Abstract:Objective To investigate the utility of the quantitative detection of PCA3 mRNA and T1E4 mRNA(the isoform of TMPRSS2:ERG fusion gene) in urine after prostate massage for the diagnosis of prostate cancer.Methods T1E4 mRNA,PCA3 mRNA and PSA mRNA were measured by real-time fluorescent quantitative reverse transcription polymerase chain reaction(FQ-RT-PCR) based on taqman technique in urine samples of 53 PCa and 37 BPH patients after prostate massage.PSA mRNA was used to normalize the number of prostate cells and T1E4 mRNA/PSA mRNA ratio was then used as a diagnostic marker for prostate cancer.The diagnostic efficacies of T1E4 mRNA and PCA3 mRNA in urine samples were evaluated by the ROC curve.Then the diagnostic efficacies of detection of PCA3 mRNA and T1E4 mRNA were analyzed.Results AUC of T1E4 mRNA/PSA mRNA ratio was 0.802(95%CI:0.709～0.895).The diagnostic sensitivity and specificity of T1E4 mRNA/PSA mRNA ratio were 56.6% and 94.6% at a cut off of 0.007.AUC of PCA3 mRNA/PSA mRNA ratio was 0.646(95%CI:0.533～0.758).The diagnostic sensitivity and specificity of T1E4 mRNA/PSA mRNA ratio were 47.2% and 73.0%at a cut off of 0.001.The T1E4 mRNA expression in PCa group was significantly higher than that in the BPH group(P=0.000).The PCA3 mRNA expression in PCa group was significantly higher than that in the BPH group(P=0.009).The combination of both markers remarkably increased the sensitivity from 47.2%(PCA3 test alone) and 56.6%(T1E4 test alone) to 81.1%.The diagnostic efficacies increased by 33.9% and 24.5%,respectively.Conclusion T1E4 mRNA can be detected in the urine of the prostate cancer patients.The two makers can improve the diagnosis prostates cancer and may potentially be used in the early diagnosis of prostate cancer.

Abstract:OBJECTIVE:To analyze the expression of DD3 mRNA in the prostate tissues.METHODS:DD3 mRNA was detected by realtime fluorescent quantitative reverse transcription polymerase chain reaction (FQ-RT-PCR) based on the Taqman technique in the tissues of 27 patients with non-prostate cancer( NPCa), 21 prostate cancer( PCa), 39 benign prostatic hyperplasia (BPH) and 15 normal prostate (NP). The ROC curve was used to evaluate the diagnostic value of DD3 mRNA.RESULTS:DD3 mRNA expression was not detected in the NPCa tissues. The median expressions of DD3 mRNA in PCa, BPH and NP tissues were 7. 2 x 10(6), 2. 5 x 10(4) and 1.5 x 10(4) copies/mg tissue, respectively. The DD3 mRNA expression levels were significantly different between nonmalignant and malignant tissues (P < 0.01). No significant differences in DD3 mRNA expression were detected between the NP and BPH tissues and no significant correlation was found between the DD3 mRNA expression and clinical pathological parameters. The AUC-ROC was 0.937 (95% CI: 0.879 - 0.995) at cutoff value 1.4 x 10(5) copies/mg tissue. The sensitivity, specificity, accuracy, positive predictive value, negative predictive value, positive likelihood ratio and negative likelihood ratio for DD3 were 90.5%, 85.0%, 86.7%, 76.0%, 94.3%, 6.03 and 0.11 respectively.CONCLUSION:The DD3 mRNA expression is confined to prostate tissues and highly upregulated in PCa tissues. It has a potential application value in the early diagnosis of prostate cancer and the follow-up of the patient.

Abstract:Objective To detect the expression of peripheral blood free prostate specific antigen (fSPA),total prostate specific antigen (tSPA),prostate specific antigen density (PSAD) and prostate cancer gene 3 (PCA3) in prostate disease,and the significance of combined detection of fSPA,tSPA and PCA3.Methods 67 patients with prostate cancer,75 patients with prostatic hyperplasia and 70 healthy male were selected as the research objects from Dec.2014 to Jul.2016.The serum level of fSPA and tSPA was detected by chemiluminescence immune staining method.The prostate volume was tested by ultrasonic sound and PSAD value was calculated.The total RNA was extracted by Trizol,and the serum PCA3 mRNA expression was detected by RT-PCR.The specificity and sensitivity of combined detection of fSPA,tSPA and PCA3 were analyzed.Results The serum levels of fSPA,tSPA,PSAD and PCA3 in prostate cancer were significantly higher than those in patients with prostatic hyperplasia and healthy male,and they were higher in patients with hyperplasia of prostate than in healthy male,and the differences had statistical significance (P＜0.01).The serum levels of fSPA,tSPA,PSAD and PCA3 were higher in patients with Gleason score ≥7 points and in T3-T4 stage than in patients with Gleason score ＜7 and in T1-T2 stage,and the difference had statistical significance (P＜0.01).The serum levels of fSPA,tSPA,PSAD and PCA3 were positively correlated with Gleason score and TMN pathological stage,and the difference had statistical significance (P＜0.01).The AUC value of fSPA,tSPA,PSAD and PCA3 in diagnosis of prostate cancer was 0.53,0.57,0.63 and 0.75,and the AUC value of combined detection was 0.92.The combined detection efficiency was higher than the single index.The specificity of fSPA,tSPA,PSAD and PCA3 was 67.16％,68.66％,73.13％ and 85.07％,and the sensitivity was 71.64％,70.15％,74.63％ and 82.09％ respectively.The specificity of combined detection was 97.01％,the sensitivity was 92.54％,and the difference had statistical significance (P＜0.01).Conclusion The serum level of fSPA,tSPA,PSAD and PCA3 is increased in prostate disease,and is negatively correlated with Gleason score and TMN pathological stage.The combined detection of fSPA,tSPA,PSAD and PCA3 can improve the sensitivity and specificity of prostate disease diagnosis,and is of high clinical value.
Expression of Trefoil Factor in Bladder Cancer and Its Clinical Significance

Abstract:Objective To explore the expressions of trefoil factors (TFFs) in bladder cancers and their clinical significance.Methods Using real-time fluorescence quantitative PCR and immunohistochemical staining from January 2013 to December 2013 in our hospital underwent surgical resection in 50 cases of superficial bladder cancer (including 20 cases of primary and 30 cases of recurrent bladder cancer) expression of TFF1,TFF2 and TFF3 and 10 cases of adjacent normal tissues.There were 32 males and 18 females,aged 45-73 years,with an average of 57 years.Pathological T staging:25 cases (50％) in pTa,25 cases in pT1 phase (50％),pathological grade:low grade 24 cases (48％),26 high grade (52％).Results TFF1,TFF2,TFF3 and mRNA in primary bladder cancer tissue expression levels were 2.04 (1.86-2.22) 、0.88 (0.11-1.6) 、0.92 (0.11-1.73) the recurrence of bladder cancer tissue expression levels were 2.22 (1.94-2.22) 、2.32 (0.22-4.42) 、2.45 (0.35-4.55) while in the adjacent normal tissues were 0.60(0.52-0.68) 、0.58(0.18-0.98) 、0.56(0.16-0.96).The adjacent normal tissues of TFF1,TFF2,TFF3 mRNA was lower than that of bladder cancer (P ＜ 0.05) and the recurrent bladder cancer,the relative expression of TFF2,TFF3 of mRNA was higher than that of primary bladder cancer (P ＜ 0.05),while the recurrent bladder cancer and primary bladder carcinoma TFF1 mRNA expression showed no significant difference (P ＞ 0.05).There was no significant difference in the positive expression rate of TFF1 between high-grade and low-grade bladder cancer [71％ (17/24) vs.77％ (20/26),P =0.88].And the positive rates of TFF2 and TFF3 in high-grade bladder cancer were 83％ (20/24) and 83 ％ (20/24) respectively,which were higher than those in low-grade bladder cancer [65 ％ (17/26),77％ (20/26),all P ＜0.05].The results of follow-up showed that the median recurrence-free survival (RFS) for 50 patients was 31 (18-38) months.The median RFS of patients with strong positive TFF1 and with weak positive (25 months and 27 months) were shorter than TFF1 negative (31 months,all P ＜0.05).The median RFS of patients with strong positive TFF2 (24 months) and TFF2 negative (30 months) were shorter than those of the weakly positive patients (median 33 months,all P ＜ 0.05).The median RFS of patients with TFF3 strongly positive (24 months) and weakly positive (28 months) were shorter than that of TFF3 negative patients (31 months,all P ＜ 0.05),and TFF3 strong positive RFS was shorter than TFF3 weak positive (P ＜ 0.05).Conclusions The expression rates of TFF1,TFF2 and TFF3 in bladder cancer tissues are higher than that of normal tissue,and they are higher than that of the primary bladder cancer tissue in the recurrent bladder cancer.TFF1 and TFF2 may be involved in the recurrence and progression of bladder cancer,and the role of TFF3 needs to be further verified.
Expressions of EGFR and TGF-α in Prostatic Adenocarcinoma and Prostatic Intraepithelial Neoplasia Tissues and Their Clinical Significances

Abstract:Objective To study the expressions of epidermal growth factor(EGFR) and transforming growth factor-α(TGF-α) in human prostatic adenocarcinoma(PAC),high grade prostate intraepithelial neoplasia(HGPIN) and benign prostatic hyperplasia(BPH) tissues,and to explain the effect of the expressions in the development of PAC.Methods 41 specimens of PAC and specimens of BPH and 32 specimens of HGPIN were selected,the expressions of EGFR and TGF-α were determined by immuohistochemisitry staining.The relationship between the expressions of EGFR and TGF-α and histopathologic classification was analyzed.Results ①The EGFR positive rates were 56.6%,53.7% and 85.4% in PAC,HGPIN and BPH,the differences between BPH and HGPIN,BPH and PAC were statistically significant(P0.01).There was no significant difference in EGFR positive rate between PAC and HGPIN. The EGFR positive rates were 53.9%,55.6% and 52.6% in well-differentiated,moderately differentiated and poorly differentiated PAC,the expressions of EGFR had no correlation with different Gleason grades in PAC(P0.05).②The TGFA positive rates in PAC and BPH were 56.1% and 85.4%,there was significant difference between them(P0.01).The TGF-α positive rate were 53.8%,66.7% and 52.6% in well differentiated,moderately differentiated and poorly differentiated PAC,the expressions of TGF-α had no correlation with different Gleason grades in PAC (P0.01).The TGF-α positive rate in HGPIN was 56.3%,there was no significant difference compared with PAC(P0.05),but there was significant difference compared with BPH(P0.01).The collective positive rate of EGFR and TGF-α was 81.8%,the respective positive rate of EGFR was 18.2% and the respective positive rate of TGF-α was 26.3% in PAC,they had positive correlation(r=0.558,P0.001) and they had no correlation in HGPIN(r=0.111,P=5.450) and BPH(r=0.024,P=0.883).Conclusion The EGFR and TGF-α expression levels in PAC,HGPIN and BPH are different,combined detection of them can be useful for the diagnosis,differential diagnosis and evaluation in benign and malignant prostate disease.The expressions of EGFR and TGF-α play centain roles in the pathogenesis of prostatic adenocarcinoma.
Expression and significance of follicle-stimulating hormone receptor in prostate cancer

Abstract:OBJECTIVE: To explore the significance of follicle-stimulating hormone receptor in prostate cancer throngh the research of the expression of FSHR in normal human prostate, benign prostatic hyperplasia and prostate cancer. METHODS: FSHR expressions were observed in 12 cases of normal specimens, 14 cases of BPH and 45 cases of PCa tissues by immunohistochemical staining. RESULTS: There were 2 positive in normal prostate, 4 positive in benign prostatic hyperplasia and 38 positive in prostate cancer. There was no significant difference between normal prostate and benign prostatic hyperplasia. But there was a significant difference between benign prostatic hyperplasia and prostate cancer. The postive rate of FSHR corresponded with the increase of tumor's pathological grading and its clinical staging, and furthermore with deeper color in cytoplasm. CONCLUSIONS: The expression of FSHR is higher in prostate cancer than that in normal human prostate and in benign prostatic hyperplasia, and the expression in prostate cancer presents a positive correlation with tumor's, sgrading and staging. The expression presents a close relation with the carcinogenesis, progression and metastasis of prostate cancer.

Abstract:Objective To investigate the differential expression and significance ofα-1,6 fuco-cyltransferase (FUT8)in prostate cancer cell lines with different biological characteristics and tis-sues. Methods The normal epithelial cells of prostate (RWPE-1),four types of prostate cancer cells (22RV1,LNCap,PC-3,DU145)and the other two types of urological cancer cells (T-24,786-O) were cultured.Fifteen pairs of prostatic cancerous and paracancerous tissues were collected.The ex-pression levels of FUT8 mRNA in the different cell lines and tissues were detected and evaluated by reverse transcription-polymerase chain reaction (RT-PCR). Results The expression levels of FUT8 mRNA in prostate cancer cell lines were significantly higher than that in normal epithelial cell line of prostate (RWPE-1),bladder cancer cell line (T-24)and renal cell carcinoma cell line (786-O).FUT8 mRNA was detected with higher levels of expression in PC-3 and DU145 cell lines which were both known as AI-PCa cell lines than that in LNCap cell line which was known as AD-PCa cell line.The expression levels of FUT8 mRNA in PC-3 and DU-145 cell lines which were both from me-tastases were apparently higher than that in 22RV1 cell line which was from primary tumor.It was also found that FUT8 mRNA was up-regulated in prostatic cancerous tissues than in paracancerous tissues.The differences were all significant(P<0.01). Conclusions There was remarkable differ-ence in the expression of FUT8 in prostate cancer cell lines with different biological characteristics and tissues,indicating that FUT8 may be involved in the conversion process from AD-PCa to AI-PCa and influence the progression and metastasis of prostate cancer.
Expressions of P53 and TGF-β in Prostatic Carcinoma Tissue and Their Significances

Abstract:Objective To study the expressions of TGF-β and P53 protein in prostatic carcinoma tissue and their clinical significances.Methods Immunohistochemical method was used to detect the expressions of TGF-β and P53 protein in 36 cases of prostatic carcinoma.Results 36 cases of prostatic carcinoma included 10 cases of well differentiation,16 cases of moderate differentiation,10 cases of poor differentiation.The positive expression rates of TGF-β in well,moderately,poorly differented prostatic carcinoma tissues were 30%(3/10),37.5%(6/16),and 70%(7/10);the positive expression rates of P53 protein in well,moderately,poorly differented prostatic carcinoma tissues were 20%(2/10),25%(4/16),and 50%(5/10).The expression rates of TGF-β and P53 protein in well and moderately differented prostatic carcinoma tissues were lower than those in poorly differented prostatic carcinoma tissues(P0.05).Conclusion The expressions of TGF-β and P53 have a negative correlation with the differented degree of prostatic carcinoma and the expressions of TGF-β and P53 may help to judge the pathologic grade and predict the prognosis of prostatic carcinoma.

Abstract:Prostate cancer (PCa) is the most common malignant tumor for men. But the mechanism is unclear. EIF3C was shown to be overexpressed in PCa tissues and cell lines. EIF3C overexpression was correlated to age and tumor stage in PCa patients and indicated poor survival. The proliferation, migration, and invasiveness of PC3 cells were all inhibited after EIF3C knockdown. Additionally, the phosphorylation level of PI3K and Akt was downregulated while total NF-kappa B and Myc decreased after EIF3C knockdown. But the expression of I kappa B increased reversely. Therefore, EIF3C at least partially regulates the activity of PI3K/Akt/NF-kappa B signaling pathway in PC3 cells.

Abstract:OBJECTIVE:To investigate the amplification and expression of FGF3 in bladder transitional cell carcinoma (BTCC) and its clinical significance.METHODS:Immunohistochemistry (IHC) and Fluorescence In Situ Hybridization (FISH) methods were used to examine the protein expression and amplification of FGF3 in a tissue microarray (TMA) of 100 BTCCs and 30 adjacent normal bladder mucosas, so as to analyze their correlation and association with patient's clinico-pathological features.RESULTS:In this study, none of the normal bladder mucosas were detected FGF3 positivity, while in 89 informative BTCCs, 20 (22%) cases were observed positive expression of FGF3 protein, and it was significantly more frequently to occur in BTCCs of poor-differentiation (Grade 3), later clinical stage (T2-4) and tumor in >or= 3 cm in diameter (P < 0.05). In FISH study, 10 of the 63 (16%) informative BTCCs were observed amplification of FGF3 and it was significantly associated with BTCC's tumor size and clinical stage (P < 0.05). In addition, 10 BTCCs with amplification of FGF3 in this study were all detected positive expression of FGF3 protein, while in the remaining 53 BTCCs without amplification of FGF3, only 3 (6%) cases were observed FGF3 protein positivity.CONCLUSION:The up-regulated expression of FGF3 in BTCC was associated closely with tumor's malignant clinical phenotypes, and it might be involved in the malignant progression of parts of BTCC. The amplification of FGF3 gene might be a predominant mechanism of increased expression of FGF3 protein in BTCC.
Expression of TGF Β 1 and Β 3 in Prostate Cancer

Abstract:Objective The expression of TGF β 1 and β 3 in the tissue of prostate cancer (PCa) and its correlation with the tumor grading, staging and metastasis were studied. Methods Samples of 46 ca ses with PCa were examined using immunohistochemical staining with TGF β 1 and β 3 antisera. Results Cytoplasm and extracelluar matrix (ECM) overexpression of TGF β 1 has been noted in 39 of the 46 cases with PCa while only ECM expression was noted in the other 7 samples. Statistically, there were positive correlation of the expression in ECM and cytoplasm with Gleason score, staging and metastasis ( r =0.42/0.31, 0.81/0.64, 0.68 /0.44, respectively, P 0.05 ). As for TGF β 3,45 oot of 46 samples had cytoplasm staining and 25 had weak ECM expression.There was no statistical correlation between the expression of TGF β 3 and Gleason score, staging and metastasis. Conclusions TGF β 1 may has promotive effect on the progression of cancer by autocrine with or without paracrine, and the effect of TGF β 3 in PCa is yet to be explored.
Detection of Type Ⅳ Collagen A2 and Integrin Β3 Expression in Prostate Cancer and Their Clinical Characteristics

Abstract:Objective To investigate the expression and significance of type Ⅳ collagen A2 (COL4A2) of extracellular matrix and integrin β3 (ITGB3) in prostate cancer (PC) of tumor microenvironment.Methods The specimens were collected and immunohistochemical staining (IHC) was used to detect the expression levels of COL4A2 and ITGB3 in human prostate cancer tissues and corresponding adjacent tissues.The expression levels were analyzed and clinically diagnosed in PC patients.The correlation between the eigenlogical features was further analyzed by Pearson statistical method.Results The expression levels of COL4A2 and ITGB3 in prostate cancer tissues were significantly higher than those in adjacent tissues [immunoreactive score (IRS):COL4A2:prostate cancer tissues =4.39 ± 1.16,adjacent normal tissues =3.02 ± 1.20 (P ＜ 0.01);ITGB3:prostate cancer tissue =5.46 ± 0.99,adjacent normal tissues =3.85 ± 0.96 (P＜0.01)],both expressions were consistent with the pathological stage of PC (P ＜0.05).The Gleason scores increased(P ＜ 0.05) and the expression of both was positively correlated in prostate cancer (Pearson r =0.206,P ＜ 0.05).Conclusion COL4A2 and ITGB3 were up-regulated in prostate cancer and were associated with the staging of tumor tissues.The later the staging,the higher the expression of both,suggesting that they might play an important role in the malignant progression of PCa.Csmbined defection might help to identify Helps to judge the degree of malignancy,invasion and metastasis of PC.

Abstract:Trefoil factor 3 (TFF3) is a secreted protein which functions in mucosal repair of the gastrointestinal tract. This is achieved through the combined stimulation of cell migration and prevention of apoptosis and anoikis, thus facilitating repair. Deregulated TFF3 expression at the gene and protein level is implicated in numerous cancers. In prostate cancer TFF3 has previously been reported as a potential biomarker, overexpressed in a subset of primary and metastatic cases. Here we investigated the effect of increased TFF3 expression on prostate cancer cell behaviour. Oncomine analysis demonstrated that TFF3 mRNA expression was upregulated in prostate cancer compared to normal tissue. Forced-expression models were established in the prostate cancer cell lines, DU145 and PC3, by stable transfection of an expression vector containing the TFF3 cDNA. Forced expression of TFF3 significantly increased total cell number and cell viability, cell proliferation and cell survival. In addition, TFF3 enhanced anchorage independent growth, 3-dimensional colony formation, wound healing and cell migration compared to control transfected cell lines. We also observed reduced sensitivity to ionising radiation in stably transfected cell lines. In dose response experiments, forced expression of TFF3 significantly enhanced the regrowth of PC3 cells following ionising radiation compared with control transfected cells. In addition, TFF3 enhanced clonogenic survival of DU145 and PC3 cells. These studies indicate that targeting TFF3 for the treatment of prostate cancer warrants further investigation.

Abstract:Objective:To investigate expression levels of E2F3 and AR in prostate cancer(PCa) and discuss their relationship with the clinical stage and pathologic grade.Methods:The expressions of E2F3 and AR were detected by immunohistochemistry(IHC) in tissues from 50 cases of prostate cancer and 50 cases benign prostatic hyperplasia(BPH).The relationship between expressions of E2F3 and AR with the clinical stage and pathologic grade was analyzed by SPSS 13.0.Results:The positive rate of E2F3 was 20%(2/10) and 66%(33/50) for BPH and prostate cancer respectively.The expression level of E2F3 was higher in prostate cancer than that in BPH(P < 0.05).There was a positive correlation in expression of E2F3 between the clinical stage and pathological grade(rs = 0.471 and 0.329,P < 0.05 and P < 0.01).The expression of AR was lower in PCa than that of BPH(P < 0.05).The expression of AR in PCa was negatively correlated with the clinical stage and pathological grade(rs =-0.403 and-0.391,P < 0.01).There was no correlation between the expression of E2F3 and the expression of AR in PCa(rs =-0.017,P > 0.05).Conclusion:E2F3 may play an important role in the onset and progression of prostate cancer.

Abstract:Trefoil factors (TFF), expressed physiologically in the secretory cells of gastrointestinal mucosa and other mucosal tissues, compose a unique peptide family with important biologic functions. Aberrant expression and/or continued secretion of TFF might exist concurrently with the processes of digestive system inflammation, tumor or other related pathological changes. Previous pathological studies have shown that the abnormal expression of TFF is associated with the prognosis of digestive system diseases. The results obtained from gene expression profiling, tumor secretomics and bioinformatic analysis have positively confirmed that clinical detection of TFF peptides or mRNAs has an excellent diagnostic performance to detect digestive system diseases, especially for the monitoring of recurrence or metastasis of tumors and for the estimation of therapeutic efficacy and prognosis. Therefore, TFF is a potential biomarker for diagnosis, treatment and prognosis of digestive system diseases.
Clinical Significance of PSMA, TERT and PDEF in Malignant Tumors of the Prostate.

Abstract:OBJECTIVE: To investigate the clinical significance of the expression of PSMA (prostate specific membrane antigen), TERT (telomerase reverse transcriptase), and PDEF (prostate derived Ets factor) in malignant tumors of the prostate. PATIENTS AND METHODS: The study was conducted with paraffin slices from 33 specimens of malignant tumors of the prostate and 17 of normal tissue. We found high levels of PSMA, TERT, and PDEF protein by Western blot and immunofluorescence in the malignant tumor of the prostate. We also detected upregulation of PSMA, TERT, and PDEF mRNA in the malignant tumor of the prostate, suggesting complex regulation of these three genes in prostate cancer. RESULTS: Variance analysis showed statistically significant differences comparing the expression of PSMA, TERT, and PDEF in the malignant tumor of the prostate and normal tissues. The high expression of PSMA, TERT, and PDEF in the malignant tumor of the prostate suggests the important roles of these three factors in the occurrence and development of the malignant tumors of the prostate. CONCLUSIONS: PSMA, TERT, and PDEF may serve as a reference for clinical diagnosis and as potential targets for the malignant tumor of the prostate therapeutics.
The Expression of TFDP3 in Breast Cancer and Its Role of Breast Cancer Molecular Classification

Abstract:Objective:The object of this study is to analysis the expression of cancer testis antigen TFDP3 in breast cancer,to explore the correlation between TFDP3 and development of breast cancer.Methods:The expression of TFDP3 was detected in pathological section of tumor tissues from patients with breast cancer using immunohistochemical method.And we analyzed the data with clinical information of patients to discover the correlation of TFDP3 and the molecular classification of breast cancer.We also detected the expression of TFDP3 protein in five breast cancer cell lines,including MDA-MB-231,MCF-7,SK-BR-3,T47D and MCF-10A,using Western Blot.Then we verified the location of TFDP3 protein in cells through immumofluorescence method.Results:TFDP3 is greatly and broadly expressed in many cell lines and breast cancer tissues,such as breast,lymph node,hypothyroid,appendix and so on,while TFDP3 is 49％ percent positive in breast cancer.The expression of TFDP3 is unrelated with clinical stage,pathology type and molecular classification of breast cancer,but related with special tumor marker HER2.Conclusion:TFDP3 highly expresses in breast cancer is related with the expression of HER2.It suggests that TFDP3 be a new potential target molecule of immunotherapy for breast cancer.
Folate-Receptor Positive Circulating Tumor Cell Is a Potential Diagnostic Marker of Prostate Cancer

Abstract:Folate-receptor positive circulating tumor cells (FR+CTCs) shows an important role in the diagnosis and dynamic monitoring for many solid tumors; however, the application of FR+CTCs in prostate cancer remains unclear. We explored the potential application of FR+CTCs in this retrospective study. The levels of FR+CTCs were detected in 30 prostate cancer patients and 7 bladder cancer patients in Peking University Cancer Hospital from August 2017 to August 2021. Clinical and pathology data were collected. One-way ANOVA was used to compare the difference in FR+CTCs levels in patients with prostate cancer, bladder cancer, and benign disease. The area under the receiver operating curve (AUROC) was used to compare the accuracy of FR+CTCs and tPSA in the diagnosis of prostate cancer. We found that levels of FR+CTCs were significantly higher in cancer patients (both prostate and bladder cancer) than in patients with benign urinary disease ( p < 0.001). Besides, FR+CTCs level was consistently high in the prostate cancer patients with different tPSA levels ( p < 0.001), and it was significantly higher in the patients with f/tPSA levels <0.16 than in those patients with f/tPSA levels >0.16 (12.20 ± 1.31 vs. 8.73 ± 0.92 FU/3 ml, p = 0.043). The diagnosis efficiency of FR+CTCs is better than the tPSA in prostate cancer patients with tPSA <10 ng/ml (0.871 vs. 0.857). In the prostate cancer patients with tPSA <10 ng/ml and f/tPSA <0.16, a combination of FR+CTCs and tPSA (AUROC, 0.934) further increased the diagnosis efficiency of each of these biomarkers alone (FR+CTCs, 0.912; tPSA, 0.857). Therefore, FR+CTCs could serve as an early diagnosis marker in the prostate cancer patients with uncertain tPSA levels.

Abstract:Objective To investigate the expressions of transforming growth factor-β type I receptor (TGF-BRI )and fibroblast growth factor type I receptor (FGFR I ) in human prostatic tissue and benign prostatic hyper-plasia (BPH) .Methods The expressions of TFGBR I and FGFR I were detected by immunohistochemistry in human prostatic tissue of 31 patients with BPH and 22 normal subjects for studying the relativity between them. Results The positive expressions of TGFBR I in epithelial and interstitial tissue in BPH were significantly higher than those in normal prostatic tissue (P<0.05).The positive expression of FGFR I in epithelial tissue in BPH was much higher than that in normal prostatic tissue ( P < 0.05).however, which was not in interstitial tissue.The expression of TGFBR I was closely related with that of FGFR I in interstitial tissue in BPH ( r =0.398, P < 0.05) .Conclusion The aberrant expression of TGFBR I and FGFR I might contribute to the pathogenesis and pathological progress of BPH.
