Abstract:Size-controlled, low-dispersed calcium carbonate microparticles were synthesized in the presence of the amphiphilic block copolymer polystyrene-b-poly(acrylic acid) (PS-b-PAA) by modulating the concentration of block copolymer in the reactive system. This type of hybrid microparticles have acid-resistant properties. By investigating the aggregation behaviors of PS-b-PAA micelles by transmission electron microscopy (TEM), the mechanism of hybrid calcium carbonate formation illustrated that the block copolymer served not only as "pseudonuclei" for the growth of calcium carbonate nanocrystals, but also forms the supramicelle congeries, a spherical framework, as templates for calcium carbonate nanocrystal growth into hybrid CaCO(3) particles. Moreover, this pilot study shows that the hybrid microparticle is a novel candidate as a template for fabricating multilayer polyelectrolyte capsules, in which the block copolymer is retained within the capsule interior after core removal under soft conditions. This not only facilitates the encapsulation of special materials, but also provides "micelles-enhanced" polyelectrolyte capsules.

Abstract:Environmentally friendly and outstanding pH responsiveness cationic starch nanoparticles (CSNP) were prepared through ethanol precipitation from pH-sensitive starch, which preparation of cationic starch (CS) by grafting copolymerization with dimethylaminoethyl methacrylate (DMAEMA). In this work, CSNP showed a nanometer size and regular sphere, highly free-flowing molecular chains, and outstanding pH responsiveness which was proved by the high stability of its stabilized emulsion through 6 emulsification/ demulsification transition. The result of the SEM and particle size distribution indicated that the size of the CSNP-0 was about 800 nm, and decreased with the DMAEMA increased. Moreover, the CSNP-stabilized emulsion was stable at pH = 7 and pH = 12. However, this emulsion exhibited breakage at pH = 2. In addition, the CSNP-stabilized Pickering emulsion achieved an emulsification/demulsification switching by cycling the pH at least 6 times, during which the average droplet size gradually increased. At pH ≥ 7, the emulsions exhibit shear thinning behavior.

Abstract:Abstract Background Nanoparticles assembled from food-grade calcium carbonate have attracted attention because of their biocompatibility, digestibility, particle and surface features (such as size, surface area, and partial wettability), and stimuli-responsiveness offered by their acid-labile nature. Results Herein, a type of edible oil-in-water Pickering emulsion was structured by calcium carbonate nanoparticles (CaCO 3 NPs; mean particle size: 80 nm) and medium-chain triglyceride (MCT) for delivery of lipophilic drugs and simultaneous oral supplementation of calcium. The microstructure of the as-made CaCO 3 NPs stabilized Pickering emulsion can be controlled by varying the particle concentration ( c ) and oil volume fraction ( φ ). The emulsification stabilizing capability of the CaCO 3 NPs also favored the formation of high internal phase emulsion at a high φ of 0.7–0.8 with excellent emulsion stability at room temperature and at 4 °C, thus protecting the encapsulated lipophilic bioactive, vitamin D3 (VD3), against degradation. Interestingly, the structured CaCO 3 NP-based Pickering emulsion displayed acid-trigged demulsification because of the disintegration of the CaCO 3 NPs into Ca 2+ in a simulated gastric environment, followed by efficient lipolysis of the lipid in simulated intestinal fluid. With the encapsulation and delivery of the emulsion, VD3 exhibited satisfying bioavailability after simulated gastrointestinal digestion. Conclusions Taken together, the rationally designed CaCO 3 NP emulsion system holds potential as a calcium-fortified formulation for food, pharmaceutical and biomedical applications.

Abstract:Amphiphilic starches incorporating fatty acid ester chains of varying lengths and degrees of substitution (DS) were synthesized to fabricate starch-based micelles for oral delivery of hydrophobic bioactive molecules. The assembly of the amphiphilic starches is influenced by the concentration, temperature, and the chain length and DS of their fatty acid ester chain. Highly acidic environment can hydrolyze the amphiphilic starches, resulting in the formation of small-sized micelles. Conversely, high ionic concentration hinders the self-assembly of amphiphilic starches and the digestive fluids can dilute the amphiphilic starches concentration, leading to the micelle dissociation. However, amphiphilic starches with longer chain length and/or higher DS of the fatty acid ester chain possess greater hydrophobicity, enhancing the stability of starch-based micelles under varying conditions and favouring the protection of Trp-2 peptides during storage. The micelles demonstrate high cell bioaccessibility for Trp-2 peptides, with 59.25 % of Trp-2 peptides being transferred by the intestinal epithelium. These findings suggest a potential starch-based micelle system can be adjusted for the oral delivery of hydrophobic bioactive molecules.

Abstract:The purpose of this study was to construct a delivery system using a microfluidic chip to protect procyanidins (PCs) and to achieve their pH-controlled release in simulated gastrointestinal fluid. The microfluidic chip was designed and fabricated to generate water-in-water-in-oil (W/W/O) templates for the preparation of sodium alginate/chitosan microparticles with a uniform size and core-shell structure, using an internal-external gelation method. Compared with free PCs, the stability of PCs embedded in microparticles was improved and a pH stimulus-responsive release of PCs from microparticles was observed under neutral pH conditions. The delivery system of microparticles was nontoxic and showed an inhibitory effect on the decrease of mitochondrial membrane potential in Caco-2 cells caused by H2O2 and acrylamide. This work provided a method for fabricating compact microfluidic chips to prepare a pH stimulus-responsive PCs delivery system with improved stability, which may have potential applications in the delivery of other nutrients.

Abstract:In this study, a system was designed that can encapsulate and deliver gallic acid (GA), which was composed of polysaccharide polymers based on sodium alginate (SA), carboxymethyl chitosan (CCT), and cellulose nanofibers (CN) and was assisted by porous starch. The compositions were characterized by rheology and zeta potentials, and the results showed that the materials used in this study could effectively guarantee the stability of the system. The morphology and chemical structure of the beads were characterized by SEM and FT-IR, the results indicated that the addition of CCT could effectively reduce the cracks and pores on the surface of the beads, which was beneficial to the encapsulation and delivery of GA. Moreover, the results of the swelling rate, release tests, and antioxidant tests also proved the effectiveness of the system. The pH response effect of SA/CN/CCT (SCC) beads and the protection of GA were superior, and the release rate of GA in simulated gastric fluid (SGF) was only 6.95%, while SA and SA/CN (SCN) beads reached 57.94% and 78.49%, respectively. In conclusion, the interpenetrating network polymers constructed by SA, CCT, and CN, which, combined with porous starch as a coating layer, can achieve the embedding and the delivery of GA.

Abstract:Hydrophilic and hydrophobic modified nanomicelles might be more conducive to passage of the gastrointestinal barrier than walnut peptide (WP). In this study, a novel double modified starch polymer, SB-CST-DCA, was synthesized by grafting sulfabetaine (SB) and deoxycholic acid (DCA) onto corn starch (CST) molecules through etherification and esterification. The modification mechanism was discussed to determine its chemical structure, morphological properties, and thermal stability. Peptide-loaded nanomicelles (SB-CST-DCA-WP) were prepared using WP as the core material. The encapsulation efficiency and peptide loading amount reached 76.90 ± 1.52% and 18.27 ± 0.53%, respectively, with good stability and pH-responsive release behavior observed to effectively control WP release and enhance its antioxidant activity. The composite exhibited safety, non-toxicity, and good blood compatibility at concentrations below 125 μg/mL. Duodenum was identified as the main absorption site with an absorption ratio of 41.16 ± 0.36%.

Abstract:The hierarchically porous property of CaCO 3 has attracted considerable attention in the field of active delivery ingredients due to its high adsorption capacity. Here, a facile and high-efficient approach to control the calcification processes of CaCO 3 ending with calcite microparticles with superior porosity and stability is reported and evaluated. In this work, a series of quercetin promoted CaCO 3 microparticles, using soy protein isolate (SPI) as entrapment agent, was synthesized, characterized, and their digestive behavior and antibacterial activity were evaluated. Results obtained indicated that quercetin showed good ability to direct the calcification pathway of amorphous calcium carbonate (ACC) with the formation of flower- and petal-like structures. The quercetin-loaded CaCO 3 microparticles (QCM) had a macro-meso-micropore structure, which was identified to be the calcite form. The macro-meso-micropore structure provided QCM with the largest surface area of 78.984 m 2 g −1 . The loading ratio of SPI to QCM was up to 200.94 μg per mg of QCM. The protein and quercetin composite microparticles (PQM) were produced by simply dissolving the CaCO 3 core, and the obtained PQM was used for the delivery of quercetin and protein. Thermogravimetric analysis showed PQM presented with good thermal stability without the CaCO 3 core. Furthermore, minor discrepancy was noted in protein conformational structures after removing the CaCO 3 core. In vitro digestion revealed that approximately 80% of the loaded quercetin was released from PQM during intestinal digestion, and the released quercetin exhibited efficient transportation across the Caco-2 cell monolayer. More importantly, the PQM digesta retained enhanced antibacterial activities to inhibit growth of Escherichia coli and Staphylococcus aureus . Porous calcites show a high potential as a delivery system for food applications.

Abstract:We fabricated CaCO3-coated vesicles as drug carriers that release their cargo under a weakly acidic condition. We designed and synthesized a peptide lipid containing the Val-His-Val-Glu-Val-Ser sequence as the hydrophilic part, and with two palmitoyl groups at the N-terminal as the anchor groups of the lipid bilayer membrane. Vesicles embedded with the peptide lipids were prepared. The CaCO3 coating of the vesicle surface was performed by the mineralization induced by the embedded peptide lipid. The peptide lipid produced the mineral source, CO32−, for CaCO3 mineralization through the hydrolysis of urea. We investigated the structure of the obtained CaCO3-coated vesicles using transmission electron microscopy (TEM). The vesicles retained the spherical shapes, even in vacuo. Furthermore, the vesicles had inner spaces that acted as the drug cargo, as observed by the TEM tomographic analysis. The thickness of the CaCO3 shell was estimated as ca. 20 nm. CaCO3-coated vesicles containing hydrophobic or hydrophilic drugs were prepared, and the drug release properties were examined under various pH conditions. The mineralized CaCO3 shell of the vesicle surface was dissolved under a weakly acidic condition, pH 6.0, such as in the neighborhood of cancer tissues. The degradation of the CaCO3 shell induced an effective release of the drugs. Such behavior suggests potential of the CaCO3-coated vesicles as carriers for cancer therapies.

Abstract:Nano-sized calcium carbonate, as a multifunctional material, is widely used in various industrial fields. The morphology, particle size, surface property of CaCO3 particles have great impact on the application effect. This study presents an energy-saving and efficient approach to controllably prepare CaCO3 nanoparticles and conduct surface modification simultaneously. Ca(OH)(2)/CO2 carbonation method was adopted to prepare CaCO3 nanoparticles using a membrane dispersion microreactor in the presence of calcium stearate (CaSt(2)) surfactant. The effects of gas and liquid flow rates, temperature and the dosage of surfactant were systematically investigated. The effect of surface modification was evaluated by contact angle and active ratio. The as-prepared hydrophobic CaCO3 nanoparticles were of good dispersity, with an average particle size of 34 nm, a specific surface area of above 30 m(2)/g and a contact angle of 107.8 degrees. In this work, the effect of surfactant on the carbonation reaction was greatly diminished by adding CaSt(2) surfactant to a portion of Ca(OH)(2) slurry. Additionally, the modification process was carried out without organic solvents and high temperature. As compared to post-modification method, the in-situ modification process is of significant practical importance because of high production efficiency, excellent process stability, and high product quality. Abstract:Calcium carbonate (CaCO3) has long been used as a delivery system owing to its wide availability, biocompatibility, and degradability. However, it often suffers from many challenges toward rapid dissolution at stomach acid environment, low retention ability, and lack of sustained release. Many of these issues can be addressed by modifying the CaCO3 particles or integrating them with other encapsulation systems, generating advanced CaCO3-derived systems. This review article presents a recent progress (2015–2022) in the utilization of CaCO3 particles in the exploration of various advanced delivery systems, including polymer-doped CaCO3, surface-coated CaCO3, CaCO3-stabilized Pickering emulsions, CaCO3-in-hydrogels, and liposomal CaCO3. Some emerging systems templated on CaCO3 sacrificial cores, such as hollow capsules, matrix-type capsules, and core-shell capsules, are also reviewed. A brief description of each system is given, and then their formation, structure, and properties are described. Particular emphasis is given to the applications and challenges of these advanced systems for the protection and controlled delivery of bioactive compounds in the biological, pharmaceutical, and functional food fields.

Abstract:Zeolitic imidazolate framework-8 (ZIF-8) is a promising material for drug delivery, but its poor stability limits its wide application. In this work, homogeneous dodecahedral ZIF-8 was prepared by the room-temperature solution stirring method, and to improve the stability of ZIF-8, ZIF-8@mSiO 2 and DZIF-8 (ZIF-8 after calcination) @PMO composites with the core-shell structure were obtained using mesoporous silica (mSiO 2 ) and periodic mesoporous organic silica (PMO) coatings. Folic acid-coupled chitosan (CS-FA) was modified on nanoparticles, and curcumin (CUR) was used as a model anti-tumor drug to obtain the drug delivery systems with targeting and pH stimulation response: ZIF-8@mSiO 2 @CUR@CS-FA and DZIF-8@PMO@CUR@CS-FA, which exhibited good dispersion and high drug loading capability. In addition, we used the MTT assay to assess the cytotoxicity of the drug-carrying systems and selected human cervical cancer cells (Hela) as model cells. The results of the in vitro experiments suggested that the final CUR-loaded drug delivery systems had significantly increased anticancer efficiency compared with the free drug CUR, which improved the therapeutic effect of the hydrophobic CUR on tumors. In conclusion, the pH-responsive nanocarriers provided potential for the targeted delivery of anticancer medication, resulting in a platform for drug-controlled release and cancer therapy.

Abstract:Oral digestion reduces calcium bioavailability in peptide-calcium supplements, and the construction of a nano-delivery system can solve this problem and protect peptide-calcium absorption. In this study, the delivery carrier of chitosan nanoparticles (CS NPs) was firstly optimized by particle size, polydispersity index, and Zeta potential. Then, the nano-delivery system loaded with phosvitin phosphopeptide-calcium chelate (PPP-Ca NPs) was optimized by particle size and encapsulation efficiency (EE). The results showed that the optimal EE was 87.80% +/- 0.66%, and the particle size was 272.3 +/- 2.4 nm. The structure characterization demonstrated that PPP-Ca NPs was mainly formed by electrostatic interaction, and the nano-delivery system was spherical particles (<100 nm). The results of in vitro digestion demonstrated that PPP-Ca NPs could improve the stability and control release of PPP-Ca. Moreover, PPP-Ca NPs could significantly improve mucoadhesion propensity, prolong residence time in the intestine, and thus enhance calcium bioavailability. Furthermore, PPP-Ca NPs could be stored for at least 90 days. This study demonstrated that this nano-delivery system with high EE of PPP-Ca NPs was stable and could protect peptide-calcium absorption. Therefore, PPP-Ca NPs have the potential to serve as a new oral calcium supplement.
Structural changes of layer-by-layer self-assembled starch-based nanocapsules in the gastrointestinal tract: Implications for their M cell-targeting delivery and transport efficiency

Abstract:Characterizing the structural changes of cell-targeting delivery carriers in gastrointestinal tract (GIT) is crucial for understanding their effectiveness in cell targeting and transport. Herein, RGD peptide-grafted carboxymethyl starch (CMS) and cationic quaternary ammonium starch (QAS) were utilized to fabricate quintet-layered nanocapsules loaded with ovalbumin (OVA). The aim was to improve delivery and transportation efficiency, specifically targeting M cells. The research analyzed the impact of pH and enzyme variations in GIT on the structure of nanocapsules, interactions between carriers and the release behavior of OVA. Results showed that the size of nanocapsules increased from 229.2 to 479.8 nm and the zeta potential decreased from -1.08 to -33.33 mV during oral delivery. This was evident in TEM images, showing a more relaxed core-shell structure. Isothermal titration calorimetry and molecular dynamic simulation indicated that pH changes primarily affected the electrostatic interaction between carriers. Increasing pH led to reduced affinity constants, and around 84.42 % of OVA was successfully delivered to M cells. Moreover, the transport efficiency of nanocapsules to M cells was five times greater than that of Caco-2 cells. This suggests the feasibility of developing a nanocapsules delivery system capable of adapting to pH changes in GIT by regulating electrostatic interactions between carriers.

Abstract:Colon-targeting delivery of insulin is surging great interests in revolutionizing diabetes. Herein, insulin-loaded starch-based nanocapsules developed by layer-by-layer self-assembly technology were rationally structured. Interactions between starches and the structural changes of the nanocapsules were unraveled to understand in vitro and in vivo insulin release properties. By increasing the deposition layers of starches, the structural compactness of nanocapsules increased and in turn retarded insulin release in the upper gastrointestinal tract. Spherical nanocapsules deposited at least five layers of starches could deliver insulin to the colon in a high efficiency according to the in vitro and in vivo insulin release performance. The underlying mechanism of the insulin colon-targeting release should ascribe to the suitable changes in compactness of the nanocapsules and the interactions between deposited starches after multi-response to the changes in pH, time and enzymes in gastrointestinal tract. Starch molecules interacted with each other much stronger at the intestine than that at the colon, which guaranteed a compact structure in the intestine but a loose structure in the colon for the colon-targeting nanocapsules. It suggested that rather than controlling the deposition layer of the nanocapsules, controlling the interaction between starches could also regulate the structures of the nanocapsules for colon-targeting delivery system.

Abstract:Abstract This study investigated the chemical, wettability, thermal and structural properties of starch nanocrystals (SNCs) modified through crosslinking modification using citric acid in aqueous medium. The results of Fourier transform infrared spectroscopy analysis suggested that new interactions occurred between starch and citric acid after modification and an increase of the reaction time increased the degree of substitution. X-ray diffraction analysis showed that the crystalline structure of SNCs was maintained after the crosslinking modification by adjusting the pH to 3.5, and destroyed after the modification by pretreatments without adjusting the pH of the citric acid solution. Crosslinking modification decreased the size and aggregation of SNCs and longer reaction time gave rise to the smaller particle size of SNCs modified by the pretreatments of adjusting the pH of the citric acid solution. The crosslinked SNCs had lower polarity and showed stronger hydrophobicity and dispersion stability which allowed them be used as reinforcements in hydrophobic polymer matrices.

Abstract:A porous starch-based carrier coated with chitosan-phytic acid was designed for oral administration to improve drug delivery to the colon. Using hydrophobic paclitaxel as a model drug, improved drug loading (15.12% ± 0.31%) and entrapment efficiency (86.63 ± 1.30%) of porous starch were achieved by size/shape matching and adsorption force. Fluorescent paclitaxel particles inside starch were captured clearly. Furthermore, chitosan-phytic acid was added as a second protection since porous starch showed a dissolution rate of only 14.98–20.27% during the simulated digestion in stomach and small intestine, which was far lower than that of raw paclitaxel in porous starch (59.65 ± 2.57%). The release curve in the colon was also obtained and showed that 86.98 ± 2.90% of the drug was released. Finally, we verified the non-covalent interactions between starch and paclitaxel. This showed that the retention of paclitaxel into porous starch decreased once hydrogen bonding stopped. The hydrophobic CH-π effect provides a binding complementing contribution.

Abstract:The importance of fertilizers in boosting crop production has motivated the development of novel high-performance systems capable of improving the phosphorus release in the soil. For instance, methods capable of increasing the surface area of fertilizer particles and promoting the solubilization of low-solubility compounds, including phosphates, are highly pursued. This study was aimed at synthesizing hydroxyapatite nanoparticles and investigating their solubility in relation to crystallinity, size, and morphology for phosphorous fertilizer applications. To improve the phosphate ions release, the hydroxyapatite nanoparticles were storage in biodegradable sachets composed of thermoplastic starch/pectin blends with different polymer ratios. The results showed that the smallest and less crystalline hydroxyapatite nanoparticles presented the highest solubility. After storage in polymeric thermoplastic starch:pectin sachets, solubility for all samples was greatly improved, enhancing the phosphorus release due to pH decrease, independent on the nanoparticle size, shape, and crystallinity. The results highlight that the use of acidic sachets is a valuable approach for enhancing phosphorus and other macronutrients release from fertilizers with basic surface properties, aiming at increasing agricultural crop productivity.Graphical Abstract

Abstract:Monodispersed poly(styrene-co-N,N′-dimethylaminoethyl methacrylate) nanoparticles (P(St-co-DMAEMA) NPs) were synthesized by emulsion polymerization. Zeta potential, volume swelling ratio and in vitro release of fluorescer coumarin-6 from the NPs were determined in buffer of various pH. With an optimized formulation, the diameter of NPs is 100nm approximately and the polydispersity index (PI) is less than 0.1. The NPs have a hydrophilic surface and alterable surface charge which is almost neutral at normal physiological pH, but becomes much more positive under acidic conditions. The volume swelling ratios and in vitro release of coumarin-6 are highly dependent on pH, which are significantly increased at the lower pH and higher DMAEMA/St molar ratio. More than 70% of the loaded coumarin-6 could be released in 24h at pH2, which is 2.3-folds higher than that at normal physiological pH. The P(St-co-DMAEMA) NPs show promising applications to targeted drug delivery to tumors.
