Abstract:We developed a facile strategy to obtain a new kind of mesoporous core-shell structured up-conversion nanoparticles (mUCNPs), composed of a NaYbF4:2%Er core and a mesoporous NaGdF4 shell. This mesoporous shell not only enhanced the up-conversion luminescence but also endowed many other functionalities of the nanoparticles such as drug delivery and bio-imaging capabilities. Moreover, after being conjugated with polyethylenimine (PEI) and folic acid (FA), core-shell mUCNPs exhibited good water dispersibility, enhanced drug delivery efficiency, and remarkable targeting ability to cancer cells. To certify the folate receptors (FR)-mediated targeted drug delivery, cell viability assay, cell up-conversion luminescence imaging and flow cytometry analysis were carried out. Furthermore, apart from the application for targeted drug delivery, the as-prepared core-shell mUCNPs could also be employed as the contrast agents for X-ray computed tomography (CT) and magnetic resonance (MR) imaging, because of the strong X-ray attenuation ability of Yb and high longitudinal molar relaxivity (r1) of Gd in the nanoparticles, providing the potential for simultaneously bio-imaging and cancer-targeting therapy.
Sub-10 Nm Water-Dispersible Β-Nagdf4:X% Eu3+ Nanoparticles with Enhanced Biocompatibility for in Vivo X-ray Luminescence Computed Tomography

Abstract:As a novel molecular and functional imaging modality, X-ray luminescence computed tomography (XLCT) has shown its potentials in biomedical and preclinic applications. However, there are still some limitations of X-ray-excited luminescent materials, such as low luminescence efficiency, poor biocompatibility, and cytotoxicity, making in vivo XLCT imaging quite challenging. In this study, for the very first time, we present on using sub-10 nm β-NaGdF4:X% Eu3+ nanoparticles with poly(acrylic acid) (PAA) surface modification, which demonstrate outstanding luminescence efficiency, uniform size distribution, water dispersity, and biosafety, as the luminescent probes for in vivo XLCT application. The pure hexagonal phase (β-) NaGdF4 has been successfully synthesized and characterized by X-ray powder diffraction (XRD) and transmission electron microscopy (TEM), and then the results of X-ray photoelectron spectroscopy (XPS), energy-dispersive X-ray spectrometry (EDX), and elemental mapping further confirm Eu3+ ions doped into NaGdF4 host. Under X-ray excitation, the β-NaGdF4 nanoparticles with a doping level of 15% Eu3+ exhibited the most efficient luminescence intensity. Notably, the doping level of Eu3+ has no effect on the crystal phase and morphology of the NaGdF4-based host. Afterward, β-NaGdF4:15% Eu3+ nanoparticles were modified with PAA to enhance the water dispersity and biocompatibility. The compatibility of in vivo XLCT imaging using such nanoparticles was systematically studied via in vitro cytotoxicity, physical phantom, and in vivo imaging experiments. The ultralow cytotoxicity of PAA-modified nanoparticles, which is confirmed by over 80% cell viability of SH-SY5Y cells when treated by high nanoparticle concentration of 200 μg/mL, overcome the major obstacle for in vivo application. In addition, the high luminescence intensity of PAA-modified nanoparticles enables the location error of in vivo XLCT imaging less than 2 mm, which is comparable to that using commercially available bulk material Y2O3:15% Eu3+. The proposed nanoparticles promote XLCT research into an in vivo stage. Further modification of these nanoparticles with biofunctional molecules could enable the potential of targeting XLCT imaging.

Abstract:New core-shell structured NaYF(4):Yb(3+),Tm(3+)@Fe(x)O(y) nanocrystals, with 20 nm Yb(3+),Tm(3+)-co-doped NaYF(4) nanocrystals as a core and 5 nm Fe(x)O(y) nanocrystals as a shell, have been synthesized and characterized by TEM and XRD analysis. These core-shell nanocrystals exhibit excellent near-infrared upconversion luminescence (UCL) emission at 800 nm under excitation by a continuous-wave 980 nm laser and superparamagnetic properties with a saturation magnetization (Ms) of ∼12 emu/g. Water-soluble nanocrystals were obtained by surface ligand exchange of oleic acid-coated precursor nanocrystals, and their internalization within living cells has been investigated by laser scanning UCL microscopy. Furthermore, the obtained core-shell nanocrystals have been applied in dual-modality T(2)-enhanced magnetic resonance (MR) and UCL imaging in vivo of the lymphatic system. Moreover, the toxicity of NaYF(4):Yb(3+),Tm(3+)@Fe(x)O(y) nanocrystals has also been evaluated by MTT assay, IC50 values, and histological analysis of lymphatic node sections.

Abstract:To circumvent the defects of different bioimaging techniques, the development of multifunctional probes for multimodality bioimaging is required. In the present study, a lanthanide-based core-shell-shell nanocomposite NaYbF4:Tm@CaF2@NaDyF4 composed of an ∼9.5 nm NaYbF4:Tm nanocrystal as the core, ∼2 nm CaF2 as the middle layer, and 1-2 nm NaDyF4 as the outermost shell was designed and synthesized. Following surface modification with the ligand, citrate acid, this nanocomposite was hydrophilic, emitted intense upconversion luminescence (UCL), and displayed a high X-ray computed tomography (CT) value of ∼490 Hounsfield units (HU) and excellent r2 relaxivity of 41.1 mM(-1) s(-1). These results confirmed that the introduction of a middle CaF2 layer was necessary as a barrier to reduce cross-relaxation and the surface quenching effect, thus enhancing the upconversion emission of Tm(3+). This citrate-modified NaYbF4:Tm@CaF2@NaDyF4 nanocomposite was used as a multifunctional contrast agent for trimodal lymphatic bioimaging with T2-weighted magnetic resonance imaging (MRI), CT, and UCL imaging. The concept of fabricating a core-multishell nanostructure and the introduction of a Dy(3+)-based host as an outer layer is a useful strategy and can be used to develop a novel multifunctional nanoprobe for multimodality bioimaging.

Abstract:Core–shell Fe3O4@NaLuF4:Yb,Er/Tm nanostructure (MUCNP) with multifunctional properties has been developed using a step-wise synthetic method. The successful fabrication of MUCNP has been confirmed by transmission electron microscopy, powder X-ray diffraction, energy-dispersive X-ray analysis and X-ray photoelectron spectroscopy. The MUCNP exhibits superparamagnetic property with saturation magnetization of 15 emu g−1, and T2-enhanced magnetic resonance (MR) effect with an r2 value of 21.63 s−1 mM−1 at 0.5 T, resulting from the Fe3O4 cores. Moreover, the NaLuF4-based MUCNP provides excellent X-ray attenuation and upconversion luminescence (UCL) emission under excitation at 980 nm. In vivo MR, computed tomography (CT) and UCL images of tumor-bearing mice show that the MUCNP can be successfully used in multimodal imaging. In vitro tests reveal that the MUCNP is non-cytotoxic. These results suggest that the developed MUCNP could be served as an MR, CT and UCL probe for tri-modality imaging.

Abstract:Multimodal imaging can compensate for the deficiencies and incorporate the advantages of individual imaging modalities. In this paper, we demonstrated the synthesis of core-shell nanocomposites LiLuF4@LiGdF4:Yb,Er/Tm constituted of tetragonal LiLuF4 nanoparticles as core and Yb,Er/Tm-codoped LiGdF4 as shell. LiLuF4@LiGdF4:Yb,Er/Tm nanoparticles display brighter upconversion luminescence (UCL) than NaGdF4:Yb,Er/Tm nanoparticles with the same size under continuous-wave excitation at 980 nm. The active shell layer of LiGdF4:Yb,Er/Tm not only provide the UCL center, but also serve as magnetic resonance (MR) imaging contrast agent. To further improve the UCL intensity, the inert LiGdF4 shell was coated on the LiLuF4@LiGdF4:Yb,Er/Tm nanoparticles. Furthermore, LiLuF4@LiGdF4:Yb,Tm@LiGdF4 nanoparticles have been successfully applied to UCL/X-ray computed tomography (CT)/MR tri-modal imaging on the modal of tumor-bearing mice.

Abstract:Molecular imaging modalities provide a wealth of information that is highly complementary and rarely redundant. To combine the advantages of molecular imaging techniques, F-18-labeled Cd3+/Yb3+/Er3+ co-doped NaYF4 nanophosphors (NPs) simultaneously possessing with radioactivity, magnetic, and upconversion luminescent properties have been fabricated for multimodality positron emission tomography (PET), magnetic resonance imaging (MRI), and laser scanning upconversion luminescence (UCL) imaging. Hydrophilic citrate-capped NaY0.2Gd0.6Yb0.18Er0.02F4 nanophosphors (cit-NPs) were obtained from hydrophobic oleic acid (OA)-coated nanoparticles (OA-NPs) through a process of ligand exchange of OA with citrate, and were found to be monodisperse with an average size of 22 x 19 nm. The obtained hexagonal cit-NPs show intense UCL emission in the visible region and paramagnetic longitudinal relaxivity (r(1) = 0.405 s(-1) -(mM)(-1)). Through a facile inorganic reaction based on the strong binding between Y3+ and F-, F-18-labeled NPs have been fabricated in high yield. The use of cit-NPs as a multimodal probe has been further explored for T-1-weighted MR and PET imaging in vivo and UCL imaging of living cells and tissue slides. The results indicate that F-18-labeled NaY0.2Gd0.5Yb0.18Er0.02 is a potential candidate as a multimodal nanoprobe for ultra-sensitive molecular imaging from the cellular scale to whole-body evaluation. Abstract:Early diagnosis that combines the high-resolutional CT and sensitive NIR-fluorescence bioimaging could provide more accurate information for cancerous tissues, which, however, remain a big challenge. Here we report a simple bimodal imaging platform based on PEGylated NaYbF(4): Tm(3+) nanoparticles (NPs) of less than 20 nm in diameter for both CT and NIR-fluorescence bioimaging. The as-designed nanoprobes showed excellent in vitro and in vivo performances in the dual-bioimaging, very low cytotoxicity and no detectable tissue damage in one month. Remarkably, the Yb(3+) in the lattice of NaYbF(4): Tm(3+) NPs functions not only as a promising CT contrast medium due to its high X-ray absorption coefficiency, but also an excellent sensitizer contributing to the strong NIR-fluorescent emissions for its large NIR absorption cross-section. In addition, these NPs could be easily excreted mainly via feces without detectable remnant in the animal bodies.

Abstract:Multimodality imaging overcomes the shortage and incorporates the advantages of different imaging tools. Lanthanide-based nanoprobes are unique and have rich optical, magnetic, radioactive, and X-ray attenuation properties; however, simple doping of different lanthanide cations into one host can result in a material with multifunction but not the optimized properties. In this study, using NaLuF4:Yb,Tm as the core and 4 nm of (153)Sm(3+)-doped NaGdF4 (half-life of (153)Sm = 46.3 h) as the shell, we developed a lanthanide-based core-shell nanocomposite as an optimized multimodal imaging probe with enhanced imaging ability. The lifetime of upconversion luminescence (UCL) at 800 nm and relaxation rate (1/T1) were at 1044 μs and 18.15 s(-1)·mM(-1), respectively; however, no significant decrease in the attenuation coefficient was observed, which preserved the excellent X-ray imaging ability. The nanomaterial NaLuF4:Yb,Tm@NaGdF4((153)Sm) was confirmed to be effective and applicable for UCL imaging, X-ray computed tomography (CT), magnetic resonance imaging, and single-photon emission computed tomography (SPECT) in vivo. Furthermore, the NaLuF4:Yb,Tm@NaGdF4((153)Sm) nanoparticles were applied in tumor angiogenesis analysis by combining multimodality imaging of CT, SPECT, and confocal UCL imaging, which shows its value of multifunctional nanoparticles NaLuF4:Yb,Tm@NaGdF4((153)Sm) in tumor angiogenesis imaging.
Yb3+/Er3+-Codoped Bi2o3 Nanospheres: Probe for Upconversion Luminescence Imaging and Binary Contrast Agent for Computed Tomography Imaging

Abstract:In this work, water-soluble Yb(3+)/Er(3+) codoped Bi2O3 upconversion (UC) nanospheres with uniform morphology have been successfully synthesized via a solid-state-chemistry thermal decomposition process. With 980 nm near-infrared irradiation, the Bi2O3:Yb(3+)/Er(3+) nanospheres have bright UC luminescence (UCL). Moreover, multicolor UC emissions (from green to red) can be tuned by simply changing the Yb(3+) ions doping concentration. After citric acid molecules were grafted on the surface of Bi2O3:20% Yb(3+)/2% Er(3+) nanospheres, the MTT assay on HeLa cells and CCK-8 assay on osteoblasts show that the UC nanospheres exhibit excellent stability and biocompatibility. The possibility of using these nanoprobes with red UCL for optical imaging in vivo has been demonstrated. Furthermore, Bi(3+) and Yb(3+) containing nanospheres as binary contrast agent also exhibited significant enhancement of contrast efficacy than iodine-based contrast agent via X-ray computed tomography (CT) imaging at different voltage setting (80-140 kVp), indicating they have potential as CT imaging contrast agent. Thus, Yb(3+)/Er(3+) codoped Bi2O3 nanospheres could be used as dual modality probe for optical and CT imagings.
Bi 3+ -Doped BaYF 5 :Yb,Er Upconversion Nanoparticles with Enhanced Luminescence and Application Case for X-ray Computed Tomography Imaging

Abstract:In this work, BaYF5:20%Yb3+/2%Er3+/x%Bi3+ (abbreviated as BaYF5:Yb,Er,Bix, where x = 0–3.0) upconversion nanoparticles (UCNPs) with various doping concentrations of Bi3+ were synthesized through a simple hydrothermal method. The influence of the doping amount of Bi3+ on the microstructures and upconversion luminescence (UCL) properties of the BaYF5:Yb,Er,Bix UCNPs was studied in detail. The doping concentration of Bi3+ has little influence on the microstructures of the UCNPs but significantly impacts their UCL intensities. Under excitation of a 980 nm near-IR laser, the observed UCL intensities for the BaYF5:Yb,Er,Bix UCNPs display first an increasing trend and then a decreasing trend with an increase in the ratio x, giving a maximum at x = 2.5. A possible energy-transfer process and simplified energy levels of the BaYF5:Yb,Er,Bix UCNPs were proposed. The potential of the BaYF5:Yb,Er,Bix UCNPs as contrast agents for computerized tomography (CT) imaging was successfully demonstrated. An obvious accumulation of BaYF5:Yb,Er,Bix in tumor sites was achieved because of high passive targeting by the enhanced permeability and retention effect and relatively low uptake by a reticuloendothelial system such as liver and spleen. Direct Visualization of Gastrointestinal Tract with Lanthanide-Doped BaYbF5 Upconversion Nanoprobes

Abstract:Nanoparticulate contrast agents have attracted a great deal of attention along with the rapid development of modern medicine. Here, a binary contrast agent based on PAA modified BaYbF5:Tm nanoparticles for direct visualization of gastrointestinal (GI) tract has been designed and developed via a one-pot solvothermal route. By taking advantages of excellent colloidal stability, low cytotoxicity, and neglectable hemolysis of these well-designed nanoparticles, their feasibility as a multi-modal contrast agent for GI tract was intensively investigated. Significant enhancement of contrast efficacy relative to clinical barium meal and iodine-based contrast agent was evaluated via X-ray imaging and CT imaging in vivo. By doping Tm(3+) ions into these nanoprobes, in vivo NIR-NIR imaging was then demonstrated. Unlike some invasive imaging modalities, non-invasive imaging strategy including X-ray imaging, CT imaging, and UCL imaging for GI tract could extremely reduce the painlessness to patients, effectively facilitate imaging procedure, as well as rationality economize diagnostic time. Critical to clinical applications, long-term toxicity of our contrast agent was additionally investigated in detail, indicating their overall safety. Based on our results, PAA-BaYbF5:Tm nanoparticles were the excellent multi-modal contrast agent to integrate X-ray imaging, CT imaging, and UCL imaging for direct visualization of GI tract with low systemic toxicity.
Multimodal-luminescence Core-Shell Nanocomposites for Targeted Imaging of Tumor Cells.

Abstract:Uniform silica-coated NaYF(4): 20 mol % Yb, 2 mol % Er nanocomposites with good dispersibility, containing organic dye incorporated in the silica shell and folic acid conjugated on the surface of the shell, were prepared and characterized. The core-shell nanocomposites are 20-22 nm in size, water soluble, and buffer stable, with good photostability and biocompatibility. Folic acid (FA) offers a means of targeting human cells that greatly overexpress the folate receptor (FR). By the use of confocal microscopy and quantitative flow cytometry analysis, we demonstrate the receptor-mediated delivery of FA-conjugated nanocomposites targeting FR-positive cell lines, such as KB cells. The receptor-mediated targeting was confirmed by a comparison with the uptake of these nanocomposites in FR-negative cell lines, such as MCF-7. These results show that the silica-coated upconverting nanophosphor (UCNP) nanocomposites prepared by our strategy can potentially be useful as multimodal bioimaging agents.

Abstract:Rare earth doped upconversion nanoparticles (UCNPs) used as bioprobes for in vivo upconversion luminescence (UCL) deep tissue imaging have been extensively studied. Red emission possesses high penetration length in biological tissue, so it's more convenient for naked eyed red emission to be performed in biological imaging and detection. However, improved in vivo imaging depth based on the NIR-to-red UCL is still challenging and desirable. Herein, novel alpha-NaYb(Mn)F-4:Er3+@NaLuF4 core-shell nanocrystals with enhanced red emission and improved imaging depth have been synthesized. Under 980 nm excitation, there is a 220% increase in red-to-green ratio and even a 400% increase in red-emission when the Yb3+ core doping concentration is increased from 18% to 68% in the core-shell structure. And the UCL signals detection of pork muscle tissues could be visualized even at a depth of 18 mm. By the tail vein injection of PEG modified alpha-NaYb(Mn)F-4:Er3+@NaLuF4 UCNPs, the in vivo and ex vivo NIR-to-red UCL deep tissue imaging present significant UCL signals in liver and spleen. Moreover, the UCNPs could also be used as in vitro and in vivo X-ray computed tomography (CT) contrast agent owing to the large X-ray absorption efficiency and high atomic number of ytterbium and lutetium elements. The synergistic combination of the in vivo NIR-to-red UCL deep tissue and CT imaging could provide comprehensive diagnosis information for cancer.
Pegylated Hybrid Ytterbia Nanoparticles As High-Performance Diagnostic Probes for in Vivo Magnetic Resonance and X-Ray Computed Tomography Imaging with Low Systemic Toxicity

Abstract:Novel nanoparticulate contrast agents with low systemic toxicity and inexpensive character have exhibited more advantages over routinely used small molecular contrast agents for the diagnosis and prognosis of disease. Herein, we designed and synthesized PEGylated hybrid ytterbia nanoparticles as high-performance nanoprobes for X-ray computed tomography (CT) imaging and magnetic resonance (MR) imaging both in vitro and in vivo. These well-defined nanoparticles were facile to prepare and cost-effective, meeting the criteria as a biomedical material. Compared with routinely used Iobitridol in clinic, our PEG-Yb2O3:Gd nanoparticles could provide much significantly enhanced contrast upon various clinical voltages ranging from 80 kVp to 140 kVp owing to the high atomic number and well-positioned K-edge energy of ytterbium. By the doping of gadolinium, our nanoparticulate contrast agent could perform perfect MR imaging simultaneously, revealing similar organ enrichment and bio-distribution with the CT imaging results. The super improvement in imaging efficiency was mainly attributed to the high content of Yb and Gd in a single nanoparticle, thus making these nanoparticles suitable for dual-modal diagnostic imaging with a low single-injection dose. In addition, detailed toxicological study in vitro and in vivo indicated that uniformly sized PEG-Yb2O3:Gd nanoparticles possessed excellent biocompatibility and revealed overall safety.

Abstract:Dual-modality imaging with magnetic resonance (MR) and upconversion luminescence (UCL) is a promising technique for molecular imaging in biomedical research. Multifunctional lanthanide-based nanoparticles have been widely investigated as agents for contrast enhanced MR and fluorescence imaging. However, the use of rare earth fluoride nanoparticles for dual-modality imaging of T2-weighted MR and UCL is rarely reported. We find that NaYF4:Yb(3+),Tm(3+),Co(2+) (MUC) nanorods can be applied as a high-performance dual contrast agent for both T2-weighted MR and UCL dual-modality imaging. After modification with 6-O-carboxymethyl chitosan (OCC), MUC nanorods can be endocytosed by cells without showing signs of cytotoxicity. High-quality UCL images of living cells incubated with MUC-OCC nanorods were acquired on a near-infrared (NIR) confocal microscopy under the excitation at 980 nm. Moreover, MUC-OCC nanorods display high transverse (r2) relaxivities in vitro. The application of low-dose MUC-OCC nanorods for NIR-to-NIR UCL and MR dual-modality in vivo imaging was also carried out successfully. In addition, the toxicity of MUC-OCC nanorods was evaluated by MTT assay, serological tests and histological analysis of visceral organs.

Abstract:Applications of lanthanide-based nanoparticles for bioimaging have attracted increasing attention. Herein, small size PEG-EuOF:153Sm nanocrystals (∼5 nm) (PEG = poly(ethylene glycol)bis(carboxymethyl)ether) combined with the radioactive and X-ray absorption properties were synthesized. The distribution of the PEG-EuOF nanocrystals in living animals was studied by ex vivo radioassay, inductively coupled plasma-atomic emission spectrum (ICP-AES) analysis and in vivo SPECT imaging, which indicated that the small size PEG-EuOF:153Sm had long blood retention time (blood half-life (t1/2) reach to 4.65 h) and were eliminated significantly through biliary/gastrointestinal pathway in vivo. Meanwhile, benefiting from the high attenuation ability of Eu, the small size PEG-EuOF was successfully applied for lymph node CT imaging, extending the bio-applications of these small nanocrystals. The results of cytotoxicity and in vivo toxicity also showed that the PEG-EuOF nanocrystals have relatively low toxicity, which suggest their safety for in vivo imaging. The studies provide preliminary validation for the use of PEG-EuOF nanocrystals for in vivo bioimaging applications.

Abstract:By thermal decomposition in the presence only of oleylamine, sub-10 nm hexagonal NaLuF4-based nanocrystals codoped with Gd3+, Yb3+, and Er3+ (or Tm3+) have been successfully synthesized. Sub-10 nm beta-NaLuF4: 24 mol % Gd3+, 20 mol % Yb3+, 1 mol % Tm3+ nanocrystals display bright upconversion luminescence (UCL) with a quantum yield of 0.47 +/- 0.06% under continuous-wave excitation at 980 nm. Furthermore, through the use of beta-NaLuF4: Gd3+,Yb3+,Tm3+ nanocrystals as a luminescent label, the detection limit of <50 nanocrystal-labeled cells was achieved for whole-body photoluminescent imaging of a small animal (mouse), and high-contrast UCL imaging of a whole-body black mouse with a penetration depth of similar to 2 cm was achieved.

Abstract:A new upconversion luminescence (UCL) nanophosphors based on host matrix of cubic NaLuF(4) with bright luminescence have been synthesized by a solvothermal method, facilitate the nanocrystals potential candidates for imaging in vivo, especially large-animals. The sub-20 nm NaLuF(4) co-doped Yb(3+) and Er(3+) (Tm(3+)) showed about 10-fold stronger UCL emission than that of corresponding hexagonal NaYF(4)-based nanocrystals with a 20 nm diameter. Near-infrared to near-infrared (NIR-to-NIR) UCL emission of PAA-coated NaLuF(4):20%Yb,1%Tm (PAA-Lu(Tm)) can penetrate >1.5 cm tissue of pork with high contrast. Based on super-strong UCL emission and deep penetration, PAA-Lu(Tm) as optical bioprobe has been demonstrated by in vivo UCL imaging of a normal black mouse, even rabbit with excellent signal-to-noise ratio. Furthermore, such cubic NaLuF(4)-based nanophosphor was applied in lymph node imaging of live Kunming mouse with rich white fur.
