Abstract:For the solid phase preparation of various (pharmacologically important) heterocycles, cyclization/cleavage (C/C) or cyclorelease strategies proved to be superior. CC approaches take utmost advantage of the benefits of solid phase synthesis. Besides the practical benefits of solid phase reactions, cyclative release approaches are distinguished especially because of the generally found high purity of the final detached products, since only the anticipated structures cleave off the resin. Also cyclization/cleavage strategies are "traceless", as the obtained moiety after cyclization is part of the formed heterocycle. Over the last thirty years numerous approaches towards important heterocycle classes have been published. Among the successfully applied strategies are carbon-nitrogen (sulfur) bond, carbon-oxygen bond, sulfur-sulfur bond and carbon-carbon forming reactions in the final cyclorelease step.

Abstract:A linked heterocycle library of isoxazoles, 1,2,3-triazoles, bicyclo[2.2.1]hepta-2,5-diene or 4-methylcyclohexa-1,3-diene and 1,2,4-oxadiazoles was prepared by solid-phase organic synthesis. Key steps on resin-bound selenium were electrophilic additions; 1,3-dipolar cycloaddition; Porco's two-step, one-pot condensation of amidoxime and carboxylate; and Diels-Alder reaction.

Abstract:A new protocol for synthesis of thioether-linked peptide macrocycles via Pd-catalyzed intramolecular S -arylation and S -alkenylation on solid phase is developed . The use of PEGA resin in mixed solvents of CH 3 CN and H 2 O is critical to suppress the undesired S -arylation reactions with the XantPhos-Pd-G3 catalyst. Peptide macrocycles of varied ring size, composition, and with different thioether linkers can be prepared in moderate to good yield with one column chromatography purification。

Abstract:Polyamines in naturally occurring compounds are biologically active molecules that do not display a great diversity but are very abundant in almost all organisms. Their functions comprise antibiotics, immunosuppressants, toxins, and many others. In most biologically active compounds the polyamine chain is coupled to other functionalities making the polyamine an excellent template for solid phase chemistry and combinatorial approaches. This review comprises common techniques for the synthesis of polyamines and polyamine derivatives on solid support. It includes the description of basic synthesis requirements such as of suitable resins, new linkers, reaction conditions for generation of biologically active polyamines and polyamines in natural products as well as the description of protection groups. Moreover, it will focus on the modular coupling of the amine building blocks using different synthesis strategies. Thus, the advantages of performing reactions on polymer-supported substrates in terms of avoiding side reactions and facilitating product/metal separation are highlighted.

Abstract:An increasingly large number of bioactive cyclic peptides have been found in nature. The enhanced biological specificity, activity, and metabolic stability of cyclopeptides, by virtue of their conformationally constrained structural feature, have attracted much attention and made these compounds extensively studied. Cyclic peptidomimetic scaffolds and templates have been widely used to assemble various spatially defined functional groups for molecular recognition and drug discovery. These efforts are complemented optimally by NMR and X-ray based structure information, and assisted by molecular modeling based structure predictive methods. Peptide cyclization also becomes an effective and commonly employed strategy for peptide modifications. During the past several decades, great effort has been made to develop more efficient methods for the synthesis of cyclic peptides and peptidomimetics, as potential drug leads and/or as models for conformational analysis. This review is aimed at highlighting novel recently developed peptide cyclization approaches, and illustrating the profitable applications.
Constructing Cyclic Peptides Using an On-Tether Sulfonium Center

Abstract:In recent years, cyclic peptides have attracted increasing attention in the field of drug discovery due to their excellent biological activities, and, as a consequence, they are now used clinically. It is, therefore, critical to seek effective strategies for synthesizing cyclic peptides to promote their application in the field of drug discovery. This paper reports a detailed protocol for the efficient synthesis of cyclic peptides using on-resin or intramolecular (intermolecular) bisalkylation. Using this protocol, linear peptides were synthesized by taking advantage of solid-phase peptide synthesis with cysteine (Cys) and methionine (Met) coupled simultaneously on the resin. Further, cyclic peptides were synthesized via bisalkylation between Met and Cys using a tunable tether and an on-tether sulfonium center. The whole synthetic route can be divided into three major processes: the deprotection of Cys on the resin, the coupling of the linker, and the cyclization between Cys and Met in a trifluoroacetic acid (TFA) cleavage solution. Furthermore, inspired by the reactivity of the sulfonium center, a propargyl group was attached to the Met to trigger thiol-yne addition and form a cyclic peptide. After that, the crude peptides were dried and dissolved in acetonitrile, separated, and then purified by high-performance liquid chromatography (HPLC). The molecular weight of the cyclic peptide was confirmed by liquid chromatography-mass spectrometry (LC-MS), and the stability of the cyclic peptide combination with the reductant was further confirmed using HPLC. In addition, the chemical shift in the cyclic peptide was analyzed by 1H nuclear magnetic resonance (1H NMR) spectra. Overall, this protocol aimed to establish an effective strategy for synthesizing cyclic peptides.

Abstract:Many naturally occurring small peptides have significant therapeutic properties. Total synthesis of these peptides is not only helpful in the identification of their structures and providing methods to synthesize their analogues, but also contributes to their biosynthetic studies. As solid‐phase synthesis simplifies the tedious and time‐consuming isolation of intermediates, it has been widely applied in peptide synthesis. This review has highlighted the recent progress in solid‐phase total synthesis of both linear and cyclic naturally occurring small peptides from 2014 to July 2021.

Abstract:The synthesis of heterocycles is a fundamental area of organic chemistry that offers enormous potential for the discovery of new products with important applications in our daily life such as pharmaceuticals, agrochemicals, flavors, dyes, and, more generally, engineered materials with innovative properties. As heterocyclic compounds find application across multiple industries and are prepared in very large quantities, the development of sustainable approaches for their synthesis has become a crucial objective for contemporary green chemistry committed to reducing the environmental impact of chemical processes. In this context, the present review focuses on the recent methodologies aimed at preparing N-, O- and S-heterocyclic compounds in Deep Eutectic Solvents, a new class of ionic solvents that are non-volatile, non-toxic, easy to prepare, easy to recycle, and can be obtained from renewable sources. Emphasis has been placed on those processes that prioritize the recycling of catalyst and solvent, as they offer the dual benefit of promoting synthetic efficiency while demonstrating environmental responsibility.

Abstract:The solid-phase synthesis of sultams from resin-bound amino acids is described. The sulfonylation of the resin-bound primary amines afforded the requisite secondary amines, after which the intramolecular Buchwald–Hartwig-type coupling forms the C–S bond. A final alkylation on the sulfonamides followed by cleavage provided the corresponding seven-membered sultams.

Abstract:While most FDA-approved peptide drugs are cyclic, robust cyclization chemistry of peptides and the deconvolution of the cyclic peptide sequences using tandem mass spectrometry render cyclic peptide drug discovery difficult. In this chapter, the protocol for the successful synthesis of tetrazine-linked cyclic peptide library in solid phase, which shows both robust cyclization and easy sequence deconvolution, is described. The protocol for the linearization and cleavage of cyclic peptides from the solid phase by simple UV light irradiation, followed by accurate sequencing using tandem mass spectrometry, is described. We describe the troubleshooting for this dithiol bis-arylation reaction and for the successful cleavage of the aryl cyclic peptide into linear form. This method for efficient solid-phase macrocyclization can be used for the rapid production of loop-based peptides and screening for inhibition of protein–protein interactions, by using the covalent inverse electron-demand Diels Alder reaction to supplement the non-covalent interaction between a protein and its peptide binder, isolating highly selective peptides in the process.

Abstract:Heteroarene 1, n-zwitterions are powerful and versatile building blocks in the construction of heterocycles and have received increasing attention in recent years. In particular, pyridinium and quinolinium 1,4-zwitterions have been widely studied and used in a variety of cyclization reactions due to their air stability, ease of use, and high efficiency. Sulfur- and nitrogen-based pyridinium and quinolinium 1,4-zwitterions, types of emerging heteroatom-containing synthons, have attracted much attention from chemists. These 1,4-zwitterions, which contain multiple reaction sites, have been successfully used in the synthesis of three- to eight-membered cyclic compounds over the last decade. In this review, we present the exciting progress made in the field of cyclization reactions of sulfur- and nitrogen-based pyridinium and quinolinium 1,4-zwitterions. Moreover, the mechanistic insights, the transition states, some synthetic applications, and the challenges and opportunities are also discussed. We hope to provide an overview for synthetic chemists who are interested in the heterocycle synthesis from cyclization reaction with pyridinium and quinolinium 1,4-zwitterions pyridinium and quinolinium 1,4-zwitterions.

The five-membered heterocycles are pharmacophores of many natural and synthetic bioactive compounds. This review focuses on the progress of the synthetic methods of these amino acids (including the limitation, simplicity, yield and optical purity) in recent years.

Abstract:The reactive intermediates of pentacoordinated phosphoranes with amino acids (P(5)-AA) as building blocks, which were obtained by the reaction of O-phenylene phosphorochloridate with N,O-bis(trimethylsilyl)amino acids, were linked to a solid-phase support containing a hydroxymethyl polystyrene functional group. The first amino acid residue was coupled to the solid-phase support after washing the resin with organic solvent. Repeating the procedure led to oligopeptides linked on the resin. A series of free oligopeptides including tetra-Gly, di-Val, tri-Val, di-Leu, di-Phe, and Phe–Leu were obtained after cleavage from solid-phase support. The structure of these oligopeptides were determined by IR, 1H NMR, FAB-MS, and HPLC.

Abstract:Herein, we report a novel strategy for diversity-oriented lysine modification of cyclic peptides based on the orthogonal alkylation of the lysine residues. All steps can be achieved in the solid phase with satisfying conversions. Notably, we demonstrated that the tether modification could help to improve the cellular uptake of peptides.

Abstract:There are numerous pyrazine and phenazine compounds that demonstrate biological activities relevant to the treatment of disease. In this review, we discuss pyrazine and phenazine agents that have shown potential therapeutic value, including several clinically used agents. In addition, we cover some basic science related to pyrazine and phenazine heterocycles, which possess interesting reactivity profiles that have been on display in numerous cases of innovative total synthesis approaches, synthetic methodologies, drug discovery efforts, and medicinal chemistry programs. The majority of this review is focused on presenting instructive total synthesis and medicinal chemistry efforts of select pyrazine and phenazine compounds, and we believe these incredible heterocycles offer promise in medicine.
