Abstract:A label-free immunosensor was designed and fabricated for sensitive detection of alpha-fetoprotein (AFP) based on the localized surface plasmon resonance (LSPR) of gold nanorods (GNRs). GNRs were fabricated through seedmediated growth and surface activation by alkanethiols for the attachment of antibodies that could directly capture of the target. A novel strategy using the absorbance ratio of transverse and longitudinal LSPR peak of GNRs as a function of AFP contraction was performed for AFP quantification. The limit of detection of the sensor is 0.25nM in the PBS, and the dynamic range spans 0.25nM to 14.3nM. It is expected that this simple and cost-effective LSPR biosensor can also be applicable to the detection of several other tumor markers that are present at low concentrations.
Antibody modified gold nano-mushroom arrays for rapid detection of alpha-fetoprotein

Abstract:Localized surface plasmon resonance (LSPR) combined with immunoassay shows greatly potential in fast detection of tumor markers. In this paper, a highly sensitive LSPR substrate has been fabricated and modified for direct detection of alpha-fetoprotein (AFP). The biosensor was prepared by interference lithography, and modified by covalently immobilizing anti-AFP on the surface of gold nano-mushroom arrays (GNMA). The modification process was investigated by Vis–NIR reflectance spectra and cyclic voltammogram measurements. We revealed the optical properties of the modified GNMA by measuring the Vis–NIR reflectance spectra and simulating its electric intensity field distribution under light illumination. The GNMA substrate was highly sensitive, with a refractive index sensitivity of ~465nm/RIU. The substrate can be applied to label-free detection of AFP, with the linear range and the limit of detection determined to be 20–200ng/mL and 24ng/mL (S/N=3), respectively. We also demonstrated its clinical application by directly detecting AFP in human serum samples. It is expected that our biosensor could be integrated on microfluidic chips for high-throughput detection in portable early diagnosis, post-operative and point-of-care (POC) in clinical applications.
Ultrasensitive dynamic light scattering immunodetection of alpha-fetoprotein using heptamer-amplified nanoparticle crosslinking aggregation

Abstract:A novel construction strategy is introduced for an ultrasensitive dynamic light scattering (DLS) immunosensor targeting alpha fetoprotein (AFP). This approach relies on a self-assembled heptamer fusion protein (A1-C4bpα), incorporating the dual functions of multivalent recognition and crosslinking aggregation amplification due to the presence of seven AFP-specific A1 nanobodies on the A1-C4bpα heptamer. Leveraging antibody-functionalized magnetic nanoparticles for target AFP capture and DLS signal output, the proposed heptamer-assisted DLS immunosensor offers high sensitivity, strong specificity, and ease of operation. Under the optimized conditions, the designed DLS immunosensor demonstrates excellent linear detection of AFP in the concentration range 0.06 ng mL-1 to 512 ng mL-1, with a detection limit of 15 pg mL-1. The selectivity, accuracy, precision, practicability, and reliability of this newly developed method were further validated through an assay of AFP levels in spiked and actual human serum samples. This work introduces a novel approach for constructing ultrasensitive DLS immunosensors, easily extendable to the sensitive determination of other targets via simply replacing the nanobody sequence, holding great promise in various applications, particularly in disease diagnosis.

Abstract:The study of luminol electrochemiluminescence (ECL) is commonly conducted in alkaline solutions. However, many substances, especially biomolecules, are only stable in neutral or weakly acidic media. In this study, a sensitive ECL immunosensor was developed using polypyrrole-luminol (PPY-Lu) nanoparticles for the determination of alpha-fetoprotein (AFP) in a neutral buffer. Polypyrrole (PPY) nanoparticles were synthesized by chemical oxidation. Subsequently, luminol was attached to the PPY nanoparticles via electrostatic interaction, resulting in the formation of the desired nanoparticles which exhibited strong and stable electrochemiluminescence in neutral solution. Furthermore, the PPY-Lu nanoparticles have abundant amino groups suitable for immobilizing biomolecules. An ECL immunosensor for AFP was fabricated by immobilizing anti-AFP on the PPY-Lu-modified electrode surface. The electrochemiluminescence decreased when AFP was captured. The immunosensor exhibited a linear range from 1.0‚ÄČ√ó‚ÄČ10 ‚ąí11 to 1.0‚ÄČ√ó‚ÄČ10 ‚ąí6‚ÄČ mg/mL with a low detection limit of 8.5‚ÄČ√ó‚ÄČ10 ‚ąí12‚ÄČ mg/mL and excellent specificity, reproducibility, and long-term stability. This designed immunosensor provides a new method for the determination of biomarkers in clinical analysis.

Abstract:An integrated dual-labeled gold nanoparticles (GNPs) based electrochemical immunochromatographic test strip was developed for rapid and sensitive detection of Alpha-Fetoprotein (AFP) in human serum samples. In this study, the test strip was based on a sandwich-type immunoassay format between a biomarker and a pair of its antibodies. The electrochemical detection of the test strip was completed by preparing HRP and anti-AFP antibody co-immobilized GNPs (HRP/Ab-GNPs), and HRP was used as a signal probe for electrochemical readout. The strong affinities among capturing antibody, AFP, and HRP/Ab-GNPs were characterized by square-wave voltammetry (SWV) measurements. Some critical parameters that affected the electrochemical response and the background signals such as the ratio of HRP/Ab, the concentration of capturing antibody, the flow time, and the amount of the sample were investigated. The electrochemical lateral flow immunochromatographic (LFIC) test strip showed a broader linear response over the concentration of AFP from 1.0 ng/mL to 100.0 ng/mL within 15 min, with a detection limit of 0.5 ng/mL. Therefore, the integrated LFIC test strip would be of great potential for rapid, sensitive, and quantitative detection of tumor markers.

Abstract:In this paper, Pd nanoplates were used as a kind of electrode materials for fabrication of an electrochemical immunosensor, which was applied for detection of cancer biomarker alpha-fetoprotein (AFP). Thanks to the unique structure and properties of Pd nanoplates, the antibody of AFP (Ab) was effectively immobilized onto the surface of the Pd nanoplates modified glassy carbon electrode (GCE). Moreover, the good electrochemical properties of Pd nanoplates greatly improved the electronic transmission rate and enhanced the electrochemical signal, which led to an increase of the detection sensitivity. Based on the specific antibody-antigen interaction, a label-free immunosensor based on Pd nanoplates was developed for sensing of AFP. The current method allows us to detect AFP over a wide concentration range from 0.01 to 75.0 ng/mL with a detection limit of 4 pg/mL. The proposed immunosensor has been used to determine AFP in human serum with satisfactory results.

Abstract:Alpha-fetoprotein (AFP) has become a specific tumor marker of primary liver cancer in clinical diagnosis. In this work, we prepared worm-like platinum (WL Pt) nanomaterial via chemical etching without organic solvents and ultra-high temperature. Due to its small particle size and the formation of surface vacancies during the etching process, it had a large specific surface area, and thus exhibited superior electrocatalytic activity for the reduction of hydrogen peroxide. Combining the signal amplification based on hydrogen peroxide reduction and the specific recognition of antigen with antibody, we constructed a simple label-free electrochemical immunosensor with a sandwich-like structure. The developed electrochemical immunosensor showed a wide linear range (0.0001−100 ng mL−1), a low detection limit (0.028 pg mL−1), good selectivity and stability. Further, the immunosensor was comparable with enzyme-linked immunosorbent assay (ELISA) and had a good accuracy for AFP detection in human serum samples proving the feasibility of potential application, which is expect to become one of the most promising method in early diagnosis and clinical analysis for liver cancer.

Abstract:We propose an ultrasensitive leaky surface acoustic wave (LSAW) immunosensor based on molybdenum disulfide @ cuprous oxide—gold (MoS2@Cu2O-Au) nanoparticles and subsequent gold staining for the detection of alpha-fetoprotein (AFP). MoS2@Cu2O-Au nanoparticles, with their large specific surface area and good biocompatibility, not only capture the secondary antibodies (Ab2) but also amplify the mass loading effect of the acoustic wave sensor in the detection of AFP. The immunosensor signals are further amplified upon injection of gold staining solution. The developed immunosensor achieved a low detection limit of 5.5 and 25.0 pg/mL with and without gold staining, respectively. The immunosensor demonstrated its efficiency for the quantitative detection of AFP in complex biological fluids, including human serum and saliva samples, with excellent selectivity and long-term stability, showing great potential for the quantification of AFP in clinical diagnosis.

Abstract:Multifunctional nanolabels with both facile signal amplification and high selectivity are essential for the sensitive determination of the vital tumor marker alpha fetoprotein (AFP). Here, we prepared an immunosensing nanolabel (Ab2-PDA-AF) for convenient AFP detection by grafting the detection antibody (Ab2) and the electroactive redox probe aminoferrocene (AF) onto polydopamine (PDA) nanospheres through Michael addition and Schiff base reactions. The electrochemical signal is dually amplified by the Ab2-PDA-AF nanolabels when the nanolabels are selectively bound to the electrode. The electrochemical signal can be first amplified by oxidizing AF of the Ab2-PDA-AF on the electrode. The signal can be further amplified by the redox reaction of PDA nanospheres with the linked AF/AF+ on the nanospheres. More importantly, the integration of AF-PDA-Ab2 favors a quick electron transfer between AF and PDA nanospheres. Redox probes of AF/AF+ are bonded to the nanolabels instead of dispersing in an electrolyte solution, thus decreasing the electron-transfer resistance and increasing the current signal. The interfacial kinetic analysis demonstrates that the charge-transfer resistance is decreased by 80% by using Ab2-PDA-AF nanolabels. The sensor displays a low detection limit of ca. 0.01 pg/mL, as well as excellent selectivity, reproducibility, and stability. The linear calibration plot of the AFP immunosensor has been obtained in a range of 0.5–1000.0 pg/mL. The capability of the immunosensor has been verified by implementing the detection of AFP in human serum samples.

Abstract:Alpha-fetoprotein (AFP) is always employed as a biomarker for liver cancer. The detection of AFP with high sensitivity and good anti-interference property is extremely imperative for clinical cancer diagnosis and effective treatment. In this work, a novel bi-directionally amplified ratiometric electrochemical aptasensor based on exonuclease-assisted target recycling and bioconjugate probe was developed for the ultrasensitive detection of AFP. The aptasensor was fabricated by attaching ferrocene-labeled capture probe (Fc-CP) onto electrode surface through the hybridization with thiolated DNA(1). The recognition of AFP caused the disassociation of Fc-CP from the DNA(1)/Fc-CP duplex, and further triggered the cleavage process of RecJ(f) exonuclease and recycling of AFP. As a result, numerous Fc labels were released, accompanied by plenty exposed DNA(1) on the electrode surface for the hybridization with bioconjugate probe containing vast methylene blue (MB) molecules. This strategy significantly amplified the decrease of Fc signal and the increase of MB signal, finally achieving a wide detection range of seven orders of magnitude and an ultralow AFP detection limit down to 269.4 ag/mL. The anti-interference property of the aptasensor was verified by analyzing clinical human serum samples. This aptasensor is promising for the development of reliable electrochemical devices for point-of-care diagnosis.

Abstract:A simple and disposable electrochemical immunosensor for detection of 68 kDa alpha-fetoprotein (AFP) was fabricated based on films of silk fibroin protein membrane (SFPM)/Prussian blue (PB)/deposition of gold nanoparticles (DpAu). First, DpAu and PB were electrochemically deposited successively on the surface of indium tin oxide (ITO) electrode. Then, SFPM with excellent biocompatibility was modified on the surface of PB/DpAu/ITO. The SFPM could form a stable matrix on the electrode surface for the deposition of immunoactive agents. More importantly, the SFPM could prevent the possible leakage of electron mediator and enhance the stability of immunosensor. Scanning electron microscopy (SEM), electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) were used to characterize the assembly process of the modified electrode. The linear range of the proposed immunosensor extended from 1.0 to 200.0 ng/mL for detection of AFP with a detection limit of 0.6 ng/mL. Moreover, the CV test demonstrated the immunosensor exhibited acceptable reproducibility and stability. This composite membrane could be applied for the detection of different biomarkers, diagnosis, and monitoring of carcinoma.

Abstract:The labeling-free and immobilization-free homogeneous aptamer sensor offers advantages including simple operation, low cost, and high sensitivity, demonstrating great potential in rapid detection of tumor biomarkers in biological samples. In this work, a labeling-free and immobilization-free homogeneous aptamer sensor was conveniently fabricated by combining size exclusion and charge-selective penetration of a nanochannel-modified electrode and two-dimensional (2D) nanorecognition probe which can realize selective and highly sensitive detection of alpha-fetoprotein (AFP) in serum. Vertically ordered mesoporous silica film (VMSF) with ultra-small, uniform, and vertically aligned nanochannels was easily grown on the simple, low-cost, and disposable indium tin oxide (ITO) electrode. Through π-π interaction and electrostatic force, the AFP aptamer (Apt) and electrochemical probe, tris(bipyridine)ruthenium(II) (Ru(bpy)32+), were coloaded onto graphene oxide (GO) through simple incubation, forming a 2D nanoscale recognition probe (Ru(bpy)32+/Apt@GO). Owing to the size exclusion effect of VMSF towards the 2D nanoscale probe, the electrochemical signal of Ru(bpy)32+/Apt@GO could not be detected. In the presence of AFP, the specific binding of AFP to the aptamer causes the dissociation of the aptamer and Ru(bpy)32+ from GO, resulting in their presence in the solution. The efficient electrostatic enrichment towards Ru(bpy)32+ by negatively charged VMSF allows for high electrochemical signals of free Ru(bpy)32+ in the solution. Linear determination of AFP ranged from 1 pg/mL to 1000 ng/mL and could be obtained with a low limit of detection (LOD, 0.8 pg/mL). The high specificity of the adapter endowed the constructed sensor with high selectivity. The fabricated probe can be applied in direct determination of AFP in serum.

Abstract:In this paper, a novel disposable immunosensor for the detection of α -fetoprotein (AFP) based on the Indium tin oxide (ITO) modified by the sequential electro-deposition of the nanogold particles (nano-Au) and prussian blue (PB) is described. The ITO is employed to reduce the cost, instead of expensive gold electrode, glassy carbon electrode or platinum electrode. The layer-by-layer (LBL) electro-deposition of the nano-Au, PB is used for blocking the possible leakage from the substrate electrode surface and to prevent shedding of composite membrane. Under optimal conditions, the proposed immunosensor displays a broad linear response to AFP, the working range being 0·25 to 300·0 ng mL −1 with a detection limit of 0·04 ng mL −1 . The studied immunosensor exhibits high sensitivity, fast analytical time and good stability. The proposed methodology is potentially attractive for clinical immunoassays.

Abstract:In this study, a low-cost and easily prepared cancer biomarker sensor-alpha-fetoprotein (AFP) biosensor was successfully constructed on the surface of a glassy carbon electrode (GCE) by a surface imprinting procedure using available and cheap agents instead of antibodies. Under optimal detection conditions, the proposed biosensor showed specific recognition ability to free AFP by comparing with other comparative proteins and exhibited a wide linear detection range from 8.0 x 10(-4) to 10 mu g mL(-1) for AFP with a low detection limit of 9.6 x 10(-5) mu g mL(-1) (S/N = 3). Ultimately, the AFP imprinted sensor was applied to the determination of AFP in a human serum sample, giving satisfactory results.

Abstract:highly sensitive electrochemiluminescence immunosensor was originally proposed based on lead sulfide nanocrystals (PbS NCs) to detect alpha fetoprotein (AFP). Herein, PbS NCs, as a kind of nanoenzyme, was synthesized and applied to the luminol-hydrogen peroxide (L-H2O2) system for the first time. Additionally, PbS NCs with superior electrocatalytic activity toward H2O2 can enhance the electrochemiluminescence response of luminol. In this protocol, chitosan was utilized to immobilize PbS NCs through its film-forming characteristics, and further developed an ECL immunosensor by amination reaction combined with AFP antibodies. When the modified electrode was incubated with different concentrations of AFP antigen, the electrochemiluminescence intensity decreased obviously. In addition, under optimal conditions, the sensor can sensitively and selectively detect the alpha fetoprotein ranging from 1.0x10(-5) to 800 ng/mL with the detection limit of 3.0 fg/mL. On this basis, the proposed ECL immunosensor will open up a new way to sensitively detect AFP and exhibit its application prospect in biological analysis.

Abstract:A novel label free electrochemiluminescence (ECL) immunosensor based on the ECL of peroxydisulfate solution for detection of α-1-fetoprotein (AFP) has been developed. For this proposed immunosensor, l-cysteine was firstly electrodeposited on the gold electrode surface, which promoted the electron transfer and largely enhanced the ECL of peroxydisulfate solution. Subsequently, gold nanoparticles (nano-Au) were assembled onto the l-cysteine film modified electrode to improve the absorption capacity of antibody and further amplify the ECL signal. Then, antibody was immobilized onto the electrode through nano-Au. At last bovine serum albumin (BSA) was employed to block the nonspecific binding sites. As a result, a novel ECL immunosensor was firstly obtained by applying the ECL of peroxydisulfate solution without conventional luminescent reagents. The AFP was determined in the range of 0.01–100ngmL−1, with a low detection limit of 3.3pgmL−1 (S/N=3). The proposed ECL immunosensor provides a rapid, simple, and sensitive immunoassay protocol for protein detection, which might hold a promise for clinical application. Moreover, this work would open up a new field in the application of peroxydisulfate solution ECL for highly sensitive bioassays.

Abstract:Rapid and accurate detection of tumor markers at extremely low levels is crucial for the early diagnosis of cancers. In this work, we developed a portable label-free sliding electrochemical paper-based analytical device (ePAD) using copper/cuprous sulfide@N-doped C@Au nanoparticles (Cu/Cu2S@NC@Au) hollow nanoboxes as the signal amplifier for the ultrasensitive detection of alpha-fetoprotein (AFP). Cu/Cu2S@NC nanoboxes were synthesized by sacrificial template and interface reaction methods, on which Au nanoparticles were electrodeposited to construct unique heterostructure for effectively capturing anti-AFP and serving as signal amplifier. The designed ePAD incorporates sliding microfluidic paper chips to form a flexible three-electrode system, enabling highly sensitive detection of AFP with a wide linear range of 0.005-50 ng mL-1 and a low detection limit of 0.62 pg mL-1. The practicality of the prepared ePAD was validated through AFP detection in clinical human serum, which was consistent with chemiluminescence immunoassay. In addition, the developed immunosensor demonstrates excellent specificity, repeatability and stability. This novel platform exhibits significant potential for rapid on-site analysis and point-of-care diagnosis.
Conductometric Immunoassay of Alpha-Fetoprotein in Sera of Liver Cancer Patients Using Bienzyme-Functionalized Nanometer-Sized Silica Beads.

Abstract:A conductometric immunoassay protocol was designed for the sensitive detection of a liver cancer biomarker, alpha-fetoprotein (AFP), in biological fluids by using enzyme-conjugated nanometer-sized enzyme-doped silica beads. Initially, urease molecules were doped into nanosilica particles by using the reverse micelle method. Thereafter, arginase-labeled anti-AFP antibodies were covalently conjugated onto the surface of the synthesized nanoparticles. The immunoreaction was carried out in a monoclonal anti-AFP capture antibody-coated microplate with a sandwich-type assay format by using bienzyme-functionalized silica nanobeads as the recognition elements. Upon l-arginine introduction, the substrate was cleaved into urea and l-ornithine on the basis of the arginase enzymatic reaction, and the as-produced urea was then decomposed into ammonia (NH4+) and bicarbonate (HCO3-) ions by the doped urease, thus causing the variation in the local conductivity of the detection solution on an interdigitated conductometric transducer. Under optimal conditions, the developed immunosensing system exhibited good conductometric responses toward target AFP within a dynamic linear range of 0.01-100 ng mL-1 at a relatively low detection limit of 4.8 pg mL-1 based on the 3sB criterion. Importantly, good reproducibility, high specificity and acceptable method accuracy were acquired for the analysis of human serum specimens in liver cancer patients.

Abstract:In this work, a novel label-free photoelectrochemical (PEC) biosensor based on Au/CsxWO3 heterogeneous films with low background noise and excellent sensitive detection of alpha-fetoprotein (AFP) was proposed. We prepared Au/CsxWO3 heterogeneous films by different methods including template removal and spin coating. The surface plasmon resonances (SPR) of Au film and CsxWO3 semiconductor nanocrystals largely improved the photoelectrochemical properties of the electrodes and enhanced the photocurrent. Under the optimal conditions, the prepared PEC biosensor showed a linear response between photocurrent variation and the logarithm of AFP concentration in the range from 0.01 ng/mL to 500 ng/mL with a low detection limit of 7 pg/mL. In addition, the proposed PEC biosensor had excellent specificity, repeatability, long-term stability and showed satisfactory results in the analysis of human serum samples, which would have great prospect in clinical application and biological analysis in the future.
An Amperometric Immunosensor for Α-Fetoprotein Based on Antibody Coated Nano ZrO2-Au-Poly Lysine Composite Membrane Modified Electrode

Abstract:One novel amperometric immunosensor for α-fetop- rotein was fabricated which was modified by antibody immobilized on nano ZrO2-Au-polyLysine composite membrane. Firstly, the deoxyribonucleic acid – phenoxyacetic acid isoniazid- Schiff base Co(II) complex (DNA-CoL) mixture film was casted on surface of multi carbon nanotubes(MWCNTs) modified glassy carbon electrode(GCE/CNTs/DNA-CoL). Then the composite nanoparticles (ZrO2-Au-pLL) was employed to immobilize the antibody of α-fetoprotein(AFP) to produce the probes(ZrO2-Au-pLL-anti AFP) for AFP. Finally the probes were modified on GCE/CNTs/DNA-CoL through the specific connection between DNA and ZrO2 to form a novel nanocomposite membrane immunosensor for AFP(GCE/CNTs/ DNA-CoL/ZrO2-Au–pLL-anti AFP). The function of immunosensor was investigated by scanning electron microscopy (SEM), cyclic voltammetry(CV), differential pulse voltammety (DPV) and electrochemical impedance spectroscopy experiment (EIS). The results indicated that combining the advantages of the MWNTs-DNA material and ZrO2-Au-pLL-anti AFP nano probes, the immunosensor with excellent sensitivity and selectivity to AFP was prepared successfully. The modified electrode was sensitive to AFP with a linear relationship between 0.05 and 10 ng · mL−1 and a correl- ation coefficient of 0.9905. The detection limit at a signal to noise ratio of 3 was 0.01 ng·mL−1 under the optimal conditions. The described immunosensor preparation and immunoassay methods offer promise for simple and cost-effective analysis of AFP in serum samples.
