Abstract:To mimic the natural fibrous structure of the tissue extracellular matrix, a nano-fibrous silk fibroin (SF)/sodium alginate (SA) composite scaffold was fabricated by a thermally-induced phase-separation method. The effects of SF/SA ratio on the structure and the porosity of the composite scaffolds were examined. Scanning electron microscopy and porosity results showed that the 5SF/1SA and 3SF/1SA scaffolds possessed an excellent nano-fibrous structure and a porosity of more than 90%. Fourier transform infrared, X-ray diffraction, and differential scanning calorimetry results indicated the physical interaction between SF and SA molecules and their good compatibility in the 5SF/1SA and 3SF/1SA scaffolds, whereas they showed less compatibility in the 1SF/1SA scaffold. Cell culture results showed that MG-63 cells can attach and grow well on the surface of the SF/SA scaffolds. The nano-fibrous SF/SA scaffold can be potentially used in tissue engineering.

Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.

Abstract:Bombyx mori silk is of great interest to people for its outstanding mechanical and biological properties. However, the traditional electrospun regenerated silk fibroin (RSF) scaffolds from aqueous solution were weak and had limited applications. This study was to fabricate reinforced scaffolds with well-aligned RSF fibers electrospun on a layer of native extracellular matrix, bladder acellular matrix graft (BAMG). The silk fibroin fibers were well-aligned as a grill in multiple layers. Both the BAMG and the grill structure significantly improved the tensile properties and suture retention of the composite scaffolds, which can be sutured well with tissue during implantation. In vitro assay indicates that the scaffolds had a good biocompatibility. Porcine iliac endothelial cells (PIECs) attached and proliferated well on the vascular endothelial growth factor (VEGF) loaded scaffolds compared with those without VEGF. Moreover, the grill-like structure guides PIECs well along the aligned fiber.
Development of Small-Diameter Artificial Vascular Grafts Using Transgenic Silk Fibroin

Abstract:Silk fibroin (SF) is a suitable material for vascular prostheses for small arteries. SF is useful not only as a base material for artificial vascular grafts but also as a coating material. This study prepared three types of transgenic SF (vascular endothelial growth factor (VEGF), Arg-Glu-Asp-Val (REDV), and Tyr-Ile-Gly-Ser-Arg (YIGSR)) incorporating expression factors that are thought to be effective for endothelialization as coating materials. We compared the contribution of these materials to early endothelialization in vivo when using them as a porous transgenic SF coating. A porous coating of transgenic SF containing VEGF, REDV, and YIGSR was applied to a silk small-diameter artificial vascular graft base with a diameter of 1.5 mm and a length of 3 cm. Two and four weeks after implantation of these artificial grafts into the abdominal aorta of rats, they were removed and evaluated by histologic examination. Transgenic SF coating incorporating VEGF and REDV demonstrated higher tissue infiltration and continuous endothelialization in the center of the graft compared to YIGSR at 4 weeks after implantation. VEGF and REDV are capable of endothelialization as early as 4 weeks after implantation, confirming the usefulness of transgenic SF when used as a porous coating.
Fabrication and characterization of porous tubular silk fibroin scaffolds.

Abstract:Silk fibroin (SF) has been one of promising resources of biotechnology and biomedical materials due to its unique properties. Here, different sizes of porous tubular scaffolds were fabricated from a SF aqueous solution with the addition of poly(ethylene glycol diglycidyl ether) (PGDE). The scaffolds were generally flexible and transparent at the wet state with a pore size of 81-128 mu m and porosity of 90-96%, depending on the concentrations of SF and PGDE. The mechanical properties measurement showed that the tubular SF scaffolds had satisfying tensile and compression properties, especially the excellent deformation-recovery ability. FT-IR spectra indicated that the SF in the tubular scaffolds was in a beta-sheet structure, and no PGDE characteristic band was observed, suggesting that the PGDE could be removed from the scaffolds by soaking in deionized water. The cell compatibility of scaffolds was evaluated, and no obvious cytotoxicity to mouse L-929 fibroblasts was detected. (C) Koninklijke Brill NV, Leiden, 2009
Electrospun silk fibroin/fibrin vascular scaffold with superior mechanical properties and biocompatibility for applications in tissue engineering

Abstract:Electrospun scaffolds play important roles in the fields of regenerative medicine and vascular tissue engineering. The aim of the research described here was to develop a vascular scaffold that mimics the structural and functional properties of natural vascular scaffolding. The mechanical properties of artificial vascular tissue represent a key issue for successful transplantation in small diameter engineering blood vessels. We blended silk fibroin (SF) and fibrin to fabricate a composite scaffold using electrospinning to overcome the shortcomings of fibrin with respect to its mechanical properties. Subsequently, we then carefully investigated the morphological, mechanical properties, hydrophilicity, hemocompatibility, degradation, cytocompatibility and biocompatibility of the SF/fibrin (0:100), SF/fibrin (15:85), SF/fibrin (25:75), and SF/fibrin (35:65) scaffolds. Based on these in vitro results, we implanted SF/fibrin (25:75) vascular scaffold subcutaneously and analyzed its in vivo degradation and histocompatibility. The fiber structure of the SF/fibrin hybrid scaffold was smooth and uniform, and its fiber diameters were relatively small. Compared with the fibrin scaffold, the SF/fibrin scaffold clearly displayed increased mechanical strength, but the hydrophilicity weakened correspondingly. All of the SF/fibrin scaffolds showed excellent blood compatibility and appropriate biodegradation rates. The SF/fibrin (25:75) scaffold increased the proliferation and adhesion of MSCs. The results of animal experiments confirmed that the degradation of the SF/fibrin (25:75) scaffold was faster than that of the SF scaffold and effectively promoted tissue regeneration and cell infiltration. All in all, the SF/fibrin (25:75) electrospun scaffold displayed balanced and controllable biomechanical properties, degradability, and good cell compatibility. Thus, this scaffold proved to be an ideal candidate material for artificial blood vessels.

Abstract:Regenerated silk fibroin (RSF)/bladder acellular matrix (BAM)/hyaluronic acid (HA) scaffolds were electrospun from aqueous solution.The morphology and biological performances in vitro were characterized by scanning electron microscope, ELISA, MTT and fluorescent microscopy.Results showed that the concentrations of spinning solutions had limited impact on the morphology of the scaffolds.The existence, bioactivity and continuous release behaviors of vascular endothelial growth factor (VEGF), platelet-derived growth factor BB (PDGF-BB), keratinocyte growth factor (KGF) from the scaffolds were confirmed and determined.Due to the bioactivity of the growth factors, the RSF/BAM/HA scaffolds showed better performance for cell growth and proliferation than bare RSF scaffolds.
A Biomimetic Silk Fibroin/Sodium Alginate Composite Scaffold for Soft Tissue Engineering

Abstract:A cytocompatible porous scaffold mimicking the properties of extracellular matrices (ECMs) has great potential in promoting cellular attachment and proliferation for tissue regeneration. A biomimetic scaffold was prepared using silk fibroin (SF)/sodium alginate (SA) in which regular and uniform pore morphology can be formed through a facile freeze-dried method. The scanning electron microscopy (SEM) studies showed the presence of interconnected pores, mostly spread over the entire scaffold with pore diameter around 54~532 μm and porosity 66~94%. With significantly better water stability and high swelling ratios, the blend scaffolds crosslinked by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) provided sufficient time for the formation of neo-tissue and ECMs during tissue regeneration. Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) results confirmed random coil structure and silk I conformation were maintained in the blend scaffolds. What's more, FI-TR spectra demonstrated crosslinking reactions occurred actually among EDC, SF and SA macromolecules, which kept integrity of the scaffolds under physiological environment. The suitable pore structure and improved equilibrium swelling capacity of this scaffold could imitate biochemical cues of natural skin ECMs for guiding spatial organization and proliferation of cells in vitro, indicating its potential candidate material for soft tissue engineering.
Facile Incorporation of REDV into Porous Silk Fibroin Scaffolds for Enhancing Vascularization of Thick Tissues

Abstract:Rapid neovascularization within scaffolds is critical for the regeneration of thick complex tissues. The surface immobilization of peptides and other active molecules have been explored to improve the vascularization capacity of implants. However, the rapid degradation of these molecules, the reaction conditions and cross-linking usually result in decreased vascularization capability. Here, we introduced a temperate, all-aqueous process to achieve bulk porous silk fibroin (SF) scaffolds. A temperature controlled process was used to induce the water stable structure by SF self-assembly. Arg-Glu-Asp-Val (REDV) peptides were added into SF solution and fixed within SF scaffolds during the self-assembly process. The results showed that the functionalized scaffolds markedly promoted the adhesion of endothelial cells in vitro. Moreover, the in vivo studies indicated enhanced cell infiltration in the bulk functionalized SF scaffolds and impressive vascularization at 4 weeks post-implantation. The functionalized scaffolds demonstrated excellent vascularization capability, providing an exciting biomaterial option for thick tissue regeneration.
Three-dimensional silk fibroin microsphere-nanofiber scaffolds for vascular tissue engineering

Abstract:A significant limitation in the engineering of artificial small-diameter vascular scaffolds is that the number of endothelial cells (ECs) is not sufficient to generate a confluent coverage of the vascular scaffolds, so that the surfaces of vascular scaffolds form thrombus via platelet adhesion and aggregation. Thrombus decrease relies on three-dimensional (3D) scaffolds to mimic the natural extracellular matrix (ECM) as templates to regulate cell behavior and facilitate tissue maturation. Here, we developed 3D scaffolds consisting of silk fibroin (SF) nanofibers and homogeneous microspheres by electrospinning and microfluidics. The nanofibers with diameters ranging from 250 to 350 nm doped with microspheres (2–10 μm) formed bridge-shaped structures. ECs were seeded and maintained on the 3D microsphere-nanofiber scaffolds with a mean fiber diameter of 300 nm. A 10% higher ratio of cell proliferation on 3D microsphere-nanofiber SF scaffolds was noted as compared to that on microporous and sponge-like SF scaffolds with small surface network fabricated by freeze-drying. Moreover, the gene transcript levels including CD146, VE-C and PECAM-1 were better preserved on 3D microsphere-nanofiber SF scaffolds than those on freeze-dried scaffolds. Thus, the developed 3D microsphere-nanofiber structure may have a myriad of applications in vascular tissue engineering scaffolds and cardiovascular devices.

Abstract:Silk-based materials have great potentials in medical applications because of the proper mechanical properties and outstanding biocompatibility. This paper reports a novel method to prepare multi-layered silk fibroin (SF) tissue engineering scaffolds with well-aligned fibers oriented in different angles by an electrospinning process. The resulted aligned fibers construct the scaffolds with a grill-like structure, large area and improved mechanical properties, which have been demonstrated by scanning electron microscopy and tensile test. Biocompatibility analysis proves that the grill-like structure well guides the adhesion, proliferation and differentiation of porcine iliac endothelial cells along the aligned SF fibers in different directions.

Abstract:Journal of Bioactive and Compatible Polymers, Ahead of Print. Biomaterials play an important role in biomedical applications serving as scaffolds in tissue engineering and substrates for cell growth by providing structural supports. Signaling cues for cellular events can be also presented, including cell viability, adhesion, spreading, and function developments. In this study, we fabricated a hierarchical nanofibrous scaffold through electrospinning silk fibroin (SF) and decellularized extracellular matrix of the small intestine submucosa (SIS-dECM). The addition of SIS-dECM improved the hydrophilic features of the resultant composite nanofibrous substrates as compared to SF nanofibrous substrates. The biocompatibility of the nanofibrous scaffolds was thoroughly investigated by culturing meningeal fibroblasts on SF and SF/SIS-dECM nanofibrous substrates. Cell viability, proliferation, focal adhesion, and quantitative measurements of cell adhesion-related gene expression levels revealed that the addition of SIS-dECM favored the adhesion and spreading of fibroblast cells. Therefore, the SF/SIS-dECM nanofibers provided a suitable substrate for cell growth and the present study suggested that the nanofibrous substrates hold high potential for bioapplications.

Abstract:Electrospun silk fibroin (SF) scaffolds have drawn much attention because of their resemblance to natural tissue architecture such as extracellular matrix, and the biocompatibility of SF as a candidate material to replace collagen. However, electrospun scaffolds lack the physical integrity of bone tissue scaffolds, which require resistance to mechanical loadings. In this work, we propose membrane-reinforced electrospun SF scaffolds by a serial process of electrospinning and freeze-drying of SF solutions in two different solvents: formic acid and water, respectively. After wet electrospinning followed by replacement of methanol with water, SF nanofibers dispersed in water were mixed with aqueous SF solution. Freeze-drying of the mixed solution resulted in 3D membrane-connected SF nanofibrous scaffolds (SF scaffolds) with a thickness of a few centimeters. We demonstrated that the SF concentration of aqueous SF solution controlled the degree of membrane reinforcement between nanofibers. It was also shown that both increase in degree of membrane reinforcement and inclusion of hydroxyapatite (HAP) nanoparticles resulted in higher resistance to compressive loadings of the SF scaffolds. Culture of human osteoblasts on collagen, SF, and SF-HAP scaffolds showed that both SF and SF-HAP scaffolds had biocompatibility and cell proliferation superior to that of the collagen scaffolds. SF-HAP scaffolds with and without BMP-2 were used for in vivo studies for 4 and 8 weeks, and they showed enhanced bone tissue formation in rat calvarial defect models.

Growth of fibroblast and vascular smooth muscle cells in fibroin/collagen scaffold

Abstract:In our recent study, a novel fibroin/collagen scaffold with improved mechanical properties and controllable porous structure was prepared through freeze–drying method. In this research, the cyto-compatibility was further studied, using fibroblast and vascular smooth muscle cells (VSMC) as the model cells. MTT results indicated that the growth of fibroblast and VSMC both further improved in the fibroin/collagen scaffold than in pure fibroin scaffolds. The confocal and SEM results showed that fibroblast cells and VSMCs had better adhesion and spreading properties in the fibroin/collagen scaffolds. Although further studies, such as the extracellular matrix production and the functional gene expression, are necessary to clarify the biocompatibility of the fibroin/collagen scaffolds, the present results indicate that the fibroin/collagen scaffold is a new scaffold material suitable for tissue engineering. On the other hand, the mild and all-aqueous preparation processes also make it possible to embed different growth factors inside the scaffolds to maximize cell functions and the formation of specific tissues.
Preparation of recombinant human-like collagen/fibroin scaffold and its promoting effect on vascular cells biocompatibility

Abstract:A novel recombinant human-like collagen/fibroin scaffold has been prepared previously, which has high porosity, controllable pore size, and much better mechanical properties than the reported fibroin-based scaffold. In this research, the cell responses of vascular smooth muscle cells to this blend scaffold were examined in vitro. Cell morphology, adherence, and growth in scaffolds were observed by scanning electron microscopy, laser scanning confocal microscopy after staining of the cells with propidium iodide at 1, 3, 5, and 7 days, respectively. A wide range of measurements, including 3-[4,5–dimethylthiazol-2-yl]-2, 5-diphenyl tetrasodium bromide assay, and total intracellular protein content at the end of 7 days culture, were conducted. An increase of viability and protein content of vascular smooth muscle cells cultured in recombinant human-like collagen/fibroin scaffold was found. The laser scanning confocal microscopy and scanning electron microscopy results confirm that the cells readily adhered and proliferation in the blend than in fibroin scaffold, and indicate a better adhesion process. The positive effects were especially significant for vascular smooth muscle cells. The recombinant human-like collagen/fibroin scaffold could be a promising biomaterial for vascular tissue engineering.

Microstructured silk fiber scaffolds with enhanced stretchability

Abstract:Despite extensive research, current methods for creating three-dimensional (3D) silk fibroin (SF) scaffolds lack control over molecular rearrangement, particularly in the formation of β-sheet nanocrystals that severely embrittle SF, as well as hierarchical fiber organization at both micro- and macroscale. Here, we introduce a fabrication process based on electrowriting of aqueous SF solutions followed by post-processing using an aqueous solution of sodium dihydrogen phosphate (NaH2PO4). This approach enables gelation of SF chains via controlled β-sheet formation and partial conservation of compliant random coil structures. Moreover, this process allows for precise architecture control in microfiber scaffolds, enabling the creation of 3D flat and tubular macro-geometries with square-based and crosshatch microarchitectures, featuring inter-fiber distances of 400 μm and ~97% open porosity. Remarkably, the crosslinked printed structures demonstrated a balanced coexistence of β-sheet and random coil conformations, which is uncommon for organic solvent-based crosslinking methods. This synergy of printing and post-processing yielded stable scaffolds with high compliance (modulus = 0.5–15 MPa) and the ability to support elastic cyclic loading up to 20% deformation. Furthermore, the printed constructs supported in vitro adherence and growth of human renal epithelial and endothelial cells with viability above 95%. These cells formed homogeneous monolayers that aligned with the fiber direction and deposited type-IV collagen as specific marker of healthy extracellular matrix, indicating that both cell types attach, proliferate, and organize their own microenvironment within the SF scaffolds. These findings represent a significant development in fabricating organized stable SF scaffolds with unique microfiber structures and mechanical and biological properties that make them highly promising for tissue engineering applications.

Abstract:Silk Fibroin Microgel Scaffolds In article number 2313354, Fatemeh Karimi, Jelena Rnjak‐Kovacina, and co‐workers introduce a novel silk fibroin microgel platform that enhances cell growth and infiltration in vitro and in vivo. Silk microgels are fabricated using a microfluidic device and stabilized through visible light‐initiated crosslinking of native tyrosines. The microgels are covalently annealed to form microporous scaffolds. Notably, the microgel size and scaffold porosity influence the inflammatory response to these implants.

Abstract:Fibrous membranes played an important role to prepare tubular scaffolds for muscular artery regeneration. In this study, a strategy has been developed to combine silk fibroin (SF) with highly porous electrospun poly(L-lactic acid) (PLLA) fibrous membrane towards vascular scaffolds. After PLLA fibres were electrospun and collected, they were immersed into acetone to generate a porous structure with ultra-high surface area. While the pores on PLLA fibres were fulfilled with SF solution and dried, SF was coated uniformly and tightly on PLLA fibres. A multi-layer tubular structure of the tunica media was simulated by winding and stacking a strip of electrospun fibrous membrane. In vitro viability and morphology studies of A7r5 smooth muscle cells were undertaken for up to 14 days. Because the hydrophilicity of SF/PLLA composite fibres were improved dramatically, it had a positive effect on cell adhesion rate (97%) and proliferation (64.4%). Moreover, good cell morphology was observed via a multiphoton laser confocal microscope on SF/PLLA bioactive materials. These results demonstrated that the hierarchical porous SF/PLLA fibrous membranes are promising off-the-shelf scaffolds for muscular artery regeneration.

A Nontoxic and Biocompatible Method for Augmenting Mechanical Strength of Acellular Matrix by Silk Fibroin Impregnation

Abstract:Biological scaffolds are plagued by poor biomechanical properties and untimely degradation. These limitations have yet to be addressed without compromising their biocompatibility. It is desirable to avoid inflammation and have degradation with concomitant host collagen deposition or even site-appropriate in situ regeneration for the successful outcome of an implanted biological scaffold. This work aims to achieve this by utilizing a biocompatible method to modify acellular scaffolds by impregnating alkaline-catalyzed citric acid (CA) cross-linking between the extracellular matrix proteins and silk fibroin (SF)/SF-gelatin (SFG) blends. Combinatorial detergent decellularization was employed to prepare a decellularized porcine liver scaffold (DPL). After proving the decellularization efficiency, the scaffold underwent modification by vacuum impregnation with CA containing SF (SF100DPL) and SFG blends (SFG5050DPL and SFG3070DPL) following pre-cross-linking, drying, and post-cross-linking. The subsequent strength augmentation was demonstrated by significant improvement in tensile strength from 2.4 ± 0.4 MPa (DPL) to, 3.8 ± 0.7 MPa (SF100DPL), 3.4 ± 0.7 MPa (SFG5050DPL), and 3.5 ± 0.2 MPa (SFG3070DPL); Young's modulus from 8.7 ± 1.8 MPa (DPL) to 20 ± 1.9 MPa (SF100DPL), 13.3 ± 2.6 MPa (SFG5050DPL), and 16 ± 1.2 MPa (SFG3070DPL); and suture retention strength from 0.9 ± 0.08 MPa (DPL) to 2.3 ± 0.2 MPa (SF100DPL), 2.8 ± 1.2 MPa (SFG5050DPL), and 2.6 ± 0.9 MPa (SFG3070DPL). The degradation resistance of the modified scaffolds was also markedly improved. Being cytocompatible, its ability to incite tolerable inflammatory and immune responses was confirmed by rat subcutaneous implantation for 14, 30, and 90 days, in terms of inflammatory cell infiltration, neoangiogenesis, and in vitro cytokine release to assess B-cell and T-cell activation. Such ECM composite scaffolds with appropriate strength and biocompatibility offer great promise in soft tissue repair applications such as skin grafting.

Abstract:In bone tissue engineering, a qualified scaffold should provide a three-dimensional porous structure mimicking the extracellular matrix of bone except good biological properties. In this study, biomorphic silk fibroin (SF) scaffolds were fabricated from cane by an innovative biotemplating-negative mold process. The physicochemical properties, in vitro enzymatic degradation behavior and biocompatibility of the biomorphic SF scaffolds were investigated. The results showed that the scaffolds well inherited the original porous morphology of cane, and possessed good mechanical stability. The scaffolds had a compressive modulus of 1.56 +/- 0.08 MPa and a porosity of 82.73%, and exhibited a bimodal pore size distribution (15 and 172 mu m). The degradation ratio of the SF scaffolds increased with prolonging degradation time and reached 29% within 21 days when exposed to 1.0 U/mL collagenase IA. The in vitro cytocompatibility evaluation indicated that the scaffolds could support cell attachment, proliferation, and osteogenic differentiation of osteoblast- like MC3T3-E1 cells, as assessed by SEM, fluorescent staining, MTT, and ALP activity assays.
