Abstract:To mimic the natural fibrous structure of the tissue extracellular matrix, a nano-fibrous silk fibroin (SF)/sodium alginate (SA) composite scaffold was fabricated by a thermally-induced phase-separation method. The effects of SF/SA ratio on the structure and the porosity of the composite scaffolds were examined. Scanning electron microscopy and porosity results showed that the 5SF/1SA and 3SF/1SA scaffolds possessed an excellent nano-fibrous structure and a porosity of more than 90%. Fourier transform infrared, X-ray diffraction, and differential scanning calorimetry results indicated the physical interaction between SF and SA molecules and their good compatibility in the 5SF/1SA and 3SF/1SA scaffolds, whereas they showed less compatibility in the 1SF/1SA scaffold. Cell culture results showed that MG-63 cells can attach and grow well on the surface of the SF/SA scaffolds. The nano-fibrous SF/SA scaffold can be potentially used in tissue engineering.

Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.

Abstract:We present a silk biomaterial platform with highly tunable mechanical and degradation properties for engineering and regeneration of soft tissues such as, skin, adipose, and neural tissue, with elasticity properties in the kilopascal range. Lyophilized silk sponges were prepared under different process conditions and the effect of silk molecular weight, concentration and crystallinity on 3D scaffold formation, structural integrity, morphology, mechanical and degradation properties, and cell interactions in vitro and in vivo were studied. Tuning the molecular weight distribution (via degumming time) of silk allowed the formation of stable, highly porous, 3D scaffolds that held form with silk concentrations as low as 0.5% wt/v. Mechanical properties were a function of silk concentration and scaffold degradation was driven by beta-sheet content. Lyophilized silk sponges supported the adhesion of mesenchymal stem cells throughout 3D scaffolds, cell proliferation in vitro, and cell infiltration and scaffold remodeling when implanted subcutaneously in vivo.
Preparation and characterization of nano-hydroxyapatite/silk fibroin porous scaffolds.

Abstract:Novel tissue engineering scaffold materials of nano-hydroxyapatite (nHA)/silk fibroin (SF) biocomposite were prepared by freeze-drying. The needle-like nHA crystals of about 10 nm in diameter by 50-80 nm in length, which were uniformly distributed in the porous nHA/SF scaffolds, were prepared by a co-precipitation method with a size. The as-prepared nHA/SF scaffolds showed good homogeneity, interconnected pores and high porosity. XRD and FT-IR analysis suggested that the silk fibroin was in beta-sheet structure, which usually provides outstanding mechanical properties for silk materials. In this work, composite scaffolds containing as high as 70% (w/w) nHA were prepared, which had excellent compressive modulus and strength, higher than the scaffolds at low nHA content level and other porous biodegradable polymeric scaffolds often considered in bone-related tissue engineering reported previously. The cell compatibility of composite scaffolds was evaluated through cell viability by MTT assay. All these results indicated that these nHA/SF scaffold materials may be a promising biomaterial for bone tissue engineering.
Fabrication and characterization of porous tubular silk fibroin scaffolds.

Abstract:Silk fibroin (SF) has been one of promising resources of biotechnology and biomedical materials due to its unique properties. Here, different sizes of porous tubular scaffolds were fabricated from a SF aqueous solution with the addition of poly(ethylene glycol diglycidyl ether) (PGDE). The scaffolds were generally flexible and transparent at the wet state with a pore size of 81-128 mu m and porosity of 90-96%, depending on the concentrations of SF and PGDE. The mechanical properties measurement showed that the tubular SF scaffolds had satisfying tensile and compression properties, especially the excellent deformation-recovery ability. FT-IR spectra indicated that the SF in the tubular scaffolds was in a beta-sheet structure, and no PGDE characteristic band was observed, suggesting that the PGDE could be removed from the scaffolds by soaking in deionized water. The cell compatibility of scaffolds was evaluated, and no obvious cytotoxicity to mouse L-929 fibroblasts was detected. (C) Koninklijke Brill NV, Leiden, 2009

Abstract:In order to use a simple and ecofriendly method to prepare porous silk scaffolds, aqueous silk fibroin solution (ASF) was extracted from silk gland of 7-day-old fifth instar larvae of Bombyx mori (B. mori). SDS-page analysis indicated that the obtained fibroin had a molecular weight higher than 200 kDa. The fabrication of porous scaffolds from ASF was achieved by using the freeze-drying method. The pore of porous scaffolds is homogenous and tends to become smaller with an increase in the concentration of ASF. Conversely, the porosity is decreased. The porous scaffolds show impressive compressive strength which can be as high as 6.9 +/- 0.4 MPa. Furthermore, ASF has high cell adhesion and growth activity. It also exhibits high ALP activity. This implies that porous scaffolds prepared from ASF have biocompatibility. Therefore, the porous scaffolds prepared in this study have potential application in tissue engineering due to the impressive compressive strength and biocompatibility.
Silk-Based Scaffold for Ligament Tissue Engineering

Abstract:In recent years, silk has been increasingly studied as the scaffold for ligament tissue engineering due to the biocompatibility, slow degradability, and remarkable mechanical properties. Braided silk scaffold modified with short polypeptide also significantly increases collagen synthesis on it. To increase cell attachment and tissue infiltration, the braided scaffold can be incorporated with silk-gelatin microsponges. A novel silk cable-reinforced gelatin/silk fibroin hybrid scaffold was fabricated, which, apart from providing proper mechanical strength and enlarged surface area, also supported the proliferation and differentiation of MSCs on it. The knitted silk mesh is another important silk-based scaffold for its excellent mechanical properties and good nutrients transport. To prevent cells from leaking out of scaffold after seeding, freeze-dried silk microsponges were incorporated into the macro pores of knitted scaffold. In vitro culture demonstrated that MSCs on scaffolds proliferated vigorously and produced abundant collagen. The transcription levels of ligament-specific genes (collagen I, collagen III, and tenascin-C) also increased significantly with time. The comparison of MSCs and fibroblasts as cell sources for ligament tissue engineering demonstrated that MSC was the most suitable candidate for its vigorous proliferation and ECM production. The MSCs/knitted scaffolds were implanted into rabbits to regenerate ACL in vivo. After 24 weeks, histology observation showed that MSCs were distributed throughout the regenerated ligament and exhibited fibroblast morphology. The key ligament ECM components including collagen I, collagen III, and tenascin-C were produced prominently. Furthermore, direct ligament-bone insertion with typical four zones (bone, mineralized fibrocartilage, fibrocartilage, ligament) was reconstructed, which resembled the native structures of ACL-bone insertion. The tensile strength of regenerated ligament also met the mechanical requirements of daily activities. In conclusion, the results imply that silk scaffold has great potentials in future clinical applications.

Abstract:Preparation of 3D scaffolds for bone tissue engineering (BTE) is a challenging task as it required appropriate pore size and porosities, excellent mechanical properties, controlled rate of bioerosion. The focus of this work is the fabrication of silk fibroin (SF) 3D microparticle scaffolds with the incorporation of hydroxyapatite (HA) and calcium sulfate (CaS) as bioceramics. Physicochemical characterization shows ∽30% filler loading, ∽ 40% optimum porosity with >100 µm pore size for these filled/unfilled scaffolds. Nanoindentation studies show improved Young's modulus at microparticle level with the incorporation of bioceramics. There is a three‐fold increase in Young's Modulus in the case of SF‐HA while two‐fold increase for SF‐CaS. In‐vitro bioerosion study results in early bioerosion with SF‐CaS scaffold while prolonged bioerosion with SF‐HA scaffold. In‐vitro osteo‐regenerative potential is analyzed by estimating ALP, BMP‐2 and OCN. SF‐CaS supports the early stage differentiation as compared to SF‐50%HA scaffolds, which predominantly support at late stage. The expression of TNF‐α, suggesting a reduced risk of immune rejection. This work, therefore, concludes that although SF support bone tissue regeneration, the choice of bioceramic enhances the applicability in various clinical scenarios by providing a controlled rate of bioerosion, tunable speed of osteo‐regeneration and improved load bearing capacities.This article is protected by copyright. Abstract:In this study, biomimetic sodium alginate (SA)/silk fibroin (SF) scaffolds were successfully fabricated by supercritical CO2 technology. The SA/SF scaffolds exhibited an interconnected porous and extracellular matrix (ECM)-like nanofibrous structures. Moreover, the SA microparticles were embedded in the SF scaffolds. Increasing the content of SA microparticles could improve tensile strength and compressive strength of the SF scaffolds and reduce the porosity of the SF scaffolds. The addition of the SA microparticles could also regulate the degradation rate of the SA/SF scaffolds. Furthermore, the results of in vitro biocompatibility evaluation, indicated that the SA/SF scaffolds exhibited no obvious cytotoxicity and higher cell adhesion ability and were more favorable for L929 fibroblasts proliferation than pure SF scaffolds. Therefore, the SA/SF scaffolds with ECM-like nanofibrous and interconnected porous structure have potential application in skin tissue engineering.

Abstract:Journal of Bioactive and Compatible Polymers, Ahead of Print. Biomaterials play an important role in biomedical applications serving as scaffolds in tissue engineering and substrates for cell growth by providing structural supports. Signaling cues for cellular events can be also presented, including cell viability, adhesion, spreading, and function developments. In this study, we fabricated a hierarchical nanofibrous scaffold through electrospinning silk fibroin (SF) and decellularized extracellular matrix of the small intestine submucosa (SIS-dECM). The addition of SIS-dECM improved the hydrophilic features of the resultant composite nanofibrous substrates as compared to SF nanofibrous substrates. The biocompatibility of the nanofibrous scaffolds was thoroughly investigated by culturing meningeal fibroblasts on SF and SF/SIS-dECM nanofibrous substrates. Cell viability, proliferation, focal adhesion, and quantitative measurements of cell adhesion-related gene expression levels revealed that the addition of SIS-dECM favored the adhesion and spreading of fibroblast cells. Therefore, the SF/SIS-dECM nanofibers provided a suitable substrate for cell growth and the present study suggested that the nanofibrous substrates hold high potential for bioapplications.

Abstract:The expanding field of tissue engineering has required the necessity of developing biomaterials that are tissue compatible, biodegradable, and comparable in mechanical properties to that of native tissue. We propose that the blending of two natural polymers, silk fibroin (SF) and chitosan (CS), into a 3D scaffold will provide unique chemical, structural, and mechanical properties that can be utilized for in vivo tissue regeneration. SF is an attractive material for biomedical applications because it is a fibrous protein that has high permeability to oxygen and water, relatively low thrombogenicity, low inflammatory response, protease susceptibility, supports cell adhesion and growth, and, foremost, high tensile strength with flexibility. CS is a crystalline polysaccharide, with structure similar to glycosaminoglycans, that has good wound healing properties, is nontoxic, and has minimal foreign body reactions. We hypothesized that increasing the SF-to-CS ratio would increase the ultimate tensile strength and elastic modulus and decrease the water capacity of the SFCS scaffolds. With increasing content of silk fibroin, it is observed that the ultimate tensile strength and elastic modulus increase significantly. The ultimate tensile strength and the elastic modulus were significantly higher in the short axis direction for 25:75 and 50:50 SFCS blends as compared to the long axis (p<0.05), while they were similar for the 75:25 SFCS blend. However, no differences were observed in the strain at failure among blends or due to directionality of applied strain. Increasing the chitosan content resulted in an increased water capacity of SFCS blends.

Abstract:Bone tissue engineering is a powerful tool to treat bone defects caused by trauma, infection, tumors and other factors. Both silk fibroin (SF) and chitosan (CS) are non-toxic and have good biocompatibility, but are poor biological scaffolds when used alone. In this study, the microscopic structure and related properties of SF/CS composite scaffolds with different component ratios were examined. The scaffold material most suitable for osteoblast growth was determined, and these results offer an experimental basis for the future reconstruction of bone defects. First, via freeze-drying and chemical crosslinking methods, SF/CS composites with different component ratios were prepared and their structure was characterized. Changes in the internal structure of the SF and CS mixture were observed, confirming that the mutual modification between the two components was complete and stable. The internal structure of the composite material was porous and three-dimensional with a porosity above 90%. We next studied the pore size, swelling ratio, water absorption ratio, degradation and in vitro cell proliferation. For the 40% SF-60% CS group, the pore size of the scaffold was suitable for the growth of osteoblasts, and the rate of degradation was steady. This favors the early adhesion, growth and proliferation of MG-63 cells. In addition to good biocompatibility and satisfactory cell affinity, this material promotes the secretion of extracellular matrix materials by osteoblasts. Thus, 40% SF-60% CS is a good material for bone tissue engineering.

Abstract:From a structural perspective, an ideal scaffold ought to possess interconnected macro pores with sizes from 10 to 100 mu m to facilitate cell infiltration and tissue formation, as well as similar topography to the natural extracellular matrix (ECM) which would have an impact on cell behavior such as cell adhesion and gene expression. For that purpose, here we developed a fabrication process to incorporate electrospun short fibers within freeze-dried scaffolds for tissue engineering applications. Briefly, PCL short fibers were first produced from electrospun fibers with ultrasonication method. They were then evenly dispersed in gelatin solution and freeze-dried to obtain fiber incorporated scaffolds. The resulting scaffolds exhibited hierarchical structure including major pores with sizes ranging from 50 to 150 mu m and the short fibers dispersed in the thin walls of major pores, mimicking the fibrous feature of natural ECM. The short fibers were proven to modify the mechanical properties of scaffold and to facilitate cell adhesion and proliferation on the scaffold. As a promising scaffold for tissue engineering and regenerative medicine, the fiber incorporated scaffold may have further biomedical applications, in which the short fibers can act as drug release vehicles for growth factors or other biomolecules to promote vascularization.

Abstract:In the article, a bilayer nanocomposite scaffold made of oxidized alginate (OAL), gelatin (G), and silk fibroin (SF) has been prepared via combining electrospinning, in situ gas foaming, in situ crosslinking and freeze drying methods. The physicochemical and mechanical properties, as well as thermal stability of the proposed composite, have been investigated by SEM, FTIR, XRD, tensile, and TGA analysis. The data indicate that structure and degree of crosslinking play a vital role in adjusting the physical and mechanical properties of composite scaffolds. Further, the authors find a favorable adipose-derived mesenchymal stem cell's (AMSC) attachment and distribution within this novel hydrogel-electrospun composite. Such a nanocomposite structure with its promising properties and cell-material interaction may be considered as a new scaffold for different tissue engineering applications.

Abstract:In this study, three-dimensional silk fibroin/chitosan (SF/CTS) composite sponges were successfully prepared with an interconnected porous structure and proper mechanical properties. Silk fibroin and chitosan have the potential to mimic a natural extra-cellular matrix (ECM). For the sponge preparation, silk fibroin from the Bombyx mori silkworm and chitosan were used, in different proportions, as composites. Sponges were prepared by means of freezing and lyophylization of corresponding composite solutions. The properties of composite sponges, such as the microstructure as well as the chemical and physical properties were studied. In the present investigation, silk fibroin, chitosan and their mixtures were characterized by attenuated total reflection Fourier transform infra-red spectroscopy (ATR-FTIR), swelling properties, thermogravimetric analysis and scanning electron microscopy (SEM). The ATR-FTIR analysis indicated a shift of band corresponding to CO, CN, COC. Moreover, NH groups of SF and CO and NH2 groups of chitosan might have participated in a specific intermolecular interaction among themselves. Thermogravimetric results suggested a higher temperature of decomposition for SF/CTS sponges than sponges made of pure silk fibroin or chitosan. Scanning electron microscopy observations showed the sponges of SF/CTS with pore size from 20 to 150μm, which is appropriate for cell growth. Biocompatibility of sponges was confirmed by in vitro test. SF/CTS sponges have sufficient mechanical integrity to resist handling during implantation and in vivo loading. Both the compressive modulus and compressive strength for the sponge decrease with the increase of silk fibroin content in the sponge. Sponges made of silk fibroin/chitosan mixtures can be interesting materials for tissue engineering as scaffold for temporary supporting the formation of new tissue and organs.

Abstract:Biological scaffolds are plagued by poor biomechanical properties and untimely degradation. These limitations have yet to be addressed without compromising their biocompatibility. It is desirable to avoid inflammation and have degradation with concomitant host collagen deposition or even site-appropriate in situ regeneration for the successful outcome of an implanted biological scaffold. This work aims to achieve this by utilizing a biocompatible method to modify acellular scaffolds by impregnating alkaline-catalyzed citric acid (CA) cross-linking between the extracellular matrix proteins and silk fibroin (SF)/SF-gelatin (SFG) blends. Combinatorial detergent decellularization was employed to prepare a decellularized porcine liver scaffold (DPL). After proving the decellularization efficiency, the scaffold underwent modification by vacuum impregnation with CA containing SF (SF100DPL) and SFG blends (SFG5050DPL and SFG3070DPL) following pre-cross-linking, drying, and post-cross-linking. The subsequent strength augmentation was demonstrated by significant improvement in tensile strength from 2.4 ± 0.4 MPa (DPL) to, 3.8 ± 0.7 MPa (SF100DPL), 3.4 ± 0.7 MPa (SFG5050DPL), and 3.5 ± 0.2 MPa (SFG3070DPL); Young's modulus from 8.7 ± 1.8 MPa (DPL) to 20 ± 1.9 MPa (SF100DPL), 13.3 ± 2.6 MPa (SFG5050DPL), and 16 ± 1.2 MPa (SFG3070DPL); and suture retention strength from 0.9 ± 0.08 MPa (DPL) to 2.3 ± 0.2 MPa (SF100DPL), 2.8 ± 1.2 MPa (SFG5050DPL), and 2.6 ± 0.9 MPa (SFG3070DPL). The degradation resistance of the modified scaffolds was also markedly improved. Being cytocompatible, its ability to incite tolerable inflammatory and immune responses was confirmed by rat subcutaneous implantation for 14, 30, and 90 days, in terms of inflammatory cell infiltration, neoangiogenesis, and in vitro cytokine release to assess B-cell and T-cell activation. Such ECM composite scaffolds with appropriate strength and biocompatibility offer great promise in soft tissue repair applications such as skin grafting.

Abstract:IntroductionIn this study, the silk fibroin blended constructs were produced, scaffold performances of different kinds of scaffold were analyzed, and the better type for tissue engineering was optimized.MethodsThe silk fibroin/collagen (SF/C) and silk fibroin/chitosan (SF/CS) were made using a freeze-drying technique, porosity, water absorption expansion rate, mechanical properties and pore size of different scaffold was detected. Bone marrow mesenchymal stem cells (BMSCs) of 4-week-old male Wistar rats were separated by density gradient centrifugation, third generation BMSCs were seeded onto scaffolds, cultured 14 days, proliferation and metabolize of cells were detected in different time using the thiazolyl blue tetrazolium bromide (MTT) assay method, and cell morphology and distribution were observed by histological analysis and scanning electron microscopy (SEM).ResultsPorosity, water absorption expansion rate and Young’s modulus of SF/C were significantly higher than SF/CS (p < 0.05); pore size of SF/C and SF/CS was 103 ± 12 and 76 ± 11 μm and had no significant differences between two types (p > 0.05); MTT results showed that the metabolism of cells in the SF/C was better than SF/CS; after cultivation for 14 days, in the inner zone of scaffolds, cells staining were little or absent from SF/CS, lots of cells staining were existing in SF/C; pore size was consistent, holes communicated with each other better, stem cells grew well inside the scaffolds, extended fully and secreted much extracellular matrix under SEM in SF/C scaffold; internal structure of SF/CS was disorder, holes size were not consistent, and did not communicated with each other and cells were partly dead.ConclusionCompared with SF/CS, SF/C scaffold showed better porosity, water absorption expansion rate, elasticity modulus and pore size, cells grow well inside the scaffolds, and was more suitable for tissue engineering.

Abstract:Silk fibroin, extracted from the silk of the Bombyx mori silkworm, stands out as a biomaterial due to its nontoxic nature, excellent biocompatibility, and adjustable biodegradability. Porous scaffolds, a type of biomaterial, are crucial for creating an optimal microenvironment that supports cell adhesion and proliferation, thereby playing an essential role in tissue remodeling and repair. Therefore, this review focuses on 3D porous silk fibroin-based scaffolds, first summarizing their preparation methods and then detailing their regenerative effects on bone, cartilage, tendon, vascular, neural, skin, hepatic, and tracheal epithelial tissue engineering in recent years.
