Abstract:Decarbonylative borylation of carboxylic acids is reported. Carbon electrophiles are generated directly after reagent-enabled decarbonylation of the in situ accessible sterically-hindered acyl derivative of a carboxylic acid under catalyst controlled conditions. The scope and the potential impact of this method are demonstrated in the selective borylation of a variety of aromatics (>50 examples). This strategy was used in the late-stage derivatization of pharmaceuticals and natural products. Computations reveal the mechanistic details of the unprecedented C-O bond activation of carboxylic acids. By circumventing the challenging decarboxylation, this strategy provides a general synthetic platform to access arylpalladium species for a wide array of bond formations from abundant carboxylic acids. The study shows a powerful combination of experiment and computation to predict decarbonylation selectivity.

Abstract:Deoxygenative alkylation of benzyl alcohols was realized by using acetate as the alcohol activation group. The C-O bond homolysis is achieved by a boryl radical-promoted beta-scission process. The strategy is amenable to a variety of benzyl alcohols, including primary, secondary, and more challenging tertiary alcohols. The synthetic practicability was demonstrated by a gram-scale one-pot reaction.

Abstract:The effect of tBuOK on the existing state of benzylic boronates in the solution phase has been investigated in detail by NMR analysis and DFT calculations. It was determined that simply using an excess of tBuOK (2.0 equivalents) can result in the full deborylation of benzylic boronates to afford free benzyl potassium species. These mechanistic insights were leveraged for the facile construction of beta-silyl/boryl functionalized 1,1-diarylalkanes from aromatic alkenes via the combination of base-mediated silylboration or diborylation of aromatic alkenes and nucleophilic-type reactions with various electrophiles. Based on further machine-learning-assisted screening, the scope of electrophiles for this transformation can be generalized to the challenging aromatic heterocycles. Late-stage functionalization performed on several drug-relevant molecules generates the highly valuable 1,1-diaryl framework. By shifting the chemical equilibrium of the benzylic boronate-base complex to the free carbanion state, we developed a general, practical, and simple method for the construction of beta-silyl/boryl functionalized 1,1-diarylalkanes.

Abstract:AbstractOrganoborane compounds are among the most commonly employed intermediates in organic synthesis and serve as crucial precursors to alcohols, amines, and various functionalized molecules. A simple palladium‐based system catalyzes the conversion of primary C(sp3)H bonds in functionalized complex organic molecules into alkyl boronate esters. Amino acids, amino alcohols, alkyl amines, and a series of bioactive molecules can be functionalized with the use of readily available and removable directing groups in the presence of commercially available additives, simple ligands, and oxygen (O2) as the terminal oxidant. This approach represents an economic and environmentally friendly method that could find broad applications.

Abstract:A direct arylation to furnish diarylmethanes from benzyl alcohols was realized through Pd(PPh3)(4)-catalyzed Suzuki-Miyaura coupling via benzylic C-O activation in the absence of any additives. The arylation is compatible with various functional groups. This development provides an atom-and step-economic way to approach a diarylmethane scaffold under mild and environmentally benign conditions.

Abstract:By developing a mild Ni-catalyzed system, a method for direct borylation of sp(2) and sp(3) C-N bonds has been established. The key to this hightly efficient C-N bond borylative cleavage depends on the appropriate choice of the nickel catalyst Ni(COD)2, ICy·HCl as a ligand, and the use of 2-ethoxyethanol as the cosolvent. This transformation shows good functional group compatibility and can serve as a powerful synthetic tool for gram-scale synthesis and late-stage C-N borylation of complex compounds.

Abstract:Biaryl skeletons were directly constructed via palladium-catalyzed ortho-arylation of N,N-dimethyl benzylamine with aryl boronic acids with high efficiency and high regioselectivity under open-flask conditions. The N,N-dimethylaminomethyl group was first applied as a directing group in such an oxidative coupling. Various substrates proved to be efficient coupling partners, furnishing the corresponding ortho-monoarylated or -diarylated arenes in moderate to good yields under mild conditions.

Abstract:Recently, it was shown that Piers’ borane [HB(C6F5)2]2 can induce rapid oligomerization reactions of arylacetylenes and arylallenes. Using phenylallene PhCH=C=CH2 and phenylacetylene PhC≡CH as typical substrates, the detailed mechanisms of borane‐induced oligomerization are disclosed by extensive state‐of‐the‐art DFT calculations. Following initial regio‐selective hydroboration of unsaturated C=C or C≡C bonds, further oligomerization and structural rearrangement reactions may occur via unique boryl‐enabled conversions such as 1,3‐boryl‐shift, allylboration, 1,2‐carboboration, and 1,5‐electrocyclization. Together with stepwise transborylation with hydroboranes such as pinacolborane HBpin, such fundamental reactions may pave a catalytic way to borane‐induced oligomerization of arylacetylenes and arylallenes.

Abstract:Decarbonylative borylation of aryl anhydrides by rhodium catalysis has been reported. Base-free system with Rh(PPh3)3Cl as catalyst enables the efficient synthesis of various arylboronate esters from readily available aryl anhydrides. The reaction process involves the cleavage of C(O)–O bonds and the formation of C–B bonds. The experimental results demonstrated that compared with carboxylic acids, amides, and esters, anhydrides have higher reactivity in decarbonylative borylation reaction under the current conditions. Furthermore, compared with the reported palladium-catalyzed borylation reaction of aryl anhydride, the present rhodium-catalyzed method has the advantage of shorter reaction time or lower reaction temperature.

Abstract:Regioselective borylation of (hetero)arenes under mild conditions is an appealing strategy for constructing boron-containing compounds, particularly considering their broad applications in organic synthesis, medicinal chemistry and material science. In this manuscript, we developed a Fe(OTf) 3 -promoted, DG (directing group)-controlled, regioselective electrophilic C–H borylation of 2-arylphenolic compounds with BBr 3 at room temperature. C2′-borylation was dramatically accelerated by an LA-catalyst, affording DAOB (diaryloxaborin) with a broad substrate scope in moderate-to-excellent yields. The selective ortho -borylation was successfully achieved by introducing phenolic carbamate as a directing group to overcome the competing Fries rearrangement, giving desired 2-HAB ((2-hydroxyaryl)boronic acid) in good yields for most substrates.

Abstract:Direct application of benzyl alcohols (or their magnesium salts) as electrophiles in various reactions with Grignard reagents has been developed via transition metal-catalyzed sp(3) C-O bond activation. Ni complex was found to be an efficient catalyst for the first direct cross coupling of benzyl alcohols with aryl/alkyl Grignard reagents, while Fe, Co, or Ni catalysts could promote the unprecedented conversion of benzyl alcohols to benzyl Grignard reagents in the presence of (n)hexylMgCl. These methods offer straightforward pathways to transform benzyl alcohols into a variety of functionalities.

Abstract:A new strategy for the direct cleavage of the C(sp3)–OH bond has been developed via activation of free alcohols with neutral diphenyl boryl radical generated from sodium tetraphenylborate under mild visible light photoredox conditions. This strategy has been verified by cross-electrophile coupling of free alcohols and carbon dioxide for the synthesis of carboxylic acids. Direct transformation of a range of primary, secondary, and tertiary benzyl alcohols to acids has been achieved. Control experiments and computational studies indicate that activation of alcohols with neutral boryl radical undergoes homolysis of the C(sp3)–OH bond, generating alkyl radicals. Abstract:The preparation of arylboronates from unactivated aryl chlorides in a transition-metal-free manner is rather challenging. There are only few examples to achieve this goal by using ultraviolet irradiation. Based on the mechanistic understanding of the diboron/methoxide/pyridine reaction system, we achieved photoactivation of the in situ generated super electron donor and developed a visible-light-induced organocatalytic method for efficient borylation of unactivated aryl chlorides.

Abstract:The deprotonative functionalization of toluenes, for their weak acidity, generally needs strong bases, thus leading to the requirement of harsh conditions and the generation of by-products. Direct nucleophilic acyl substitution reaction of amides with organometallic reagents could provide an ideal solution for ketone synthesis. However, the inert amides and highly reactive organometallic reagents bring great challenges for an efficient and selective synthetic approach. Herein, we reported an lithium diisopropylamide (LDA)-promoted benzylic aroylation of toluenes with unactivated tertiary benzamides, providing a direct and efficient synthesis of various aryl benzyl ketones. This process features a kinetic deprotonative functionalization of toluenes with a readily available base LDA. Mechanism studies revealed that the directed ortho-lithiation of the tertiary benzamide with LDA promoted the benzylic kinetic deprotonation of toluene and triggered the nucleophilic acyl substitution reaction with the amide.

Abstract:Due to boron's metalloid properties, aromatic boron reagents are prevalent synthetic intermediates. The direct borylation of aryl C-H bonds for producing aromatic boron compounds offers an appealing, one-step solution. Despite significant advances in this field, achieving regioselective aryl C-H bond borylation using simple and readily available starting materials still remains a challenge. In this work, we attempted to enhance the reactivity of the electron-donor-acceptor (EDA) complex by selecting different bases to replace the organic base (NEt3) used in our previous research. To our delight, when using NH4HCO3 as the base, we have achieved a mild visible-light-mediated aromatic C-H bond borylation reaction with exceptional regioselectivity (rr > 40:1 to single isomers). Compared with our previous borylation methodologies, this protocol provides a more efficient and broader scope for aryl C-H bond borylation through the use of N-Bromosuccinimide. The protocol's good functional-group tolerance and excellent regioselectivity enable the functionalization of a variety of biologically relevant compounds and novel cascade transformations. Mechanistic experiments and theoretical calculations conducted in this study have indicated that, for certain arenes, the aryl C-H bond borylation might proceed through a new reaction mechanism, which involves the formation of a novel transient EDA complex.
