Abstract:There has also been a growing interest in the application of carbohydrates as pharmaceutical drugs in the field of invasive bacterial diseases, inflammatory diseases, and viral infections. Chemical oligosaccharide synthetic procedures often require tedious protection and deprotection steps. Enzymatic synthesis offers tremendous benefits because of its high regio- and stereoselectivities, stability, organic solvent compatibility, and low cost. Glycosyltransferase-catalyzed synthesis of carbohydrates has been recognized as one of the most practical approaches for its high efficiency and specificity under very mild conditions. The two main obstacles in the enzymatic synthesis of carbohydrates are the inadequate availability of glycosyltransferases and the high cost of sugar nucleotides, which serve as intermediates. Significant progress in the study of the enzymatic biosynthesis of complex carbohydrates has been made with developments in protein purification, molecular genetics, and new methods of enzymological analysis. Bioinformatics provide a large amount of putative candidates for carbohydrate active enzymes. The combined enzymatic and genetic approach has overcome formidable obstacles ubiquitous to the study of glycans, and has begun to yield new information that definitively addresses basic enzymological issues relevant to these complex carbohydrates. In fact, complex carbohydrates representing the glycan chain of gangliosides such as GT2, GM1, and GD1a were synthesized using glycosyltransferases. Although traditional cell-free enzymatic synthesis of oligosaccharides will continue to be used in laboratory scale synthesis, the future direction for large-scale preparation of complex oligosaccharides and polysaccharides will be whole-cell systems. Further development of these enzymatic methods will allow synthetic biochemists to create important molecular tools for biochemical, biophysical, and medical applications.

Abstract:Glycoproteins and glycopeptides play important roles in various physiological and pathophysiological processes. Efficient preparation of glycopeptides with a specific structure is one of the pivotal areas in current chemistry research. In this article, a new SHGPFT approach to the synthesis and efficient purification of O-glycosylated peptides is developed by combining a solid-phase glycosylation and a light-fluorous glycosyl donor protocol. The desired product is finally isolated from the side products in the cleaved mixture by an efficient fluorous solid-phase extraction (F-SPE) step.

Abstract:The development and application of commercially available automated peptide synthesizers has played an essential role in almost all areas of peptide and protein research. Recent advances in peptide synthesis method and solid-phase chemistry provide new opportunities for optimizing synthetic efficiency of peptide synthesizers. The efforts in this direction have led to the successful preparation of peptides up to more than 150 amino acid residues in length. Such success is particularly useful for addressing the challenges associated with the chemical synthesis of glycoproteins. The purpose of this review is to provide a brief overview of the evolution of peptide synthesizer and glycoprotein synthesis. The discussions in this article include the principles underlying the representative synthesizers, the strengths and weaknesses of different synthesizers in light of their principles, and how to further improve the applicability of peptide synthesizers in glycoprotein synthesis.

Abstract:Aim To design and synthesize octreotide acetate.Method Octreotide was synthesized by the solid-phase peptide synthesis method and stepwise condensation strategy.Result and conclusion Octreotide acetate was synthesized.The total yield was 30%(calculated from the first amino acid anchored to Resin).The structures of the key intermediates and target molecule were confirmed by ESI-MS,IR and NMR.

Abstract:Glycans have been hot topics in recent years due to their exhibition of numerous biological activities. However, the heterogeneity of their natural source and the complexity of their chemical synthesis impede the progress in their biological research. Thus, the development of glycan assembly strategies to acquire plenty of structurally well-defined glycans is an important issue in carbohydrate chemistry. In this review, the latest advances in glycan assembly strategies from concepts to their applications in carbohydrate synthesis, including chemical and enzymatic/chemo-enzymatic approaches, as well as solution-phase and solid-phase/tag-assisted synthesis, are summarized. Furthermore, the automated glycan assembly techniques are also outlined.

Abstract:The phosphopeptide and corresponding nonphosphopeptide was synthesized with the Fmoc strategy solid-phase synthesis method. Using N, N-diethyl-dialkyl-phosphoramidity as phosphorylating reagent, some phosphopeptides were obtained by the use of the golble phosphorylation approach, others were obtained from the phosphorylated building block. The peptides were purified with HPLC. The longer the phosphopeptide, the lower the phosphorylating efficiency. Up to now, this Fmoc strategy solid-phase synthesis method with the step-wise phosphotylation approach only can be used in the synthesis of phosphopeptide with Tyr.

Abstract:A new approach is described for the general Fmoc-based solid-phase synthesis of (glyco)peptide aryl thioesters. A peptide alkyl oxoester obtained by standard Fmoc-based chain elongation undergoes an O-to-S acyl shift, and is followed by alkyl thioester exchanges with a large excess of aryl thiol, affording the corresponding peptide aryl thioester. The newly developed methodology is useful for the chemical synthesis of post-translationally modified proteins because of its compatibility with standard Fmoc-SPPS conditions. In addition, the peptide aryl thioesters are essential intermediates for chemical synthesis of proteins by kinetically controlled convergent strategy.

Abstract:Glycoproteins account for numerous biological processes including those associated with diseases and infections. The advancement of glycopeptides has emerged as a promising strategy for unraveling biological pathways and discovering novel medicines. In this arena, a key challenge arises from the absence of efficient synthetic strategies to access glycopeptides and glycoproteins. Here, we present a highly concise approach to bridging saccharides with amino acids and peptides through an amide linkage. Our amide-linked C-glycosyl amino acids and peptides are synthesized through cooperative Ni-catalyzed and photoredox processes. The catalytic process generates a glycosyl radical and an amide carbonyl radical, which subsequently combine to yield the C-glycosyl products. The saccharide reaction partners encompass mono-, di-, and trisaccharides. All 20 natural amino acids, peptides, and their derivatives can efficiently undergo glycosylations with yields ranging from acceptable to high, demonstrating excellent stereoselectivities. As a substantial expansion of applications, we have shown that simple C-glycosyl amino acids can function as versatile building units for constructing C-glycopeptides with intricate spatial complexities.

Abstract:Glycosylation is one of the most ubiquitous post‐translational modifications. It affects the structure and function of peptides/proteins and consequently has a significant impact on various biological events. However, the structural complexity and heterogeneity of glycopeptides/proteins caused by the diversity of glycan structures and glycosylation sites complicates the detailed elucidation of glycan function and hampers their clinical applications. To address these challenges, chemical and/or enzyme‐assisted synthesis methods have been developed to realize glycopeptides/proteins with well‐defined glycan morphologies. In particular, N‐glycans are expected to be useful for improving the solubility, in vivo half‐life and aggregation of bioactive peptides/proteins that have had limited clinical applications so far due to their short duration of action in the blood and unsuitable physicochemical properties. Chemical glycosylation performed in a post‐synthetic procedure can be used to facilitate the development of glycopeptide/protein analogues or mimetics that are superior to the original molecules in terms of physicochemical and pharmacokinetic properties. N‐glycans are used to modify targets because they are highly biodegradable and biocompatible and have structures that already exist in the human body. On the practical side, from a quality control perspective, close attention should be paid to their structural homogeneity when they are to be applied to pharmaceuticals.

Abstract:Polymyxin B and E are used as a ‘last line’ therapy for infections caused by serious Gram-negative bacteria due to their highly efficient antibacterial activity and nephrotoxicity. Many research groups have been performed on designing polymyxin analogues by chemical synthesis in order to decrease the nephrotoxicity and increase the antibacterial activity simultaneously. In this study, we developed a new strategy for total solid phase synthesis of polymyxins and their analogues. This method is achieved by anchoring the amine side chain of Dab9 on resin first and on-resin cyclization at last. Comparing to the mostly used chemical synthetic method for polymyxins which involves solid phase peptide synthesis followed by liquid phase cyclization, the method reported here is more convenient and efficient. Therefore, this new strategy may be a good replacement for current chemical synthetic method in designing polymyxin analogues.
Recovery, Purification, and Reusability of Building Blocks for Solid Phase Synthesis

Abstract:Solid phase synthesis (SPS) is well established for the synthesis of biomacromolecules such as peptides, oligonucelotides, and oligosaccharides, and today is also used for the synthesis of synthetic macromolecules and polymers. The key feature of this approach is the stepwise assembly of building blocks on solid support, enabling monodispersity and monomer sequence control. However, in order to achieve such control, a high excess of building blocks is required during the reaction. Herein, the recovery, purification, and reusability of building blocks used in SPS, including representative examples of tailor-made building blocks, Fmoc-protected amino acids, and functionalized carbohydrate ligands, are reported for the first time. Results demonstrate the general applicability with recovery in high yields and high purity. Furthermore, the described recovery process can be applied in both manual and automated synthesis using a standard peptide synthesizer. Overall, this process is envisioned to be applicable for a large variety of building blocks used in the SPS of different types of molecules, and to contribute to more resourceful SPS syntheses.

Abstract:An efficient and practical method for macrocyclic lipoglycopeptide synthesis was developed and utilized to synthesize lipoglycosylated derivatives of Tyrocidine A. The method is based on solid-phase peptide synthesis using 2-chlorotrityl resin as the solid-phase support and lipoglycosyl amino acids as building blocks. This synthetic method should be generally applicable to various macrocyclic lipoglycopeptides.

Abstract:Many naturally occurring small peptides have significant therapeutic properties. Total synthesis of these peptides is not only helpful in the identification of their structures and providing methods to synthesize their analogues, but also contributes to their biosynthetic studies. As solid‐phase synthesis simplifies the tedious and time‐consuming isolation of intermediates, it has been widely applied in peptide synthesis. This review has highlighted the recent progress in solid‐phase total synthesis of both linear and cyclic naturally occurring small peptides from 2014 to July 2021.
