduing the active expansion of biofilms mediated by twitching motility. This study provided more information about the effect of Mg2+ stress on biofilm formation and may be useful for increasing the leaching rate in low-grade pentlandit.

Abstract:Biodegradable magnesium and magnesium alloys have received much attraction due to their light weight, suitable elastic modulus, biodegradability and excellent biocompatibility. However, the magnesium corrodes too fast in vivo, accompanied with hydrogen evolution and increase of pH value in local tissues around during degradation, which plays a detrimental role in the surrounding tissues and leads to hemolysis and bone tissue absorption sometimes. The poor corrosion resistance of Mg and Mg alloys restricts their clinical applications. Addition of proper alloying elements and surface modification of magnesium alloys are expected to be effective methods for retarding the corrosion rate and attaining controllable degradation behavior of Mg alloys. In this paper, investigations on antibacterial properties and hemolysis of biodegradable magnesium alloys are reviewed and research progresses of biocompatibility of biodegradable magnesium alloys in recent five years are systematically discussed. The development directions and challenges of biodegradable magnesium alloys in the future are presented.
Corrosion Resistance and Antibacterial Activity of Zinc-Loaded Montmorillonite Coatings on Biodegradable Magnesium Alloy AZ31.

Abstract:A Zinc-loaded montmorillonite (Zn-MMT) coating was hydrothermally prepared using Zn2+ ion intercalated sodium montmorillonite (Na-MMT) upon magnesium (Mg) alloy AZ31 as bone repairing materials. Biodegradation rate of the Mg-based materials was studied via potentiodynamic polarization curves, electrochemical impedance spectroscopy (EIS) and hydrogen evolution tests. Results revealed that both Na-MMT and Zn-MMT coatings exhibited better corrosion resistance in Dulbecco's modified eagle medium (DMEM) + 10% calf serum (CS) than bare Mg alloy AZ31 counterparts. Hemolysis results demonstrated that hemocompatibility of the Na-MMT and Zn-MMT coatings were 5%, and lower than that of uncoated Mg alloy AZ31 pieces. In vitro MTT tests and live-dead stain of osteoblast cells (MC3T3-E1) indicated a significant improvement in cytocompatibility of both Na-MMT and Zn-MMT coatings. Antibacterial properties of two representative bacterial strains associated with device-related infection, i.e. Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), were employed to explore the antibacterial behavior of the coatings. The measured inhibitory zone and bacterial growth rate confirmed that Zn-MMT coatings exhibited higher suppression toward both E. coli and S. aureus than that of Na-MMT coatings. The investigation on antibacterial mechanism through scanning electron microscopy (SEM) and lactate dehydrogenase (LDH) release assay manifested that Zn-MMT coating led to severe breakage of bacterial membrane of E. coli and S. aureus, which resulted in a release of cytoplasmic materials from the bacterial cells. In addition, the good inhibition of Zn-MMT coatings against E. coli and S. aureus might be attributed to the slow but sustainable release of Zn2+ ions (up to 144 h) from the coatings into the culture media. This study provides a novel coating strategy for manufacturing biodegradable Mg alloys with good corrosion resistance, biocompatibility and antibacterial activity for future orthopedic applications. STATEMENT OF SIGNIFICANCE: The significance of the current work is to develop a corrosion-resistant and antibacterial Zn-MMT coating on magnesium alloy AZ31 through a hydrothermal method. The Zn-MMT coating on magnesium alloy AZ31 shows better corrosion resistance, biocompatibility and excellent antibacterial ability than magnesium alloy AZ31. This study provides a novel coating on Mg alloys for future orthopedic applications.
Self-assembled Silane Film and Silver Nanoparticles Coating on Magnesium Alloys for Corrosion Resistance and Antibacterial Applications

Abstract:Contamination resulting from microbial adhesion on magnesium alloys is very common in many applications. Self-assembly technology was employed to prepare an antibacterial composite coating by fixing silver nanoparticles (AgNPs) onto the surface of magnesium alloys. The AgNPs were immobilized on the surface of 3-aminopropyltrimethoxysilane (APTMS)-modified magnesium alloy AZ31 (APTMS/Mg) through electrostatic inter-attraction between partially protonated amino groups and negatively charged citrate-capped AgNPs, resulting in the AgNPs attached APTMS/Mg (AgNPs/APTMS/Mg) substrate. The prepared Ag colloid and functionalized AZ31 alloy were characterized by ultraviolet-visible (UV-vis) spectroscopy, Fourier transform infrared (FTIR) spectroscopy, scanning electron microscopy (SEM), and electrochemical methods. Finally, the bactericidal activity of AgNPs/APTMS/Mg substrate against Escherichia coli was assessed by the inhibition zone. The results demonstrated that Si-O-Si covalent bonds existed on the substrate with the formation of inorganic Si-O-Mg bonds. AgNPs were immobilized and well-dispersed, forming a uniform submonolayer on the silane film in two dimensions. The AgNPs/APTMS-pretreated AZ31 alloys exhibited better corrosion resistance and excellent antibacterial performance.

Abstract:To overcome the restricted degradation, poor antiacterial and osteoindctive problems of magnesium and its alloys, this study presented the spinning coating of an antimicrobial peptide (AP)-loaded silk fibroin (SF) composite onto a corrosion-resistant MgO-coated AZ31 Mg alloy via anodization (aMgO) and electrodeposition (eMgO) methods. The composite coatings not only created a smooth and hydrophilic surface, but also obviously improved the corrosion resistance according to the test of corrosion potential and current density. The colonization of E. coli on MgO-AP composite coatings was significantly reduced as compared to the MgO layers, due to the potential synergetic effects of APs and Mg2+. Compared with the bare AZ31, the composite coating inhibited the corrosion of the substrate and the release of Mg2+, supported the adhesion, spreading and proliferation of osteoblasts, and presented a significantly improved osteogenic differentiation trend. Therefore, the MgO-AP composite coating, which had both antibacterial and bone-promoting abilities, was expected to be applied for surface modification of magnesium alloy implants to solve the clinical problems of bacterial infection and poor osseointegration.
Nano-layered magnesium fluoride reservoirs on biomaterial surfaces strengthen polymorphonuclear leukocyte resistance to bacterial pathogens

Abstract:Biomaterial-related bacterial infections cause patient suffering, mortality and extended periods of hospitalization, imposing a substantial burden on medical systems. In this context, understanding of nanomaterials-bacteria-cells interactions is of both fundamental and clinical significance. Herein, nano-MgF2 films were deposited on titanium substrate via magnetron sputtering. Using this platform, the antibacterial behavior and mechanism of the nano-MgF2 films were investigated in vitro and in vivo. It was found that, for S. aureus (CA-MRSA, USA300) and S. epidermidis (RP62A), the nano-MgF2 films possessed excellent anti-biofilm activity, but poor anti-planktonic bacteria activity in vitro. Nevertheless, both the traditional SD rat osteomyelitis model and the novel stably luminescent mouse infection model demonstrated that nano-MgF2 films exerted superior anti-infection effect in vivo, which cannot be completely explained by the antibacterial activity of the nanomaterial itself. Further, using polymorphonuclear leukocytes (PMNs), the critical immune cells of innate immunity, a complementary investigation of MgF2-bacteria-PMNs co-culturing revealed that the nano-MgF2 films improved the antibacterial effect of PMNs through enhancing their phagocytosis and stability. To our knowledge, this is the first time of exploring the antimicrobial mechanism of nano-MgF2 from the perspective of innate immunity both in vitro and in vivo. Based on the research results, a plausible mechanism is put forward for the predominant antibacterial effect of nano-MgF2in vivo, which may originate from the indirect immune enhancement effect of nano-MgF2 films. In summary, this study of surface antibacterial design using MgF2 nanolayer is a meaningful attempt, which can promote the host innate immune response to bacterial pathogens. This may give us a new understanding towards the antibacterial behavior and mechanism of nano-MgF2 films and pave the way towards their clinical applications.

Abstract:To control the degradation of magnesium alloy and enhance its osteoinduction activity and antibacterial properties, we proposed the addition of Zn and Sr ions in the process of surface modification of the magnesium alloy (ZK60) by a one-pot hydrothermal process. We found that, after surface modification, the surface of the materials formed a cluster crystal structure coating layer, and the successful incorporation of Zn and Sr ions in the surface coating did not affect the morphology of the microstructure. The corrosion resistance of the surface of the modified magnesium alloy was significantly improved, and cells grew well on the modified material surfaces. Zn and Sr ions released from the coating layer promote cell osteogenic differentiation, and Zn ions also provide a good antibacterial effect. Thus, the combined use of Zn and Sr offers antibacterial effects and promotes osteogenic differentiation of cells. To summarize, we have developed a controllable and degradable magnesium alloy material that offers both osteoinduction and antibacterial effects. The development of this material provides ideas about the preparation of a novel biodegradable magnesium alloy with better bioactivity for clinical application.

Abstract:MgO and silver-containing MgO (AgMgO, 3.3, 9.1 at% Ag) films were sol-gel coated on titanium to improve its antibacterial property. Thermal analyses of MgAc2·4H2O powder revealed that MgO was crystallized at 400 °C. X-ray diffraction analysis showed that MgO was converted to Mg(OH)2 or Mg5(CO3)4(OH)2·4H2O during the ageing in air. Silver nanoparticles at the surface of AgMgO films were identified by scanning electron microscopy and x-ray photoelectron spectroscopy. The MgO film was about 1.5 um thick. The potentiodynamic polarization test in the Ca-free Hank’s balanced salt solution showed that the coated titanium samples had better corrosion resistance than the polished one. The dissolution of silver nanoparticles resulted in a current peak in the polarization plots. In the antibacterial test against E. Coli, the inhibition zone width was 0.3, 1.6, 2.0 mm for the films with 0, 3.3, 9.1 at% Ag, respectively. The bactericidal mechanisms of the MgO and AgMgO films were discussed. The present work would provide a facile method for antibacterial surface modification of titanium based osteo-implants.
