Abstract:In our work,ester hydrolysis of chiral primary alcohols was adopted as model reaction. The results showed that those chiral primary alcohols with non-acyloxy oxygen atom were able to be resolved by Pseudomonas cepacia lipase (PcL) with a relatively high enantioselectivity,but those without the extra oxygen did not. Investigation by chemical modification of PcL by N-acetylimidazole indicated enantioselectivity towards primary alcohol with the non-acyloxy oxygen decreased significantly,but those without retained almost the same low value. Protein mass spectrometry analysis suggested that Tyr29 in PcL had a significant effect on selectivity inside the active center. Further investigation by molecular simulation indicated that phenolic hydroxyl hydrogen atom in PcL’s Tyr29 was able to serve as an extra hydrogen bond donor,and could form a hydrogen bond with the extra oxygen of primary alcohol. To a large extent,whether the substrate molecule could form a hydrogen bond with Tyr29 of PcL decided the lipase’s enantioselectivity. The mechanism of enantioselectivity towards chiral primary alcohols via study of chemical modification of PcL was proposed.
Performance enhancement of chemically modified lipase in the organic phase guided by molecular simulation

Abstract:Lipase is one of the most important biocatalysts because of its stereoselectivity towards chiral substrates and reasonable adaptiveness of non-conventional reaction media,including hydrophobic organic solvents. Increasing the selectivity of lipase in such non-conventional environments has been a continuous hotspot. However,current researches on improving lipase selectivity usually cause critical decrease of catalytic activity. In this work,a novel computation- directed approach to enhance lipase enantioselectivity in organic media without losing activity was introduced. Stereoselective acylation of chiral secondary alcohols catalyzed byPcL(Pseudomonas cepacia lipase) inn-hexane was taken as the model reaction. According to the results from molecular dynamics simulation,when the Tyr29 inside thePcL catalytic cavity was modified by N-acetylimidazole (NAI),the enantioselectivity of the lipase was significantly increased. The simulation result was agreed with the experimental data. MALDI-TOF-MS confirmed the Tyr29 modified position,and kinetic resolution of chiral secondary alcohols by NAI-modifiedPcL displayed an increase of enantioselectivity ratio from 12.6 to 48.1 and higher activity.
A New Mechanism of Enantioselectivity Toward Chiral Primary Alcohol by Lipase from Pseudomonas Cepacia

Abstract:The stereo-recognition of chiral primary alcohols by lipase from Pseudomonas cepacia was found to deviate from earlier observations. Enantioselectivity toward 14 pairs of chiral primary alcohol esters by this lipase was dependent on the existence of an Onon-alpha (oxygen at non-alpha-position of the acyloxy group) in the alcohol moiety, and decreased as the size of the acyl moiety increased. Chemical modification on the lipase and molecular dynamics simulations indicated that Tyr(29) located within the catalytic cavity forms a hydrogen bond with the Onon-alpha of the preferred enantiomer of the primary alcohol ester. However, a larger acyl moiety suffered stronger hindrance from the catalytic cavity wall of the lipase, pushing the Onon-alpha away from Tyr(29), and thus weakening the stereo-recognition. Controlling Enantioselectivity of Esterase in Asymmetric Hydrolysis of Aryl Prochiral Diesters by Introducing Aromatic Interactions.

Abstract:Aromatic interactions specific to aryl radicals were introduced into two esterases, BioH from Escherichia coli and RspE from Rhodobacter sphaeroides to control their enantioselectivity in the asymmetric hydrolysis of prochiral aryl glutaric acid diesters. As a result, the enantiomeric excess (ee) of the S-product of dimethyl 3-phenylglutarate was increased from 25% (BioH wild type) to 96% (B_L83F/L86F) and from 13% (RspE wild type) to >99% (R_Y27R), respectively, while another variant of RspE R_M121F gave a reversed ee of 50% (R-product). Similar enhancement or reversion of enantioselectivity were also observed in the hydrolysis of three other prochiral aryl diesters (dimethyl 3-(4-flouro)-phenylglutarate, dimethyl 3-(4-cholo)-phenylglutarate and dimethyl 3-(3,4-dicholo)-phenylglutarate). Especially, the mutant R_Y27R was shown to be an excellent S-selective hydrolase for prochiral aryl diesters, with ee of all S-products >99%. In the mutants with altered enantioselectivity, the successful introduction of designed aromatic interactions was confirmed by molecular dynamics simulations and binding free energy analysis. These results demonstrate that aromatic interaction is one of the origins of enzyme enantioselectivity, the tuning of which leads to dramatic change in enantioselectivity. Besides, the successful engineering of the enantioselectivity in two different proteins toward four different substrates suggests that the introduction of aromatic interactions is a generally applicable strategy in the control of enantioselectivity toward aryl substrates.
Site-specifically Incorporated Non-Canonical Amino Acids into Pseudomonas alcaligenes Lipase to Hydrolyze L-menthol Propionate among the Eight Isomers

Abstract:It remains a challenge to improve the diastereopreference of enzymes when there are multiple chiral centers in the substrate, mainly because the limited understanding of mechanism determining diastereoselectivity. Compared with natural amino acids, non-canonical amino acids (ncAAs) provide side chains with wider range of functional groups and genetically encoded ncAAs have been applied in probing the complex enzyme mechanisms, improving catalytic activity, and even designing enzymes with new catalytic mechanisms. Here, the ncAAs were site-specifically incorporated into a lipase (PaL) produced by Pseudomonas alcaligenes to explore its diastereopreference mechanism. Menthol propionate has three chiral centers, eight isomers in total. Molecular dynamics (MD) simulations were first applied to analyze the interactions between the active sites of PaL and the target substrate L-menthol propionate. Furthermore, the four ncAAs (o-bromophenylalanine, o-chlorophenylalanine, p-cyanophenylalanine and p-aminophenylalanine) were substituted for 9 amino acids sites that potentially influenced three chiral centers and several variants with significant improvement in the diastereopreference were obtained. The diastereomer selectivity of beat variant at Ala253 was 100 % higher than that of the wild-type. A linear relationship was found between volume, flexibility of the active center and diastereoselectivity.
Multi-electrolytes activation of Lipase for 4-methylhept-4-en-1-yn-3-ol resolution in organic media

Abstract:Lipase was an important biological catalyst,which was widely applied in many areas,how to improve its catalytic performance in non-natural environment has become a research hot spot gradually. Studies showed that the specific activity and enantioselectivity of lipase could be improved remarkably by adding some electrolytes into reaction system. However,relevant researches mainly focused on the role of single electrolyte,report on the types of electrolytes involved was much less,and further systematic research has not been reported yet. By utilizing Pseudomonas cepacia lipase (PcL) as the catalyst,the asymmetric transesterification resolution of 4-methylhept-4-en-1-yn-3-ol was conducted with the precursor of chiral chrysanthemum ester pesticides in hydrophobic organic media, the influences of 8 types,including halide,nitrate,sulfate,phosphate et al,35 kinds of electrolytes and their different combinations on PcL's catalytic performance were investigated systematically. The results indicate that the specific activity of PcL is 23 times higher than that of native lipase with synergistic effect of NaF,Na2HPO4 and NaH2PO4. Further study shows that the improvement of enantioselectivity has been achieved from 11 to 21,due to the different degree of activation of enantiomers by PcL.

Abstract:A combination of site-directed mutagenesis and chemical modification was employed to alter protein structure with the objective of improving diastereopreference over that achieved by simple site-directed mutagenesis. Conformational analysis using molecular dynamic (MD) simulation of Pseudomonas alcaligenes lipase (PAL) indicated that stronger steric exclusion and structural rigidity facilitated diastereopreference. A cysteine (Cys) residue was introduced using site-directed mutagenesis to construct variant A272C. The modifier 5,5′-dithiobis-(2-nitrobenzoic acid) (DTNB) was then reacted with the introduced Cys residue to provide stronger steric exclusion and structural rigidity. The modification was verified by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry. Diastereopreference was improved significantly. The diastereomeric excess (dep) of l-menthol increased from 35% with wild type PAL to 90% with A272C-DTNB modified PAL when the conversion ratio of l-menthyl propionate was nearly 100%. Conformation and kinetic parameter analysis showed that A272C-DTNB modified PAL exhibited stronger steric exclusion and increased structural rigidity around the modification site that inhibited the hydrolysis of non-targeted substrates. The combination of site-directed mutagenesis and chemical modification could be an effective method to alter protein properties and enhance diastereopreference through the combined effect of steric exclusion and structural rigidity.

Abstract:Enantioconvergent catalysis enables the conversion of racemic molecules into a single enantiomer in perfect yield and is considered an ideal approach for asymmetric synthesis. Despite remarkable advances in this field, enantioconvergent transformations of inert tertiary C-H bonds remain largely unexplored due to the high bond dissociation energy and the surrounding steric repulsion that pose unparalleled constraints on bond cleavage and formation. Here, we report an enantioconvergent Pd-catalyzed alkylation of racemic tertiary allylic C-H bonds of α-alkenes, providing a unique approach to access a broad range of enantioenriched γ,δ-unsaturated carbonyl compounds featuring quaternary carbon stereocenters. Mechanistic studies reveal that a stereoablative event occurs through the rate-limiting cleavage of tertiary allylic C-H bonds to generate σ-allyl-Pd species, and the achieved E/Z-selectivity of σ-allyl-Pd species effectively regulates the diastereoselectivity via a nucleophile coordination-enabled SN 2'-allylation pathway.

Abstract:Isoamylase catalyzes the hydrolysis of α-1,6-glycosidic linkages in glycogen, amylopectin and α/β-limit dextrins. A semi-rational design strategy was performed to improve catalytic properties of isoamylase from Bacillus lentus . Three residues in vicinity of the essential residues, Arg505, Asn513, and Gly608, were chosen as the mutation sites and were substituted by Ala, Pro, Glu, and Lys, respectively. Thermal stability of the mutant R505P and acidic stability of the mutant R505E were enhanced. The k cat /K m values of the mutant G608V have been promoted by 49 %, and the specific activity increased by 33 %. This work provides an effective strategy for improving the catalytic activity and stability of isoamylase, and the results obtained here may be useful for the improvement of catalytic properties of other α/β barrel enzymes.

Abstract:Biocatalysts with tailor-made or controllable stereoselectivity are valuable and highly desired in the precision synthesis of chiral compounds. Most lipases display excellent (R)-enantioselectivity for various sec-alcohols. In order to reverse the inherent (R)-enantioselectivity of lipase A from Bacillus subtilis (BSLA), an important amino acid position of BSLA (H76) which can reverse the (R)-enantioselectivity to (S) in the hydrolytic kinetic resolution (KR) of various esters derived from sec-alcohols was discovered, basing on the comparison with the well-known “gourd”-like pocket of Candida antarctica lipase B (CALB). After iterative mutagenesis, the best mutant H76 A/N18C was obtained, displaying high (S)-selectivity for the model reaction (45% conv. and 92% ee). Meanwhile, a single-point mutation of N18W was also found which can remarkably improve the inherent (R)-enantioselectivity of BSLA toward sec-alcohol. A series of enantiocomplementary sec-alcohols can be prepared through the KR catalyzed by WT BSLA or mutants, and the highest enantiomeric ratio (E) value is up to >300 for (R)-enantiomer and 178 for (S)-enantiomer, respectively.

In silico Mining and Identification of a Novel Lipase from Paenibacillus larvae : Rational Protein Design for Improving Catalytic Performance

Abstract:Lipases play a vital role in various biological processes, from lipid metabolism to industrial applications. However, the ever-evolving challenges and diverse substrates necessitate the continual exploration of novel high-performance lipases. In this study, we employed an in silico mining approach to search for lipases with potential high sn -1,3 selectivity and catalytic activity. The identified novel lipase, PLL, from Paenibacillus larvae subsp. larvae B-3650 exhibited a specific activity of 111.2 ± 5.5 U/mg towards the substrate p -nitrophenyl palmitate ( p NPP) and 6.9 ± 0.8 U/mg towards the substrate olive oil when expressed in Escherichia coli ( E. coli ). Computational design of cysteine mutations was employed to enhance the catalytic performance of PLL. Superior stability was achieved with the mutant K7C/A386C/H159C/K108C (2M3/2M4), showing an increase in melting temperature ( T m ) by 1.9°C, a 2.05-fold prolonged half-life at 45°C, and no decrease in enzyme activity. Another mutant, K7C/A386C/A174C/A243C (2M1/2M3), showed a 4.9-fold enhancement in specific activity without compromising stability. Molecular dynamics simulations were conducted to explore the mechanisms of these two mutants. Mutant 2M3/2M4 forms putative disulfide bonds in the loop region, connecting the N- and C-termini of PLL, thus enhancing overall structural rigidity without impacting catalytic activity. The cysteines introduced in mutant 2M1/2M3 not only form new intramolecular hydrogen bonds but also alter the polarity and volume of the substrate-binding pocket, facilitating the entry of large substrate p NPP. These results highlight an efficient in silico exploration approach for novel lipases, offering a rapid and efficient method for enhancing catalytic performance through rational protein design.

Abstract:Over the past decades, asymmetric catalysis has been intensely investigated as a powerful tool for the preparation of numerous chiral biologically active compounds. However, developing general and practical strategies for preparation of both enantiomers of a chiral molecule via asymmetric catalysis is still a challenge, particularly when the two enantiomers of a chiral catalyst are not easily prepared from natural chiral sources. Inspired by the biologic system, we report herein an unprecedented catalytic enantiodivergent Michael addition of pyridazinones to enones by subtle adjustment of achiral amino moiety of dipeptide phosphine catalysts. These two dipeptide phosphine catalysts, P5 and P8, could deliver both enantiomers of a series of N2-alkylpyridazinones in good yields (up to 99%) with high enantioselectivities (up to 99% ee) via the catalyst-controlled enantiodivergent addition of pyridazinones to enones.

Abstract:An unprecedented enantioselective allylic alkylation of readily available aldimine esters has been developed, and is catalyzed by a synergistic Cu/Pd catalyst system. This strategy provides facile access to nonproteinogenic α,α-disubstituted α-amino acids in high yield with excellent enantioselectivity. The more challenging double allylic alkylation of glycinate-derived imine esters was also realized. Furthermore, this methodology was applied for the construction of the key intermediate of PLG peptidomimetics.
Improving Pseudomonas alcaligenes lipase’s diastereopreference in hydrolysis of diastereomeric mixture of menthyl propionate by site-directed mutagenesis

Abstract:The resolutions of racemic diastereomeric mixtures of menthyl propionate was performed by Pseudomonas alcaligenes lipase (PaL) to produce (2S, 5R) L -menthol. Because of the inherently low diastereopreference of PaL, covalent docking and molecular dynamic (MD) simulations were used to investigate possible avenues of improvement. Rational site-directed mutagenesis of PaL revealed residues V180 and A272 to be the hotspots for diastereopreference. The double V180L/A272F mutant exhibited the highest degree of diastereopreference, as the diastereomeric ratio of (2S, 5R) L -menthol increased towards both (2R, 5S) L -neomenthol ( dr1 ) and (2R, 5R) D -isoneomenthol ( dr2 ) (diastereomeric ratios dr 1 and dr 2 increased to 4.65 and 2.13 times that of wild-type PaL). MD simulation analysis indicated that these mutations decrease the flexibility of the surrounding protein regions. The combination of increased steric exclusion and decreased flexibility results in less favorable binding of the non-target substrates, (2R, 5S) L -neomenthyl propionate and (2R, 5R) D -isoneomenthyl propionate, to the V180L/A272F mutant. These results confirmed and further improved our previously proposed model of the diastereomer recognition mechanism based on the combined effect of steric exclusion and regional flexibility.
Electronic Effect‐Guided Rational Design of Candida antarctica Lipase B for Kinetic Resolution Towards Diarylmethanols

Abstract:Herein, we developed an electronic effect‐guided rational design strategy to enhance the enantio‐selectivity of Candida antarctica lipase B (CALB) mutants towards bulky pyridyl(phenyl)methanols. Compared to W104A mutant previously reported with reversed S‐stereoselectivity toward sec‐alcohols, three mutants (W104C, W104S and W104T) displayed significant improvement of S‐enantioselectivity in the kinetic resolution (KR) of various phenyl pyridyl methyl acetates due to the increased electronic effects between pyridyl and polar residues. The electronic effects were also observed when mutating other residues surrounding the stereospecificity pocket of CALB, such as T42A, S47A, A281S or A281C, and can be used to manipulate the stereoselectivity. A series of bulky pyridyl(phenyl) methanols, including S‐(4‐chlorophenyl)(pyridin‐2‐yl) methanol (S‐CPMA), the intermediate of bepotastine, were obtained in good yields and ee values.

Improvement of Catalytic Properties of Lipase from Arthrobacter Sp by Encapsulation in Hydrophobic Sol-Gel Materials

Abstract:In this work, lipase from Arthrobacter sp. was immobilized by sol-gel encapsulation to improve its catalytic properties. Various silanizing agents including vinyl-trimethoxy silane, octyl-trimethoxy silane, gamma-(methacryloxypropyl)-trimethoxy silane (MAPTMS) and tetraethoxysilane (TEOS) were chosen as the precursors. Among them, MAPTMS was for the first time utilized to encapsulate lipases, and the prepared enzyme by copolymerization of MAPTMS and TEOS exhibited the highest activity in both the hydrolysis of p-nitrophenyl palmitate and the asymmetric acylation of 4-hydroxy-3-methyl-2-(2-propenyl)-2-cyclopenten-1-one. The effects of various immobilization parameters were investigated. Under the optimum conditions of MAPTMS/TEOS=1/1 (mol/mol), water/silane molar ratio (R value)=20 and lipase loading=0.01 g/mL sol, the total activity of the immobilized enzyme reached up to 13.6-fold of the free form. Moreover, the encapsulated lipase exhibited higher thermal stability than the free form and retained 54% of the original activity after uses for 60 d. Enantioselectivity of enzyme was also improved with an E value of 150 after encapsulation from 85 for the free form.
A Calcium-Ion-Stabilized Lipase from Pseudomonas stutzeri ZS04 and its Application in Resolution of Chiral Aryl Alcohols

Abstract:An extracellular organic solvent-tolerant lipase-producing bacterium was isolated from oil-contaminated soil samples and was identified taxonomically as Pseudomonas stutzeri , from which the lipase was purified and exhibited maximal activity at temperature of 50 °C and pH of 9.0. Meanwhile, the lipase was stable below or at 30 °C and over an alkaline pH range (7.5–11.0). Ca 2+ could significantly improve the lipase thermal stability which prompts a promising application in biocatalysis through convenient medium engineering. The lipase demonstrated striking features such as distinct stability to the most tested hydrophilic and hydrophobic solvents (25 %, v / v ), and DMSO could activate the lipase dramatically. In the enzyme-catalyzed resolution, lipase ZS04 manifested excellent enantioselective esterification toward the ( R )-1-(4-methoxyphenyl)-ethanol (MOPE), a crucial chiral intermediate in pharmaceuticals as well as in other analogs with strict substrate specificity and theoretical highest conversion yield. This strong advantage over other related schemes made lipase ZS04 a promising biocatalyst in organic synthesis and pharmaceutical applications.
Enhancing Biocatalysis through Chiral Supramolecular Scaffolds: Insights into the Structural Adaptability of Lipase

Abstract:The chemoselective capabilities of porcine pancreatic lipase (PPL) in tetrahydrofuran andCandida rugosalipase (CRL) in diisopropyl ether have been investigated for selective acetylation and deacetylation of hydroxymethylated phenols and hydroxyaryl alkyl ketones and their peracetylated derivatives. Both PPL and CRL exhibited exclusive selectivity for the acetylation of alcoholic hydroxyl group over the phenolic hydroxyl group(s) of the hydroxymethylated phenols1–5and aryl alkyl ketones6–9, and for the deacetylation of ester group involving the phenolic hydroxyl group over the ester group involving alcoholic hydroxyl of the peracetates19–24. The preliminary results indicate that this strategy of chemoselective acetylation can also be used in the enantiomeric resolution of racemic ketones6–9. Single crystal X-ray diffraction studies have confirmed the structures of compounds4, 15, and17.
