Abstract:Preparation and biomedical applications of aggregation induced emission (AIE) dye based fluorescent nanoprobes (FNPs) have been extensively investigated in recent years. And great efforts have been devoted toward fabrication of ALE dye based FNPs through both noncovalent and covalent strategies. However, to the best of our knowledge, the preparation of AIE dye based FNPs via dynamic bonds is rarely reported. In this work, ultrabright FNPs based on AIE dyes have been fabricated using aldehyde-terminated polyethylene glycol (CHO-PEG-CHO) with molecular weight of 2000 Da and 2,2'-diaminotetraphenyl ethylene (TPE-NH2) through formation of Schiff base. The resulting TPE contained copolymers are prone to self-assemble into stabilized core shell nanoparticles (TPE@PEG FNPs). The successful formation of TPE@PEG FNPs was confirmed by H-1 nuclear magnetic resonance spectroscopy, transmission electron microscopy, fluorescence spectroscopy and Fourier transform infrared spectroscopy. Due to their amphipihlic properties, TPE@PEG FNPs displayed high water dispersibility and strong fluorescence intensity. Furthermore, biological evaluation results demonstrated that TPE@PEG FNPs possess excellent biocompatibility and can be facilely internalized by cells. The extraordinary fluorescent properties, excellent water dispersibility as well as the desirable biocompatibility make TPE@PEG FNPs promising candidates for biomedical applications. On the other hand, because of the pH stimuli responsiveness of Schiff base, TPE@PEG FNPs are expected to exhibit some advantages for biomedical applications as compared with the FNPs fabricated by noncovalent and covalent strategies. Abstract:Diethylene glycol (miniPEG)-containing chiral gamma PNA is considered to be one of the best PNA derivatives. Its preparation is mainly based on the Boc strategy for solid phase peptide synthesis (SPPS), requiring the repeated use of trifluoroacetic acid TFA, which is not suitable for the in situ synthesis of PNA arrays and some other applications under mild conditions. Herein, Fmoc/Cbz orthogonal protected miniPEG-containing chiral gamma PNA monomers were synthesized, and a 15mer gamma PNA was prepared using the Fmoc strategy under mild conditions. A facile entrapment approach to construct PEGylated polyplexes for improving stability in physiological condition.

Abstract:PEGylated polyplexes had been proved to improve the stability of DNA complexes. However, the conjugation reaction might reduce the capacity of efficient DNA complexation. Herein we described an easy and favorable approach to construct PEGylated polyplexes via entrapping poly(ethylene glycol) cholesterol ether (CPEG) into polyplexes. It was of interest to find the addition sequence of CPEG had great effect on the stability of polyplexes in physiological salt concentration. The addition of CPEG into the formed PEI25k/DNA polyplexes had no effect to improve the stability. Whereas by the “CPEG first” method of adding CPEG and PEI25k mixture into the DNA solution, the PEI25k/CPEG/DNA polyplexes showed excellent anti-aggregation effect and enhanced transfection efficiency in physiological condition. The difference performance might be explained by the possibility of CPEG entrapment. By the “CPEG first” method, PEGylated polyplexes was constructed due to the hydrophobic interaction between the cholesterol group of CPEG and hydrophobic charged-compensated core. The PEG coating significantly improved the stability of polyplexes in physiological condition. This facile entrapment approach to prepare PEGylated polyplexes might have great potential in non-viral gene delivery research and application.

Abstract:Self-assembly of amphiphilic luminescent copolymers is a general route to fabricate fluorescent polymeric microparticles (FPMs). In this work, the FPMs with aggregation-induced emission (AIE) feature were fabricated via the combination of the chain transfer free radical polymerization and "one-pot" multicomponent reaction, which conjugated the aldehyde-containing AIE active dye AIE (CHO-An-CHO) and amino-terminated hydrophilic polymer (ATPPEGMA) using mercaptoacetic acid (MTA) as the "lock" molecule. The structure, chemical compositions, optical properties as well as biological properties of the PPEGMA-An-PPEGMA FPMs were characterized and investigated by means of a series of techniques and experiments in detail. We demonstrated the final copolymers showed amphiphilic properties, strong yellow fluorescence and high water dispersibility. Biological evaluation suggested that PPEGMA-An-PPEGMA FPMs possess low cytotoxicity and can be used for cell imaging. More importantly, many other AIE active FPMs are expected to be fabricated using the similar strategy because of the good substrate and monomer applicability of the multicomponent reaction and chain transfer living radical polymerization. Therefore, we could conclude that the strategy described in this work should be of great interest for fabrication of multifunctional AIE active nanoprobes for biomedical applications.

Abstract:Driven by the high demand for sensitive and specific tools for optical imaging, fluorescent nanoprobes with various working mechanisms and advanced functionalities are flourishing at an incredible speed. This work reports the design and fabrication of aggregation-induced emission (AIE)-active fluorescent organic nanoparticles (FNPs) via forming dynamic phenyl borate between diol containing hydrophobic AIE dye (APD-PhCHO) and phenylboronic acid pendant hydrophilic polymers (PEGMA-VPBA) within 30 min. The final AIE-active APD-PhCHO-PEGMA-VPBA FNPs display high water dispersibility and strong fluorescence emission because of their amphiphilic properties and AIE feature. Biological evaluation suggests that APD-PhCHO-PEGMA-VPBA FNPs possess negative effect on HeLa cells and desirable optical properties for biological imaging. More importantly, phenyl borate is a dynamic bond with pH and glucose responsiveness. Furthermore, different functions can be designed and introduced into these AIE-active systems through adoption of different monomers for good applicability of free radical polymerization. Therefore, this work provides a novel platform for preparation of multifunctional AIE-active nanosystems with responsiveness for various biomedical applications.

Abstract:Fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) property have received increasing attention and possess promising biomedical application potential in the recent years. Many efforts have been devoted to the fabrication methodologies of FPNs and significant advance has been achieved. In this contribution, a novel strategy for the fabrication of AIE-active amphiphilic copolymers is reported for the first time based on the Ce(IV) redox polymerization. As an example, ene group containing AIE-active dye (named as Phe-alc) is directly grafted onto a water soluble polymer polyethylene glycol (PEG) in H2 O/THF system under low temperature. Thus-obtained amphiphilic fluorescent polymers will self-assemble into FPNs with ultra-low critical micelle concentration, ultra-brightness, and great water dispersibility. Biological evaluation results suggest that the PEG-poly(Phe-alc) possess excellent biocompatibility and can be used for tracing their behavior in cells using confocal laser scanning microscope. These features make PEG-poly(Phe-alc) FPNs promising candidates for many biomedical applications, such as cell imaging, drug delivery vehicles, and targeted tracing. More importantly, many other functional groups can also be incorporated into these AIE-active FPNs through the redox polymerization. Therefore, the redox polymerization should be a facile and effective strategy for fabrication of AIE-active FPNs.

Abstract:Fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) characteristics have attracted much attention for biomedical applications due to their remarkable AIE feature, high water dispersity and desirable biocompatibility. The development of facile and effective strategies for fabrication of these AIE-active FPNs therefore should be of great importance for their biomedical applications. In this work, we reported that a catalyst-free thiol-yne click reaction can be utilized for fabrication of AIE-active FPNs in short reaction time and even without protection of inert gas. The results indicated that the obtained AIE-active amphiphilic copolymers (PEGMA-PhE) can readily self-assemble into luminescent nanoparticles (PEGMA-PhE FPNs) with high water dispersity, uniform size and morphology, red fluorescence. Cell viability examination and cell uptake behavior of PEGMA-PhE FPNs confirmed that these AIE-active FPNs possess low toxicity towards cells and can be easily internalized by cells through non-specific route. Therefore the remarkable properties of PEGMA-PhE FPNs such as high water dispersity, AIE-active fluorescence and nanoscale size as well as excellent biocompatibility make them promising for biomedical applications.

Abstract:Fluorescent organic nanoparticles (FNPs) with intensive luminescence are widely applied in various biomedical fields, but the malicious aggregation caused quenching effect of conventional organic dyes hampers their biomedical application. In this work, we report a facile and highly efficient method for fabrication of FNPs with a stable cross-linked structure and enhanced luminescence via a one-pot mercaptoacetic acid locking imine reaction, in which a hydrophobic aggregation induced emission (AIE) dye (AHAn) and hydrophilic polylysine (Plys) were used as two reaction components, while mercaptoacetic acid acted as the lock to conjugate AHAn and Plys. The final amphiphilic fluorescent materials (Plys&AHAn&Plys) are readily formed FNPs in aqueous solution and exhibit high water dispersibility and strong luminescence. Furthermore, Plys&AHAn&Plys FNPs display low toxicity and good biological imaging performance. Altogether, we have developed a rather facile one-pot multicomponent reaction (MCR) to fabricate AIE active FNPs, which shows many promising properties for biomedical applications. More importantly, the MCR strategy could also be used for fabrication of many other multifunctional AIE active polymeric nanoprobes due to its good substrate adaptability and high efficiency.
One-Step Preparation of Branched Peg Functionalized Aie-Active Luminescent Polymeric Nanoprobes

Abstract:The synthesis of amphiphilic aggregation-induced emission (AIE) dyes based organic nanoparticles has recently attracted increasing attention in the biomedical fields. These AIE dyes based nanoparticles could effectively overcome the aggregation caused quenching effect of conventional organic dyes, making them promising candidates for fabrication of ultrabright organic luminescent nanomaterials. In this work, AIE-active luminescent polymeric nanoparticles (4-NH 2 -PEG-TPE-E LPNs) were facilely fabricated through Michael addition reaction between tetraphenylethene acrylate (TPE-E) and 4-arm-poly(ethylene glycol)-amine (4-NH 2 -PEG) in rather mild ambient. The 4-NH 2 -PEG can not only endow these AIE-active LPNs good water dispersibility, but also provide functional groups for further conjugation reaction. The size, morphology and luminescent properties of 4-NH 2 -PEG-TPE-E LPNs were characterized by a series of techniques in detail. Results suggested that these AIE-active LPNs showed spherical morphology with diameter about 100–200 nm. The obtained 4-NH 2 -PEG-TPE-E LPNs display high water dispersibility and strong fluorescence intensity because of their self assembly and AIE properties of TPE-E. Biological evaluation results demonstrated that 4-NH 2 -PEG-TPE-E LPNs showed negative toxicity toward cancer cells and good fluorescent imaging performance. All of these features make 4-NH 2 -PEG-TPE-E LPNs promising candidates for biological imaging and therapeutic applications.

Abstract:Novel cross-linked copolymers of PEG-IM-PhNH2 are successfully synthesized through PEGylation via radical polymerization of 2-isocyanatoethyl methacrylate and poly(ethylene glycol) monomethyl ether methacylate and subsequent cross-linking with an amino-terminated aggregation-induced emission fluorogen. Such obtained amphiphilic copolymers can self-assemble to form uniform fluorescent polymeric nanoparticles (FPNs) and be utilized for cell imaging. These cross-linked FPNs are demonstrated good water dispersibility with ultralow critical micelle concentration (∼ 0.002 mg mL(-1)), uniform morphology (98 ± 2 nm), high red fluorescence quantum yield, and excellent biocompatibility. More importantly, this novel strategy of fabricating cross-linked FPNs paves the way to the future development of more robust and biocompatible fluorescent bioprobes.

Abstract:Aggregation-induced emission (AIE)-active fluorescent organic nanoparticles (FNPs) have been extensively explored for fluorescence "turn-on" bio-imaging applications with the unique advantages over conventional FNPs. Transformation of AIE-active molecules into FNPs can greatly expand their biomedical application potential. Here we reported a novel "one-pot" strategy for fabricating AIE-active FNPs through an ultrasonic-assisted, catalysts-free and solvent-free Kabachnik-Fields (KF) reaction for the first time. The KF reaction can be completed within 10min to generate AIE-active PTH-CHO-PEI-DEP FNPs through mixing polyethylenimine and aldehyde group containing AIE dyes and diethyl phosphate. These PTH-CHO-PEI-DEP FNPs were confirmed by proton nuclear magnetic resonance (1H NMR) spectroscopy, transmission electron microscopy (TEM) and fluorescence spectroscopy etc. The cell uptake behavior as well as cell viability of PTH-CHO-PEI-DEP FNPs was examined to evaluate their potential for biomedical application. We demonstrated that the amphiphilic α-aminophosphonate polymers could self-assemble into PTH-CHO-PEI-DEP FNPs in aqueous solution and showed excellent water dispersibility. TEM image shows the size of PTH-CHO-PEI-DEP FNPs is 100-200nm. More importantly, the PTH-CHO-PEI-DEP FNPs emit strong green fluorescence and desirable biocompatibility, making them very suitable for biomedical applications. Finally, thus smart FNPs design together with their excellent performance will open a new avenue in the development of FNPs for following biological processes such as carcinogenesis.

Abstract:The development of simple and effective methods for synthesis of fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) plays an important role for the biomedical applications of AIE-active FPNs. In present work, we developed a facile strategy for the fabrication of AIE-active FPNs by a post-polymerization method based on the microwave-assisted Kabachnik–Fields (KF) reaction, which can conjugate with poly(PEGMA-NH2), AIE-active dye (TPE-CHO) and diethyl phosphate (DP) under microwave irradiation within 5min. The characterization results confirm that PEGMA-TPE FPNs are successfully prepared through the microwave-assisted KF reaction. The resultant AIE-active FPNs show high water dispersity, intensive fluorescence and low cytotoxicity. These features make these AIE-active FPNs great potential for biomedical applications. Moreover, the microwave-assisted KF reaction is simple, fast, atom economy that should be a general strategy for the fabrication of various multifunctional AIE-active FPNs. We believe this work will open up a new avenue for the preparation of AIE-active functional materials with great potential for different applications.

Abstract:The fluorescent organic nanoparticles (FNPs) with aggregation-induced emission (AIE) feature have received increasing attention for their advanced optical properties. Although many efforts have been devoted to the fabrication and biomedical applications of AIE-active FNPs, the preparation of branched AIE-active FNPs with degradability through formation of dynamic bonds have rarely been reported. In this work, branched AIE-active FNPs were fabricated via dynamic linkage of hydrophobic hyperbranched and degradable Boltorn H40 (H40) with phenylboronic acid terminated AIE dye (PhB(OH)2) and mPEG (mPEG-B(OH)2), which relied on a facile one-pot strategy between phenylboronic acid and diol group of H40. The branched H40-star-mPEG-PhB(OH)2 FNPs were characterized using nuclear magnetic resonance spectroscopy, Fourier transform infrared spectroscopy, and fluorescence spectroscopy. Benefiting from their highly branched structure and amphiphilic properties, H40-star-mPEG-PhB(OH)2 could self-assemble into micelles and emit strong orange-red fluorescence. More importantly, cell viability results demonstrated that H40-star-mPEG-PhB(OH)2 FNPs showed good biocompatibility and promising candidates for bio-imaging. Taken together, we developed a one-pot strategy for preparation of branched AIE-active FNPs through the formation of dynamic phenyl borate. The resultant H40-star-mPEG-PhB(OH)2 FNPs should be promising biomaterials for different applications for biodegradability of H40 and responsiveness of phenyl borate.

Abstract:In recent years, the fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) feature have been extensively exploited in various biomedical fields owing to their advantages, such as low toxicity, biodegradation, excellent biocompatibility, good designability and optical properties. Therefore, development of a facile, efficient and well designable strategy should be of great importance for the biomedical applications of these AIE-active FPNs. In this work, a novel method for the fabrication of AIE-active FPNs has been developed through the self-catalyzed photo-initiated reversible addition fragmentation chain transfer (RAFT) polymerization using an AIE dye containing chain transfer agent (CTA), which could initiate the RAFT polymerization under light irradiation. The results suggested that the final AIE-active FPNs (named as TPE-poly(St-PEGMA)) showed great potential for biomedical applications owing to their optical and biological properties. More importantly, the method described in the work is rather simple and effective and can be further extended to prepare many other different AIE-active FPNs owing to the good monomer adoptability of RAFT polymerization.

Abstract:Over the past several years, the biomedical applications of fluorescent organic nanoparticles (FONs) with aggregation-induced emission (AIE) feature have been extensively explored because the AIE-active FONs could effectively overcome the aggregation caused quenching (ACQ) effect of FONs based on conventional organic dyes. The development of novel methods for synthesis of AIE-active FONs plays a centre role for their biomedical applications. In this work, we reported a facile one-step thiol-ene click reaction for fabrication of AIE-active FONs through conjugation of acrylated PEG and AIE-active tetraphenylethylene (TPE) with two ene bonds using pentaerythritol tetra(3-mercaptopropionate) as the linkage. The successful synthesis of TPE containing AIE-active copolymers was evidenced by various characterization techniques. The particle size and fluorescence properties of the resultant TPE-S-PEG copolymers were evaluated by transmission electronic microscopy and fluorescence spectroscopy. Moreover, the cell viability and cell uptake behavior was also examined to evaluate their potential for biological imaging. We demonstrated that the cross-linked TPE-S-PEG show small size, high water dispersibility, low cytotoxicity and strong fluorescence for tracing. All of these advantages endow the TPE-S-PEG FONs great potential for biological imaging applications. Furthermore, this novel click reaction can take place under mild experimental conditions with high efficiency. It could be also further expanded for preparation of multifunctional AIE-active materials due to the universality of the thiol-ene click reaction and good precursor applicapability. Taken together, we have developed a novel and effective thiol-ene click reaction to fabricate the cross-linked AIE-active FONs, which display excellent physicochemical and biological properties and are promising for biomedical applications.

Abstract:<p>Aggregation-induced emission (AIE) materials have attracted more and more attention for their broad applications in cell imaging, fluorescent detection, photodynamic therapy and so on. Obvious different from traditional fluorescence quenching materials, AIE-active materials could emit stronger fluorescence in concentrated and solid state. In this work, an AIE-active molecule (named as TPE-DB) was synthesized and applied to prepare amphiphilic fluorescent copolymers through light-induced RAFT polymerization in the present of air. The poly(ethylene oxide) monomethacrylate (PEGMA), itaconic acid (IA) and TPE-DB were selected as monomers and Ir<sup>3+</sup> complex was used as photoredox catalyst. The samples were carefully characterized and analyzed by different techniques. More interesting, we demonstrated that TPE-PEGMA-IA could form nanoparticles with small size and high water dispersibility. The results from cell viability evaluation and confocal laser scanning microscopy imaging suggested that these fluorescent nanoparticles possess low cytotoxicity and are promising for biological imaging. Owing to the introduction of IA, these fluorescent polymeric nanoparticles (FPNs) could also be utilized for drug delivery. Taken together, we believe that the light-induced polymerization should be a simple and useful route for synthesizing AIE-active fluorescent copolymers, which could greatly advance the biomedical applications of AIE-active functional materials.</p>

Abstract:Aggregation-induced emission (AIE) active fluorescence polymeric nanoparticles (FPNs) have recently received increasing interests for biomedical applications such as cell imaging, drug delivery, disease diagnosis and treatment. Fabricated strategies of AIE-active FPNs with high efficiency, simplification and tenderness are still passionately pursued to promote the development of theranostic systems. In this work, we develop a facile method for the preparation of AIE-active FPNs by adopting Ce(IV)/HNO3 redox polymerization under near room temperature. Thus-prepared FPNs (named as PEG-PLC-1) possess unique AIE feature, great water dispersity, excellent biocompatibility and biodegradability because of the conjugation of ultra-bright AIE dye (PhE-alc) and biodegradable PEG-PCL linear copolymers. The 1H nuclear magnetic resonance (NMR) spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, transmission electron microscopy (TEM), UV–Visible and fluorescence spectrometers were used to confirm the successful fabrication of AIE-active FPNs. Cell viability and cellular uptake behavior of PEG-PLC-1 FPNs were further investigated for their potential biomedical applications. Results demonstrated that PEG-PLC-1 FPNs are high water dispersity, intensive luminescence and low cytotoxicity, making them very attractive for biomedical applications. More importantly, the method for the fabrication of AIE-active biodegradable FPNs can be occurred under rather facile conditions (e.g., low temperature, free of metal catalysts, common chain transfer agent and aqueous solution) and are specially used for fabrication of AIE-active polysaccharides with poor organic solubility.

Abstract:The development of effective strategies for fabrication of fluorescent organic nanoparticles (FONs) with an aggregation-induced emission (AIE) feature has an important impact on the biomedical applications of these AIE-active FONs. In the current work, an ultrafast strategy for fabricating AIE-active FONs is developed through a one-pot microwave-assisted, catalysts-free, and solvent-free Kabachnik-Fields (KF) reaction for the first time. It is demonstrated that such organophosphorous-containing AIE-active block polymers can be synthesized within 2 min under air atmosphere through the microwave-assisted KF reaction. These polymers show amphiphilic properties and can self-assemble into mPEG-CHO-Phe-NH2-DEP FONs, which display high water dispersibility and desirable optical properties. Biological evaluation results suggest that the mPEG-CHO-Phe-NH2-DEP FONs exhibit low toxicity and are potential for biological imaging applications. More importantly, many other multifunctional AIE-active FONs can also be fabricated through the strategy described in this work owing to the universality of KF reaction. Besides, combined with the excellent properties of mPEG-CHO-Phe-NH2-DEP FONs, it is believed that such microwave-assisted KF reaction shall be an effective route for designing various AIE-active nanomaterials for different biomedical applications.

Abstract:In recent years, aggregation-induced emission (AIE) dyes based fluorescent organic nanoparticles (FONs) have achieved significant progress in various biomedical applications. In this work, we developed a covalent strategy to prepare biocompatible AIE-active dyes based cross-linked copolymers (MPC-POSS-PhE) via controllable reversible addition fragmentation chain transfer (RAFT) polymerization using zwitterionic 2-methacryloyloxyethyl phosphorylcholine (MPC), polymerizable AIE dye (named as PhE) and 8-vinyl polyoctahedral silsesquioxanes (POSS) as monomers. Due to the existence of hydrophilic MPC and hydrophobic PhE, the resultant copolymers will self-assemble into core-shell nanoparticles in aqueous solution with ultra-low critical micelle concentration (CMC). This could effectively overcome the drawbacks of non-crosslinked micelles and show more attractive properties and better performance for biomedical applications. Furthermore, the characterization results and biological assays demonstrated that the final MPC-POSS-PhE FONs show stable aqueous stability, uniform size and morphology, high water dispersity, desirable optical properties and low cytotoxicity. These remarkable properties make the resultant AIE-active nanoprobes great potential for biomedical applications.
