Abstract:Aliphatic polyesters, which can be obtained from bacterial or chemical synthesis, are emerging as an important class of biodegradable and biocompatible polymers for applications as biomedical and conventional materials. The last two decades have witnessed explosive growth in this subject. Most biodegradable polyesters are crystallizable. The crystallization and thermal behavior of biodegradable polyesters have been extensively studied in recent years. The physical properties, such as the mechanical, thermal properties, and biodegradability (of biodegradable polymers) of a semicrystalline polymer generally depend on the crystalline structure. The polymorphic crystallization of biodegradable polyesters, including poly(3-hydroxybutyrate) [P(3HB)], poly(lactide) (PLA), poly(3-hydroxypropionate) (PHP), and poly(butylene adipate) (PBA), as well as isomorphic crystallization in many kinds of random copolyesters, such as poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)], have been drawing more and more attention from both academic and industrial viewpoints. This article aims to provide a comprehensive review of the current understanding on (i) polymorphic crystallization and phase transition of biodegradable polyesters, (ii) isomorphic crystallization of biodegradable random copolyesters, where the effects of comonomer composition and crystallization conditions are highlighted. The relationships between the solid-state structure and physical properties as well as the biodegradability of biodegradable polyesters are also addressed.

Abstract:Poly(aspartic acid) is a water-soluble polymer of great importance.This polymer is known to be biodegradable and thus can be used in medical,comestic.fabric and metal absorbents.In this paper,we mainly review recent research on poly(aspartic acid),polyaspartamide and its copolymers,including synthesis of these polymers.The use of polyaspartamide,especially as drug carriers and bioscaffold materials is also described in detail.

Abstract:Biodegradable and biocompatible biomaterials have offered much more opportunities from an engineering standpoint for treating diseases and maintaining health. Poly(ester amide)s (PEAs), as an outstanding family among such biomaterials, have risen overwhelmingly in the past decades. These synthetic polymers have easily and widely available raw materials and a diversity of synthetic approaches, which have attracted considerable attention. More importantly, combining the superiorities of polyamides and polyesters, PEAs have emerged with better functions. They could have improved biodegradability, biocompatibility, and cell-material interactions. The PEAs derived from α-amino acids even allow the introduction of pendant sites for further modification or functionalization. Meanwhile, it is gradually recognized that the chemical structures are closely related to the physiochemical and biological properties of PEAs so that their properties can be precisely controlled. PEAs therefore become significant materials in the biomedical fields. This review will attempt to summarize the recent progress in the development of PEAs with respect to the preparation materials and methods, structure-property relationships along with their latest biomedical accomplishments, especially for drug delivery and tissue engineering.

Abstract:AbstractPolypeptoids, with similar structure to polypeptides, are nowadays rising stars due to their excellent solubility, processibility, enzymatic stability, and biocompatibility. Attractive properties of polypeptoid‐containing copolymers endow them with various applications in surface antifouling, biosensing, microreactor, drug delivery, and stimuli–response. The syntheses of block copolymers containing polypeptoids have attracted a great amount of interest so far. The review summarizes the synthetic strategies of polypeptoid‐containing copolymers, that is, polypeptoid‐b‐polypeptoid, −polypeptide, −polyester, −polyether and ‐polyolefin developed recently, as well as their phase‐separation, assembly and stimuli–response properties.

Abstract:Poly(alkylene dicarboxylate)s constitute a family of biodegradable polymers with increasing interest for both commodity and speciality applications. Most of these polymers can be prepared from biobased diols and dicarboxylic acids such as 1,4-butanediol, succinic acid and carbohydrates. This review provides a current status report concerning synthesis, biodegradation and applications of a series of polymers that cover a wide range of properties, namely, materials from elastomeric to rigid characteristics that are suitable for applications such as hydrogels, soft tissue engineering, drug delivery systems and liquid crystals. Finally, the incorporation of aromatic units and α-amino acids is considered since stiffness of molecular chains and intermolecular interactions can be drastically changed. In fact, poly(ester amide)s derived from naturally occurring amino acids offer great possibilities as biodegradable materials for biomedical applications which are also extensively discussed.

Abstract:Poly(ethylene glycol) (PEG) segments were introduced into a polyanhydride main chain by copolymerization of terminal-carboxylated poly(ethylene glycol) with diacidic monomers (sebacic acid and trimellitylimidoglycine). IR and 1H NMR spectroscopy confirmed the copolymer structures. DSC analysis showed that these polyanhydrides have low Tg and low crystallinity. In vitro degradation tests indicated that introducing PEG segments accelerated the degradation rate of these polymers and the degradation duration could be manipulated from 3 days to 3 weeks. The pH of the environment caused by the polymer degradation was lower than 5.0; therefore, the polyanhydrides could be used as components of a newly designed pulsed-release device for peptide and protein delivery. © 1999 Society of Chemical Industry
Synthesis, characterization and in vitro degradation of a new family of alternate poly(ester-anhydrides) based on aliphatic and aromatic diacids

Abstract:A new family of alternate poly(ester-anhydrides) containing aliphatic and aromatic diacids were synthesized. The dicarboxylic acids were obtained by derivatization of p-hydroxy benzoic acid at the hydroxy terminus with cyclic anhydride (adipic anhydride and succinic anhydride) and subsequently polymerized via the corresponding mixed anhydrides by melt polycondensation. DSC traces revealed that the polymers had low Tg (<40°C) and no crystallinity. The static contact angle measurements indicated that the poly(ester-anhydrides) were more hydrophobic than poly(d,l-lactide) and poly(adipic anhydride). In vitro degradation of the polymers was also investigated in pH 7.4 PBS at 37°C. It was found that degradation rate of the poly(ester-anhydrides) increased with p-carboxy phenyl adipic monoester (CPA) content in the polymers and the degradation duration could be adjusted from ca. 20 days to ca. 2 months. Erosion curve of poly(p-carboxy phenyl adipic monoester anhydride) (PCPA) was characterized by a linear region of weight loss at nearly constant rate in the first 7 days (ca. 80% of weight loss) followed by a gradual decrease region. IR and SEM analysis showed that significant erosion of PCPA occurred in the outer layer and no apparent erosion could be seen in the inner layer of the degrading sample after 7-day degradation. The poly(ester-anhydrides) may be used as either anti-infective polymeric prodrugs or matrices for drug delivery.

Abstract:Polypeptides, a kind of very promising biomaterial, have shown a wide range of applications due to their excellent biocompatibility, easy accessibility, and structural variability. To synthesize polypeptides with desired functions, post-polymerization modification (PPM) plays an important role in introducing novel chemical structure on their side-chains. The key to PPM strategy is to develop highly selective and efficient reactions that can couple the additional functional moieties with pre-installed side-chain functionalities on polypeptides. In this minireview, classic PPM approaches and their recent progresses are summarized and categorized into five sections, including various reactions on unsaturated alkyl, oxygen-, nitrogen-, sulfur-containing and other special functional groups. In addition, we also highlight the applications of the resultant structure-diversified polypeptides in the field of biomaterial.

Abstract:There are some persons who stand out prominently in any review of the history of polyesters. This chapter talks about two giant polymer scientists Staudinger and Carothers and their contributions to the origin and further development of polymer science as well as some historical records on the synthesis of polyamides and polyesters. Three well‐known biodegradable polyesters, namely, Poly(lactic acid) (PLA), Poly(glycolic acid) (PGA), and poly(caprolactone) were selected to describe their synthesis, structures, properties, and applications. Owing to the similarity of the biodegradable polyesters in synthesis, properties, and applications, the chapter gives the readers a general outline about biodegradable polyesters. Biodegradable polyesters have intensively been researched in the last two decades because of their biodegradability and superb physical properties. Recently, nanomaterials, nanotechnology, and the opportunities for wider medical applications have resulted in much more attention being directed to this class of polyesters.

Abstract:Polyphosphoesters (PPEs) represent an innovative class of biodegradable polymers, with the phosphate ester serving as the core repeating unit of their polymeric backbone. Recently, biomaterials derived from functionalized PPEs have garnered significant interest in biomedical applications because of their commendable biocompatibility, biodegradability, and the capacity for functional modification. This review commences with a brief overview of synthesis methodologies and the distinctive properties of PPEs, including thermoresponsiveness, degradability, stealth effect, and biocompatibility. Subsequently, the review delves into the latest applications of PPEs-based nanocarriers for drug or gene delivery and PPEs-based polymeric prodrugs and scaffolds in the biomedical field, presenting several illustrative examples for each application. By encapsulating the advancements of recent years, this review aims to offer an enhanced understanding and serve as a reference for the synthesis and biomedical applications of functional PPEs.
Biodegradable polyester copolymers: synthesis based on the Biginelli reaction, characterization, and evaluation of their application properties

Abstract:The Biginelli reaction, a three-component cyclocondensation reaction, is an important member of the multicomponent reaction (MCR) family. In this study, we conducted end-group modifications on a variety of biodegradable polyesters, including poly(1,4-butylene adipate) (PBA), poly(ε-caprolactone) (PCL), polylactic acid (PLA), and poly(p-dioxanone) (PPDO), based on the precursor polyethylene glycol (PEG). By combining two polymers through the Biginelli multi-component reaction, four new biodegradable polyester copolymers, namely DHPM-PBA, DHPM-PCL, DHPM-PLA, and DHPM-PPDO, were formed. These Biginelli reactions demonstrated exceptional completeness, validating the efficiency of the synthesis strategy. Although the introduction of various polyesters lead to different properties, such as crystallinity and cytotoxicity, the newly synthesized 3,4-dihydro-2(H)-pyrimidinone compounds (DHPMs) exhibit enhanced hydrophilicity and can self-assemble in water and N,N-dimethylformamide (DMF) solution to form micelles with a controllable size. Furthermore, DHPM-PPDO promotes cellular growth and has potential applications in wound healing and tissue engineering. In conclusion, this method demonstrates great universality and methodological significance and offers insights into the medical applications of polyethylene glycol.

Abstract:Polyphosphoesters are a class of biodegradable polymers with repeated phosphoester linkage in the backbone. Due to their biocompatibility and structural similarities to naturally occurring nucleic and teichoic acids, functionalized polyphosphoesters have attracted more and more attention in the last decade and have currently been studied extensively. The phosphorus is a pentavalent atom and can be modified by the incorporation of various substituents, which leads to formation of versatile structures and the easy side-chain functionalization, thus, it allows effective modification of their physical and chemical properties. Therefore polyphosphoesters have been applied in drug/gene delivery, tissue engineering and so on. Among them, the most popular and widely used of such polymers is poly (ethylene phosphate). This review briefly summarizes the structural characteristics of poly (ethylene phosphate) and the controlled synthetic methods, which provide access to novel and complex polymer structures. The article focuses on the various types of synthetic poly(ethylene phosphate) such as linear, star and hyperbranched derivatives, particularly those poly(ethylene phosphate) derivatives that applied in biological fields are highlighted. In addition, on the basis of current research in the fields, this paper presents several representative topologies of poly(ethylene phosphate) and their application in drug/gene deliveries and tissue engineering. At last we propose the prospects of poly(ethylene phosphate) development in life sciences, and we look forward to better communication with other scientists in this field.
Biocompatibility of Fully Biodegradable Polylactic Acid and Its Copolymers: Present and Future Applications

Abstract:BACKGROUND:Polylactic acid and its copolymers have been widely used in biomedical fields because of their good biocompatibility and biodegradability, which have become a hotspot in the research of biomaterials. OBJECTIVE:To review the biocompatibility of fully degradable polylactic acid and its copolymers. METHODS:A computer-based online retrieval of PubMed and CNKI was performed to search relevant papers published from 2006 to 2016, with the key words of "polylactide, polylactic acid, copolymer, biodegradability, biocompatibility, animal" in English and Chinese, respectively. RESULTS AND CONCLUSION:Polylactic acid and its copolymers as polyester compounds are approved by the US Food and Drug Administration for use in the biomedical field because of their good biocompatibility mainly as drug delivery carriers and temporary implants. Moreover, their side effects in clinical application have attracted attentions. Polylactide copolymers can cause some adverse reactions when used in drug delivery systems, orthopedic and skin care, and other clinical medical fields. These copolymers are deemed to have no impact on the central nervous system, eyes, cardiovessels and other tissues and organs. They also have no virtually genotoxic and carcinogenic effects. Currently, the polylactide copolymer implant mainly results in local reactions in the surrounding tissues, and no systemic reactions have been found.

Abstract:In the last two decades, the biodegradable polycarbonates have attracted much attention in biomedical application due to their excellent properties such as mechanical properties and biocompatibility and so on. Usually, biodegradable polycarbonats are divided into two classes: aliphatic polycarbonates and aromatic polycarbonates in view of their main chain structure. Recently, increasing effort was taken to adjust the structure and properties of the polycarbonates to meet a range of different demands in biomedical application. In this paper, the development of the biodegradable polycarbonates was thoroughly reviewed and the application in the biomedical field was also chiefly illustrated.

Abstract:Biodegradable polyphosphazenes,because of their unusual structures and versatility in terms of chemical and physical properties,have been investigated for different biomedical and pharmaceutical applications.The types of biodegradable polyphosphazenes are briefly introduced in this paper.The recent progress of the synthetic procedures used for the preparation of biodegradable polyphosphazenes with two same side groups including amine-substituted polyphosphazenes,alkoxy(or aryloxy)-substituted polyphosphazenes,glycery-substituted polyphosphazenes and side groups-derived polyphosphazenes is reviewed.The purpose and method of synthesizing biodegradable mixed substituent polyphosphazenes with two(or more) different side groups are summarized.The factors that affect the reaction of synthesizing polyphosphazenes and modification of polyphosphazenes and other polymers are also discussed.

Abstract:Biodegradable and biocompatible biomaterials have several important applications in drug delivery. The biomaterial family known as poly(ester amide)s (PEAs) has garnered considerable interest because it exhibits the benefits of both polyester and polyamide, as well as production from readily available raw ingredients and sophisticated synthesis techniques. Specifically, α-amino acid-based PEAs (AA-PEAs) are promising carriers because of their structural flexibility, biocompatibility, and biodegradability. Herein, we summarize the latest applications of PEAs in drug delivery systems, including antitumor, gene therapy, and protein drugs, and discuss the prospects of drug delivery based on PEAs, which provides a reference for designing safe and efficient drug delivery carriers.
