Abstract:Measuring the three dimension nanoscale organization of protein or cellular structures is challenging, especially when the structure is dynamic. Owing to the informative total internal reflection fluorescence (TIRF) imaging under varied illumination angles, multi-angle (MA) TIRF has been examined to offer a nanoscale axial and a sub-second temporal resolution. However, conventional MA-TIRF still performs badly in lateral resolution and fail to characterize the depth-image in densely-distributed regions, leaving a huge contrast between the nanoscale axial resolution and the diffraction limited lateral resolution. Moreover, the previous reconstructions are highly restricted by the efficiency with the increased amount of illumination angles. Here, we for the first time, emphasize the lateral super-resolution in the MA-TIRF and exampled by simply introducing polarization modulation into the illumination procedure. Equipped with a sparsity and accelerated proximal algorithm, we examine a more precise 3D sample structure compared with previous methods, enabling live cell imaging with temporal-resolution of 2 seconds, recovering high-resolution mitochondria fission and fusion process. Since the introduced vortex half wave retarder is an add-on component to the existing MA-TRIF system and the algorithm is the first open sourced and the fastest, we anticipate that the method and algorithm introduced here would be adopted rapidly by the biological community.

Abstract:We report a widefield super-resolution (SR) fluorescence microscopy technique termed total internal reflection fluorescence pattern-illuminated Fourier ptychographic microscopy (TIRF-piFPM). It employs pattern-illuminated Fourier ptychography (piFP) under the total internal reflection fluorescence (TIRF) mode, providing a lateral resolution of similar to 100 nm, reducing the background level, and correcting unknown optical aberrations. Like the total internal reflection fluorescence structure illuminated microscopy (TIRF-SIM), the illumination field of TIRF-piFPM is modulated by sinusoidal patterns generated by evanescent wave interference. It differs from TIRF-SIM in that TIRE-piFPM reconstructs the SR image by FP iteration. To demonstrate the performance of TIRF-piFPM, we compare it with TIRF-SIM by conducting simulations and experiments and prove that TIRF-piFPM can provide a better result than TIRF-SIM in terms of its robustness to noise and aberration correction ability. In addition, dynamic changes of mitochondria in U2OS cells are captured with a temporal resolution of 2 s, demonstrating its live-cell imaging capability. The addvantages may enable TIRF-piFPM to serve as an alternative to SR fluorescence microscopes in the TIRF family, with promising applications in biological and biomedical imaging.

Abstract:Three-dimensional structured illumination microscopy (3D-SIM) plays an essential role in biological volumetric imaging with the capabilities of improving lateral and axial resolution. However, the traditional linear 3D algorithm is sensitive to noise and generates artifacts, while the low temporal resolution hinders live-cell imaging. In this paper, we propose a novel 3D-SIM algorithm based on total variation (TV) and fast iterative shrinkage threshold algorithm (FISTA), termed TV-FISTA-SIM. Compared to conventional algorithms, TV-FISTA-SIM achieves higher reconstruction fidelity with the least artifacts, even when the signal-to-noise ratio (STIR) is as low as 5 dB, and a faster reconstruction rate. Through simulation, we have verified that TV-FISTA-SIM can effectively reduce the amount of required data with less deterioration. Moreover, we demonstrate TV-FISTA-SIM for high-quality multi-color 3D super-resolution imaging, which can be potentially applied to live-cell imaging applications. Abstract:Spatial frequency shift (SFS) microscopy with evanescent wave illumination shows intriguing advantages, including large field of view (FOV), high speed, and good modularity. However, a missing band in the spatial frequency domain hampers the SFS superresolution microscopy from achieving resolution better than 3 folds of the Abbe diffraction limit. Here, we propose a novel tunable large-SFS microscopy, making the resolution improvement of a linear system no longer restricted by the detection numerical aperture (NA). The complete wide-range detection in the spatial frequency domain is realized by tuning the illumination spatial frequency actively and broadly through an angle modulation between the azimuthal propagating directions of two evanescent waves. The vertical spatial frequency is tuned via a sectional saturation effect, and the reconstructed depth information can be added to the lateral superresolution mask for 3D imaging. A lateral resolution of λ /9, and a vertical localization precision of ∼ λ /200 (detection objective NA = 0.9) are realized with a gallium phosphide (GaP) waveguide. Its unlimited resolution enhancing capability is demonstrated by introducing a designed metamaterial chip with an unusual large refractive index. Besides the great resolution enhancement, this method shows better anti-noise capability than classical structured illumination microscopy without SFS tunability. This method is chip-compatible and can potentially provide a mass-producible illumination chip module achieving the fast, large-FOV, and deep-subwavelength 3D nanoscopy.

Abstract:Total-internal-reflection fluorescence (TIRF) microscopy provides high optical-sectioning capability and a good signal-contrast ratio for structures near the surfaces of cells. In recent years, several improvements have been developed, such as variable-angle TIRF (VA-TIRF) and spinning TIRF (sp-TIRF), which permit quantitative image analysis and address non-uniform scattering fringes, respectively. Here, we present a dual-color DMD-based shadowless-illuminated variable-angle TIRF (siva-TIRF) system that provides a uniform illumination field. By adjusting the incidence angle of the illuminating laser on the back focal plane (BFP) of the objective, we can rapidly illuminate biological samples in layers of various thicknesses in TIRF or hollow-cone epi-fluorescence mode. Compared with other methods of accomplishing VA-TIRF/sp-TIRF illumination, our system is simple to build and cost-effective, and it provides optimal multi-plane dual-color images. By showing spatiotemporal correlated movement of clathrin-coated structures with microtubule filaments from various layers of live cells, we demonstrate that cortical microtubules are important spatial regulators of clathrin-coated structures. Moreover, our system can be used to prove superb axial information of three-dimensional movement of structures near the plasma membrane within live cells.

Abstract:Total internal reflection fluorescence (TIRF) microscopy is widely used in fluorescent imaging. Evanescent wave fields generated by the internal reflection are used to illuminate the sample, and only fluorophores within a thickness of 100 nm thick from the surface are activated, improving the signal-to-noise ratio (SNR) of the image. Sub-diffraction imaging with TIRF microscopy by stochastic photobleaching is studied. Each fluorophore can be localized from the recorded streaming followed by image subtraction. This method can yield contrast-enhanced images with a higher SNR and improve the lateral resolution to approximately 120 nm.

Abstract:To investigate fine structure in biological samples, over recent decades, super-resolution methods in light microscopy have been honed. Here, we report a technique integrating nonlinearity arising from fluorophore saturation with a new version of I5S to produce quasi-isotropic three-dimensional (3D) wide-field (WF) super-resolution imaging. This extended technique—saturated I5S (SI5S)—promises lateral and axial resolutions below 60 nm when the illuminating light intensity approaches the saturation threshold in simulation. A linear processing algorithm is used to acquire SI5S image reconstructions, and comparisons with 3D structured illumination microscopy (3D-SIM) and I5S show that a substantial improvement in resolution can be realized using SI5S. The notable improvement in 3D resolution means that SI5S could facilitate volumetric visualization of intracellular fine structure within biological samples.

Abstract:We present an alternative approach to realize structured illumination microscopy (SIM), which is capable for live cell imaging. The prototype utilizes two sets of scanning galvo mirrors, a polarization converter and a piezo-platform to generate a fast shifted, s-polarization interfered and periodic variable illumination patterns. By changing the angle of the scanning galvanometer, we can change the position of the spots at the pupil plane of the objective lens arbitrarily, making it easy to switch between widefield and total internal reflection fluorescent-SIM mode and adapting the penetration depth in the sample. Also, a twofold resolution improvement is achieved in our experiments. The prototype offers more flexibility of pattern period and illumination orientation changing than previous systems. Abstract:Imaging and tracking of near-surface three-dimensional volumetric nanoscale dynamic processes of live cells remains a challenging problem. In this paper, we propose a multi-color live-cell near-surface-volume super-resolution microscopy method that combines total internal reflection fluorescence structured illumination microscopy with multi-angle evanescent light illumination. We demonstrate that our approach of multi-angle interference microscopy is perfectly adapted to studying subcellular dynamics of mitochondria and microtubule architectures during cell migration.

The performance of our method was validated in simulated microtubules with variable signal-to-noise ratios (SNR) and the axial resolution and accuracy of reconstruction were evaluated in selecting different numbers of illumination angles or in different SNR conditions. In U373 cells, we reconstructed the 3-D localization of microtubules near the cell surface with high resolution using over a hundred different illumination angles. Theoretically, the presented TIRFM setup and 3D reconstruction method can achieve similar to 40 nm axial resolution in experimental conditions where SNR is as low as 2, with similar to 35 different illumination angles. Moreover, our system and reconstruction method have the potential to be used in live cells to track membrane dynamics in 3-D.

Abstract:Structured illumination microscopy (SIM) is one of the most widely used imaging tools in live-cell imaging because of the fast imaging speed and low-intensity requirement. In order to observe the interactions between cell structures, it is necessary to achieve multi-color simultaneous rapid imaging. However, existing systems collect multi-color results in a time-sequential manner. In live-cell imaging, this method brings distortions of the result. In this paper, we provide a dual-color simultaneous SIM system based on galvanometers, which can control the structured pattern of the two-channel flexibly. The resolution capability and simultaneous imaging capability of this system are verified, and the imaging results of living cells are obtained.

Abstract:We propose an elliptical mirror-based total-internal-reflection fluorescence (e-TIRF) microscopy with shadowless illumination and adjustable penetration depth. The elliptical mirror is used to produce a hollow-cone illumination with all azimuthal directions and a large range of incident angle, so as to attenuate the potential shadow effects when utilizing a single-direction illumination, such as asymmetries and low contrast. The experiment demonstrates that the e-TIRF method can realize shadowless imaging with symmetric intensity distribution. Meanwhile, the penetration depth of e-TIRF can be theoretically adjusted from 58 nm to 250 nm by adjusting the size of the aperture or the position of an opaque mask. This method extends the minimum penetration depth, which is useful for high axial resolution. (C) 2017 Optical Society of America

Abstract:Fourier ptychographic microscopy (FPM), as an emerging computational imaging method, has been applied to quantitative phase imaging with resolution bypassing the physical limit of the detection objective. Due to the weak illumination intensity and long image acquisition time, the achieved imaging speed in current FPM methods is still low, making them unsuitable for real-time imaging applications. We propose and demonstrate a high-speed FPM method based on using laser illumination and digital micro-mirror devices for illumination angle scanning. In this new, to the best of our knowledge, FPM method, we realized quantitative phase imaging and intensity imaging at over 42 frames per second (fps) with around 1 µm lateral resolution. The quantitative phase images have revealed membrane height fluctuations of red blood cells with nanometer-scale sensitivity, while the intensity images have resolved subcellular features in stained cancer tissue slices.

Fast two-snapshot structured illumination for temporal focusing microscopy with enhanced axial resolution

Abstract:We present a new two-snapshot structured light illumination (SLI) reconstruction algorithm for fast image acquisition. The new algorithm, which only requires two mutually {\pi} phase-shifted raw structured images, is implemented on a custom-built temporal focusing fluorescence microscope (TFFM) to enhance its axial resolution via a digital micromirror device (DMD). First, the orientation of the modulated sinusoidal fringe patterns is automatically identified via spatial frequency vector detection. Subsequently, the modulated in-focal-plane images are obtained via rotation and subtraction. Lastly, a parallel amplitude demodulation method, derived based on Hilbert transform, is applied to complete the decoding processes. To demonstrate the new SLI algorithm, a TFFM is custom-constructed, where a DMD replaces the generic blazed grating in the system and simultaneously functions as a diffraction grating and a programmable binary mask, generating arbitrary fringe patterns. The experimental results show promising depth-discrimination capability with an axial resolution enhancement factor of 1.25, which matches well with the theoretical estimation, i.e, 1.27. Imaging experiments on pollen grain samples have been performed. The results indicate that the two-snapshot algorithm presents comparable contrast reconstruction and optical cross-sectioning capability with those adopting the conventional root-mean-square (RMS) reconstruction method. The two-snapshot method can be readily applied to any sinusoidally modulated illumination systems to realize high-speed 3D imaging as less frames are required for each in-focal-plane image restoration, i.e., the image acquisition speed is improved by 2.5 times for any two-photon systems.

Abstract:Interferometric Scattering Microscopy (iSCAT) enables prolonged and high frame rate Single Particle Tracking (SPT) for single molecule dynamics studies. Typically, iSCAT setups employ scanning illumination schemes to achieve uniform sample illumination. However, this implementation limits the field of view (FoV) and maximum sampling rate, while increasing hardware requirements and setup size. We demonstrate the realization of a large (60μm x 60μm) uniformly illuminated FoV through a passive refractive optical element in the iSCAT illumination path. This scanning-free iSCAT microscopy setup is further combined with an objective based Total Internal Reflection Fluorescence Microscopy (TIRF) channel for a complementary fluorescence readout, a focus-lock system, and a tailored control platform via the open-source ImSwitch software, and has a compact footprint. As a proof-of-principle, we highlight the performance of the setup through the acquisition of iSCAT images with a uniform contrast and a ≤10 nm localization precision throughout the whole FoV. The performance is further demonstrated through dynamic iSCAT SPT and imaging Fluorescence Correlation Spectroscopy of lipid diffusion in a model membrane system. Our iSCAT setup thus depicts an accurate and improved way of recording fast molecular dynamics in life sciences.

Abstract:Total internal reflection fluorescence microscopy (TIRF microscopy) uses a rapid decay of evanescent waves to excite fluorophores within several hundred nanometers (nm) beneath the plasma membrane, which can effectively suppress excitation of fluorescence signals in the deep layers. From image stacks obtained with a plurality of different incident angles, a three-dimensional spatial structure of the observed sample can be reconstructed by a Multi-Angle-TIRF (MA-TIRF) algorithm that provides an axial resolution of ~50 nm. Taking into account the point spread function (PSF) of the TIRF microscopes, we further increase its lateral resolution by introducing a fast deconvolution algorithm into the reconstruction of MA-TIRF data (DMA-TIRF), which is approached in just one step of minimizing the reconstruction function. We also introduce a TV regularization term in the deconvolution algorithm to suppress artifacts induced by the excessive noise. Therefore, based on the hardware of existing MA-TIRF microscopes, the proposed DMA-TIRF algorithm has achieved lateral and axial resolutions of ~200 and ~50 nm, respectively.

Abstract:Two-photon microscopy (TPM) based on two-dimensional micro-electro-mechanical (MEMS) system mirrors shows promising applications in biomedicine and the life sciences. To improve the imaging quality and real-time performance of TPM, this paper proposes Lissajous scanning control and image reconstruction under a feed-forward control strategy, a dual-parameter alternating drive control algorithm and segmented phase synchronization mechanism, and pipe-lined fusion-mean filtering and median filtering to suppress image noise. A 10 fps frame rate (512 × 512 pixels), a 140 µm × 140 µm field of view, and a 0.62 µm lateral resolution were achieved. The imaging capability of MEMS-based Lissajous scanning TPM was verified by ex vivo and in vivo biological tissue imaging.

Abstract:We report a resolution-enhanced lensless color shadow imaging microscopy (RELCSIM) system based on large field-of-view (FOV) submicron-pixel imaging sensors. The physical pixel size of our custom made imaging chip is 0.95um x 0.95um, and the pixel-count is 25 millions (5120H x 5120V). By directly recording the shadow of the samples without any postprocssing, we have realized a microscope with a half-pitch resolution of similar to 1um and a FOV of similar to 25mm(2) simutaneously. To verify the resolution of our system, the grating samples coated on the surface of the chip are imaged. We further demonstrate the monochromatic and color shadow imaging of muscle tissue specimens with the prototype, which show the potential for applications such as diagnostic pathology.
