Abstract:Nacre powder suspensions in alcohol added with acetic acid (Suspension I) or calcium chloride (Suspension II) were prepared. Titanium substrates were coated with nacre powder by EPD in the suspensions. The result shows that Suspension II is more stable than Suspension I, and deposition rate in Suspension II is much higher than that in Suspension I. XRD and FTIR results show that the nacre coatings exhibit the same characteristics as the raw nacre powder. SEM morphologies show that uniform coatings are achieved. Scratch test shows that bonding strength of coatings from Suspension I is much higher than that of Suspension II. The higher bonding strength from Suspension I was confirmed to be closely related to the concurrent deposition of organic molecules dissolved in the suspension.

Abstract:Background For dental implants, early infection and peri-implantitis after implant restoration are major reasons for implant failure. Plaque biofilm formation is the initiator for implant-related infection. Therefore, it is urgent to develop a new antibacterial implant materials or treatment methods. Aim/Hypothesis In this study, Sr- and Zn- incorporated micro nano-structured titanium surface with nanorods (MNT-Sr Zn) was fabricated by a two-step hydrothermal process, aiming to improve osseointegration and reduce implant failure caused by implant-related infection and broaden new ideas for the future research Material and Methods The Mod Ti surfaces were used as a control. The characterization of different surfaces was detected by a field emission scanning electron microscope (FE-SEM) equipped with energy dispersive spectra (EDS) detector, x-ray diffractometer (XRD) and inductively coupled plasma mass spectroscopy (ICP-MS). Meanwhile, Staphylococcus aureus (S. aureus) was selected to evaluate antibacterial properties through Live Dead Staining and watching the adhesive bacterial morphology under FE-SEM. What's more, rat bone marrow-derived mesenchymal stem cells (rBMSCs) were cultured on the surfaces to detect their early osteogenic properties, like the alkaline phosphatase(ALP) activity and the expressions of osteogenesis related genes. Results The results confirmed that nanorod-like particles with a diameter of about 30-50 nm of MNT-Sr Zn composed of well-crystallized ZnO and SrTiO3 phases. In the EDS spectra, the elements of Zn and Sr were evenly distributed, and approximately 1.49 ± 0.16 wt% and 21.69 ± 2.74 wt% respectively. Besides, the release of Sr2+ and Zn2+ was a controlled and long-term process. More importantly, Zn-containing nanorod-like particles and the morphology could work together, slightly decreasing adhesion of S. aureus but increasing the proportion of dead cells, thus inhibiting subsequent biofilm formation, compared with the controlled group. In addition to this, MNT-Sr Zn could enhance the osteogenic differentiation via the upregulation of the transcription of osteogenic related genes and the expression of ALP in vitro. Conclusion and Clinical Implications This research provides a new surface modification method on titanium substrate for multi-functional implant material development. Due to the co-doping of Sr and Zn in the coating, biomechanical stability, antibacterial capability and early osteogenic inductive effect are enhanced, which has the potential to be applied in dental implantation in the future.
Fabrication and in vitro evaluation of the collagen/hyaluronic acid PEM coating crosslinked with functionalized RGD peptide on titanium.

Abstract:Surface modification of titanium (Ti) using biomolecules has attracted much attention recently. In this study, a new strategy has been employed to construct a stable and bioactive coating on Ti. To this end, a derivative of hyaluronic acid (HA), i.e. HA-GRGDSPC-(SH), was synthesized. The disulfide-crosslinked Arg–Gly–Asp (RGD)-containing collagen/hyaluronic acid polyelectrolyte membrane (PEM) coating was then fabricated on Ti through the alternate deposition of collagen and HA-GRGDSPC-(SH) with five assembly cycles and subsequent crosslinking via converting free sulphydryl groups into disulfide linkages (RGD–CHC-Ti group). The assembly processes for PEM coating and the physicochemical properties of the coating were carefully characterized. The stability of PEM coating in phosphate-buffered saline solution could be adjusted by the crosslinking degree, while its degradation behaviors in the presence of glutathione were glutathione concentration dependent. The adhesion and proliferation of MC3T3-E1 cells were significantly enhanced in the RGD–CHC-Ti group. Up-regulated bone specific genes, enhanced alkaline phosphatase activity and osteocalcin production, the increased areas of mineralization were also observed in the RGD–CHC-Ti group. These results indicate that the strategy employed herein may function as an effective way to construct stable, RGD-containing bioactive coatings on Ti.

Abstract:Titanium implants are surface-modified to achieve bioactivity and often antibacterial properties. Such surface coatings may increase corrosion degradation and be weakly attached to the substrate. In the present research, biodegradable composite coatings, investigated so far as smart environment-sensitive, slowly releasing silver to the implant neighborhood, were produced as a combination of chitosan (CS) with Eudragit E 100 (EE100) or poly(4-vinylpyridine) (P4VP) polymers incorporating mesoporous bioactive glass nanoparticles (MBGNs). Such coatings were designed to positively affect corrosion resistance, bioactivity in vitro , and the bonding to the surface. The coating was obtained in a single electrophoretic deposition (EPD) process on the surface of chemically etched grade 2 titanium. The deposition was carried out using an ethanol-based suspension for 5 min at 60 V. Suspension stability was assessed through the quantification of zeta potential. The fabricated coatings underwent microstructural analysis, chemical composition profiling, surface roughness measurement, wettability assessment, thickness determination, evaluation of corrosion resistance, and investigation of their adherence to metallic substrates. Additionally, an immersion test extending up to 14 days was employed to assess the coatings' capacity for calcium phosphate formation. The resulting coatings tightly covered the metallic substrate; the CS/EE100/MBGNs coating exhibited superior uniformity, which can be attributed to the increased stability of this suspension, indicated by a higher zeta potential value. The deposited MBGNs showed a diameter of <100 nm. The prepared coatings have contributed to improved corrosion resistance of the system and facilitated the deposition of calcium phosphates. All investigated surfaces showed hydrophilic properties. Substrate-coating adhesion was notably enhanced for the CS/EE100/MBGNs coating. The proposed coatings, after in vitro and in vivo cytotoxicity studies, can successfully be applied on long-term load-bearing titanium bone implants.

Abstract:Implants with antibiotic drug loaded bioactive coatings have been increasingly applied in orthopedic operations. Here we report the drug release behavior of gentamycin loaded chitosan/calcium phosphate coatings on titanium. Chitosan/calcium phosphate coatings with different component ratios and surface topographies were prepared by electrochemical deposition method. Our results showed that the drug release from these coatings was controlled by their component ratio and surface topography, and the former ratio played a more significant role. The present coatings could provide an effective way to create both good bioactivity and antibacterial activity.

Abstract:The bacterial infection is one of the main reasons for the failure of implant surgery. In this paper, catechol functionalized chitosan (CSCT) was used as a reducing agent and stabilizer to synthesize CSCT-Ag NPs. The composite coating of titanium surface was then electrodeposited by a CSCT layer initially, followed by spin-coating of a CSCT/gelatin (G)/CSCT-Ag NPs layer. The results of XRD, UV–Vis, and TEM showed that the silver nanoparticles were successfully synthesized with uniform distribution, and the particle size was 4–22 nm. The cytotoxicity test results of CSCT-Ag NPs on A549 and EA.Hy926 cells showed when the concentration of CSCT-Ag NPs was less than 25 μg/mL, the survival rate of the two kinds of cells was greater than 80%. First electrodeposition and then spin-coating is feasible through the coating adhesion test and the adhesion level up to 5B. Compared with the pure Ti substrate, the corrosion resistance and hydrophilicity of the composite coating surface are significantly improved. The antibacterial rate of the composite coating against E. coli and S. aureus can reach 99% and 96%. In conclusion, CSCT-Ag NPs and the method of preparing coatings by electrodeposition and then spin-coating have good clinical application potential.

Abstract:The purpose of the study was to investigate the electrophoretic deposition (EPD) route, microstructure and surface properties of composite curcumin/chitosan coatings on commercially pure titanium substrates for biomedical applications. Multiple routes of preparation of the dispersed systems for the EPD process and their electrokinetic properties have been investigated to obtain homogeneous coatings. The zeta potential of solutions with various curcumin content in ethanol or isopropanol proved their relatively low electrophoretic mobility. Thus, curcumin was co-deposited with chitosan molecules on the cathode. The surface morphology of the coatings consisted of submicrometric curcumin particles embedded in the chitosan matrix. The increase in the curcumin content in the ethanol caused large agglomerates and undissolved curcumin particles to appear on the coating surface. The coatings were characterized by high adhesion to the substrate and a water contact angle in the range of 85° to 95°. The coatings changed the zeta potential of the titanium surface from significantly negative (-46.7 ± 2.3 mV) to less negative values (-20.6 ± 2.6 mV). The developed coatings are promising for mitigating biofilm formation on the surface of titanium bone implants.

Abstract:As local gene therapy has received attention, immobilizing functional gene onto irregular oral implant surface has become an advanced challenge. Electrostatic layer-by-layer (LBL) assembly technique could achieve this goal and allow local and efficient administration of genes to the target cells. In this study, multilayers of cationic lipid/plasmid DNA (pEGFP-C1) complex (LDc) and anionic hyaluronic acid were assembled onto sandblasted-dual acid etched titanium disks by the LBL technique. Surface characteristics of the coatings were performed by x-ray photospectroscopy (XPS), contact angle measurements, and scanning electron microscopy (SEM). The cell biological characteristics of the coatings were evaluated by in vitro experiments. SEM results demonstrated that the porous titanium surface was gradually flattened with the increase of the multilayer. The XPS survey indicated that the N element was found from the coating. The coating degradation and pEGFP-C1 releasing kinetics showed that the more assembled layer numbers were, the larger the amount of DNA released in the first 30 h. MC3T3-E1 cells were cultured directly on the DNA-loaded surface. Higher enhanced green fluorescent protein (EGFP) expression efficiency was achieved by increasing the number of layers when cells were cultured after 24 or 72 h. The MC3T3-E1 cell viability on the surface of multilayer DNA coatings was significantly higher than that on control porous titanium surface. It was concluded that the approach established by the LBL technique had great potential in immobilizing gene coatings onto the porous titanium surface and subsequently influenced the function of the cultured cell. Abstract:Recently, surface micron/nano-topographical modifications have attracted a great deal of attention because it is capable of mimicking the hierarchical characteristics of bone. In the current work, a novel titania/calcium silicate hydrate (CSH) bi-layer coating with hierarchical surface topography was successfully prepared on titanium substrate through micro-arc oxidation (MAO) and subsequent hydrothermal treatment (HT). MAO treatment could lead to a micron-scale topographical surface with numerous crater-like protuberances. The subsequent HT process enables the in situ nucleation and growth of CSH nanoplates on MAO-fabricated titania surface. The nucleation of CSH nanoplates is considered to follow a dissolution-precipitation mechanism. Compared to MAO-fabricated coating with single-scale surface topography, MAO-HT-fabricated coating with hierarchical surface topography exhibits enhanced hydrophilicity, fibronectin adsorption and initial MG-63 cell attachment. The process of cell-material interactions is considered to be triggered by surface properties of the coated layer and indirectly mediated by protein adsorption on coating surface. These results suggest that MAO-HT treatment is an efficient way to prepare coatings with hierarchical surface topography on titanium surface, which is essential for altering protein adsorption and initial cell attachment. Abstract:The bacterial colonization and poor osseointegration of Ti implants significantly compromise their applications in load-bearing bone repair and replacement. To endorse the Ti with both excellent bioactivity and antibacterial ability, we developed a microarc oxidation coating that was modified uniformly by hydroxyapatite (HA) nanodots arrays and loaded regionally with chitosan hydrogel containing ciprofloxacin. The bonding between the HA nanodots covered coating and the chitosan hydrogel is further enhanced via silanization and chemical grafting of glutaraldehyde. Benefiting from the regionally loaded structure of the chitosan hydrogel, the chitosan hydrogel unloaded area can promote the cell adhesion and proliferation with excellent bioactivity, though relatively low OD value of cck8 has been observed at the beginning of the cell culturing. Whereas, the OD value of cck8 rises with the prolongation of the cell culturing time due to the degradation of the regionally loaded chitosan hydrogel. With the help of the laden ciprofloxacin in chitosan hydrogels, the sample effectively sterilizes the bacterial with a bacteriostatic ring. Therefore, regional loading of chitosan hydrogel containing ciprofloxacin on the modified microarc oxidation coating is a good approach to endorse Ti with both excellent bioactivity and antibacterial ability.

Abstract:A novel double-layered antibacterial coating was fabricated on pure titanium (Ti) via a simple three-step electrodeposition process. Scanning electronic microscopy (SEM) images show that the coating was constructed with the inner layer of TiO2 nanotubes doped with silver nanoparticles (TNTs/Ag) and the outer layer of chitosan-gelatin mixture with zinc oxide and silver nanoparticles (CS-Gel-Ag-ZnO). In comparison, we also investigated the composition, structure and antibacterial properties of pure Ti coated with TNTs, TNTs/Ag or TNTs/Ag + CS-Gel-Ag-ZnO, respectively. The TNTs was about 100 nm wide and 240 nm to 370 nm tall, and most Ag nanoparticles (Ag NPs) with diameter smaller than 20 nm were successfully deposited inside the tubes. The CS-Gel-Ag-ZnO layer was continuous and uniform. Antibacterial activity against planktonic and adherent bacteria were both investigated. Agar diffusion test against Staphylococcus aureus (S. aureus) shows improved antibacterial capacity of the TNTs/Ag + CS-Gel-Ag-ZnO coating, with a clear zone of inhibition (ZOI) up to 14.5 mm wide. Dead adherent bacteria were found on the surface by SEM. The antibacterial rate against planktonic S. aureus was as high as 99.2% over the 24 h incubation period.

Abstract:Electroneuronographic study of the spontaneous course of Bell's palsy shows that chance of a satisfactory spontaneous return of facial function is reduced by 50% when 95% or more maximal degeneration is reached within two weeks of onset. For this reason immediate surgical decompression (including the meatal foramen) was performed on 14 patients with 90% or more degeneration within three weeks of palsy onset. These patients and 13 who refused treatment under similar conditions were reviewed one to three years later by three physicians familial with facial nerve problems. Results show that to obtain satisfactory (80% to 100%) return of facial movements in all cases of Bell's palsy, surgical decompression should be performed within 24 hours when degeneration reaches 90% to 94% within one to 21 days after onset. By using these guidelines, the number of unnecessary operations will be kept under 16%.

Abstract:Chitosan/drug (cefazolin) coatings were synthesized on alkali-treated Ti surfaces via electrophoretic deposition to improve the antibacterial activities and corrosion resistances of the surfaces. The coatings exhibited desirable adhesion to the alkali-treated Ti due to the penetration of chitosan and drug into the pores of the substrates. The chitosan were cationic molecules which carried drug and both of them migrated toward the cathode during electrophoretic deposition. At a low potential, the drug dissolved in the chitosan film, whereas at a high potential, the drug aggregated into sheets on the surface of chitosan. The high potential provided the coating with high drug loading and releasing capacity. The total drug release of chitosan/drug coatings with 4.3 and 6.9 μm thickness in Hank's balanced salt solution (HBSS) was about 3 and more than 14 days, respectively. The coatings exhibited increased corrosion resistance than the Ti in HBSS evaluated via potentiostat. Additionally, the coating with high drug loading demonstrated approximate 100% antibacterial activities against Escherichia coli; thus, the proposed coatings can be used as promising Ti coatings to limit bacterial adhesion and proliferation.

Abstract:Burst release of antibacterial agents generated by the surface antibacterial modification of Ti implants frequently results in a reduction in their biocompatibility and substantial toxic side effects. In this study, core-shelled nanoparticles namely AgNR@MSN, were synthesized successfully by mesoporous silica shell in-situ encapsulated Ag nanorods, and then co-deposited with chitosan to form a AgNR@MSN-chitosan composite coating on Ti substrate by electrophoretic deposition . The developed composite coating, due to the presence of AgNR, presented excellent photothermal effect under the irradiation of an 808 nm near-infrared laser, leading to both rapid increase of local temperature and triggered release of Ag ions for bacterial-killing effect against both E. coli and S. aureus . The addition of AgNR@MSN also improved the wettability of the coating and the MSN shell with mesoporous tunnels could barrier the contact and release rate of Ag to improve the biocompatibility. The developed photothermal responsive coating could be considered as a promising platform for achieving controlled release of Ag ions with outstanding antibacterial and biocompatible properties.

Abstract:In the current work, TiO2/silicate hierarchical coatings with various nanostructure morphologies were successfully prepared on titanium substrates through micro-arc oxidation (MAO) and subsequent hydrothermal treatment (FIT). Moreover, the nucleation mechanism and growth behavior of the nanostructures, hydrophilicity, protein adsorption and apatite-inducing ability of various coatings were also explored. The novel TiO2/silicate hierarchical coatings comprised calcium silicate hydrate (CSH) as an outer-layer and TiO2 matrix as an inner-layer.According to the morphological features, the nanostructures were classified as nanorod, nanoplate and nanoleaf. The morphology, degree of crystallinity and crystalline phases of CSH nanostructures could be controlled by optimizing the HT conditions. The nucleation of CSH nanostructures is caused by release and re-precipitation mechanism. The TiO2/CSH hierarchical coatings exhibited some enhanced physical and biological performances compared to MAO-fabricated coating. The improvement of the hydrophilicity, fibronectin adsorption and apatite-inducing ability was found to be morphological dependent according to the following trend: nanoleaf coating > nanoplate coating > nanorod coating > MAO coating. The results indicate that the tuning of physical and morphological properties of nanostructures coated on biomaterial surface could significantly influence the hydrophilicity, protein adsorption and in vitro bioactivity of biomaterial. Abstract:Titanium is widely used as surgical bone implants due to its excellent mechanical properties, corrosion resistance, and good biocompatibility. However, due to chronic inflammation and bacterial infections caused by titanium implants, they are still at risk of failure in interfacial integration of bone implants, severely limiting their broad clinical application. In this work, chitosan gels crosslinked with glutaraldehyde were prepared and successfully loaded with silver nanoparticles (nAg) and catalase nanocapsules (n (CAT)) to achieve functionalized coating on the surface of titanium alloy steel plates. Under chronic inflammatory conditions, n (CAT) significantly reduced the expression of macrophage tumor necrosis factor (TNF-α), increased the expression of osteoblast alkaline phosphatase (ALP) and osteopontin (OPN), and enhanced osteogenesis. At the same time, nAg inhibited the growth of S. aureus and E. coli . This work provides a general approach to functional coating of titanium alloy implants and other scaffolding materials.

Abstract:Infection-related titanium implant failure rates remain exceedingly high in the clinic. Functional surface coating represents a very promising strategy to improve the antibacterial and bioactive properties of titanium alloy implants. Here, we describe a novel bioactive surface coating that consists of a mussel-inspired carboxymethyl chitosan hydrogel loaded with silver nanoparticles (AgNPs) to enhance the bioactive properties of the titanium alloy. The preparation of hydrogel is based on gallic acid grafted carboxymethyl chitosan (CMCS-GA) catalyzed by DMTMM (4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride). To build a firm bonding between the hydrogel and titanium alloy plate, a polydopamine layer was introduced onto the surface of the titanium alloy. With HRP/H2O2 catalysis, CMCS-GA can simply form a firm gel layer on the titanium alloy plate through the catechol groups. The surface properties of titanium alloy were characterized by scanning electron microscope (SEM), X-ray photoelectron spectroscopy (XPS), and water contact angle. Silver nanoparticles were loaded into the gel layer by in situ reduction to enhance the antibacterial properties. In vitro antibacterial and cell viability experiments showed that the AgNPs-loaded Ti-gel possesses excellent antibacterial properties and did not affect the proliferation of rabbit mesenchymal stem cells (MSCs).

Abstract:Electrophoretic deposition (EPD) is a facile and feasible technique to prepare functional nanocomposite coatings for application in orthopedic-related implants. In this work, a ternary graphene oxide–chitosan–hydroxyapatite (GO–CS–HA) composite coating on Ti substrate was successfully fabricated by EPD. Coating microstructure and morphologies were investigated by scanning electron microscopy, contact angle test, Raman spectroscopy, Fourier transform infrared spectroscopy and thermogravimetric analysis. It was found GO–CS surface were uniformly decorated by HA nanoparticles. The potentiodynamic polarization test in simulated body fluid indicated that the GO–CS–HA coatings could provide effective protection of Ti substrate from corrosion. This ternary composite coating also exhibited good biocompatibility during incubation with MG63 cells. In addition, the nanocomposite coatings could decrease the attachment of Staphylococcus aureus.

Abstract:Cathodic electrophoretic deposition (EPD) was used to fabricate novel porous chitosan/pectin/hydroxyapatite (CS/P/HA) nanocomposite coatings on Ti6Al4V substrates. Rietveld refinement results revealed variations in peak intensity, position, and width, reflecting structural changes resulting from the reduction of the applied voltage from 30 to 10 V. The deposits obtained at 10 V after 1 h of presedimentation exhibited a denser microstructure with narrow pore size distribution compared to the others. The wettability and adhesion strength of coated Ti6Al4V improved with increasing HA content. Release of antibiotic from the composite coating confirmed 40% initial burst release, 50% semi-steady release and 10% residual release. A thick and uniform porous apatite layer was formed on Ti6Al4V after 21 days of immersion in simulated body fluid (SBF). Therefore, CS/P/HA coated Ti6Al4V has the potential for practical antiinfection performance.

Abstract:In the field of tissue engineering, a promising approach to obtain a bioactive, biomimetic, and antibiotic implant is the functionalization of a "classical" biocompatible material, for example, titanium, with appropriate biomolecules. For this purpose, we propose preparing self-assembling films of multiple components, allowing the mixing of different biofunctionalities "on demand". Self-assembling peptides (SAPs) are synthetic materials characterized by the ability to self-organize in nanostructures both in aqueous solution and as thin or thick films. Moreover, ordered layers of SAPs adhere on titanium surface as a scaffold coating to mimic the extracellular matrix. Chitosan is a versatile hydrophilic polysaccharide derived from chitin, with a broad antimicrobial spectrum to which Gram-negative and Gram-positive bacteria and fungi are highly susceptible, and is already known in the literature for the ability of its derivatives to firmly graft titanium alloys and show protective effects against some bacterial species, either alone or in combination with other antimicrobial substances such as antibiotics or antimicrobial peptides. In this context, we functionalized titanium surfaces with chitosan grafted to EAK16-II (a SAP), obtaining layer-by-layer structures of different degrees of order, depending on the preparative stoichiometry and path. The chemical composition, molecular structure, and arrangement of the obtained biofunctionalized surfaces were investigated by surface-sensitive techniques such as reflection-absorption infrared spectroscopy (RAIRS) and state-of-the-art synchrotron radiation-induced spectroscopies as X-ray photoemission spectroscopy (SR-XPS), and near-edge X-ray absorption fine structure (NEXAFS). Furthermore, was demonstrated that surfaces coated with EAK and Chit-EAK can support hNPs cell attachment and growth.
