Abstract:β-Thiolated-α-arylated ketones are perversive in bioactive molecules and serve as potential bidentate ligands for catalysis. Herein, a straightforward protocol to access β-thiolated ketones from aldehydes, alkenes, and disulfides enabled by the combination of photocatalysis and N-heterocyclic carbene catalysis is reported. The sequential radical addition to alkenes and subsequent radical-radical coupling cascade process simultaneously forge C-S and C-C bonds. The mild conditions allow for radical relay coupling with a broad functional group tolerance.

Abstract:The radical‐mediated reductive functionalization of aryl halides has been extensively studied. However, the related radical‐mediated intermolecular reductive 1,2‐diarylation of alkenes, using aryl halides as aryl radical sources, remains unexplored. Herein, a new electrophotocatalytic intermolecular reductive 1,2‐diarylation of alkenes is reported using aryl halides and cyanoaromatics to produce polyarylated alkanes. Using synergistic cathodic reduction and visible‐light photoredox catalysis, various electron‐rich and electron‐deficient aryl halides are combined with various alkenes and cyanoaromatics to characterize the broad substrate scope, excellent functional group compatibility, and excellent selectivity of this reaction. Mechanistic investigations reveal that this reaction may proceed via a radical process initiated by the reductive generation of aryl radicals from aryl halides and terminated by radical‐radical coupling with cyanoaromatic radical anions. This method provides an efficient approach to the synthesis of pyrrolidines, structural units prevalent in bioactive compounds and organocatalysts, from readily available starting materials. Abstract:The direct coupling of alkene feedstocks with aldehydes represents an expedient approach to the generation of new and structurally diverse C(sp3)-hybridized alcohols that are primed for elaborated into privileged architectures. Despite their abundance, current disconnection strategies enabling the direct coupling of carbon-carbon π-bonds and aldehydes remain challenging because contemporary methods are often limited by substrate or functional group tolerance, and compatibility in complex molecular environments. Here, we report a coupling between simple alkenes, heteroarenes and unactivated aliphatic aldehydes via an electrochemically induced reductive activation of C–C π bonds. The cornerstone of this approach is the discovery of a rapid alternating polarity (rAP) electrolysis to access and direct, highly reactive radical anion intermediates derived from conjugated alkenes and heterocyclic compounds. Our developed catalyst-free protocol enables direct access to new, and structurally diverse C(sp3)-hybridized alcohol products. This is achieved by the controlled reduction of conjugated alkenes and the C2–C3 π-bond in heteroarenes via an unprecedented reductive dearomative functionalization for heterocyclic compounds. Experimental mechanistic studies demonstrate that rAP electrolysis is necessary for the controlled generation of radical anion intermediates and promotes a kinetically biased single-electron reduction of the C–C π-bonds over aldehydes. Overall, this technology provides a versatile approach to the reductive coupling of olefin and heterocycle feedstocks with aliphatic aldehydes, offering straightforward access to diverse C(sp3)-rich oxygenated scaffolds.

Abstract:Density functional theory investigations revealed that the pyridine-boryl radical generated in situ using 4-cyanopyridine and bis(pinacolato)diboron could be used as a bifunctional "reagent", which serves as not only a pyridine precursor but also a boryl radical. With the unique reactivity of such radicals, 4-substituted pyridine derivatives could be synthesized using α,β-unsaturated ketones and 4-cyanopyridine via a novel radical addition/C-C coupling mechanism. Several controlled experiments were conducted to provide supportive evidence for the proposed mechanism. In addition to enones, the scope could be extended to a wide range of boryl radical acceptors, including various aldehydes and ketones, aryl imines and alkynones. Lastly, this transformation was applied in the late-stage modification of a complicated pharmaceutical molecule.

Abstract:Alkene dicarbofunctionalizations enable the streamlined construction of aliphatic structures and have thus been the subject of intense research efforts. Despite significant progress, catalytic asymmetric variants remain scarce. Inspired by the advantages of reductive cross-coupling approaches, we present here a highly efficient asymmetric intermolecular Ni-catalyzed reductive dicarbofunctionalization of alkenes. Two distinct readily available electrophiles, namely, Csp2- and Csp3-halides, are added simultaneously across a variety of olefins (vinyl amides, vinyl boranes, vinyl phosphonates) at room temperature in a highly regio- and enantioselective manner. The reaction, devoid of sensitive organometallic reagents, takes advantage of an in situ generated chiral alkyl Ni(III)-intermediate to ensure a stereodefined outcome in the Csp3-Csp2 bond-forming reaction. An (l)-(+)-isoleucine chiral bisoxazoline ligand and the presence of coordinating sites on the alkene are key for the successful outcome in these "asymmetric radical relayed reductive couplings" (ARRRCs). Further, multiple transformations of the chiral amides obtained in this process showcase the potential of this new methodology for the straightforward assembly of chiral building blocks such as primary and secondary amines and oxazolines, highlighting its synthetic utility.

Abstract:An NHC organocatalytic radical aminoacylation of alkenes has been developed by using aldehydes as the acylating reagents and activated aryloxy-amides as the N-radical precursors, providing a new platform for modular access to highly functionalized β-aminoketones. The key to the success of this reaction relies on the single electron transfer (SET) event between the enolates of Breslow intermediates and carefully screened N-radical precursors, followed by a radical addition and radical-radical coupling relay process. The protocol features simple and readily available materials (abundant and feed-stock aldehydes and alkenes), mild reaction conditions (metal-, photo- and oxidant-free, in EA at room temperature), easily removable protecting group for late-stage functionalization.

Abstract:In this Communication, we report an unprecedented β-regioselective radical inverse hydroboration (compared with ionic hydroboration) of α,β-unsaturated amides with NHC-BH3 enabled by photoredox catalysis. Density functional theory (DFT) calculations show that the unique photoredox cycle is a key factor to control the β-regioselective radical hydroboration, by lowering the energy barrier in comparison with other pathways. This protocol provides a general and convenient route to construct a wide range of structurally diverse β-borylated amides in synthetically useful yields under mild conditions.

Abstract:A Co-catalyzed highly regio- and enantioselective reductive coupling of alkynes and aldehydes has been developed under visible light photoredox dual catalysis. A variety of enantioenriched allylic alcohols have been obtained by using unsymmetrical internal alkynes and commercially available catalyst, chiral ligand, and reagents. Abstract:The asymmetric cross-coupling of unsaturated bonds, hampered by their comparable polarity and reactivity, as well as the scarcity of efficient catalytic systems capable of diastereo- and enantiocontrol, presents a significant hurdle in organic synthesis. In this study, we introduce a highly adaptable photochemical cobalt catalysis framework that facilitates chemo- and stereoselective reductive cross-couplings between common aldehydes with a broad array of carbonyl and iminyl compounds, including N-acylhydrazones, aryl ketones, aldehydes, and α-keto esters. Our methodology hinges on a synergistic mechanism driven by photoredox-induced single-electron reduction and subsequent radical-radical coupling, all precisely guided by a chiral cobalt catalyst. Various optically enriched β-amino alcohols and unsymmetrical 1,2-diol derivatives (80 examples) have been synthesized with good yields (up to 90% yield) and high stereoselectivities (up to >20:1 dr, 99% ee). Of particular note, this approach accomplishes unattainable photochemical asymmetric transformations of aldehydes with disparate carbonyl partners without reliance on any external photosensitizer, thereby further emphasizing its versatility and cost-efficiency.

Abstract:Radical reactions play an important role in modern organic synthetic chemistry. The generation of carbon radicals from the homolytic cleavage of carbon–metal bonds has been widely studied as a fundamental step in organometallic chemistry. However, the implementation of this phenomenon in catalysed cascade reactions, that is, an organometallic-radical relay, is a highly challenging undertaking due to transient radicals generated from these thermodynamically disfavoured endergonic processes. Here we disclose a set of catalytic alkene 1,1-difunctionalization reactions conceptually based on an organometallic-radical relay. This strategy enables a diversity of sp 3 / sp 2 fragments and aryl groups to be simultaneously introduced to the same carbon of both terminal and internal alkenes with outstanding chemo- and regioselectivity. This study provides a concept for the generation of functionalized benzylic radicals, which are difficult to access by classical protocols, from non-radical feedstock alkenes and arylboronic acids.

Abstract:The mild catalytic generation of ketyl radicals for organic transformations remains an unsolved issue, although it facilitates the discovery of metal-catalyzed reactions in feature of high functional group tolerance. We report here the generation of ketyl radicals and coupling with alkynes that were enabled by cost-effective chromium catalysis, allowing for the formation of valuable E-exocyclic allyl alcohols in high stereo- and chemoselectivity. A broad range of synthetically useful functional groups that are sensitive to strong reductants are compatible by the catalytic system, providing access to diverse substituted E-exocyclic allyl alcohols under mild conditions. The appended hydroxyl groups in the products are facilely late-stage functionalized in accessing numerous derivatives, as well as the enantioenrichement of exocyclic allyl alcohol using chiral ligands. Mechanistic studies suggest that bipyridine-ligated Cr(II) complex serves as reactive catalyst with enabling the generation of ketyl radical for the coupling, giving vinyl radical and followed by the combination of Cr and transmetalation with Cp2ZrCl moiety in affording oxazirconiumacycle. This reaction provides a new entry for the mild formation of transient ketyl radicals from widely accessible aliphatic aldehydes for couplings by earth-abundant metal catalysis.

Abstract:The stable α-boryl aldehydes are important amphoteric synthetic intermediates that act as linchpins in organic synthesis. However, current approaches to these important building blocks typically rely on oxidative processes, which might lead to issues such as lack of product diversity and functional group compatibility. Here, we report the design and utilization of 2-(BMIDA) acrylaldehyde as a suitable coupling partner toward photoredox-catalyzed intermolecular radical addition reaction for the rapid synthesis of functionalized α-boryl aldehydes. With a commercially available photocatalyst and readily accessible alkyltrifluoroborates as radical precursors, a wide range of α-boryl aldehydes were obtained in up to 90% yield. Preliminary mechanistic studies suggested that the reaction might proceed through intermolecular radical addition and radical-polar crossover processes. The synthetic potential of the method was further demonstrated by scale-up reactions, several synthetic transformations, and derivatization of bioactive molecules. Moreover, by simply switching the radical precursors to α-amino acids, diverse 4-boryl 2,3-dihydropyrroles could be accessed via the intermolecular radical addition/cyclization sequence. Furthermore, the MIDA boronate-bearing six- or seven-membered heterocycles could also be readily obtained when the corresponding β- or γ-amino acid was used. This strategy provides a facile, robust, and diversity-oriented synthetic approach to functionalized α-boryl aldehydes or borylated heterocycles.
