Abstract:In this study, a relatively facile porous magnetic hollow fiber engineering electrospinning method is demonstrated, which modulates fiber morphology based on secondary solvents (at variable temperatures). To demonstrate this, polycaprolactone (PCL) polymer and iron oxide nanoparticles (NPs) were used as the fibrous composite matrix. Fiber pore size increased with increasing immersed secondary solvent temperature. By contouring the surface morphology (via modulation of secondary collection solvents) of hollow magnetic fibers, drug (ketoconazole) release kinetics from spun mats were tuned. Furthermore, applying an external AC magnetic field to NP embedded porous fibers enhanced drug release. These findings are promising for alternative engineering, tuning and controlling fiber morphology and drug release behavior. (C) 2017 Elsevier B.V. All rights reserved.
Fabrication of Patterned Three-Dimensional Micron Scaled Core-Sheath Architectures for Drug Patches

Abstract:Recent advances in selective integration of micro and nano-scaled features towards material design have paved way to enhance desirable properties or functions of biomaterials. For drug delivery applications these include improved active component encapsulation, controlled drug release and managed interaction with the intended host environment. Electrohydrodynamic (EHD) direct-printing technique is a one-step on demand fiber deposition method which enables precise micron-scaled topographic and structural enhancement during material fabrication. In this study, core-sheath composite fibers comprising polycaprolactone, polyvinyl pyrrolidone and the drug tetracycline hydrochloride were prepared using the coaxial format of EHD direct-printing. Once positioned and aligned; multi-stacked fibers gave rise to patches. Coaxial fiber (diameter range similar to 13-25 mu m) optimization (deposition and integrity) involved parameter-structure (e.g. collector speed, flow rate, working distance and applied voltage) impact analysis. Water contact angle measurements, tensile testing and Fourier transform infrared spectroscopy were used to analyze core-sheath integrated patches. In-vitro drug release studies clearly elude to the impact of core-shell and patterned architectures; demonstrating their viability and the forming method as emerging tools for advanced drug delivery system design and fabrication.

Abstract:Within the past few years electrospinning has been paid much attention due to the increasing interests of the researchers in the nanotechnology and the unique properties of the nanomaterials. This review focused on the electrospinning behaviors of biomacromolecules including synthetic biomedical polymers, natural-occurring biopolymer and the composite nanofibrous membranes. The electrospinning parameters of the biomacromolecules and the properties of the electrospun membranes such as the morphological stability, mechanical properties and degradation behavior were discussed in details. In addition, the application of the electrospun membranes for controlled drug release was also included. The incorporation method and the release behavior of the drugs were discussed and summarized, according to the solubility of the drugs.

Abstract:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.

Abstract:Coaxial electrospinning is a robust technique for one-step encapsulation of fragile, water-soluble bioactive agents, including growth factors, DNA and even living organisms, into core–shell nanofibers. The coaxial electrospinning process eliminates the damaging effects due to direct contact of the agents with organic solvents or harsh conditions during emulsification. The shell layer serves as a barrier to prevent the premature release of the water-soluble core contents. By varying the structure and composition of the nanofibers, it is possible to precisely modulate the release of the encapsulated agents. Promising work has been done with coaxially electrospun non-woven mats integrated with bioactive agents for use in tissue engineering, in local delivery and in wound healing, etc. This paper reviews the origins of the coaxial electrospinning method, its updated status and potential future developments for controlled release of the class of fragile, water-soluble bioactive agents.

Abstract:Application of electrospun fibers for the purpose of loading and controlled release of water-soluble drugs remains a challenge due to their low carrying effect as well as quick and unstable drug release. In this study, we have developed a novel nanogel-electrospinning technology through which more stable loading and prolonged release of water-soluble drugs was achieved. In brief, nanogel particles synthesized from a chloroquine (CQ)-loaded bovine serum albumin (BSA) solution were prepared and then combined with genipin, a crosslinking agent. The nanogel solution was then crosslinked to prepare an electrospinning solution with an inner mesh structure. Finally, the microfibrous membranes were fabricated by electrospinning the solution. Uniform BSA nanogel particles were wrapped in the fiber membrane and the number of particles increased with the increase of BSA and genipin concentrations. In addition to being loaded within the BSA nanogel particles, CQ was distributed in the fibers as well, which could be clearly identified using ultraviolet-visible spectroscopy (UV-Vis). The physical, chemical, and mechanical properties of nanogel-electrospun microfibers were similar to those of microfibers formed through a conventional electrospinning approach. The drug release tests indicated that with the same number of BSA nanogel particles, increased CQ loading resulted in increased initial release of the same. The duration of a single drug release cycle lasted up to 40 days. In conclusion, findings from this study have indicated that nanogel-electrospinning is a convenient and effective technology to achieve controlled long-term release of water-soluble drugs.

Abstract:A one-step, mild procedure based on coaxial electrospinning was developed for incorporation and controlled release of two model proteins, BSA and lysozyme, from biodegradable core-shell nanofibers with PCL as shell and protein-containing PEG as core. The thickness of the core and shell could be adjusted by the feed rate of the inner dope, which in turn affected the release profiles of the incorporated proteins. It was revealed that the released lysozyme maintained its structure and bioactivity. The current method may find wide applications for controlled release of proteins and tissue engineering.

Abstract:In this paper, we prepared composite fibers via electrospinning from either W/O or O/W emulsion. SEM images demonstrate the beads-in-string structures in these fibers and proved this technique to be an effective method for microencapsulation. As a practical application, Ca-alginate microspheres, which serve as reservoirs for hydrophilic drugs, were prepared in a reverse emulsion and then incorporated into poly (L-lactic acid) (PLLA) fibers by electrospinning. With the bovine serum albumin (BSA) loaded into the microspheres, the beads-in-string structure thus entrapped hydrophilic proteins in hydrophobic polymeric matrix. In the in vitro release test, BSA, which was released from composite fibers, achieved prolonged release profiles and lower burst release rates than those from naked Ca-alginate microspheres. In comparison with other well-established techniques to prepare microcapsules, such as solvent evaporation and spray-drying techniques, emulsion electrospinning features partly competing, partly complementary characteristics. Extension to other emulsion systems will be able to fabricate new types of functional structures.

Abstract:Electrospun zein membranes were prepared using DMF as solvent. By changing the solution concentration, the electrospinning voltage and the distance between the spinneret and collector, nanofibrous meshes without bead defects could be obtained. In order to improve the mechanical strength of the hydrated zein meshes, core-shell-structured nanofibrous membranes with PCL as the core material and zein forming the shell were prepared by coaxial electrospinning. The core-shell structure of the composite fibers was confirmed by SEM characterization of the fibers, either extracted with chloroform to remove the inner PCL, or elongated to expose their cross-section. The composition and average diameter of the composite fibers could be modulated by the feed rate of the inner PCL solution. It was found that the core-shell fibrous membranes have similar wettability to the electrospun zein mesh. The presence of PCL in the fibers could significantly improve the mechanical properties of the zein membrane.
Feedback System Control Optimized Electrospinning for Fabrication of an Excellent Superhydrophobic Surface

Abstract:Superhydrophobic surface, as a promising micro/nano material, has tremendous applications in biological and artificial investigations. The electrohydrodynamics (EHD) technique is a versatile and effective method for fabricating micro- to nanoscale fibers and particles from a variety of materials. A combination of critical parameters, such as mass fraction, ratio of N, N-Dimethylformamide (DMF) to Tetrahydrofuran (THF), inner diameter of needle, feed rate, receiving distance, applied voltage as well as temperature, during electrospinning process, to determine the morphology of the electrospun membranes, which in turn determines the superhydrophobic property of the membrane. In this study, we applied a recently developed feedback system control (FSC) scheme for rapid identification of the optimal combination of these controllable parameters to fabricate superhydrophobic surface by one-step electrospinning method without any further modification. Within five rounds of experiments by testing totally forty-six data points, FSC scheme successfully identified an optimal parameter combination that generated electrospun membranes with a static water contact angle of 160 degrees or larger. Scanning electron microscope (SEM) imaging indicates that the FSC optimized surface attains unique morphology. The optimized setup introduced here therefore serves as a one-step, straightforward, and economic approach to fabricate superhydrophobic surface with electrospinning approach.

Abstract:Aqueous microdroplets are of significant interest in biological fields, while the employment of water-in-oil (w/o) emulsion and lack of spatial control precluded its widespread application. Herein a novel microfluidic approach is developed to generate water-in-water (w/w) microdroplets embedded in hydrogel microfibers. Aqueous two phase system (ATPS) is applied to generate w/w droplets, and alginate is introduced to continuous phase to form microfibers, which offers spatial restriction and manipulation possibility to droplets. The size and pattern of aqueous droplets can be precisely controlled, and immobilization within hydrogel fiber facilitates easy manipulation and observation. The microdroplets surrounded by hydrophilic environment can act as a cell-cell interaction model, and their potential for biological and environmental applications are demonstrated by long-term culture of encapsulated cells and water remediation of Bacillus subtilis.

Abstract:This study leverages the ancient craft of weaving to prepare membranes that can effectively treat oil/water mixtures, specifically challenging nanoemulsions. Drawing inspiration from the core-shell architecture of spider silk, we have engineered fibers, the fundamental building blocks for weaving membranes, that feature a mechanically robust core for tight weaving, coupled with a CO 2 -responsive shell that allows for on-demand wettability adjustments. Tightly weaving these fibers produces membranes with ideal pores, achieving over 99.6% separation efficiency for nanoemulsions with droplets as small as 20 nm. They offer high flux rates, on-demand self-cleaning, and can switch between sieving oil and water nanodroplets through simple CO 2 /N 2 stimulation. Moreover, weaving can produce sufficiently large membranes (4800 cm 2 ) to assemble a module that exhibits long-term stability and performance, surpassing state-of-the-art technologies for nanoemulsion separations, thus making industrial application a practical reality.

Abstract:A core-sheath structure is one of the methods developed to overcome the challenges often faced when using monolithic fibers for drug delivery. In this study, fibers based on polyvinylpyrrolidone (core) and ethyl cellulose (sheath) were successfully produced using a novel core-sheath pressure-spinning process. For comparison, these two polymers were also processed into as blend fibers. All samples were then investigated for their performances in releasing water-soluble ampicillin (AMP) and poorly water-soluble ibuprofen (IBU) model drugs. Scanning electron,digital and confocal microscopy confirmed that fibers with a core-sheath structure were successfully made. Fourier transform infrared spectroscopy showed the success of the pressure-spinning technique in encapsulating AMP/IBU in all fiber samples. Compared to blend fibers, the core-sheath fibers had better performance in encapsulating both water-soluble and poorly water-soluble drugs. Moreover, the core-sheath structure was able to reduce the initial burst release and provided a better sustained release profile than the blend fiber analog. In conclusion, the pressure-spinning method was capable of producing core-sheath and blend fibers that could be used for the loading of either hydrophilic or hydrophobic drugs for controlled drug delivery systems.

Abstract:Electrospun nanofibers have been extensively studied for encapsulated drugs releasing from the inside of the fiber matrix, but have been barely looked at for their potential to control release as a semi-permeable membrane. This study investigated molecular transport behaviors across nanofiber membranes with different micro-structure sizes and compositions. Four types of membranes were made by 5% and 10% poly (ε-caprolactone) (PCL) solutions electro-spun with or without 50 nm calcium carbonate (CaCO3) nanoparticles. The membranes were tested for thickness, fiber diameter, pore size, porosity, tensile strength and elongation, contact angle of water and their impacts on molecular transport behaviors. The presence of the CaCO3 nanoparticles made the 5% membranes stronger and stiffer but the 10% membranes weaker and less stiff due to the different (covering or embedded) locations of the nanoparticles with the corresponding fibers. Solute transport studies using caffeine as the model drug found the 5% membranes further retarded release from the 10% membranes, regardless of only half the amount of material being used for synthesis. The addition of CaCO3 nanoparticles aided the water permeation process and accelerated initial transports. The difference in release profiles between 5% and 10% membranes suggests different release mechanisms, with membrane-permeability dominated release for 5% PCL membranes and solute-concentration-gradient dominated release for 10% PCL membranes.

Evolution of Core–shell Structure: from Emulsions to Ultrafine Emulsion Electrospun Fibers

Abstract:Due to the relative ease for fabricating fibers with a core–shell structure, emulsion electrospinning has been investigated intensively for making nanofibrous delivery vehicles for local and sustained release of biomolecules such as growth factors. In this investigation, water-in-oil (w/o) emulsions consisting of deionized (DI) water or phosphate buffer saline (PBS), poly(lactic-co-glycolic acid) (PLGA) solution and a surfactant were electrospun into fibers for studying the evolution of core–shell structures in emulsion electrospun fibers. Optical microscopy, high speed imaging, fluorescence microscopy and transmission electron microscopy were employed to examine the morphology of the water phase in fibers at different locations of the jet (or fiber) trajectory during emulsion electrospinning. The results indicated that the water phase in emulsion jets underwent multi-level stretching and breakup. When the jet (or fiber) diameter was large (i.e., for the jet near the Taylor cone), the water phase within the jet (or fiber) exhibited a discrete morphology whereas in ultrafine fibers collected beyond certain distance from the Taylor cone, a mostly continuous water phase core was formed. The Rayleigh/capillary instability determined the breakup point of the deformed water phase. When the water phase in emulsions for electrospinning was changed from DI water to PBS, it changed from a continuous state to a discontinuous state in electrospun nanofibers.
The bubble electrostatic spraying a new technology for fabrication of superhydrophobic nanofiber membranes

Abstract:Researchers are excited about the latest advances in the long needle electrospinning and the bubble electrospinning, which have triggered wide-spread concern. This paper offers a new angle for modifying both methods, the former is developed into a modified one with an auxiliary helix needle, which is used for fabrication of super-hydrophobic polyvinylidene difluoride-copolypolyhexafluoropropylene nanofiber membrane (PH-E membrane for short), and the latter is extended to a bubble electrostatic spaying, which is used for spraying PDMS microspheres on the PH-E membrane surface, and the resultant membrane is called as the PDMS-PH-E membrane. Both membranes hydrophobicity, surface roughness, porosity, and wetting property are measured and compared, and the drop impact dynamical property of the PDMS-PH-E membrane has opened the path for a new way to design of self-clearing and anti-fouling and anti-wetting surfaces. Far-reaching applications of the membranes include energy harvesting devices and sensors. We anticipate this article to be a starting point for more sophisticated study of the bubble electrostatic spaying and PDMS-PH-E membrane advanced applications.

Abstract:Herein, it is demonstrated that coaxial electrohydrodynamic atomization can be used for the fabrication of microspheres with distinct core/shell structure. This allows the encapsulation of two different types of drugs in different compartments in one single step. In Group A, we prepared microspheres in which the core and the shell contain hydrophobic and hydrophilic drugs, respectively. In contrast, in Group B, the opposite is prepared. While the former can be achieved by using amphiphilic polymers in aqueous environment, the latter is difficult to be prepared. The release patterns of the two groups are significantly different. The release of drugs from Group A microspheres is rather sequential, whereas group B microspheres release drugs in a parallel (co-release) manner. Nevertheless, in both groups, we found that the release of drugs can be easily tailored by altering outer/inner flow ratios. These findings present the advantages and possible application of this multi-drug release system in chemotherapy. Moreover, cell culture experiments have been performed to testify the performances of different microspheres in cytotoxicity and cellular apoptosis in vitro.

Abstract:Electrospinning is a popular technique for constructing nanofibrous tissue engineering scaffolds. Electrospinning is also amenable to the incorporation of drugs or biomolecules in fibers, which can provide local and sustained delivery of biological signals, such as growth factors, for the seeded cells. Drugs can normally be dissolved in polymer solutions for electrospinning, forming nanofibrous drug delivery systems. However, simply blending biomolecules in polymer solutions can result in denaturation of biomolecules and large initial burst release. Therefore, emulsion electrospinning, which can provide protection for biomolecules during electrospinning, is of great interest. In this investigation, biomolecule-containing scaffolds were emulsion electrospun using bovine serum albumin (BSA) as the model protein. Two polymers, poly (lactic-co-glycolic acid) and poly (D,L-lactic acid), were used due to their different degradation characteristics. Nanofibers with core-shell structures were electrospun from water-in-oil emulsions formulated by polymer solution, BSA-containing deionized water and a surfactant. By changing the polymer concentration and water phase volume, the fiber diameter and core-shell structure were varied. With different polymers and different fiber structures, the in vitro BSA release behaviours from fibrous scaffolds were different.

Abstract:Physically crosslinked microscale biomembranes synthesized from pure chitosan are designed and demonstrated for pH-triggered release of embedded functionalized mesoporous silica nanoparticles. Nanoparticle-loaded membranes are formed in a microfluidic channel at the junction between accurately controlled co-flowing streams to achieve highly tuneable membrane properties. After formation, the loaded membranes remain stable until contact with physiological acidic conditions, resulting in controlled nanoparticle release. Furthermore, nanoparticle-loaded membranes with complex layered architectures are synthesized using different flow schemes, thus enabling customized nanoparticle release profiles. These novel materials are well-suited for integration within small medical devices as well as off-chip applications.
