Abstract:Linear and miktoarm star-shaped diblock copolymers consisting of single-stranded DNA and poly(N-isopropylacrylamide) (PNIPAAm) with various compositions were synthesized via atom transfer radical polymerization and click chemistry. The temperature-responsive phase transition behavior, micellization, was systematically examined using UV vis spectrometry, high-sensitivity differential scanning calorimetry, dynamic light scattering, and small-angle X-ray scattering. The lower critical solution temperature (LCST) increased, and its enthalpy decreased with decreasing PNIPAAm content The copolymers self-assembled into well-defined nanoparticles having a core composed of PNIPAAm and a coronal layer of DNA above LCST. The particle size and micellar aggregation number of copolymer chains depended on the macromolecular composition and chain architecture. On the other hand, regardless of their factors, the surface area occupied by one DNA strand was found to be almost unchanged. The hybridization of DNA on the nanoparticles with fully complementary one induced the aggregation of the particles in a non-cross-linking configuration. The nanoparticle composed of miktoarm star copolymer showed a quicker DNA-hybridization response in this non-cross-linking aggregation compared with the case of a linear analogue.

Abstract:A series of pH-sensitive and thermosensitive amphiphilic pentablock copolymers, PNIPAM(x)-b-P(tBA-co-AA)(90)-bPPO(36)-b-P(tBA-co-AA)(90)-PNIPAM(x) (x-DH) with different lengths x of PNIPAM blocks (x = 58, 95, and 111) and various degrees of hydrolysis of PtBA blocks (DH approximate to 10, 60, and 100%), were prepared and self-assembled into micelles in dilute aqueous solutions. By dynamic light scattering (DLS) and UV-vis spectrophotometry characterizations, at pH = 2, x-DH micelle aqueous solutions show an irreversible thermosensitive process due to the hydrogen (H) bonding between PNIPAM blocks and protonated AA groups. At pH = 12, x-DH micelle aqueous solutions show a reversible thermosensitive process when the DH is low (10%) but are thermoinsensitive when the DH is high (similar to 60 and 100%) due to the electrostatic repulsion between deprotonated AA groups. The morphologies and structures of x-DH micelles are directly visualized by cryogenic transmission electron microscopy (cryo-TEM) at 20 degrees C. Under acid conditions, with increasing hydrophilicity, the transformation of x-DH micelles from large compound micelles (LCMs) with a bent corona, through LCMs with extended corona, core-corona irregularities, core-shell-corona spheres, coexistence of core-shell-corona spheres and core-corona cylinders, core-corona cylinders, and coexistence of core-corona spheres and core-corona cylinders, to core-corona spheres is identified. Under alkaline conditions, the deprotonated AA groups introduce electrostatic repulsion into micelles, and four nanostructures of x-DH micelles, i.e., LCMs with an uneven surface and inhomogeneous core, core-corona spheres, core-corona cylinders, and core-shell-corona spheres, are observed.
Thermosensitive Self-Assembly Behaviors of Novel Amphiphilic Polyphosphazenes

Abstract:A series of amphiphilic thermosensitive polyphosphazenes (PNIPAm-g-PPP) bearing N-isopropylacrylamide oligomers (oligo-PNIPAm) and glycine ethyl groups (GlyEt) as co-substituents have been synthesized via polymer substitute reaction. UV-visible spectra indicated that the aqueous solution of PNIPAm-g-PPP exhibited the lower critical solution temperature (LCST). Furthermore, the LCST was seriously influenced by the substitution ratios of PNIPAm to GlyEt in the copolymer. The more GlyEt the copolymer contained, the lower LCST it had. The critical association concentration (cac) of copolymers was determined by fluorescence probe method. It was found that cac was decreased with increasing GlyEt content of polyphosphazene. Also the formation of self-assembled micelles or nano-particles was confirmed by TEM.

Abstract:A novel thermosensitive amphiphilic copolymer of PNIPAm grafted polyphosphazene (PNIPAm-g-PPP) has been synthesized through a co-substitution reaction of poly(dichlorophosphazene) with amino terminated NIPAm oligomer and glycine ethyl ester (GlyEt). The polymer obtained had a molar ratio of 1:5.25 PNIPAm to GlyEt and had a lower critical solution temperature (LCST) near 30 degreesC. The formation of polymeric micelles was confirmed by fluorescence, dynamic light scattering and transmission electron microscopy measurements.

Abstract:A series of linear thermosensitive amphiphilic pentablock terpolymers PNIPAM(x)-b-PtBA(90)-b-PPO36-b-PtBA(90)-b-PNIPAM(x) (N(x)T(90)o(36)T(90)N(x)) with various lengths x of PNIPAM block (x = 111, 107, 95, 58, 33, and 26) were synthesized via a two-step atom transfer radical polymerization (ATRP) using modified poly(propylene oxide) (PPO36) as the macroinitiator, tert-butyl acrylate (tBA) as the first monomer, and N-isopropylacrylamide (NIPAM) as the second monomer. The NxT90O36T90Nx pentablock terpolymers form micelles in dilute aqueous solution, whose structure and morphology are dependent on the length x of PNIPAM block and solution temperature. At 14 degrees C, below the lower critical solution temperature (LCST) of PNIPAM block, the morphology of NxT90O36T90Nx micelles changes from core corona spherical micelles with homogeneous hydrophobic core to rodlike micelles, core-shell-corona spherical micelles with clear boundaries, core shell corona spherical micelles with blurry boundaries, and finally large compound spherical micelles with embedded hydrophilic chains in the hydrophobic region when x decreases from 111 to 26. This morphology change might be attributed to the different stretching factors of hydrophobic PPO and PtBA blocks in the micelles, which varied differently with length x of PNIPAM block and hence resulted in the change of compatibility of the two hydrophobic blocks. A scaling relationship between the length L-0 of hydrophilic corona and the length x of PNIPAM block is found to be L-0 similar to x(0.77 +/- 0.08), which falls in the range 0.5-0.86 theoretically predicted for the micelles of diblock copolymers. The size of hydrophobic regions increases with decreasing x. At 60 degrees C above the LCST of PNIPAM block, all NxT90O36T90Nx pentablock terpolymers form spherical micelles with uniform structure and size distribution in aqueous solution regardless of the length x of PNIPAM block.

Abstract:Acrylonitrile (AN) was copolymerized with N-isopropylacrylamide (NIPA) to synthesize thermosensitive hydrogels, and the on-off switch behavior of poly(NIPA-co-AN) hydrogels with different fraction of hydrophobic component (AN) was investigated. It is found that the lower critical solution temperature (LCST), the swelling ratio at certain temperature and the reswelling rate of poly(NIPA-co-AN) hydrogels decreased as AN unit fraction in copolymers increased. In order to improve the responsive rate of poly(NIPA-co-AN) hydrogels, they were further treated by surface crosslinking using N,N'-methylene bisacrylamide (BIS) as a crosslinking agent. The swelling and deswelling behaviors of these copolymers were compared with those of the untreated hydrogels. The results indicated that the responsive rate of poly(NIPA-co-AN) hydrogel was improved by surface crosslinking. The resulting hydrogels bearing cyano groups with fast response have potential applications in the field of drug-controlled release and immobilization of biomolecules.

Abstract:The assembly morphologies of amphiphilic graft polyphosphazene containing oligopoly (N-isopropyl-acrylamide) and ethyl 4-aminobenzoate as co-side groups (PNIPAm/EAB-PPPs) with different hydrophobic/hydrophilic balance, were illustrated. Multimorphological assemblies ranging from network structure and nanospheres to high-genus particles were observed for the copolymer containing a relatively high EAB content. It was observed that temperature induced morphological changes of aggregates, which was resulted from the thermosensitivity of PNIPAm segments in the copolymers, The results show that self-assembly morphologies of PNIPAm/EAB-PPPs are regulated along with the variation of copolymer composition.

Abstract:A series of poly(N-isopropylacrylamide-co-N-hydroxymethylacrylamide) P(NIPAM-co-NHMA) copolymers were firstly synthesized via free radical polymerization. Then, the hydrophobic, photosensitive 2-diazo-1,2-naphthoquinone (DNQ) molecules were partially and randomly grafted onto P(NIPAM-co-NHMA) backbone through esterification to obtain a triple-stimuli (photo/pH/thermo) responsive copolymers of P(NIPAM-co-NHMA-co-DNQMA). UV-vis spectra showed that the lower critical solution temperature (LCST) of P(NIPAM-co-NHMA) ascended with increasing hydrophilic comonomer NHMA molar fraction and can be tailored by pH variation as well. The LCST of the P(NIPAM-co-NHMA) went down firstly after DNQ modification and subsequently shifted to higher value after UV irradiation. Meanwhile, the phase transition profile of P(NIPAM-co-NHMA-co-DNQMA) could be triggered by pH and UV light as expected. Thus, a triple-stimuli responsive copolymer whose solution propel-ties could be, respectively, modulated by temperature, light, and pH, has been achieved. Abstract:The chain conformations and adsorption behaviors of four thermo-sensitive poly(N-isopropylacrylamide)(x) poly(propylene oxide)(36)-poly(N-isopropylacrylamide)(x) (PNIPAmx-PPO36-PNIPAmx) triblock copolymers with x values of 15, 33, 75, and 117 in dilute aqueous solutions were investigated by combined techniques of micro-differential scanning calorimetry (micro-DSC), static and dynarhic light scattering (SLS & DLS), and the quartz crystal microbalance (QCM). PNIPAm15-PPO36-PNIPAm15 only exhibited the lower critical solution temperature (LCST) of the PPO block, i.e. 25 degrees C, because the PNIPAm block with x = 15 was too short to maintain its own LCST. With middle lengths x of 33 and 75, the LCSTs of PPO and PNIPAm blocks were observed, respectively. For the longest PNIPAm block with x = 117, only LCST of PNIPAm block dominated, i.e. 32.3 degrees C. DLS results revealed that the four PNIPAmx-PPO36-PNIPAmx triblock copolymers formed "associate" structures in their dilute aqueous solutions at 20 degrees C, which was well below the LCSTs of the PPO and PNIPAm blocks. QCM results indicated that the adsorption time constant decreased with increasing adsorption temperature but tended to increase with increasing length x of the PNIPAm block. A complex adsorption behavior with large adsorption amounts was only observed at the corresponding LCST of the PNIPAm block for PNIPAmx-PPO36-PNIPAmx with longer PNIPAm blocks with x = 33, 75, and 117. Furthermore, the adsorbed PNIPAmx-PPO36-PNIPAmx layers obtained at 20 degrees C were rigid with less energy dissipation.

Abstract:Grafting of poly(N-isopropylacrylamide) (PNIPAAm) having carboxylic groups at one end onto poly(allylamine) (PAH) in the presence of water soluble carbodiimide has yielded PAH-g-PNIPAAm copolymers with grafting ratios of 50, 29 and 18, respectively. These thermosensitive copolymers exhibit a lower critical solution temperature (LCST) at 34°C at a temperature increase cycle regardless of their grafting ratios, a temperature identical to that of PNIPAAm-COOH oligomers. Temperature cycling reveals completely reversible polymer aggregation and dissolution above and below the LCST, respectively. Much smaller particle sizes are observed by scanning force microscopy and transmission electron microscopy compared to dynamic light scattering. A porous sphere model is suggested to depict the structure of the particles formed above the LCST, by which the dependence of the particle sizes on their grafting ratios is interpreted taking into account the surface tension and the spatial aggregation distance. Finally, to demonstrate the capability of the copolymers being used as thermosensitive polyelectrolytes, assembly onto multilayers is conducted and the increase of layer thickness is confirmed by small angle X-ray scattering and ellipsometry characterizations.
Charged End-Group Terminated Poly(N-Isopropylacrylamide)-B-Poly(Carboxylic Azo) With Unusual Thermoresponsive Behaviors

Abstract:We report unusual thermoresponsive behaviors of one poly(N-isopropylacrylamide) (PNIPAm)-based diblock copolymer containing carboxylic azo moieties due to the introduction of one charged end-group. On heating, the ionized end-group prevented macroscopic phase separation of the block copolymer aqueous solution under basic conditions, leading to the absence of cloud point. The micelle solution showed a quick micelle-to-vesicle morphological transition via the end-group stabilized large micelles as a transition state when being stored at 50 degrees C for 2 h. Such a quick micelle-to-vesicle transition can be attributed to the joint effect of the charged end-group stabilization and the changed hydrophilic-hydrophobic balance of PNIPAm. When adding a tiny amount of Ca2+, macroscopic phase transition reoccurred by breaking the end-group protection layer. This study represents a unique separation of microscopic dehydration and macroscopic phase transition, providing a new way for design of stimuli-responsive polymers.
Effect of Structural Constraint on Dynamic Self-Assembly Behavior of PNIPAM-based Nonlinear Multihydrophilic Block Copolymers

Abstract:The effects of structural constraint on the thermosensitivity and dynamic self-assembly behaviors of PNIPAM-based nonlinear multihydrophilic block copolymers (MHBCs) are investigated by a combination of calorimetric, NMR, FTIR, DLS and zeta potential measurements. Two novel miktoarm star MHBCs, (PNIPAM)(2)-(PVP-b-PAA)(2) with PNIPAM segments constrained at one end and (PNIPAM-b-PAA)(2)-(PVP)(2) with PNIPAM segments constrained at two ends, are mainly analyzed to make comparison. As we previously reported (Soft Matter, 2012, 8, 3980), in (PNIPAM)(2)-(PVP-b-PAA)(2) the three polymeric segments have relatively independent phase behaviors during the formation of PNIPAM-core micelles. However, by introducing more structural constraints to PNIPAM segments, (PNIPAM-b-PAA)(2)-(PVP)(2) exhibits a much smoother and weaker phase transition. Additional analysis by IR spectroscopy, in combination with perturbation correlation moving window (PCMW) and two-dimensional correlation spectroscopy (2DCOS), indicates that PVP segments in (PNIPAM-b-PAA)(2)-(PVP)(2) show phase transition-like behavior by participating in the self-association process of PNIPAM segments, and there are both unimers and small associates existing in the solution below the LCST due to the slight interaction between PVP and PNIPAM segments. Moreover, due to the presence of PVP segments in the core, the micelles formed by (PNIPAM-b-PAA)(2)-(PVP)(2) should have a loosely organized superstructure, which may account for the weak phase transition and low degree of phase separation. Finally, we propose a mechanism of the self-assembly process of (PNIPAM-b-PAA)(2)-(PVP)(2), which is further confirmed by DLS and zeta potential measurements.
Synthesis and Self-Assembly Behavior of A Thermo-/Photo-Dual Responsive Triblock Copolymer

Abstract:A novel triblock copolymer containing azobenzene (AZO) and N-isopropylacrylamide (NIPAM) was synthesized by atom transfer radical polymerization (ATRP) using a poly(ethylene oxide) bifunctional macroinitiator (Br-PEO-Br). The triblock copolymer was characterized by H-1 NMR and gel permeation chromatography (GPC). Uniformly distributed polymeric micelles were obtained in aqueous solution by self-assembly method. Transmission electron microscope (TEM), fluorescence spectroscopy and dynamic light scattering (DLS) were used for the investigation of the self-assemblies and their stimuli-responsive behavior. Photoinduced release behavior was achieved with the as-prepared triblock copolymer micelles, which has not been observed in our previously-reported diblock copolymer micelles: the fluorescence intensity of Nile Red trapped in the triblock copolymer micelles dropped by approximately 40% after UV and Vis light alternative irradiation, indicating that a portion of the Nile Red was released to the aqueous media. Besides, the micelles shrank during the heating process.
The Preparation of Hydrophilic Temperature Sensitive Fluorescent Polymers P(NIPAM-co-NBDAE) by Atom Transfer Radical Polymerization

Abstract:Firstly,A monomerized fluorescence molecular 4-(2-acryloyloxy-ethylamino)-7-nitro-2,1,3-benzoxadiazole(NBDAE) was prepared with NBD-Cl(4-Chloro7-nitro-2,1,3-benzoxadiazole) as raw material.Then,well-defined thermoresponsive water-soluble random copolymers,poly(N-isopropylacrylamide-co-NBDAE)(P(NIPAM-co-NBDAE)),were prepared by atom transfer radical polymerization (ATRP),with ethylbutyl-2-bromoiso-butyrate(t-EBiB) as initiator,and CuCl/Me6TREN as catalyst.The thermoresponsive copolymers were characterized by 1H-NMR,GPC,UV-Vis,and fluorescence measurement.The results indicate that the well-defined thermoresponsive hydrophilic copolymers are narrow molecular-weight distribution with the polydispersity index of less than 1.20.The copolymers possess good fluorescence performance.The phase transition temperature of PNIPAM is 30 ℃.At >30 ℃,The fluorescence emission of hydrophilic copolymers is weak.While at <30 ℃,the thermoresponsive hydrophilic copolymers exhibit 2.5 times increased fluorescence intensity.It is observed that temperature induce polymers collapse at elevated temperatures and can be employed as a novel temperature sensor.

Abstract:In this work, a new smart copolymer of poly(N-isopropylacrylamide)-b-polyacrylamide-b-poly(N-isopropylacrylamide) (PNIPAm-b-PAM-b-PNIPAm) was designed as an efficient thermo-sensitive polymer with a potential for formation a physical network structure above a certain temperature in water. To design the desired copolymer, molecular behaviors of the copolymer blocks were investigated in water at different blocks fractions and temperatures via atomistic simulation. The blocks were evaluated from the interactional aspects via estimating the electrostatic and van der Waals contributions of their interaction energies with the media to consider the amount of hydrophilicity variation of the blocks with temperature variation. To find the origin of changes in blocks hydrophilicity, the structure and orientation of interfacial water were determined via calculating coordination number and order parameters of the water molecules in the first and second hydration shell of the blocks. In this regard, a novel parameter based on water ordering and hydration level was presented. The PAM block in the copolymer with shorter PNIPAm blocks, particularly 8 mol% (percentage by mole) PNIPAm, enforced the thermoresponsive end blocks to behave like a water-soluble polymer above their lower critical solution temperature. In contrast, increasing the PNIPAm content to 32 mol% caused an interactive competition between the blocks. Finally, the copolymer with the PNIPAm/PAM=0.19 was found as the most efficient copolymer composition for the mentioned goal via using the analysis of variance (ANOVA) of the calculated responses.

Abstract:A novel amphiphilic diblock copolymer composed of a hydrophilic poly(ethylene oxide) block and a hydrophobic block copolymerized by azobenzene-containing methacrylate and N-isopropylacrylamide was synthesized using ATRP. The polymer micelles showed dual responsiveness to heat and light. The size of the micelles was dependent on temperature and the encapsulated substance in the hydrophobic cores was released during heating and cooling processes. The hydrophobicity of the micellar cores appeared as a reversible change in response to light with neither disruption of the micelles nor leakage of the encapsulated substance while H-aggregation of the azobenzene moieties was detected.

Abstract:Thermoresponsive statistical copolymers of zwitterionic sulfobetaine methacrylate (SBMA) and nonionic N-isopropylacrylamide (NIPAAm) were prepared with an average molecular weight of about 6.0 kDa via homogeneous free radical copolymerization. The aqueous solution properties of poly(SBMA-co-NIPAAm) were measured using a UV-visible spectrophotometer. The copolymers exhibited controllable lower and upper critical solution temperatures in aqueous solution and showed stimuli-responsive phase transition in the presence of salts. Regulated zwitterionic and nonionic molar mass ratios led to poly (SBMA-co-NIPAAm) copolymers having double-critical solution temperatures, where the water-insoluble polymer microdomains are generated by the zwitterionic copolymer region of polySBMA or nonionic copolymer region of polyNIPAAm depending on temperature. A high content of the nonionic polyNIPAAm in poly(SBMA-co-NIPAAm) exhibits nonionic aggregation at high temperatures due to the desolvation of polyNIPAAm, whereas relatively low content of polyNIPAAm in poly(SBMA-co-NIPAAm) exhibits zwitterionic aggregation at low temperatures due to the desolvation of polySBMA. Plasma protein adsorption on the surface coated with poly(SBMA-co-NIPAAm) was measured with a surface plasmon resonance (SPR) sensor. The copolymers containing polySBMA above 29 mol% showed extremely low protein adsorption and high anticoagulant activity in human blood plasma. The tunable and switchable thermoresponsive phase behavior of poly(SBMA-co-NIPAAm), as well as its high plasma protein adsorption resistance and anticoagulant activity, suggests a potential for blood-contacting applications.

Abstract:A novel thermo-sensitive a-cyclodextrin pendent poly(N-isopropylacrylamide) (PnipamCD) was prepared, which had a higher lower critical solution temperature(LCST) than that of pure poly (N-isopropylacrylamide) ( PNIPAM). In its aqueous solution, spherical micelles were obtained as temperature increased rapidly while hollow vesicles were obtained as temperature increased slowly. In addition, rod-like aggregates were achieved in acetone, a selective solvent of PNIPAM.
