Abstract:Silkworms have been developed over thousands years to optimize folding and crystallization of fibroin under highly controlled conditions which have resulted in their efficient fiber formation. In this paper, we reconstructed the three-dimensional architecture of the spinneret of a wild silkworm Samia cynthia ricini from approximately 1000 optical micrographs of the semi-thin cross sections. The chitin plates and muscles were observed in the silk press part together with large change in the diameter of the spinneret lumen at the press part by large shear stress. This is similar to the case of the spinneret of Bombyx mori silkworm, indicating that the structural change in the silk fibroin of S.c. ricini silkworm occurs exclusively at the silk press part due to large shear stresses. Molecular dynamics (MD) calculations were then performed to study the structural change that occurs in the crystalline region of S.c. ricini silk fibroin under shear stress. Namely, using the peptide AGGAGG(A)12GGAGAG as a model of the crystalline part of the silk fibroin under different shear stresses in the presence of water molecules and followed by molecular mechanics (MM) calculation after removal of water molecules. The simulation indicates that the Ala residues in the model peptides adopt a predominantly β-sheet structure under shear stresses of above 1.0GPa.

Abstract:In order to develop new silk-like materials in the form of fiber and non-woven nano-fiber, this study synthesized a new silk-like protein by selecting the sequence from the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(6), to imitate the processing condition of the silk fibroin with the combination of the sequence YGGLGSQGAGRG, the hydrophilic motif of spider dragline silk which was considered as the origin of supercontraction of spider dragline silk (Yang Z, et al. J Am Chem Soc 2000;122: 9019-25). The CD pattern of the silk-like protein in hexafluoroacetone (HFA) indicates that it takes helical structure. The solid-state structures of the silk-like protein before and after methanol treatment were determined to be random coil and Silk II structure (mainly beta-sheet structure), respectively, using C-13 CP/MAS NMR. The C-13 CP/MAS NMR spectra of the protein after methanol treatment in hydrated state showed that the random coil peaks from Ala C beta and Set C beta carbons become sharper compared with the corresponding peaks in the dry state. The 2D-WISE spectra in the hydrated state also showed increase of the narrow components for these carbons. Thus, water molecules are relatively easy to access at the random coil regions of the protein. The fiber formation of the silk-like protein was possible with wet-spinning or electrospinning methods using HFA as dope solvent and methanol as coagulation solvent. (C) 2008 Elsevier Ltd. Design, Expression and Solid-State Nmr Characterization of Silk-Like Materials Constructed from Sequences of Spider Silk, Samia Cynthia Ricini and Bombyx Mori Silk Fibroins

Abstract:Silk has a long history of use in medicine as sutures. To address the requirements of a mechanically robust and biocompatible material, basic research to clarify the role of repeated sequences in silk fibroin in its structures and properties seems important as well as the development of a processing technique suitable for the preparation of fibers with excellent mechanical properties. In this study, three silk-like protein analogs were constructed from two regions selected from among the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(2), the crystalline region of Samia cynthia ricini silk fibroin, (Ala)(12,) the crystalline region of spider dragline silk fibroin, (Ala)(6), and the Gly-rich region of spider silk fibroin, (GGA)(4). The silk-like protein analog constructed from the crystalline regions of the spider dragline silk and B. mori silk fibroins, (A(6)SCS)(8), that constructed from the crystalline regions of the S. c.ricini and B. mori silk fibroins, (A(12)SGS)(4), that constructed from and the crystalline region of S. c.ricini silk fibroin and the glycine-rich region of spider dragline silk fibroin, (A(12)SGS)(4),were expressed their molecular weights being about 36.0 kDa, 17.0 kDa and 17.5 kDa, respectively in E. coli by means of genetic engineering technologies. (A(12)-SCS)(4) and (A(12)SGS)(4) undergo a structural transition from alpha-helix to beta-sheet on a change in the solvent treatment from trifluoroacetic acid (TFA) to formic acid (FA). However, (A(6)SCS)(4) takes on the P-sheet structure predominantly on TFA treatment and FA treatment. Structural analysis was performed on model peptides selected from spider dragline and S. c.ricini silks by means of C-13 CP/MAS NAIR.

Abstract:As a natural protein polymer, Bombyx mori silk fibroin (SF) has superior characteristics such as excellent mechanical properties, biocompatibility and biodegradation which make SF to be a potential candidate in the field of biomaterials. It has been reported that the mineralized SF can be used for bone repair and regeneration in the bone tissue engineering. In order to map out the growth process of hydroxyapatite (HAp) induced by SF in the mineralized buffer solution, the co-solution of SF and supersaturated simulated body fluid (1.5 SBF) was incubated at 37.2 degrees C for 7 days and observed by using TEM, DSC, DTA and TGA in this study. The TEM results indicated that nanoscale crystal nuclei were found along the surface of SF in the initial stage of mineralization and finally these crystals aggregated into three-dimensional network. DSC and DTA results showed that the thermal stability of HAp/SF was greatly improved by increasing incubation duration and that the phase transition of HAp occurred from metastable state to steady state. TGA results suggested the organic content of HAp/SF complex was about 21% which was similar to the natural bone.

Abstract:Bombyx mori (B. mori) silk sericin is a protein with features desirable as a biomaterial, such as increased hydrophilicity and biodegradation, as well as resistance to oxidation, bacteria, and ultraviolet light. In contrast to other widely studied B. mori silk proteins such as fibroin, sericin is still unexplored as a building block for fabricating biomaterial, and thus a facile technique of processing it into a material is needed. Here, electrospinning technology was used to fabricate it into biomaterials from two forms of B. mori silk sericin with different molecular weights, one is a low (12.0 kDa) molecular sericin (LS) form and another is a high (66.0 kDa) molecular weight sericin (HS) form. Circular dichroism (CD) spectra showed that LS in hexafluoroacetone (HFA) solvent adopted a predominantly random coil conformation, whereas HS tended to form a β-sheet structure along with a large content of random coils. In addition, LS and HS in HFA solvent were found to form cylinder-like smaller nanoparticles and larger irregular aggregates before electrospinning, respectively. As a result, biomaterials based on microparticles and nanofibers were successfully fabricated by electrospinning of LS and HS dissolved in HFA, respectively. The cell viability and differentiation assay indicated that nanofibers and microparticles improved cell adhesion, growth, and differentiation, proving that the scaffolds electrospun from sericin are biocompatible regardless of its molecular weight. The microparticles, not common in electrospinning of silk proteins reported previously, were found to promote the osteogenic differentiation of mesenchymal stem cells in comparison to the nanofibers. This study suggested that molecular weight of sericin mediates its secondary structure and assembly structure, which in turn leads to a control of final morphology of the electrospun materials. The microparticles and nanofibers of sericin can be potentially used as building blocks for fabricating the scaffolds for tissue engineering.

Abstract:In this paper,we investigated the effect of different structure silk fibroin on crystallization of hydroxyapatite(HA),so as to provide the evidences for HA/silk fibroin composite used as a scaffold in the bone tissue engineering.A kind of silk fibroin(SF) with coil/helical structure was prepared through neutral salt dissolution method,and the other kind of silk fibroin(ALSF) with β-sheet structure was obtained from the former SF by treatment of alcohol solution.HA/SF and HA/ALSF composites were respectively deposited with the organic template of those two silk fibroin.The second structure,phase composition and morphology of HA,HA/SF and HA/ALSF composites were investigated by Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD) and transmission electron microscopy(TEM).These results showed that nucleation and growth of HA were affected by silk fibroin structure,which induced different morphology and particle size of HA.

Abstract:Multi-walled carbon nanotubes (MWCNTs) were homogeneously dispersed in silk fibroin (SF) solutions at different compositions, and a simple solvent-casting method was used to fabricate SF/MWCNT films. Structure, viscosity, and mechanical properties of the SF/MWCNT nanocomposites were characterized by FTIR (Fourier Transform Infrared Spectroscopy), viscometry, and tensile testing. Fibroblast cells were used to examine cell viability and attachment to nanocomposite films. Compared to a pure SF film, adding just 0.5 % (w/w) of MWCNT to the SF matrix could enhance the Young’s modulus and ultimate tensile strength by approximately 24 % and 39 %, respectively. In addition, with increasing MWCNTs content, the percentage of β -sheet structure increased significantly. When the content of MWCNTs increased from 0 to 1 % (w/w), the β -sheet content improved from 17.8 % to 53.6 %. Furthermore, adding up to 1 % (w/w) MWCNT increased the viscosity of the SF solution ten fold. From a biological point of view, no toxic effect was observed on fibroblast cells in the presence of MWCNTs after 3 and 7 days. This investigation suggests that SF/MWCNT nanocomposite films with improved mechanical properties and biocompatibility are well-suited as novel biomaterials.

Abstract:As an analog of the main protein contained in nacre regenerated Bombyx mori silk fibroin has a significant influence on the morphology and polymorphic nature of CaCO3 in the biomineralization process. A number of studies have implied that the self-assembling aggregate structure of silk fibroin is a key factor in controlling CaCO3 aggregation. Further insight into this role is necessary with a particular need to prepare silk fibroin aggregates with homogeneous structures to serve as templates for the mineralization process. Here we have prepared homogeneous silk microspheres to serve as templates for CaCO3 mineralization in order to provide an experimental insight into silk-regulated crystallization processes. CaCO3 particles with different nano- and microstructures, and their polymorphs, were successfully formed by controlling the mineralization process. The key function of silk aggregation in controlling the morphology and polymorphic nature of CaCO3 particles was confirmed. A regulating effect of silk on the spatial features was also observed. A two-step process for silk fibroin-regulated biomineralization was found, with different levels of heterogeneous nucleation and aggregation. A full understanding of silk fibroin-regulated biomineralization mechanisms would help in understanding the function of organic polymers in natural biomineralization, and provide a way forward in the design and synthesis of new materials in which organic-inorganic interfaces are the keys to function, biological interfaces and many related material features.

Abstract:Controlled deposition of calcium carbonate crystals can be obtained on degummed Bombyx mori silk fibers through the use of a silk fibroin solution; aragonite crystallites are found on the surface of the fiber with consistent orientation along the longitudinal axis; the results indicate that the combination of the ordered surface structure on the silk fiber and the directing-effect of silk fibroin solution are the key factors in the orientative deposition process of the mineral.

Abstract:Silk sericin(SS) contains a large number of carboxyl,carbonyl and other active groups,which have close relationship with biological mineralization.In this study,silkworm SS films were used as the template to study the mineral ized substance deposited on SS film after treatment with different alternate dipping mineration cycles and the change of SS film structure.Micro-morphology and structure of SS films deposited with mineralized substance was characterized by using Fourier transform infrared spectrometer(FTIR),X-ray diffraction(XRD),scanning electronic microscopy(SEM),and X-ray energy disperse spectroscopy(EDX).The results indicated that the mineralized substance could be quickly and efficiently deposited and accumulated on surface of the SS film.With the increase of alternate mineralization cycles,the amount of mineralized substance deposited on SS films increased and the crystallinity of SS film surface slightly improved.The mineralized substance was in lamellar form with cracks on it.The main component of mineralized substance was amorphous calcium phosphate.It was shown that alternately dipping SS film in CaCl2 and Na2HPO4 solutions and increase of mineralization cycles can promote the depo-sition of amorphous calcium phosphate on SS film sur-face,being promising in developing sericin/calcium phosphate salt composite scaffold material for bone tissue engineering.

Abstract:Mollusk shell is one of the best studied of all calcium carbonate biominerals. Its silk-like binder-matrix protein plays a pivotal role during the formation of aragonite crystals in the nacre sheets. Here, we provide novel experimental insights into the interaction of mineral and protein compounds using a model system of reconstituted Bombyx mori silk fibroin solutions serving as templates for the crystallization of calcium carbonate (CaCO3). We observed that the inherent (self-assembling) aggregation process of silk fibroin molecules affected both the morphology and crystallographic polymorph of CaCO3 aggregates. This combination fostered the growth of a novel, rice-grain-shaped protein/mineral hybrid with a hollow structure with an aragonite polymorph formed after ripening. Our observations suggest new hypotheses about the role of silk-like protein in the natural biomineralization process, but it may also serve to shed light on the formation process of those 'ersatz' hybrids regulated by artificially selected structural proteins.

Abstract:Silk is extensively investigated in bone tissue engineering due to its extraordinary mechanical properties and ability to regulate biomineralization. Protein templates regulate biomineralization process through chemical interaction with ions. However, the effect of structural differences in silk fibroin on biomineralization has not been studied in detail. In this study, Antheraea pernyi silk fibroin (ASF) and Bombyx mori silk fibroin (BSF) fibers were used as templates to study the effect of silk species on biomineralization. The results showed that silk fibroin could induce the formation of calcium-deficient hydroxyapatite in simulated body fluid (SBF), and the SBF treatment resulted in the formation of silk I crystals. Compared with BSF, ASF exhibited a higher ability to induce mineralization, which may depend on the differences in hydrophilic amorphous fractions between ASF and BSF. The amorphous fractions of ASF contain more acidic amino acids, which can provide more nucleation sites in the initial stage of mineralization, resulting in faster mineralization process and more mineral deposits. This study decodes the key role of silk fibroin fractions on biomineralization, and provides deeper insights for the study of silk fibroin as biomineralization template and bone repair materials.

Abstract:Silk fibers have many inherent properties that are suitable for their use in biomaterials. In this study, the silk fibroin was genetically modified by including a Ca-binding sequence, [(AGSGAG)6ASEYDYDDDSDDDDEWD]2 from shell nacreous matrix protein. It can be produced as fibers by transgenic silkworm. The Ca-binding activity and mineralization of the transgenic silk fibroin were examined in vitro. The results showed that this transgenic silk fibroin had relatively higher Ca-binding activity than unmodified silk fibroin. The increased Ca-binding activity could promote the usage of silk fibroin as a biomaterial in the pharmaceutical industry. This study shows the possibility of using silk fibroin as a mineralization accelerating medical material by generating genetically modified transgenic silkworm.

Abstract:Abstract Fibroins’ transition from liquid to solid is fundamental to spinning and underpins the impressive native properties of silk. Herein, we establish a fibroin heavy chain fold for the Silk-I polymorph, which could be relevant for other similar proteins, and explains mechanistically the liquid-to-solid transition of this silk, driven by pH reduction and flow stress. Combining spectroscopy and modelling we propose that the liquid Silk-I fibroin heavy chain (FibH) from the silkworm, Bombyx mori , adopts a newly reported β-solenoid structure. Similarly, using rheology we propose that FibH N-terminal domain (NTD) templates reversible higher-order oligomerization driven by pH reduction. Our integrated approach bridges the gap in understanding FibH structure and provides insight into the spatial and temporal hierarchical self-assembly across length scales. Our findings elucidate the complex rheological behaviour of Silk-I, solutions and gels, and the observed liquid crystalline textures within the silk gland. We also find that the NTD undergoes hydrolysis during standard regeneration, explaining key differences between native and regenerated silk feedstocks. In general, in this study we emphasize the unique characteristics of native and native-like silks, offering a fresh perspective on our fundamental understanding of silk-fibre production and applications.

Abstract:Significant difference in microscopic structure of cocoon filaments from three silkworm varieties which shows extremely fine,coarse or normal size of silk were obseved in the present paper by scanning electron microscope.Results attained in this study showed that the hybrid Xinmiao×Mingri was not only much thicker in filament size but also better in tensile strength,which were confirmed by the physical properties of raw silk and fabric test.

Abstract:As an analogue of the main protein contained in naturally formed nacre, reconstituted silk fibroin (SF) from the Bombyx mori silkworm silk shows a strong preference for the formation of the aragonite form of CaCO3 crystals and allows fine control over their size and morphology. The aragonite phase could be generated via two different routes: direct growth or dissolution and recrystallization, depending on the concentration of Ca2+ and SF. Generally, lower concentrations of Ca2+ and SF favor the formation of aragonite needles and their aggregates, of which the lattice structure of the precursor is similar to that of the organic matrix in natural shell. Higher concentrations lead to the formation of aragonite aggregates via a dissolution and recrystallization process through intermediates of lens-like vaterite. Molecular modeling shows that the beta-strand conformers of silk fibroin molecules has an excellent match with the ionic spacing in the aragonite (010) plane, which can promote growth along the (001) long axis of aragonite crystals. This synergy between silk fibroin and the aragonite phase may help our understanding of the function of organic matrices involved in the biomineralization process, and facilitate the fabrication of synthetic materials with the potential for high performance mechanical properties.
Effect of Metallic Ions on Silk Formation in the Mulberry Silkworm, Bombyx Mori.

Abstract:A protein conformation transition from random coil and/or helical conformation to beta-sheet is known to be central to the process used by silk-spinning spiders and insects to convert concentrated protein solutions to tough insoluble threads. Several factors including pH, metallic ions, shear force, and/or elongational flow can initiate this transition in both spiders and silkworms. Here, we report the use of proton induced X-ray emission (PIXE), inductively coupled plasma mass spectroscopy (ICP-MS) and atomic adsorption spectroscopy (AAS) to investigate the concentrations of six metal elements (Na, K, Mg, Ca, Cu, and Zn) at different stages in the silk secretory pathway in the Bombyx mori silkworm. We also report the use of Raman spectra to monitor the effects of these six metallic ions on the conformation transition of natural silk fibroin dope and concentrated regenerated silk fibroin solution at concentrations similar to the natural dope. The results showed that the metal element contents increased from the posterior part to the anterior part of silk gland with the exception of Ca which decreased significantly in the anterior part. We show that these changes in composition can be correlated with (i) the ability of Mg2+, Cu2+, and Zn2+ to induce the conformation transition of silk fibroin to beta-sheet, (ii) the effect of Ca2+ in forming a stable protein network (gel), and (iii) the ability of Na+ and K+ to break down the protein network.
