Abstract:Fluorescent nanoparticles used to detect important biological events in living cells or animals are in increasing demand in the biological and biomedical fields, and have attracted much attention from chemists and biologists in the past decade. Here, one aggregation-induced emission (AIE) bioconjugate, tetraphenylethene labelled chitosan (TPE-CS), is synthesized, which could be strongly emissive in the solid state. TPE-CS is used as a coating agent for negatively charged hydroxyapatite (HA) nanoparticles, and TPE-CS/HA nanocomposites with positive charges are well dispersed in their aqueous solution with a diameter of 111.9 nm. MTT assay indicates that the fluorescent TPE-CS/HA nanoparticles have good cytocompatibility. 293T cells are imaged by TPE-CS/HA nanoparticles. First, the nanoparticles are adhered to the cell membrane, and then many more particles are endocytosed through phagocytotic vesicles by culturing for a long time, resulting in a much stronger fluorescence emission. TPE-CS/HA bioprobes could strongly bind the cell cytoplasmic region, and might have promising applications in tumor diagnosis, long-term cell tracing, drug metabolism and drug delivery systems.

Abstract:Self-assembly of amphiphilic luminescent copolymers is a general route to fabricate fluorescent polymeric microparticles (FPMs). In this work, the FPMs with aggregation-induced emission (AIE) feature were fabricated via the combination of the chain transfer free radical polymerization and "one-pot" multicomponent reaction, which conjugated the aldehyde-containing AIE active dye AIE (CHO-An-CHO) and amino-terminated hydrophilic polymer (ATPPEGMA) using mercaptoacetic acid (MTA) as the "lock" molecule. The structure, chemical compositions, optical properties as well as biological properties of the PPEGMA-An-PPEGMA FPMs were characterized and investigated by means of a series of techniques and experiments in detail. We demonstrated the final copolymers showed amphiphilic properties, strong yellow fluorescence and high water dispersibility. Biological evaluation suggested that PPEGMA-An-PPEGMA FPMs possess low cytotoxicity and can be used for cell imaging. More importantly, many other AIE active FPMs are expected to be fabricated using the similar strategy because of the good substrate and monomer applicability of the multicomponent reaction and chain transfer living radical polymerization. Therefore, we could conclude that the strategy described in this work should be of great interest for fabrication of multifunctional AIE active nanoprobes for biomedical applications.

Abstract:Fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) characteristics have attracted much attention for biomedical applications due to their remarkable AIE feature, high water dispersity and desirable biocompatibility. The development of facile and effective strategies for fabrication of these AIE-active FPNs therefore should be of great importance for their biomedical applications. In this work, we reported that a catalyst-free thiol-yne click reaction can be utilized for fabrication of AIE-active FPNs in short reaction time and even without protection of inert gas. The results indicated that the obtained AIE-active amphiphilic copolymers (PEGMA-PhE) can readily self-assemble into luminescent nanoparticles (PEGMA-PhE FPNs) with high water dispersity, uniform size and morphology, red fluorescence. Cell viability examination and cell uptake behavior of PEGMA-PhE FPNs confirmed that these AIE-active FPNs possess low toxicity towards cells and can be easily internalized by cells through non-specific route. Therefore the remarkable properties of PEGMA-PhE FPNs such as high water dispersity, AIE-active fluorescence and nanoscale size as well as excellent biocompatibility make them promising for biomedical applications.

Abstract:Novel aggregation-induced emission (AIE) polymers (NS1 and NS2) were prepared through reversible addition fragmentation chain transfer polymerization of poly(ethylene glycol) monomethyl ether methacylate and a new AIE monomer with a vinyl group. The NS1 and NS2 copolymers were characterized by gel permeation chromatography, H-1 NMR spectroscopy, FT-IR spectroscopy, X-ray photoelectron spectroscopy, which proved their successful syntheses. Such amphiphilic copolymers could self assemble into fluorescent polymeric nanoparticles (FPNs) in water with good dispersibility, which were spherical in shape. The prepared FPNs of NS1 had a size range of 50-150 nm, strong green fluorescence in aqueous solution With fluorescence quantum yield of 31%, and low critical micelle concentration (CMC) of 0.030 mg mL(-1). The NS2 FPNs exhibited size range of 200-300 nm, intense green fluorescence with quantum yield of 38%, and CMC of 0.037 mg mL(-1). These FPNs were demonstrated highly biocompatible for cell imaging. By virtue of excellent biocompatibility via PEG modification, good particle stability with low CMC, and AIE fluorescent polymers with high photostability, the FPNs constructed in this work showed obvious advantages. Abstract:Fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) property have received increasing attention and possess promising biomedical application potential in the recent years. Many efforts have been devoted to the fabrication methodologies of FPNs and significant advance has been achieved. In this contribution, a novel strategy for the fabrication of AIE-active amphiphilic copolymers is reported for the first time based on the Ce(IV) redox polymerization. As an example, ene group containing AIE-active dye (named as Phe-alc) is directly grafted onto a water soluble polymer polyethylene glycol (PEG) in H2 O/THF system under low temperature. Thus-obtained amphiphilic fluorescent polymers will self-assemble into FPNs with ultra-low critical micelle concentration, ultra-brightness, and great water dispersibility. Biological evaluation results suggest that the PEG-poly(Phe-alc) possess excellent biocompatibility and can be used for tracing their behavior in cells using confocal laser scanning microscope. These features make PEG-poly(Phe-alc) FPNs promising candidates for many biomedical applications, such as cell imaging, drug delivery vehicles, and targeted tracing. More importantly, many other functional groups can also be incorporated into these AIE-active FPNs through the redox polymerization. Therefore, the redox polymerization should be a facile and effective strategy for fabrication of AIE-active FPNs.

Abstract:Here we report the ring-opening crosslinking PEGylation of an AIE epoxy monomer and a 4-arm PEG-amine to prepare a new cross-linked fluorescent polymer (PEG-EP3). When PEG-EP3 was dispersed in aqueous solution, the AIE components formed the hydrophobic cores and the PEG parts covered the surfaces, resulting in fluorescent polymeric nanoparticles (FPNs) with good dispersibility. PEG-EP3 and the resulting FPNs were characterized by gel permeation chromatography, H-1 NMR spectroscopy, FT-IR spectroscopy, X-ray photoelectron spectroscopy, dynamic light scattering, transmission electron microscopy, UV-Visible absorption and fluorescence spectra. The results confirmed the successful synthesis of PEG-EP3, which showed high water dispersibility with a size distribution of 249 +/- 1 nm, intense yellow-green fluorescence in aqueous solution with a fluorescence quantum yield of 35%, and a low critical micelle concentration (CMC) of 0.039 mg mL(-1). The cell uptake behaviour and cell imaging of the PEG-EP3 FPNs proved their high biocompatibility for biomedical applications. Owing to their excellent biocompatibility by the introduction of PEG as the main component, good colloidal stability with low CMC, and high fluorescence stability, the strategy in this work would provide a new approach to prepare novel biocompatible and robust cross-linked FPNs for biomedical applications.

Abstract:Glycosylated cross-linked red fluorescent amphiphilic polymer has been facilely synthesized in one pot, which would readily self-assemble into nanoparticles with high water dispersibility in aqueous media. The resulted nanoparticles have surplus carboxyl groups and glycosyl groups on the surface, which can be further functionalized, and they can demonstrate strong bio-favorable red fluorescence with a fluorescence quantum yield of 18% due to the aggregation induced emission (AIE) dyes aggregated in the core. Moreover, the nanoparticles also revealed great photostability and structure stability with an ultralow critical micelle concentration of 0.0018 mg mL(-1). Abstract:Aggregation-induced emission (AIE) dyes based fluorescent polymeric nanoparticles (FNPs) have been intensively explored for biomedical applications. However, many of these AIE-active FNPs are relied on the self-assembly of amphiphilic copolymers, which are not stable in diluted solution. Therefore, the introduction of cross-linkages into these micelles has demonstrated to be an efficient route to overcome this stability problem and endow ultra-low critical micelle concentrations (CMC) of these AIE-active FNPs. In this work, we reported the fabrication of cross-linked AIE-active FNPs through controllable reversible addition fragmentation chain transfer polymerization by using commercially available octavinyl-T8-silsesquioxane (8-vinyl POSS) as the cross-linkage for the first time. The resultant cross-linked amphiphilic copolymers (named as PEG-POSS-PhE) are prone to self-assemble into stable core-shell nanoparticles with well water dispersity, strong red fluorescence and low CMC (0.0069 mg mL(-1)) in aqueous solution. More importantly, PEG-POSS-PhE FNPs possess some other properties such as high water dispersity, uniform morphology and small size, excellent biocompatibility and cellular internalization, providing great potential of PEG-POSS-PhE FNPs for biological imaging application. Abstract:Driven by the high demand for sensitive and specific tools for optical imaging, fluorescent nanoprobes with various working mechanisms and advanced functionalities are flourishing at an incredible speed. This work reports the design and fabrication of aggregation-induced emission (AIE)-active fluorescent organic nanoparticles (FNPs) via forming dynamic phenyl borate between diol containing hydrophobic AIE dye (APD-PhCHO) and phenylboronic acid pendant hydrophilic polymers (PEGMA-VPBA) within 30 min. The final AIE-active APD-PhCHO-PEGMA-VPBA FNPs display high water dispersibility and strong fluorescence emission because of their amphiphilic properties and AIE feature. Biological evaluation suggests that APD-PhCHO-PEGMA-VPBA FNPs possess negative effect on HeLa cells and desirable optical properties for biological imaging. More importantly, phenyl borate is a dynamic bond with pH and glucose responsiveness. Furthermore, different functions can be designed and introduced into these AIE-active systems through adoption of different monomers for good applicability of free radical polymerization. Therefore, this work provides a novel platform for preparation of multifunctional AIE-active nanosystems with responsiveness for various biomedical applications.

Abstract:Fluorescent materials with aggregation-induced emission (AIE) have standout as one of the most interesting candidates for biomedical applications. The development of new methods for fabrication of AIE-active materials will surely advance their applications in related fields. In this work, we reported a novel method for preparation of AIE-active luminescent polymer (Lys-PEG-TPE) through a three-component reaction involving carbon dioxide (CO 2 ) using lysine, Polyethylene glycol (PEG) and AIEgen as the reactants. On account of the introduction of PEG, Lys-PEG-TPE could well disperse in aqueous solution, while hydrophobic TPE will aggregate in the core of self-assembled nanoparticles, endowing the obvious enhancement of fluorescence intensity. Results from biological assays suggested that Lys-PEG-TPE exhibits almost no toxicity after incubation with HepG2 cells and its fluorescence can be clearly observed in cytoplasm after cell internalization. More importantly, the AIE feature of Lys-PEG-TPE could not only effectively solve the aggregation-induced quenching effect of traditional fluorescent molecules, the carboxyl groups of lysine could also be used for loading functional cationic ions (e.g., anticancer drugs or imaging agents) to fabricate multifunctional theranostic and imaging composites. Considering the benign reaction conditions, high atom economy, and well tunability of reactants of multicomponent reactions, this strategy should be of broad interesting for fabrication of multifunctional AIE-active materials for biomedical applications.

Abstract:Over the past several years, the biomedical applications of fluorescent organic nanoparticles (FONs) with aggregation-induced emission (AIE) feature have been extensively explored because the AIE-active FONs could effectively overcome the aggregation caused quenching (ACQ) effect of FONs based on conventional organic dyes. The development of novel methods for synthesis of AIE-active FONs plays a centre role for their biomedical applications. In this work, we reported a facile one-step thiol-ene click reaction for fabrication of AIE-active FONs through conjugation of acrylated PEG and AIE-active tetraphenylethylene (TPE) with two ene bonds using pentaerythritol tetra(3-mercaptopropionate) as the linkage. The successful synthesis of TPE containing AIE-active copolymers was evidenced by various characterization techniques. The particle size and fluorescence properties of the resultant TPE-S-PEG copolymers were evaluated by transmission electronic microscopy and fluorescence spectroscopy. Moreover, the cell viability and cell uptake behavior was also examined to evaluate their potential for biological imaging. We demonstrated that the cross-linked TPE-S-PEG show small size, high water dispersibility, low cytotoxicity and strong fluorescence for tracing. All of these advantages endow the TPE-S-PEG FONs great potential for biological imaging applications. Furthermore, this novel click reaction can take place under mild experimental conditions with high efficiency. It could be also further expanded for preparation of multifunctional AIE-active materials due to the universality of the thiol-ene click reaction and good precursor applicapability. Taken together, we have developed a novel and effective thiol-ene click reaction to fabricate the cross-linked AIE-active FONs, which display excellent physicochemical and biological properties and are promising for biomedical applications.

Abstract:In recent years, the fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) feature have been extensively exploited in various biomedical fields owing to their advantages, such as low toxicity, biodegradation, excellent biocompatibility, good designability and optical properties. Therefore, development of a facile, efficient and well designable strategy should be of great importance for the biomedical applications of these AIE-active FPNs. In this work, a novel method for the fabrication of AIE-active FPNs has been developed through the self-catalyzed photo-initiated reversible addition fragmentation chain transfer (RAFT) polymerization using an AIE dye containing chain transfer agent (CTA), which could initiate the RAFT polymerization under light irradiation. The results suggested that the final AIE-active FPNs (named as TPE-poly(St-PEGMA)) showed great potential for biomedical applications owing to their optical and biological properties. More importantly, the method described in the work is rather simple and effective and can be further extended to prepare many other different AIE-active FPNs owing to the good monomer adoptability of RAFT polymerization.

Abstract:Chitosan with tetraphenylethene pendants (TPE-CS) are synthesized by reaction between amine and isothiocyanate groups of chitosan and tetraphenylethene (TPE), respectively. Nanoparticles of TPE-CS (TPE-CS NPs) are fabricated by ionic gelation method. The NPs are uniform in size, spherical in shape, monodispersed, and positive in surface charge. The suspension of TPE-CS NPs emits strong blue fluorescence under photoexcitation due to the aggregation-induced emission characteristics of the TPE moieties. The NPs can be internalized into cytoplasm through endocytosis pathway and retain inside the live cells to image the cells. Cytotoxicity assay reveals that TPE-CS NPs are cytocompatible and thus can be used for long-term live cell imaging.

Abstract:<p>Aggregation-induced emission (AIE) materials have attracted more and more attention for their broad applications in cell imaging, fluorescent detection, photodynamic therapy and so on. Obvious different from traditional fluorescence quenching materials, AIE-active materials could emit stronger fluorescence in concentrated and solid state. In this work, an AIE-active molecule (named as TPE-DB) was synthesized and applied to prepare amphiphilic fluorescent copolymers through light-induced RAFT polymerization in the present of air. The poly(ethylene oxide) monomethacrylate (PEGMA), itaconic acid (IA) and TPE-DB were selected as monomers and Ir<sup>3+</sup> complex was used as photoredox catalyst. The samples were carefully characterized and analyzed by different techniques. More interesting, we demonstrated that TPE-PEGMA-IA could form nanoparticles with small size and high water dispersibility. The results from cell viability evaluation and confocal laser scanning microscopy imaging suggested that these fluorescent nanoparticles possess low cytotoxicity and are promising for biological imaging. Owing to the introduction of IA, these fluorescent polymeric nanoparticles (FPNs) could also be utilized for drug delivery. Taken together, we believe that the light-induced polymerization should be a simple and useful route for synthesizing AIE-active fluorescent copolymers, which could greatly advance the biomedical applications of AIE-active functional materials.</p>

Abstract:Fluorescent organic nanoparticles (FNPs) with intensive luminescence are widely applied in various biomedical fields, but the malicious aggregation caused quenching effect of conventional organic dyes hampers their biomedical application. In this work, we report a facile and highly efficient method for fabrication of FNPs with a stable cross-linked structure and enhanced luminescence via a one-pot mercaptoacetic acid locking imine reaction, in which a hydrophobic aggregation induced emission (AIE) dye (AHAn) and hydrophilic polylysine (Plys) were used as two reaction components, while mercaptoacetic acid acted as the lock to conjugate AHAn and Plys. The final amphiphilic fluorescent materials (Plys&AHAn&Plys) are readily formed FNPs in aqueous solution and exhibit high water dispersibility and strong luminescence. Furthermore, Plys&AHAn&Plys FNPs display low toxicity and good biological imaging performance. Altogether, we have developed a rather facile one-pot multicomponent reaction (MCR) to fabricate AIE active FNPs, which shows many promising properties for biomedical applications. More importantly, the MCR strategy could also be used for fabrication of many other multifunctional AIE active polymeric nanoprobes due to its good substrate adaptability and high efficiency.
Preparation of biocompatible and photostable PEGylated red fluorescent nanoparticles for cellular imaging

Abstract:PEGylated red fluorescent nanoparticles with excellent biocompatibility and photostablity were facilely prepared from nonluminous materials: first red fluorescent nanoparticles (GCC NPs) were robustly prepared from chitosan and glutaraldehyde; then the amino groups and hydroxyl groups on the GCC NP surfaces were modified with 2-bromoisobutyryl bromide to obtain GCC-Br, which was employed as the nanoparticles' initiator in the ATRP system to finally obtain the GCC-poly(PEGMA) conjugates. The resulting GCC-poly(PEGMA) can emit a biofavorable deep red luminescence, which is quite stable and antiphotobleaching. Furthermore, GCC-poly(PEGMA) demonstrated excellent biocompatibility and water dispersibility due to the PEGMA chains conjugated on the surface; and the abundant hydroxyl groups on the surface can be further functionalized with diverse other groups or large biomolecules. This construction method demonstrated great advantages due to its simple strategy and economic value, and the NPs' successful cellular imaging endows them with a lot of potential for various biomedical applications.

Abstract:Aggregation-induced emission (AIE) polymeric nanoparticles (NPs) with a size of several tens of nanometers were successfully prepared by miniemulsion copolymerization of methyl methacrylate (MMA) with an AIE monomer, 1-allyl-1-methyl-2,3,4,5-tetraphenylsilole (AMTPS). The prepared AIE poly(MMA-co-AMTPS) NPs exhibited strong photoluminescence with high quantum yields. Promisingly, the brightness of AIE poly(MMA-co-AMTPS) NPs, which increased linearly with an increase in AMTPS content, could be accurately modulated via the AMTPS content in the range of 0-20 wt%. In vitro cell experiments indicated that the AIE poly(MMA-co-AMTPS) NPs were promising cell staining reagents with good biocompatibility and high photostability.
