Abstract:Novel cross-linked copolymers of PEG-IM-PhNH2 are successfully synthesized through PEGylation via radical polymerization of 2-isocyanatoethyl methacrylate and poly(ethylene glycol) monomethyl ether methacylate and subsequent cross-linking with an amino-terminated aggregation-induced emission fluorogen. Such obtained amphiphilic copolymers can self-assemble to form uniform fluorescent polymeric nanoparticles (FPNs) and be utilized for cell imaging. These cross-linked FPNs are demonstrated good water dispersibility with ultralow critical micelle concentration (∼ 0.002 mg mL(-1)), uniform morphology (98 ± 2 nm), high red fluorescence quantum yield, and excellent biocompatibility. More importantly, this novel strategy of fabricating cross-linked FPNs paves the way to the future development of more robust and biocompatible fluorescent bioprobes.

Abstract:Facile one-pot preparation of cross-linked amphiphilic fluorescent polymer based on aggregation induced emission (AIE) dyes and 2-isocyanatoethyl methacrylate (IM) has been developed. This was carried out first by free radical polymerization between AIE monomer (PhE) and IM, and then polyethyleneimine (PEI) was introduced to obtain the cross-linked fluorescent polymer. The resulted cross-linked amphiphilic polymer was prone to self-assemble into stable nanoparticles in aqueous solution with surplus amino groups on the surface which made them highly water dispersible and can be further functionalized. The as-prepared fluorescent polymer nanoparticles (PhE-IM-PEI FPNs) were fully characterized by a series of techniques including (1)H NMR spectrum, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, transmission electron microscopy, dynamic light scattering, UV-vis absorption spectrum, and fluorescence spectra. Such FPNs demonstrated intense orange fluorescence with a high quantum yield of about 40%. Biocompatibility evaluation and cell uptake behavior of the nanoparticles were further investigated to explore their potential biomedical applications; the demonstrated excellent biocompatibility made them promising for cell imaging.

Abstract:Fluorescent probes have long been regarded as tools for imaging living organisms with advantages such as high sensitivity, good designability and multifunctional potential. Many fluorescent probes, especially the probes based on aggregation-induced emission (AIE) dyes, have received increasing attention since the AIE phenomenon was discovered. These AIE dye-based fluorescent probes could elegantly overcome the notorious quenching effect caused by aggregation of conventional organic dyes. However, it is still difficult to directly apply these AIE-active dyes for biomedical applications owing to their hydrophobic nature. Therefore, the development of novel and facile strategies to endow them with water dispersibility is of critical importance. In this work, we exploit an efficient and simple strategy to fabricate an AIE dye-based fluorescent copolymer through the combination of reversible addition-fragmentation chain transfer and the Biginelli reaction. Moreover, the copolymer can self-assemble to fluorescent polymeric nanoparticles (FPNs) in water solution. Hydrophilic poly(PEGMA-co-AEMA) was reacted with the AIE-active dye 4',4‴-(1,2-diphenylethene-1,2-diyl)bis([1,1'-biphenyl]-4-carbaldehyde (CHO-TPE-CHO) to form amphiphilic luminescent polymers using urea as the connection bridge. The successful synthesis of the final products (poly(PEGMA-co-AEMA-TPE) FPNs) was confirmed by various instruments. Furthermore, Transmission electron microscopy (TEM) images manifest that poly(PEGMA-co-AEMA-TPE) copolymers will self-assemble into spherical nanoparticles in aqueous environments with sizes between 100 nm and 200 nm. The cell uptake and bioimaging experiment confirm that poly(PEGMA-co-AEMA-TPE) FPNs have excellent biocompatibility and emit strong green fluorescence in a cellular environment. Thus, poly(PEGMA-co-AEMA-TPE) FPNs are excellent candidates for biomedical applications.

Abstract:Aggregation-induced emission (AIE) dyes based fluorescent polymeric nanoparticles (FNPs) have been intensively explored for biomedical applications. However, many of these AIE-active FNPs are relied on the self-assembly of amphiphilic copolymers, which are not stable in diluted solution. Therefore, the introduction of cross-linkages into these micelles has demonstrated to be an efficient route to overcome this stability problem and endow ultra-low critical micelle concentrations (CMC) of these AIE-active FNPs. In this work, we reported the fabrication of cross-linked AIE-active FNPs through controllable reversible addition fragmentation chain transfer polymerization by using commercially available octavinyl-T8-silsesquioxane (8-vinyl POSS) as the cross-linkage for the first time. The resultant cross-linked amphiphilic copolymers (named as PEG-POSS-PhE) are prone to self-assemble into stable core-shell nanoparticles with well water dispersity, strong red fluorescence and low CMC (0.0069 mg mL(-1)) in aqueous solution. More importantly, PEG-POSS-PhE FNPs possess some other properties such as high water dispersity, uniform morphology and small size, excellent biocompatibility and cellular internalization, providing great potential of PEG-POSS-PhE FNPs for biological imaging application. Abstract:Application of organic dyes is limited in biomedical fields due to the rapid self-quenching, poor stability and water solubility. In this work, polymer modified organic dyes is developed through the dyes initiating polymerization. The polymer length is studied by tuning the amount of monomer. With an optimal molecule weight of the polymer, the pi-pi stacking from pi-conjugated organic fluorescent dyes and fluorescence quenching are inhibited, resulting in an enhancement of fluorescence intensity and photostability. Nanoparticles are further fabricated to be used for cell uptake based on the optimized organic dyes. Then, long term cellular fluorescence imaging is realized. This work highlights the potential of polymer modification to improve the performance of organic dyes and expand their applications.

Abstract:A water-soluble polymeric fluorescent nanoparticle (PTCN) has been proposed for two-photon fluorescence (TPF) imaging of living cells. PTCN was firstly prepared by chemical crosslinking of polyethyleneimine using a dual carboxyl 1, 8-naphthalimide derivative as the crosslinker. Subsequently, the investigations on its structure, morphology, one- and two-photon fluorescence properties and cytotoxicity were carried out. It was found that PTCN was spherical with the diameters of 5-10 nm. It could emit fluorescence at 536 nm with the excitation wavelengths of 443 nm and 800 nm, respectively. PTCN possessed good pH stability and photostability. Its fluorescence did not change in the pH range of 4.0-9.0. In pH 7.4 buffer solution, when it was detected at lambda(ex)/lambda(em) = 443 nm/536 nm for 12000 times, the decrease in fluorescence intensity was less than 1%. Cytotoxic assays indicated it was low toxic upon interactions with Hela cells for 24 h at a concentration of 15.0 mg/L. Finally, the application of PTCN for imaging live cells was assessed by TPF microscopy technique. After Hela cells were incubated with PTCN for 2 h, the green fluorescence was observed in the cellsap at the excitation wavelength of 800 nm. The proposed nanoparticle is potential in the development of targeting TPF probes.

Abstract:Luminescent polymeric nanoparticles (LPNs) with aggregation-induced emission (AIE) feature have emerged as the most promising candidates for biological imaging owing to their unique AIE feature, great water dispersity, strong fluorescence, low cytotoxicity and biocompatibility. Although numerous successful strategies for construction of AIE-active LPNs have been developed, the preparation of dynamic linkages containing AIE-active LPNs based on multicomponent reactions has been rarely reported. In this work, we report a facile method for the formation of AIE-active LPNs via a one-pot conjugation of PEG-B(OH)2, 1-thioglycerol and AIE-active dye PhE-alc in short time under rather mild reaction conditions (e.g. ambient temperature, air atmosphere, absent of metal catalysts and in the present of water). The successful formation of AIE-active mPEG-PhE LPNs was confirmed by different characterization techniques in details. The great optical and biological properties certified their applicable for biological imaging application. More importantly, the novel method for the formation of AIE-active LPNs is rather simple, high efficiency and atom economy, which greatly enriched their practical biomedical applications.
An Amphiphilic Fluorescent Polymer Combining Aggregation-Induced Emission Monomer and Ε-Polylysine for Cell Imaging

Abstract:An amphiphilic fluorescent polymer (Ply-EP3) combining an aggregation-induced emission monomer and e-polylysine was prepared through epoxy ring-opening polymerization. H-1 NMR, FT-IR, and X-ray photoelectron spectra were conducted to characterize its chemical property. Such amphiphilic Ply-EP3 could assemble into nanoparticles in water and form fluorescent polymeric nanoparticles (FPNs) with high water dispersibility. Dynamic light scattering result of PIy-EP3 FPNs demonstrated the size distribution of 106 +/- 8 nm with the polydispersity index of 0.268. Transmission electron microscopy image of Ply-EP3 FPNs showed spherical nanoparticle morphology with a size range of 50-100 nm. The optical property of FPNs indicated intense green fluorescence with a fluorescence quantum yield-of 33%. The PlyEP3 FPNs further proved excellent stability with low critical micelle concentration of 0.042 mg mL(-1). The prepared FPNs also revealed high photostability after UV light irradiation for 120 min. Such FPNs showed excellent biocompatibility as the cells retained high viability at the concentration range of 10 -120 mu g mL(-1) of Ply-EP3 for an incubated period of 12 h or 24 h, and exhibited excellent performance for cell imaging. Thanks to the high-performance Ply-EP3 FPNs, including bright fluorescent emission, high water dispersibility and particle stability, and excellent photostability, this work provide an approach to prepare robust cross-linked biocompatible fluorescent polymers for cell imaging. Abstract:Fluorescent nanoparticles used to detect important biological events in living cells or animals are in increasing demand in the biological and biomedical fields, and have attracted much attention from chemists and biologists in the past decade. Here, one aggregation-induced emission (AIE) bioconjugate, tetraphenylethene labelled chitosan (TPE-CS), is synthesized, which could be strongly emissive in the solid state. TPE-CS is used as a coating agent for negatively charged hydroxyapatite (HA) nanoparticles, and TPE-CS/HA nanocomposites with positive charges are well dispersed in their aqueous solution with a diameter of 111.9 nm. MTT assay indicates that the fluorescent TPE-CS/HA nanoparticles have good cytocompatibility. 293T cells are imaged by TPE-CS/HA nanoparticles. First, the nanoparticles are adhered to the cell membrane, and then many more particles are endocytosed through phagocytotic vesicles by culturing for a long time, resulting in a much stronger fluorescence emission. TPE-CS/HA bioprobes could strongly bind the cell cytoplasmic region, and might have promising applications in tumor diagnosis, long-term cell tracing, drug metabolism and drug delivery systems.

Abstract:A new type of Cy5-encapsulated photostable fluorescent silica nanoparticles (FSNPs) bearing positive charges have been successfully fabricated by a reverse microemulsion synthesis in one-pot. The Cy5 dye containing four primary amines are embedded into silica via covalent bonds through a silane coupling agent (GPTMS), followed by co-condensation with tetraethylorthosilicate. The uniform-sized, spherical and monodispersed FSNPs have high fluorescence intensity and photostability. The FSNPs exhibit high stability, good biocompatibility as well as low cytotoxicity. These FSNPs can be internalized into live cells and thus fluorescently label the cells. This study provides a simple synthesis approach that can be applied to other water-soluble and amino-modified organic dye molecules for biological targeting and fluorescent cell imaging.

Abstract:Glycosylated cross-linked red fluorescent amphiphilic polymer has been facilely synthesized in one pot, which would readily self-assemble into nanoparticles with high water dispersibility in aqueous media. The resulted nanoparticles have surplus carboxyl groups and glycosyl groups on the surface, which can be further functionalized, and they can demonstrate strong bio-favorable red fluorescence with a fluorescence quantum yield of 18% due to the aggregation induced emission (AIE) dyes aggregated in the core. Moreover, the nanoparticles also revealed great photostability and structure stability with an ultralow critical micelle concentration of 0.0018 mg mL(-1). Abstract:A novel water-soluble fluorescent amphiphile based on amino polyethylene glycol (PEG-NH2) substituted oligo (p- phenyleneethynylene) (OPE) was designed and synthesized successfully. Taking anion OPE amphiphile as a comparison, the photophysical features were investigated through ultraviolet absorption (UV) and photoluminescence (PL) analyses. Due to the hydrophobic-hydrophilic property of the OPE conjugated molecule, self-assembled nanoparticles with the size ranging from 19.6 to 93.5 nm along with the change of morphology from "grain" to "strawberry" were conveniently prepared via adjusting concentrations of OPE aqueous solution. Interestingly, after aging for a period of time, homogeneous hollow nanospheres were spontaneously constructed with a diameter of about 200 nm. Cytotoxicity test and cellular uptake behavior of the nanoparticles were further investigated to evaluate their potential biomedical applications. Subsequently, the promising applications of two-photon cell imaging were explored using human pancreatic cancer cells (PANC-1 cells), which indicated that the nanoparticles were mainly located within the cell cytoplasm.

Abstract:Fluorescent carbon nanoparticles (FCNs) have gradually become the most promising alternative candidates to other traditional fluorescent nanomaterials for biological applications on account of their excellent fluorescence property and remarkable biocompatibility. Although many methods have reported on the preparation of FCNs, to date, no studies have reported the preparation of polymers of functionalized FCNs. A high-efficiency method was developed in this work to synthesize high-quality poly(ethylene oxide) (PEG)-functionalized FCNs from cigarette ash and thiol group-containing PEG via a facile one-pot ultrasonic irradiation treatment. A series of characterization techniques demonstrated the uniform nanoscale size, good fluorescence stability, high water dispersibility and remarkable biocompatibility of the generated FCNs. Furthermore, cell imaging was easily achieved at high resolution using the synthetic FCNs as probes, which validates their potential for bioimaging applications. In summary, an efficient one-pot strategy is reported for the first time on the preparation of PEG-functionalized FCNs with the assistance of ultrasonic irradiation. This method should be of great research interest for the fabrication of other polymer-functionalized FCNs with designable properties and functions.

Abstract:Novel amphiphilic organic asymmetrically oligo(p-phenyleneethynylene) (OPE) conjugated molecules, ending with a hydrophobic alkyl chain and a hydrophilic methoxypolyethyleneglycol (MPEG) segment, were designed and synthesized. Due to the hydrophilic-hydrophobic nature of the asymmetrically amphiphilic OPE conjugated molecules, monodispersed fluorescent nanoparticles were obtained easily by self-assembly in aqueous solution. Through adjusting the length of MPEG chains in amphiphilic OPE, a series of nanoparticles with different water solubility were prepared. The aggregation behaviors of these dyes were investigated by dynamic light scattering (DLS) and transmission electron microscopy (TEM), which indicated that when the MPEG chains bonding with amphiphilic OPE are longer, smaller sized nanoparticles will be formed at the same concentration of 10(-5) mol L-1. Besides, taking OPE-PEG(1900) as an example, diversified aggregation behaviors under different concentrations can be inferred through DLS and TEM, which were also supported by ultraviolet absorption (UV) and photoluminescence (PL) analyses. Compared with previously reported conjugated-polymer nanoparticles consisting of amphiphiles without photoelectric features and oil-soluble conjugated-polymers through non-covalent encapsulation, the introduction of OPE chromophores into amphiphiles by covalent linkage efficiently enhanced the structural stability of nanoparticles. A photobleaching test under UV excitation revealed the high photostability of these OPE nanoparticles. In view of the good water solubility, biocompatibility, structural stability, photostability, and excellent photoelectric features of these nanoparticles, cellular imaging of human pancreatic cancer cells (PANC-1 cells) was conducted. Confocal microscopy results showed that the nanoparticles (OPE-PEG(1000) and OPE-PEG(1900)) were located specifically within the cell cytoplasm. We believe that the OPE nanoparticles would play an important rote as fluorescent biomarkers for long-term bioimaging, and offer new opportunities for good applicability in cell imaging and sensing in biomedical science.

Abstract:A polyvinylpyrrolidone (PVP)-based fluorescent film with stable optical properties is successfully prepared in one pot without any additive. The reaction mechanism of ring-opening and self-crosslinking of linear PVP is proposed and demonstrated. The morphologies and the nanostructures of the fluorescent film as well as the unmodified film are investigated. The dye is incorporated into the film networks via covalent linkages, thus leading to the highly stable optical properties. The facile and effective synthesis approach opens a new way for the design of other multi-functional composite materials based on linear PVP.

Abstract:Water dispersion aggregation-induced emission (AIE) dyes based nanomaterials have recently attracted increasing attention in the biomedical fields because of their unique optical properties, outstanding performance as imaging and therapeutic agents. The methods to conjugate hydrophilic polymers with AIE dyes to solve the hydrophobic nature of AIE dyes and makeS them widely used in biomedicine, which have been extensively explored and paid great effort previously. Although great advance has been made in the fabrication and biomedical applications of AIE-active polymeric nanoprobes, facile and efficient strategies for fabrication of biodegradable AIE-active nanoprobes are still high desirable. In this work, amphiphilic biodegradable fluorescent organic nanoparticles (PLL-TPE-O-E FONs) have been fabricated for the first time by conjugation of AIE dye tetraphenylethene acrylate (TPE-O-E) with Poly-l-Lysine (PLL) through a facile one-step Michael addition reaction, which was carried out under rather mild conditions, included air atmosphere, near room temperature and absent of metal catalysts or hazardous reagents. Due to the unique AIE properties, these amphiphilic copolymers tend to self-assemble into high luminescent water dispersible nanoparticles with size range from 400 to 600nm. Laser scanning microscope and cytotoxicity results revealed that PLL-TPE-O-E FONs can be internalized into cytoplasm with negative cytotoxicity, which implied that PLL-TPE-O-E FONs are promising for biological applications.
