Abstract:A method for intermolecular [3+2] cycloaddition between aryl cyclopropyl ketones and alkenes involving the combination of Lewis acid and photoredox catalysis is reported. In contrast to other more common methods for [3+2] cycloaddition, these conditions operate using a broad range of both electron-rich and electron-deficient reaction partners. The critical factors predicting the success of these reactions is the redox potential of the cyclopropyl ketone and the ability of the alkene to stabilize a key radical intermediate.

Abstract:The selective coupling of two different radicals is a long-standing challenge due to the process occurring statistically. Generally, the cross-coupling selectivity is achieved by the persistent radical effect (PRE), resulting in limited radical precursors. In this paper, a highly selective cross-coupling of alkyl radicals with acyl radicals to construct C(sp2)-C(sp3) bonds, or with alkyl radicals to construct C(sp3)-C(sp3) bonds have been achieved with the readily available carboxylic acids and their derivatives (NHPI ester) as coupling partners. The success originates from the use of tridentate ligand (L1) to enable radical cross-coupling process to Ni-mediated organometallic mechanism. This protocol offers a facile and flexible access to structurally diverse ketones (up to 90% yield), and also a new solution for the challenging double decarboxylative C(sp3)-C(sp3) coupling. The broad utility and functional group tolerance are further illustrated by the late-stage functionalization of natural-occurring carboxylic acids and drugs.

Abstract:A photoinduced copper-catalyzed alkoxyl triggered C−C bond cleavage/aminocarbonylation cascade is presented. Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:We developed a chemoselective catalytic activation of carboxylic acid for a 1e- radical process. α-Oxidation of a variety of carboxylic acids, which preferentially undergo undesired decarboxylation under radical conditions, proceeded efficiently under the optimized conditions. Chemoselective enolization of carboxylic acid was also achieved even in the presence of more acidic carbonyls. Extensive mechanistic studies revealed that the cooperative actions of iron species and alkali metal ions derived from 4 Å molecular sieves substantially facilitated the enolization. For the first time, catalytic enolization of unprotected carboxylic acid was achieved without external addition of stoichiometric amounts of Brønsted base. The formed redox-active heterobimetallic enediolate efficiently coupled with free radical TEMPO, providing synthetically useful α-hydroxy and keto acid derivatives.

Abstract:We developed the Ti‐catalyzed regioselective reductive coupling of readily available aldehydes and racemic propargyl bromides to rapidly access a wide range of α‐allenols. This method proceeds efficiently in a reductive radical‐polar crossover manner featuring mild conditions, excellent regioselectivity control, broad substrate scope, and eco‐friendliness. Preliminary mechanistic studies support the radical‐involved catalytic cycle. DFT calculations demonstrate that the regioselectivity is determined by the Zimmerman‐Traxler‐type transition states. Abstract:The mild catalytic generation of ketyl radicals for organic transformations remains an unsolved issue, although it facilitates the discovery of metal-catalyzed reactions in feature of high functional group tolerance. We report here the generation of ketyl radicals and coupling with alkynes that were enabled by cost-effective chromium catalysis, allowing for the formation of valuable E-exocyclic allyl alcohols in high stereo- and chemoselectivity. A broad range of synthetically useful functional groups that are sensitive to strong reductants are compatible by the catalytic system, providing access to diverse substituted E-exocyclic allyl alcohols under mild conditions. The appended hydroxyl groups in the products are facilely late-stage functionalized in accessing numerous derivatives, as well as the enantioenrichement of exocyclic allyl alcohol using chiral ligands. Mechanistic studies suggest that bipyridine-ligated Cr(II) complex serves as reactive catalyst with enabling the generation of ketyl radical for the coupling, giving vinyl radical and followed by the combination of Cr and transmetalation with Cp2ZrCl moiety in affording oxazirconiumacycle. This reaction provides a new entry for the mild formation of transient ketyl radicals from widely accessible aliphatic aldehydes for couplings by earth-abundant metal catalysis.

Abstract:Direct functionalizations of two distinct inert C–H bonds represent the most ideal ways to construct C–C bonds. Herein, we report an intermolecular vinylation of aldehydes using alkenes as the vinylating reagents through sequential two-fold C–H functionalizations. The merging of visible light and N-heterocyclic carbene catalysis allows for the coupling of alkenes with aldehydes through a dual catalysis relay enabled cross-dehydrogenative coupling mechanism. Abstract:A new strategy for the direct cleavage of the C(sp3)–OH bond has been developed via activation of free alcohols with neutral diphenyl boryl radical generated from sodium tetraphenylborate under mild visible light photoredox conditions. This strategy has been verified by cross-electrophile coupling of free alcohols and carbon dioxide for the synthesis of carboxylic acids. Direct transformation of a range of primary, secondary, and tertiary benzyl alcohols to acids has been achieved. Control experiments and computational studies indicate that activation of alcohols with neutral boryl radical undergoes homolysis of the C(sp3)–OH bond, generating alkyl radicals. Abstract:An electrophotochemical dual metal-catalyzed protocol for decarboxylative arylation of simple aliphatic carboxylic acids with aryl halides is reported. The relative stabilities of catalytically active metal complexes simultaneously generated at anode and cathode are the key design elements for the success of this convergent paired electrolysis. This new electrophotocatalytic method is mild, robust, and most importantly, capable of accommodating simple primary aliphatic acids including acetic acid - ubiquitous and variegated structural motifs yet remain oddly challenging substrates - directly as native functional groups for decarboxylative C(sp2 )-C(sp3 ) bond formation.

Abstract:Empowered by the ubiquity of carbonyl functional groups in organic compounds, decarbonylative functionalization was prevalent in the construction of complex molecules. Under this context, asymmetric decarbonylative functionalization has emerged as an efficient pathway to accessing chiral motifs. However, ablation of enantiomeric control in a conventional 2e transition metal–catalyzed process was notable because of harsh conditions (high temperatures, etc.) that are usually required. To address this challenge and use readily accessible aldehyde directly, we report the asymmetric radical decarbonylative azidation and cyanation. Diverse aldehydes were directly used as alkyl radical precursor, engaging in the subsequent inner-sphere or outer-sphere ligand transfer where functional motifs (CN and N 3 ) could be incorporated in excellent site- and enantioselectivity. Mild conditions, broad scope, excellent regioselectivity (driven by polarity-matching strategy), and enantioselectivity were shown for both transformations. This radical decarbonylative strategy using aldehydes as alkyl radical precursor has offered a powerful reaction manifold in asymmetric radical transformations to construct functional motifs regio- and stereoselectively.

Abstract:A silver oxidant/Lewis acid catalyst cooperatively promoted three-component carbo-heterofunctionalization of alkenes is reported. A broad range of nucleophiles, including H2O, alcohols, and anilines, can combine with a variety of 1,3-dicarbonyl compounds or analogues as radical precursors to realize difunctionalization of various styrene derivatives. Interestingly, mechanistic investigations demonstrate that the Ag(I) complex of bidentate phosphine ligand quickly oxidizes the radical intermediate generated from the addition of initial radical across alkene double bonds to form carbocation intermediates, which may be the reason behind the excellent yields and high selectivity of such reactions.

Abstract:The asymmetric cross-coupling of unsaturated bonds, hampered by their comparable polarity and reactivity, as well as the scarcity of efficient catalytic systems capable of diastereo- and enantiocontrol, presents a significant hurdle in organic synthesis. In this study, we introduce a highly adaptable photochemical cobalt catalysis framework that facilitates chemo- and stereoselective reductive cross-couplings between common aldehydes with a broad array of carbonyl and iminyl compounds, including N-acylhydrazones, aryl ketones, aldehydes, and α-keto esters. Our methodology hinges on a synergistic mechanism driven by photoredox-induced single-electron reduction and subsequent radical-radical coupling, all precisely guided by a chiral cobalt catalyst. Various optically enriched β-amino alcohols and unsymmetrical 1,2-diol derivatives (80 examples) have been synthesized with good yields (up to 90% yield) and high stereoselectivities (up to >20:1 dr, 99% ee). Of particular note, this approach accomplishes unattainable photochemical asymmetric transformations of aldehydes with disparate carbonyl partners without reliance on any external photosensitizer, thereby further emphasizing its versatility and cost-efficiency.

Abstract:Radical reactions play an important role in modern organic synthetic chemistry. The generation of carbon radicals from the homolytic cleavage of carbon–metal bonds has been widely studied as a fundamental step in organometallic chemistry. However, the implementation of this phenomenon in catalysed cascade reactions, that is, an organometallic-radical relay, is a highly challenging undertaking due to transient radicals generated from these thermodynamically disfavoured endergonic processes. Here we disclose a set of catalytic alkene 1,1-difunctionalization reactions conceptually based on an organometallic-radical relay. This strategy enables a diversity of sp 3 / sp 2 fragments and aryl groups to be simultaneously introduced to the same carbon of both terminal and internal alkenes with outstanding chemo- and regioselectivity. This study provides a concept for the generation of functionalized benzylic radicals, which are difficult to access by classical protocols, from non-radical feedstock alkenes and arylboronic acids.

Abstract:Given the importance and beneficial characteristics of aliphatic CF3 chiral compounds in modern chemistry, efficient strategies for their synthesis are highly sought after. While α-CF3 carboxylic acid is an emerging and easily accessible CF3-containing synthon, its use as a source of fluoroalkyl is highly challenging due to its high oxidation potential. Herein, we disclose a photoelectrocatalytic method for the direct and enantioselective decarboxylative cross-coupling of α-CF3 carboxylic acids. Key to our approach is the strategic integration of LMCT-induced decarboxylative process with classical nickel catalysis. This strategy enables the efficient synthesis of aliphatic chiral CF3 compounds with a broad range of substrates.
